RESUMO

New 4d-4f heterometallic complexes with a one-dimensional structure, (1)∞[{Ru(valen)(CN)2KRu(valen)(CN)2}{Ln(O2NO)2(CH3OH)3}]·2CH3OH (Ln = Gd, Tb, Dy), have been assembled from the reaction of [K(H2O)2Ru(III)(valen)(CN)2]·H2O with lanthanide nitrates. The exchange interaction between Ru(III) and Gd(III) mediated by the cyanido ligand was determined for the first time and found to be weak and of antiferromagnetic nature.

The aim of the study was to evaluate the influence of metal [Zn(II), Cu(II), Ni(II)] complexes with ursodeoxycholic acid (UDCA) on the viability and proliferation of tumour and non-tumour cells. Cell lines established from retrovirus-transformed chicken hepatoma (LSCC-SF-Mc29) and rat sarcoma (LSR-SF-SR) as well as from human cancers of the breast (MCF-7), uterine cervix (HeLa), lung (A549) and liver (HepG2) were used as model systems. Non-tumour human embryo (Lep-3) cells were also included in some of the experiments. The investigations were carried out by the thiazolyl blue tetrazolium bromide (MTT) test, neutral red uptake cytotoxicity assay, crystal violet staining, double staining with acridine orange and propidium iodide and the colony-forming method. The results obtained revealed that: (1) UDCA and its metal complexes in the tested concentrations decreased (to a varying degree) the viability and proliferation of the treated cells in a time- and concentration-dependent manner; (2) chicken hepatoma (LSCC-SF-Mc29) cells were most sensitive to the cytotoxic and antiproliferative action of the compounds tested, followed by rat sarcoma (LSR-SF-SR) cells; (3) Cu‒UDCA and Ni‒UDCA were more effective against animal LSCC-SF-Mc29 and LSR-SF-SR cells, while Zn‒UDCA significantly decreased the viability and proliferation of human tumour cell lines; (4) applied independently, UDCA expressed lower cytotoxic/cytostatic activity as compared to metal complexes; and (5) the sensitivity of the non-tumour embryonic Lep-3 cells to the effects of UDCA and its metal complexes was comparable or even higher than those of the human tumour cells.

Recent epidemiologic studies carried out in Romania confirmed an ascending trend for cardiovascular disease (CVD) risk factor prevalence such as diabetes mellitus (DM), obesity and dyslipidemia. The aim of this study is to describe the CVD risk factor profile and preventative behavior in a representative sample of the general adult population of an Eastern Romanian urban area. More than 70% of the studied population had a body mass index (BMI) above the normal range for their age, with 36.7% of the subjects residing in obesity and severe obesity clusters. For overweight and obese subjects, the number of comorbidities (CVD, arterial hypertension and DM type 2) was higher than in the population with normal weight (44% vs. 31%, 22% vs. 14% and 18% vs. 10%, respectively). The prevalence of high blood pressure was almost double that reported in previous Romanian studies (69.3% vs. 36.6%) and higher than expected, based on self-reported known CVD diagnoses (37.5%). There was a visible difference between the results obtained for quantifiable CVD risk factors and self-reported lifestyle ones. Routine blood test monitoring may be an easy and inexpensive tool to guide educational and medical interventions to address modifiable CV risk factors in the adult population in order to prevent the fatal consequences of cardiovascular disease.

Inorganic nanoparticles can be embedded within gels by selectively preloading them with suitable molecular precursors followed by reduction or another suitable reaction. Here, we exploit the selective sorption properties of cross-linked ß-cyclodextrin/poly(ethylene glycol) hydrogels, in analogy with polyurethane foams, to preconcentrate metal salts (HAuCl4 and K2[Co(SCN)4]) and subsequently generate gel-embedded metal nanoparticles (10-50 nm). The nanoparticles are shown to be immobilized within the gel network as a consequence of their large dimensions in comparison to the gel network pore size. We suggest this is a useful approach for the generalized synthesis of hybrid soft-hard nanomaterials.

A new series of nanostructured materials was obtained by functionalization of SBA-15 mesoporous silica with Ru(II) and Ru(III) complexes bearing Schiff base ligands derived from salicylaldehyde and various amines (1,2-diaminocyclohexane, 1,2-phenylenediamine, ethylenediamine, 1,3-diamino-2-propanol, N,N-dimethylethylenediamine, 2-aminomethyl-pyridine, and 2-(2-aminoethyl)-pyridine). The incorporation of ruthenium complexes into the porous structure of SBA-15 and the structural, morphological, and textural features of the resulting nanostructured materials were investigated by FTIR, XPS, TG/DTA, zeta potential, SEM, and N2 physisorption. The ruthenium complex-loaded SBA-15 silica samples were tested against A549 lung tumor cells and MRC-5 normal lung fibroblasts. A dose-dependent effect was observed, with the highest antitumoral efficiency being recorded for the material containing [Ru(Salen)(PPh3)Cl] (50%/90% decrease in the A549 cells' viability at a concentration of 70 µg/mL/200 µg/mL after 24 h incubation). The other hybrid materials have also shown good cytotoxicity against cancer cells, depending on the ligand included in the ruthenium complex. The antibacterial assay revealed an inhibitory effect for all samples, the most active being those containing [Ru(Salen)(PPh3)Cl], [Ru(Saldiam)(PPh3)Cl], and [Ru(Salaepy)(PPh3)Cl], especially against Staphylococcus aureus and Enterococcus faecalis Gram-positive strains. In conclusion, these nanostructured hybrid materials could represent valuable tools for the development of multi-pharmacologically active compounds with antiproliferative, antibacterial, and antibiofilm activity.

Two families of homo- and heterometallic complexes, [Zn2L1(µ-OH)(H2O)2](ClO4)2, [Zn2L2(µ-OH)(H2O)2](ClO4)2, [Zn2L3(µ-OH)(H2O)2](ClO4)2, 1∞[{L1Zn2(µ-OH)}{µ-[Ag(CN)2]}](ClO4), [{L1Zn2(µ-OH)}2{µ-[Au(CN)2]}{[Au(CN)2]2}](ClO4)·H2O, 1∞[{L2Zn2(µ3-OH)}2(H2O){µ-[Ag(CN)2]}](ClO4)3·THF·0.5MeOH, 1∞[{L2Zn2(µ3-OH)}2(H2O){µ-[Au(CN)2]}](ClO4)3·THF·H2O, and 1∞[{L3Zn2(µ-OH)}{µ-[Ag(CN)2]}][Ag(CN)2]·H2O, respectively, have been synthesized and characterized. The Schiff bases used as ligands were obtained by condensation reactions of 2,6-diformyl-p-cresol with N,N-dimethyl-ethylenediamine (HL1), 2-aminomethyl-pyridine (HL2), and 2-aminoethyl-pyridine (HL3), respectively. The cytotoxic/cytostatic and genotoxic effects in cultured human MCF-7 (luminal type A breast cancer), MDA-MB-231 (triple negative breast cancer), HeLa (cervical carcinoma), and Lep-3 (non-tumor embryonal fibroblastoid cells) were studied. The investigations were performed by thiazolyl blue tetrazolium bromide test (MTT test), neutral red uptake cytotoxicity assay, crystal violet staining, hematoxylin and eosin staining, double staining with acridine orange and propidium iodide, AnnexinV/FITC, and Comet assay in short-term experiments (24-72 h, with monolayer cell cultures) as well as by 3D colony-forming method in long-term experiments (28 days, with 3D cancer cell colonies). The results obtained revealed that: (i) applied at a concentration range of 0.1-100 µg mL-1, the compounds investigated decrease in a time- and concentration-dependent manner the viability and/or proliferation of the treated cells; (ii) complexes of {Zn(II)Au(I)} show relatively higher cytotoxic/genotoxic activity and antitumor potential as compared to {Zn(II)Ag(I)}; (iii) some of the complexes demonstrate more pronounced cytotoxic potential than commercially available antitumor agents cisplatin, oxaliplatin, and epirubicin.

In this study, three novel magnetic nanocomposites based on carboxyl-functionalized SBA-15 silica and magnetite nanoparticles were prepared through an effective and simple procedure and applied for methylene blue (MB) and malachite green G (MG) adsorption from single and binary solutions. Structure, composition, morphology, magnetic, and textural properties of the composites were thoroughly investigated. The influence of the amount of carboxyl functional groups on the physicochemical and adsorptive properties of the final materials was investigated. The capacity of the synthesized composites to adsorb MB and MG from single and binary solutions and the factors affecting the adsorption process, such as contact time, solution pH, and dye concentration, were assessed. Kinetic modelling showed that the dye adsorption mechanism followed the pseudo-second-order kinetic model, indicating that adsorption was a chemically controlled multilayer process. The adsorption rate was simultaneously controlled by external film diffusion and intraparticle diffusion. It was evidenced that the molecular geometry of the dye molecule plays a major role in the adsorption process, with the planar geometry of the MB molecule favoring adsorption. The analysis of equilibrium data revealed the best description of MB adsorption behavior by the Langmuir isotherm model, whereas the Freundlich model described better the MG adsorption.

BACKGROUND: The incidence of cancer in patients with neurological diseases, who have been treated with LiCl, is below average. LiCl is a well-established inhibitor of Glycogen synthase kinase-3, a kinase that controls several cellular processes, among which is the degradation of the tumour suppressor protein p53. We therefore wondered whether LiCl induces p53-dependent cell death in cancer cell lines and experimental tumours. RESULTS: Here we show that LiCl induces apoptosis of tumour cells both in vitro and in vivo. Cell death was accompanied by cleavage of PARP and Caspases-3, -8 and -10. LiCl-induced cell death was not dependent on p53, but was augmented by its presence. Treatment of tumour cells with LiCl strongly increased TNF-α and FasL expression. Inhibition of TNF-α induction using siRNA or inhibition of FasL binding to its receptor by the Nok-1 antibody potently reduced LiCl-dependent cleavage of Caspase-3 and increased cell survival. Treatment of xenografted rats with LiCl strongly reduced tumour growth. CONCLUSIONS: Induction of cell death by LiCl supports the notion that GSK-3 may represent a promising target for cancer therapy. LiCl-induced cell death is largely independent of p53 and mediated by the release of TNF-α and FasL.Key words: LiCl, TNF-α, FasL, apoptosis, GSK-3, FasL.

Five new cyanido-bridged heterometallic coordination polymers have been obtained by reacting PPh4[RuIII(salpn)(CN)2]·H2O (1) and AsPh4[RuIII(valen)(CN)2]·8.5H2O (2) (H2salpn and H2valen being Schiff-base proligands resulting from the condensation reaction of salicylaldehyde with 1,3-propanediamine and, respectively, o-vanillin with 1,2-ethanediamine) with divalent transition metal perchlorate salts: ∞1[{RuIII(salpn)(CN)2}3{MII(DMF)3}2](ClO4)·4DMF (MII = Mn, 3; Co, 4) and ∞2[{RuIII(valen)(CN)2}4{MII(DMF)3}2{MII(DMF)4}](ClO4)2·4DMF (MII = Mn, 5; Co, 6; Ni, 7), respectively. The dicyanido species, trans-[Ru(salpn)(CN)2]- and trans-[Ru(valen)(CN)2]-, act as metalloligands with the 3d metal ions. Compounds 3 and 4 are isostructural one-dimensional (1D) coordination polymers with a ladder topology. Each MII ion is hexacoordinated by three cyanido groups arising from three {Ru(salpn)(CN)2} units and by the oxygen atoms from three DMF molecules, which are coordinated at meridional positions. Compounds 5-7 are also isostructural, their structures consisting of 2D networks with a herringbone topology. The magnetic susceptibility measurements of these 1D and 2D systems reveal the presence of dominating RuIII-MII antiferromagnetic (AF) interactions in compounds 3, 4, 5 and 6, while ferromagnetic RuIII-NiII interactions are observed in 7. All these compounds stay in their paramagnetic state down to 1.8 K except compound 4 which possesses a 3D ordered AF ground state.

The in situ combination of M(II) cations (Co, Ni, Cu or Zn) with 2,4,6-pyridinetricarboxylic acid as a ligand, a bisamidinium dication as a H-bond donor tecton and NaOH leads to the formation of anionic metal complexes ML2(2-) and their interconnection into isomorphous 3D H-bonded networks displaying different colours which were used as preformed seed crystals for the formation of crystals of crystals by 3D epitaxial growth.

Upon combining the bis-amidinium dication 1-2H(+) with thiocyanatometallate M(SCN)(4)(2-) (M = Pd, Hg) or isothiocyanatometallate Cu(NCS)(4)(2-) anions (behaving as H-bond donor and acceptors, respectively) three new hybrid molecular networks have been obtained in the crystalline phase and structurally characterized by X-ray diffraction on single crystals. Whereas for the combination of tecton 1-2H(+) and both Pd(SCN)(4)(2-) and Hg(SCN)(4)(2-) anions analogous 1-D H-bonded networks were observed, for the Cu(NCS)(4)(2-) anion a 2-D network was obtained. Based on structural features of both components, the formation of the two types of networks is discussed.
