RESUMO

Infants born preterm face an increased risk of deleterious effects on lung and brain health that can significantly alter long-term function and quality of life and even lead to death. Moreover, preterm birth is also associated with a heightened risk of diabetes and obesity later in life, leading to an increased risk of all-cause mortality in young adults born prematurely. While these preterm-birth-related conditions have been well characterized, less is known about the long-term effects of preterm birth on skeletal muscle health and, specifically, an individual's skeletal muscle hypertrophic potential later in life. In this review, we discuss how a confluence of potentially interrelated and self-perpetuating elements associated with preterm birth might converge on anabolic and catabolic pathways to ultimately blunt skeletal muscle hypertrophy, identifying critical areas for future research.

The weight, urine colour and thirst (WUT) Venn diagram is a practical hydration assessment tool; however, it has only been investigated during first-morning. This study investigated accuracy of the WUT Venn diagram at morning and afternoon timepoints compared with blood and urine markers. Twelve men (21 ± 2 years; 81·0 ± 15·9 kg) and twelve women (22 ± 3 years; 68·8 ± 15·2 kg) completed the study. Body mass, urine colour, urine specific gravity (USG), urine osmolality (UOSM), thirst and plasma osmolality (POSM) were collected at first-morning and afternoon for 3 consecutive days in free-living (FL) and euhydrated states. Number of markers indicating dehydration levels were categorised into either 3, 2, 1 or 0 WUT markers. Receiver operating characteristics analysis calculated the sensitivity and specificity of 1, 2 or 3 hydration markers in detecting dehydration or euhydration. Specificity values across morning and afternoon exhibited high diagnostic accuracy for USG (0·890-1·000), UOSM (0·869-1·000) and POSM (0·787-0·990) when 2 and 3 WUT markers were met. Sensitivity values across both timepoints exhibited high diagnostic accuracy for USG (0·826-0·941) and UOSM (0·826-0·941), but not POSM in the afternoon (0·324) when 0 and 1 WUT markers were met. The WUT Venn diagram is accurate in detecting dehydration for WUT2 and WUT3 based off USG, UOSM and POSM during first-morning and afternoon. Applied medical, sport and occupational practitioners can use this tool in field settings for hydration assessment not only at various timepoints throughout the day but also in FL individuals.

This investigation examined the effect of rest- redistribution (RR) on the circulating cytokine response to resistance exercise in resistance- trained women. Participants (n = 13; 24 ± 4y; 65.0 ± 10.6 kg; 160.5 ± 5.1 cm) completed two sessions of back squats at 70% of 1- repetition maximum: Traditional sets (TS: 4x10, 120-s inter- set rest) and RR (4x 2x5) with 30-s intra- set rest after 5 repetitions and 90-s inter- set rest. Serum creatine kinase (CK), IL-1ß, IL-6, IL-8, IL-10, IL-15, and IL-6/IL-10 were determined before (PRE), immediately- post (IP), and 5, 15, 30, 60 min, 24 h, and 48 h post- exercise. CK increased significantly (p < 0.05) from PRE through 24 h for TS and through 48 h for RR. IL-15 was greater for RR than TS at 48 h. A condition effect was found for IL-10 and IL-6/IL-10 with a lower IL-10 and a greater IL-6/IL-10 for RR. A time effect demonstrated that IL-1ß and IL-10 peaked at IP. IL-8 was increased from 5 min to 60 min and 24 h. IL-6/IL-10 was increased from PRE to 30 min, 60 min, and 24 h. These results suggested that rest period configuration- specific differences exist for the inflammatory response in trained women during intramuscular homoeostatic disruption.

The purpose of this work was to determine the effect of resistance exercise (RE)-induced hormonal changes on the satellite cell (SC) myogenic state in response to muscle damage. Untrained men (n = 10, 22 ± 3 yr) and women (n = 9, 21 ± 4 yr) completed 2 sessions of 80 unilateral maximal eccentric knee extensions followed by either an upper body RE protocol (EX) or a 20-min rest (CON). Muscle samples were collected and analyzed for protein content of Pax7, MyoD, myogenin, cyclin D1, and p21 before (PRE), 12 h, and 24 h after the session was completed. Serum testosterone, growth hormone, cortisol, and myoglobin concentrations were analyzed at PRE, post-damage, immediately after (IP), and 15, 30, and 60 min after the session was completed. Testosterone was significantly (P < 0.05) higher immediately after the session in EX vs. CON for men. A significant time × sex × condition interaction was found for MyoD with an increase in EX (men) and CON (women) at 12 h. A significant time × condition interaction was found for Pax7, with a decrease in EX and increase in CON at 24 h. A significant time effect was found for myogenin, p21, and cyclin D1. Myogenin and p21 were increased at 12 and 24 h, and cyclin D1 was increased at 12 h. These results suggest that the acute RE-induced hormonal response can be important for men to promote SC proliferation after muscle damage but had no effect in women. Markers of SC differentiation appeared unaffected by the hormonal response but increased in response to muscle damage.

RESUMO

Muscle satellite cells can proliferate and differentiate into myocytes; this biological function has important implications for muscle development, aging, repair, and wasting diseases. Established analytical methods, including western blotting, PCR, and immunohistochemistry, has been used to characterize the stages of satellite cells growth and development but there is currently a lack of methods to simultaneously monitor cell proliferation and identify changes in cell properties (i.e., expression of myogenic regulatory factors) for each individual cell during the progression to become myocytes. Image-based flow cytometry has the capability to simultaneously obtain morphometric data, monitor cell division, and detect expression of multiple proteins of interest. In this article we demonstrate the use of image-based flow cytometry and cell-trace dye to monitor satellite cells proliferation and to identify different stages of satellite cell differentiation.

PURPOSE: To investigate the effect of acute alcohol consumption on muscular performance recovery, assessed by maximal torque production, and on inflammatory capacity, assessed by lipopolysaccharide (LPS)-stimulated cytokine production, following muscle-damaging resistance exercise in women. METHODS: Thirteen recreationally resistance-trained women completed two identical exercise bouts (300 maximal single-leg eccentric leg extensions) followed by alcohol (1.09 g ethanol kg-1 fat-free body mass) or placebo ingestion. Blood was collected before (PRE), and 5 (5 h-POST), 24 (24 h-POST), and 48 (48 h-POST) hours after exercise and analyzed for LPS-stimulated cytokine production (TNF-α, IL-1ß, IL-6, and IL-8 and IL-10). Maximal torque production (concentric, eccentric, isometric) was measured for each leg at PRE, 24 h-POST, and 48 h-POST. RESULTS: Although the exercise bout increased LPS-stimulated production of TNF-α (%change from PRE: 5 h-POST 109%; 24 h-POST 49%; 48 h-POST 40%) and decreased LPS-stimulated production of IL-8 (5 h-POST -40%; 24 h-POST -50%; 48 h-POST: -43%) and IL-10 (5 h-POST: -37%; 24 h-POST -32%; 48 h-POST -31%), consuming alcohol after exercise did not affect this response. Regardless of drink condition, concentric, eccentric, and isometric torque produced by the exercised leg were lower at 24 h-POST (concentric 106 ± 6 Nm, eccentric 144 ± 9 Nm, isometric 128 ± 8 Nm; M ± SE) compared to PRE (concentric 127 ± 7 Nm, eccentric 175 ± 11 Nm, isometric 148 ± 8 Nm). Eccentric torque production was partially recovered and isometric torque production was fully recovered by 48 h-POST. CONCLUSIONS: Alcohol consumed after muscle-damaging resistance exercise does not appear to affect inflammatory capacity or muscular performance recovery in resistance-trained women. Combined with previous findings in men, these results suggest a gender difference regarding effects of alcohol on exercise recovery.

PURPOSE: To examine the effect of post-resistance exercise alcohol ingestion on lipopolysaccharide (LPS)-stimulated production of IFNγ, TNF-α, IL-1ß, IL-6, IL-8, and IL-10. METHODS: Recreationally resistance-trained men (n = 10, 25 ± 3 year, 177 ± 7 cm, 83.8 ± 15.7 kg, 14.8 ± 8.5% body fat) and women (n = 8, 23 ± 2 year, 161 ± 3 cm, 59.5 ± 6.0 kg, 26.5 ± 3.0% body fat) completed two identical heavy back squat sessions (6 × 10 at 80% 1 repetition maximum) followed by ingestion of either an alcohol (ALC; 1.09 g ethanol · kg fat-free mass(-1)) or water (PLA) drink. Blood samples were collected before exercise (PRE), and at 3 h (3 h), and 5 h (5 h) after exercise, stimulated with LPS, and analyzed for IFNγ, TNF-α, IL-1ß, IL-6, IL-8, and IL-10 concentrations. RESULTS: There were no drink conditions by time effects for IFNγ, TNF-α, IL-1ß, or IL-10. Regardless of condition, resistance exercise induce an increase in IFNγ, TNF-α, and IL-1ß at 5 h compared to PRE but a decrease in IL-10 at 3 and 5 h compared to PRE. For ALC, IL-8 was reduced at 5 h compared to PLA. From PRE to 3 h, IL-6 was reduced for ALC but increased for PLA; resistance exercise induced an increase in IL-6 for both conditions at 5 h. CONCLUSIONS: Heavy resistance exercise increased production of IFNγ, TNF-α, IL-1ß, and Il-6 and decreased production of IL-10. Alcohol ingestion after resistance exercise affected aspects of inflammatory capacity (IL-6 and IL-8 production). It appears that some of the effects previously observed for alcohol ingestion alone on the LPS-stimulated cytokine production were overwhelmed by the response to resistance exercise.

PURPOSE: The purpose of this study was to examine the circulating cytokine response to a recreational 164-km road cycling event in a high ambient temperature and to determine if this response was affected by self-paced exercise time to completion. METHODS: Thirty-five men and five women were divided into tertiles based on time to complete the cycling event: slowest (SLOW), moderate (MOD), and fastest (FAST) finishers. Plasma samples were obtained 1-2 h before (PRE) and immediately after (IP) the event. A high-sensitivity multiplex assay kit was used to determine the concentration of plasma anti-inflammatory cytokines (IL-4, IL-5, IL-10, IL-13) and pro-inflammatory cytokines (IL-1ß, IL-2, IL-6, IL-7, IL-8, IL-12, GM-CSF, IFN-γ, and TNF-α). RESULTS: The concentration of plasma IL-10 increased significantly (p < 0.05) in FAST and MOD groups and had no change in the SLOW group in response to a 164-km cycling event in the hot environment. Other cytokine responses were not influenced by the Time to completion. Pro-inflammatory cytokines IL-1ß, IL-2, GM-CSF, and TNF-α decreased; whereas, IL-6 and IL-8 increased from PRE to IP. Additionally, anti-inflammatory cytokines IL-4 and IL-13 decreased. CONCLUSIONS: Completion of a 164-km cycling event induced substantial changes in circulating pro- and anti-inflammatory cytokine concentrations. Time to completion appears to have a greater influence on the systemic IL-10 response than the environmental condition; however, it is possible that a threshold for absolute intensity must be reached for environmental conditions to affect the IL-10 response to exercise. Thus, cyclists from the FAST/MOD groups appear more likely to experience an acute transient immune suppression than cyclists from the SLOW group.

This study investigated the acute endocrine responses to a 164-km road cycling event in a hot environment. Thirty-four male experienced cyclists (49.1 ± 8.3 years, 86.8 ± 12.5 kg, 178.1 ± 5.1 cm) participating in a 164-km road cycling event were recruited. Blood samples were collected within 0.3-2.0 h before the start (PRE: ~0500-0700 h) and immediately following the ride (POST). Samples were analysed for testosterone, growth hormone (GH), cortisol and interleukin-6 (IL-6). The temperature and humidity during the event were 35.3 ± 4.9°C and 47.2 ± 14.0%, respectively. Based on the finishing time, results for the fastest (FAST, 305 ± 10 min) and the slowest (SLOW, 467 ± 31 min) quartiles were compared. At POST, testosterone concentration was significantly (P < 0.05) lower (PRE, 20.8 ± 8.6; POST, 18.2 ± 6.7 nmol · L(-1)), while GH (PRE, 0.3 ± 0.1; POST, 2.3 ± 0.3 µg · L(-1)), cortisol (PRE, 661 ± 165; POST, 1073 ± 260 nmol · L(-1)) and IL-6 (PRE, 4.0 ± 3.4; POST, 22.4 ± 15.2 pg · mL(-1)) concentrations were significantly higher than those at PRE. At POST, GH and cortisol were significantly higher for the FAST group than for the SLOW group (GH, 3.6 ± 2.0 and 1.0 ± 0.8 µg · L(-1); cortisol, 1187 ± 209 and 867 ± 215 nmol · L(-1)). Participation in an ultra-endurance road cycling event in a hot environment induced significant acute changes in concentrations of circulating hormones, with a greater augmentation of GH and cortisol in those completing the ride fastest.

J Strength Cond Res 30(8): 2286-2291, 2016-The warm-up routine preceding a training or athletic event can affect the performance during that event. Whole-body vibration (WBV) can increase muscle performance, and thus the inclusion of WBV to the warm-up routine might provide additional performance improvements. The purpose of this investigation was to examine the acute effect of a WBV warm-up, using a vertical oscillating platform and a more traditional warm-up protocol on feet quickness in physically active men. Twenty healthy and physically active men (18-25 years, 22 ± 3 years, 176.8 ± 6.4 cm, 84.4 ± 11.5 kg, 10.8 ± 1.4% body fat) volunteered for this study. A 2 × 2 factorial design was used to examine the effect of 4 warm-up scenarios (no warm-up, traditional warm-up only, WBV warm-up only, and combined traditional and WBV warm-up) on subsequent 3-second Quick feet count test (QFT) performance. The traditional warm-up consisted of static and dynamic exercises and stretches. The WBV warm-up consisted of 60 seconds of vertical sinusoidal vibration at a frequency of 35 Hz and amplitude of 4 mm on a vibration platform. The WBV protocol significantly (p ≤ 0.0005, η = 0.581) augmented QFT performance (WBV: 37.1 ± 3.4 touches; no-WBV: 35.7 ± 3.4 touches). The results demonstrate that WBV can enhance the performance score on the QFT. The findings of this study suggest that WBV warm-up should be included in warm-up routines preceding training and athletic events which include very fast foot movements.

PURPOSE: We assessed the impact of completing the Hotter'n Hell Hundred (HHH), an annual 164 km road cycling event performed in a hot environment, on hemostatic balance in men. METHODS: Sixteen men who completed the ride in <6 h were included in this study. Plasma samples were collected on that morning of the ride (PRE) and immediately on the completion of the ride (IP). Primary hemostasis was assessed by platelet count and von Willebrand factor antigen (vWF:Ag). Coagulation was assessed by measuring prothrombin fragment 1 + 2 (PTF 1 + 2) and thrombin-antithrombin complex (TAT), whereas fibrinolysis was assessed by plasminogen activator inhibitor antigen (PAI-1 Ag), tissue plasminogen activator (tPA Ag), and D-Dimer analyses. RESULTS: Compared to PRE, increases (p < 0.001) were observed at IP for platelets (39 %), vWF:Ag (65 %), PTF 1 + 2 (47 %), TAT (81 %), tPA Ag (231 %), PAI-1 Ag (148 %), and D-Dimer (54 %). PRE PAI-1 Ag concentrations were directly related to BMI and waist circumference (p < 0.05). D-Dimer concentrations at IP correlated positively with age (p < 0.05). CONCLUSIONS: Completing the HHH activated the coagulation and fibrinolytic systems in balance. Age was positively correlated with IP D-Dimer concentrations. Additionally, participants displaying a larger BMI and waist circumference exhibited a positive correlation with PRE PAI-1 Ag concentrations.

The purpose of the study was to examine the bilateral and unilateral force production difference in powerlifters (bilateral) and field jumpers (unilateral) to determine the existence of leg dominance. Nineteen powerlifters (PL; n = 11) and field jumpers (J; n = 8) were included in the study. Five different no arm swing countermovement jumps were randomized for testing: (a) double-leg jump; (b) dominant leg-specified double-leg jump; (c) nondominant leg-specified double-leg jump; (d) dominant leg-specified single-leg jump; and (e) nondominant leg-specified single-leg jump. The force, velocity, and power were measured via a forceplate. The Limb Symmetry Index (LSI% = (1 - ND limb/D limb) × 100%) was calculated for force imbalance between the dominant (N) and nondominant (ND) limb between PL and J. Based on the analysis, PL (mean = 2.75 ± 2.45%) had a lower LSI (p < 0.05) than J (mean = 6.81 ± 5.16%). The single-leg jumps show a (p < 0.05) significantly higher force and power and a significantly lower velocity than double-leg jump for both D and ND leg than double-leg jump. Finally, the ratio between bilateral and unilateral exercise for PL and J is 2.36:1.36 and 2.38:1, respectively. In summary, sport-specific demands between PL and J may contribute to the occurrence of the imbalance force production between limbs. This phenomenon is important for the strength coach to acknowledge, to perform an appropriate strength balance test during the off-season, and to implement a training program to reduce the force disparity between limbs. Neglecting the development of force imbalance between limbs may predispose healthy players to injury.

New technology offers potential advantages in physically demanding environments where convenience and comfort are important and accurate and reliable data collection is challenging. Nevertheless, it is important to validate the accuracy and reliability of such biological monitoring systems (BMS) before they are adopted. The purpose of this investigation was to assess the concurrent validity of a new heart rate monitor across a range of exercise intensities and with a large and diverse group of male subjects in a large cohort with diverse physical fitness characteristics. Seventy-five men (age, 23 ± 4 years; height, 181 ± 8 cm; body mass, 83 ± 12 kg; estimated V[Combining Dot Above]O2peak, 3.16 ± 0.63 [L·min]) volunteered and completed a graded cycle ergometer exercise protocol while heart rate was continuously monitored before, during, and after exercise with the new device (Armour39) and the gold standard (electrocardiogram). The 2-minute stages included sitting, standing, and cycling with 35 W increments until volitional fatigue. The coefficient of determination between mean heart rate values at each stage was R = 0.99, whereas Pearson correlations (r) at each stage were ≥ 0.99. Heart rates during exercise were typically within 1 beat of each other. The Armour39 BMS, therefore, is an acceptable means for the valid and reliable determination of heart rate under various bodily positions and levels of exertion, including maximal exercise intensity.

BACKGROUND/AIM: Oxidative stress, regulated by SOD2 and mitochondrial dynamics, contributes to muscle atrophy in diabetes. Ginger root extract (GRE) reduces oxidative stress. However, its effect on oxidative stress, mitochondrial dynamics, and muscle atrophy is not known in the diabetic muscle. This study examined the effect of GRE on intramuscular oxidative stress, mitochondrial dynamics, and muscle size in diabetic rats. MATERIALS AND METHODS: Twenty-six male Sprague-Dawley rats were randomly divided into control diet (CON; n=10), high-fat diet with one dose of 35 mg/kg streptozotocin (HFD; n=9), and high-fat diet with one dose of 35 mg/kg streptozotocin and 0.75% w/w GRE (GRE; n=7) fed for seven weeks. Subsequently, the muscle was analyzed for cross-sectional area (CSA), H2O2 concentration, and DRP-1, MFN2, Parkin, PINK1, SOD2 mRNA. Additionally, the protein levels of SOD2, DRP-1, DRP-1ser616, LC3AB, MFN2, OPA1, Parkin, and PINK1 were analyzed. CSA, H2O2 concentration, and gene and protein expression levels were analyzed using a one-way ANOVA. Correlations among intramuscular H2O2, CSA, and SOD2 protein were assessed using Pearson's bivariate correlation test. RESULTS: In the soleus, the GRE group had a greater CSA and lower intramuscular H2O2 concentration compared to the HFD group. Compared to the HFD group, the GRE group had higher SOD2 and DRP-1 mRNA levels and lower MFN2 and total OPA1 protein levels. H2O2 concentration was negatively correlated with CSA and positively correlated with SOD2. CONCLUSION: GRE attenuated intramuscular H2O2, mitochondrial fusion, and muscle size loss. These findings suggest that GRE supplementation in diabetic rats reduces oxidative stress, which may contribute to muscle size preservation.

Muscle-damaging exercise (e.g., downhill running [DHR]) or heat exposure bouts potentially reduce physiological and/or cellular stress during future exertional heat exposure; however, the true extent of their combined preconditioning effects is unknown. Therefore, this study investigated the effect of muscle-damaging exercise in the heat on reducing physiological and cellular stress during future exertional heat exposure. Ten healthy males (mean ± Standard Definition; age, 23 ± 3 years; body mass, 78.7 ± 11.5 kg; height, 176.9 ± 4.7 cm) completed this study. Participants were randomly assigned into two preconditioning groups: (a) DHR in the heat (ambient temperature [Tamb], 35 °C; relative humidity [RH], 40%) and (b) DHR in thermoneutral (Tamb, 20 °C; RH, 20%). Seven days following DHR, participants performed a 45-min flat run in the heat (FlatHEAT [Tamb, 35 °C; RH, 40%]). During exercise, heart rate and rectal temperature (Trec) were recorded at baseline and every 5-min. Peripheral blood mononuclear cells were isolated to assess heat shock protein 72 (Hsp72) concentration between conditions at baseline, immediately post-DHR, and immediately pre-FlatHEAT and post-FlatHEAT. Mean Trec during FlatHEAT between hot (38.23 ± 0.38 °C) and thermoneutral DHR (38.26 ± 0.38 °C) was not significantly different (P = 0.68), with no mean heart rate differences during FlatHEAT between hot (172 ± 15 beats min-1) and thermoneutral conditions (174 ± 8 beats min-1; P = 0.58). Hsp72 concentration change from baseline to immediately pre-FlatHEAT was significantly lower in hot (-51.4%) compared to thermoneutral (+24.2%; P = 0.025) DHR, with Hsp72 change from baseline to immediately post-FlatHEAT also lower in hot (-52.6%) compared to thermoneutral conditions (+26.3%; P = 0.047). A bout of muscle-damaging exercise in the heat reduces cellular stress levels prior to and immediately following future exertional heat exposure.

INTRODUCTION: To assess hydration status, hydration markers [urine color, osmolality, and urine-specific gravity (USG)] are used. Urine color, osmolality, and USG have shown to be stable for 7, 7, and 3 days, respectively, at 4 °C. However, refrigeration could produce a dry environment which enhances evaporation and potentially affects urine hydration markers. PURPOSE: To examine the effect of duration and moisture on urine markers with refrigeration. METHODS: 24 participants provided urine samples between 9 and 10 AM. Urine color, osmolality, and USG were analyzed within 2 h (baseline). Then, each urine sample was divided into two urine cups and placed in a storage container with (moisture condition) and without (no moisture condition) water bath at 3 °C. Hydration markers were analyzed at day 1(D1), D2, D7, D10, D14, and D21. A two-way ANOVA (time x condition) and repeated-measures ANOVA on time were performed to examine differences. RESULTS: No significant (p > 0.05) condition x time effect was observed for urine color (p = 0.363), urine osmolality (p = 0.358), and USG (p = 0.248). When urine samples were stored in moisture condition, urine color (p = 0.126) and osmolality (p = 0.053) were stable until D21, while USG was stable until D2 (p = 0.394). CONCLUSION: When assessing hydration status, it appears that the urine color and osmolality were stable for 21 days, while USG was stable for 2 days when stored with moisture at 3 °C. Our results provide guidelines for practitioners regarding urine storage duration and conditions when urine cannot be analyzed immediately.

Previous research has shown reduced tissue disruption and inflammatory responses in women as compared to men following acute strenuous exercise. While the mechanism of this action is not known, estrogen may reduce the inflammatory response through its interaction with granulocytes. The purpose of this study was to determine if estrogen receptor ß expression on granulocytes is related to sex differences in tissue disruption in response to an acute heavy resistance exercise protocol. Seven healthy, resistance-trained, eumenorrheic women (23 ± 3 years, 169 ± 9.1 cm, 66.4 ± 10.5 kg) and 8 healthy, resistance-trained men (25 ± 5 years, 178 ± 6.7 cm, 82.3 ± 9.33 kg) volunteered to participate in the study. Subjects performed an acute resistance exercise test consisting of six sets of five squats at 90% of the subject's one repetition maximum. Blood samples were obtained pre-, mid-, post-, and 1-, 6-, and 24-h postexercise. Blood samples were analyzed for 17-ß-estradiol by ELISA, creatine kinase by colorimetric enzyme immunoassay, and estradiol receptors on circulating granulocytes through flow cytometry. Men had higher CK concentrations than women at baseline/control. Men had significantly higher CK concentrations at 24-h postexercise than women. No significant changes in estradiol ß receptors were expressed on granulocytes after exercise or between sexes. While sex differences occur in CK activity in response to strenuous eccentric exercise, they may not be related to estradiol receptor ß expression on granulocytes. Thus, although there are sex differences in CK expression following acute resistance exercise, the differences may not be attributable to estrogen receptor ß expression on granulocytes.

Although mouth guards were originally designed for injury prevention, even elite athletes are now using performance mouth guards to improve athletic success. Both expensive custom models and over-the-counter models are available, but the efficacy is not well known. Some athletes remain wary of the perceived potential for detriments using a mouth guard to their performance. Thus, the purpose of this study was to examine various physical performance tests when using a mouth guard including a customized over-the-counter mouth guard. Twenty-six trained men (25 ± 4 years; 1.78 ± 0.07 m; 83.3 ± 11.4 kg) and 24 trained women (23 ± 3 years; 1.65 ± 0.08 m; 62.6 ± 7.8 kg) volunteered for the investigation. The subjects completed a familiarization period and then balanced and randomized treatment conditions that included: (a) a customized Power Balance performance mouth guard (PB MG); (b) a regular over the counter boil-and-bite mouth guard (Reg MG); and (c) a no mouth guard (No MG) treatment condition. At each visit, the subjects completed a testing protocol that was sequenced in the following order: sit-and-reach flexibility, medial-lateral balance, visual reaction time, vertical jump, 10-m sprint, bench throw, and plyo press power quotient (3PQ). Heart rate and rating of perceived exertion (RPE) were recorded around the 3PQ. Significance was set at p ≤ 0.05. Expected significant sex differences existed for all power, strength, and speed variables. Bench throw power (watts) and force (newtons) were significantly higher under PB MG than either Reg MG or No MG or in both men and women. The 3PQ power and force production were higher than that for the other 2 treatments for the PB MG for men only. There were no significant differences for treatment conditions in the heart rate or RPE after the 3PQ test. Men were better able to maintain significantly higher 3PQ power production under PB MG treatment condition compared with the other 2 treatment conditions. Rate of power development was significantly higher in men for the vertical jump when using the PB MG compared with that for other treatment conditions in men only. No differences were observed in flexibility, balance, visual reaction time, or sprint time. The PB MG performance mouth guard improves performance of upper-body loaded power exercises in both men and women and lower body power exercise in men without compromising performance on any other performance parameters.

Skeletal muscle mass is determined by the balance between muscle protein synthesis (MPS) and degradation. Several intracellular signaling pathways control this balance, including mammalian/mechanistic target of rapamycin (mTOR) complex 1 (C1). Activation of this pathway in skeletal muscle is controlled, in part, by nutrition (e.g., amino acids and alcohol) and exercise (e.g., resistance exercise (RE)). Acute and chronic alcohol use can result in myopathy, and evidence points to altered mTORC1 signaling as a contributing factor. Moreover, individuals who regularly perform RE or vigorous aerobic exercise are more likely to use alcohol frequently and in larger quantities. Therefore, alcohol may antagonize beneficial exercise-induced increases in mTORC1 pathway signaling. The purpose of this review is to synthesize up-to-date evidence regarding mTORC1 pathway signaling and the independent and combined effects of acute alcohol and RE on activation of the mTORC1 pathway. Overall, acute alcohol impairs and RE activates mTORC1 pathway signaling; however, effects vary by model, sex, feeding, training status, quantity, etc., such that anabolic stimuli may partially rescue the alcohol-mediated pathway inhibition. Likewise, the impact of alcohol on RE-induced mTORC1 pathway signaling appears dependent on several factors including nutrition and sex, although many questions remain unanswered. Accordingly, we identify gaps in the literature that remain to be elucidated to fully understand the independent and combined impacts of alcohol and RE on mTORC1 pathway signaling.

Muscle damage imposes stress on mitochondria resulting in mitochondrial fusion, fission, and mitophagy. Testosterone is a regulator of these processes. However, no study has examined the effect of sex-specific resistance exercise (RE)-induced hormonal response on mitochondrial dynamics and mitophagy after muscle damage in untrained men and women. Untrained men and women performed two sessions of 80 unilateral maximal eccentric knee extensions (ECC) followed by upper-body RE (ECC+RE) aimed to induce hormonal changes and maintain a similar lower body demands between conditions or 20 min seated rest (ECC+REST). Vastus lateralis samples were analyzed for gene and protein expression of OPA1, MFN1, DRP1, PINK1, and Parkin at baseline (BL), 12 and 24 h. Testosterone area under the curve was greater for ECC+RE than ECC+REST in men and was greater in men than women for both conditions. A significant time × sex × condition effect was found for Parkin protein expression. At 12 and 24 h, Parkin was lower for ECC + REST than ECC + RE for men; whereas, Parkin was increased at 24 h for women regardless of condition. A significant time effect was found for OPA1 protein expression increasing at 12 and 24 h. A significant time × sex × condition effects were found for MFN1, DRP1, and PINK1 gene expression with increases at 12 h in men for ECC + RE. A significant time × sex effect was found for OPA1 gene expression with a decrease at 12 h in men, and 12 h expression in men was lower than women. RE-induced hormonal changes promoted expression of fission, fusion, and mitophagy markers in men. With muscle damage, regardless of condition, expression of inner mitochondrial membrane fusion markers are promoted in both sexes; whereas, those for mitophagy were promoted in women but reduced in men.
