RESUMO

A rabbit serum (0013) used to identify pericentriolar proteins from isolated centrosomes (Gosti-Testu, F., Marty, M.C., Berges, J., Maunoury, R. and Bornens, M. (1986) EMBO J. 5, 2545-2550) was shown also to react through the same epitope with several non-centrosomal proteins including a major 36 kDa cytosolic antigen. This protein was identified to be human lactate dehydrogenase and the co-distribution of 0013 epitope on the centrosomal protein and on lactate dehydrogenase (LDH) was shown to be specific for human cells (Gosti, F., Marty, M.C., Courvalin, J.C., Maunoury, R. and Bornens, M. (1987) Proc. Natl. Acad. Sci. USA 84, 1000-1004). Human hepatic cells constitute, so far, the only exception to this co-distribution rule. By using this cell type which expresses only the LDH-A4 isozyme, we demonstrate that 0013 epitope is specific for the human LDH-B subunit, making serum 0013 the strongest anti-LDH-B available so far. The evolutionary and physiological significance of this situation is discussed.

Hyaluronan (HA) and the hyaluronan-binding glycoprotein hyaluronectin (HN) were measured in 23 gliomas and 8 meningiomas and their location was revisited in 35 tumours. A clear-cut difference was found in the HN/HA ratio values of glioblastomas (below 0.5) and that of astrocytomas (above 0.5 P < 0.001). Besides their location in the intercellular part of gliomas, HA and HN displayed a perivascular location in 1/3 astrocytomas, 17/24 glioblastomas, and 3/7 meningiomas, suggesting they could be produced also by the vascular stroma of tumours and that they would characterise the neoangiogenesis. All cultivated glioma cells tested produced HA in vitro, whereas only 1/11 cell lines produced HN, at a low level. The results obtained suggest that glioma HA and HN are produced by both cancer cells and vascular stroma cells, which contribute to the edification of the extracellular matrix. In meningiomas only the stroma would be responsible for HA and HN production.

In an immunocytochemical investigation of the expression of glial fibrillary acidic protein (GFAP) in non-nervous system tissues ten anti-GFAP antibodies were used on a range of normal adult organs from different species. All four polyclonal and six monoclonal antibodies revealed the expression of GFAP in cells of the zona fasciculata and reticularis of the adrenal cortex and Leydig cells of the Syrian hamster. The Chinese hamster, mole, rat, mouse, guinea pig, rabbit, pig, duck and man were negative. Co-expression of immunoreactivity for GFAP and vimentin was observed in adrenocortical and Leydig cells of the Syrian hamster but there were differences in the staining patterns of these intermediate filament proteins. Expression of GFAP in adrenal cortex of Syrian hamster is confirmed by immunoblot and limited proteolysis analysis which reveal a light form which is immunochemically indistinguishable from its counterpart in the central nervous system. The results presented here suggest a new model for the study of the possible role of GFAP expression in cells known to be sites of steroid synthesis.

Astrocytic cells of unusual aspect can be detected in the cerebellum of normal mice during the first 4 weeks of life. They are visualized with anti-GFAP (glial fibrillary acidic protein), anti-S100 and anti-vimentin immune sera. Their perikaryons, located in the white matter or in the granular layer, extend long processes which are inserted onto the pial surface. These cells may be transitional forms between the radial glial cells and some of the differentiated astroglial elements. These unusual astrocytes are more numerous and heavily stained in the reeler mutant than in the normal mouse and it is suggested that our observations signify some degree of glial immaturity in the cerebellum of the mutant.

A polypeptide of about 120 kDa, called KIN, has been identified in rat FR 3T3 cells by immunoblotting using affinity-purified antibodies against the RecA protein of Escherichia coli (38 kDa). The KIN protein as shown by fluorescent light microscopy and electron microscopy is essentially concentrated in the nucleus. Its level is higher in proliferating than in quiescent cells. Cell treatment with mitomycin C increases the level of the KIN protein. We sought similar proteins in other mammalian cells. Proteins with the same electrophoretic mobility were detected in mouse, monkey and human cell lines as well as in rat and mouse embryos.

RESUMO

Six cell lines originated from foetal mouse brain were maintained in vitro for more than two years. The morphology of most of these cultures suggests their glial character. This is corroborated, for some lines, by the presence of specific glial protein (Glial fibrillary acidic protein, or GFAP). Five out of these lines remained not transplantable in animals during all the experiments. However one of them went through a spontaneous transformation at the eleventh population doubling. This culture, after transformation remained glial and when injected in animals, induced GFAP containing tumors.

RESUMO

Investigations performed by various workers during the last ten years suggest that some forms of mental illness are associated with a variety of immunological reactions. Significant variations in immunoglobulin ratios have been observed in patients with psychotic illness, and cellular and humoral cytotoxic activities against cerebral tissue have been demonstrated in vitro. At the molecular level, several subjects can be proposed for investigation into immunological phenomena in neuropathology. Thus, the neurospecific antigens, which have been isolated by biochemical methods, but the physiological role of which is largely unknown, open news ways to the experimental exploration of psychopathology.

A histological study of lymphocytic infiltration has been undertaken following removal at operation of one hundred human gliomas. This study has been completed with eight cases of tumour recurrence, six attempted autologous grafts, twenty one post-mortems and fourteen explorations of delayed hypersensitivity reactions. About half of the glioma specimens showed lymphocytic infiltration and no reaction was present in the other. Within this series of glioblastomas a correlation existed between the presence of infiltrating lymphocytes and the duration both of pre and post operative tumour evolution. The rejection of autologous grafts seemed to depend directly on the presence of lymphocytic infiltrations associated with the primary tumour. The retarded hypersensitivity reactions obtained after intradermic inoculations of cellular lyophilizates did not seem to be directly related either to the phenomenon of autologous graft rejection or to the presence of lymphocytic infiltrations. The results suggest that there is a complex system represented by two antigenic groups of transplantation type and sensitization type.

RESUMO

Three cell lines derived in our laboratory from human malignant gliomas (SA 130, SA 132, SA 134) were injected subcutaneously into pathogen-free nude thymus less mice. These three cell lines gave origine to malignant tumors which, as original tumors, were positive for the glial fibrillary acidic protein (GFAP) revealed by immunoperoxidase method.

On the basis of studies utilizing antibody to GFAP (glial fibrillary acidic protein) in the indirect immunofluorescent and immunoperoxydase methods we report the presence of GFAP in 5 astrocytomas, 1 ependymoma and 1 medulloblastoma. The GFAP was evidenced on cryostat sections of frozen material and in short-term tissue culture. Five others tumors including 1 oligodendrocytoma, 1 choroid-plexus papilloma, 1 meningioma and 2 secondary sarcomas were negative. Possible applications of antibodies to GFAP for localization and therapy of brains tumors were considered.

A series of 50 human primary intracranial tumors were cultivated in vitro in an attempt to establish cell lines with the trypsinization technique. During the in vitro adaptation period, cultures were maintained at high cell density to avoid rapid over-growth by connective tissue. Five lines were established from 5 tumors including 4 malignant gliomas (SA4, SA44, SA45, SA101) and 1 meningeal sarcoma (SA39). These 5 lines were compared for the following investigations and criteria: light and electron microscopy, chromosomal patterns, saturation density, plating efficiency, doubling time, synthesis of collagen, S-100 and GFA proteins, response to d b-c AMP and tumorigenicity into nude mice. For one or more of the characteristics mentioned above each line was distinct from the others. None of the growth parameters that serve to define the abnormal state in vitro correlated consistently with cellular tumorigenicity in athymic nude mice. The nature of lines derived from gliomas remain uncertain and the collagen synthesis in vitro is not sufficient to affirm that these lines are mesenchymal rather than glial.

RESUMO

Twelve human tumors were cultivated in vitro by the trypsinization technique and 3 cell lines were established. Each of these 3 lines showed individual characteristics which were maintained during passages. After inoculation into nude mice, 2 lines induced tumors which possessed the histologic features of the original human tumors (melanoma and carcinoma of stomach), the other line derived from a breast carcinoma was rejected.

RESUMO

A rabbit showing natural centrosphere-immunoreactivity was immunized with streptococcus pyogenes type 24 (S 24). During the immunization, an additional NOR-immunoreactivity appeared. This reactivity was not inhibited after the serum absorption by S 24 and seems fortuitous. The simultaneous immunoperoxidase technique staining centrosphere and NOR is described and illustrated during successive phases of mitosis. Unique as of this date, this serum may serve as a useful tool to study the centrosphere and NOR.

Human serum SH2172, obtained from a girl suffering from bullous dermatosis, showed a natural immunoreactivity against the peripheral nervous system (PNS) of rat, mouse and hamster. Immunocytochemical staining and examination by light and electron microscopy demonstrated an intracellular neurofibrillary network localized in neurites and neuronal pericaryons. Comparative testing clearly showed that SH2172 immunoreactivity is different from that of the antibodies against the triplet of proteins NFP70, 160 and 200 kD. This unique serum should be a useful probe to study PNS neurocytoskeleton.

By immunofluorescent and immunoperoxidase techniques and with the use of highly purified antibodies, tubulin and its associated proteins, MAP1 and MAP2 have been identified and precisely localized in human spermatozoa. In relation to well known structural elements, tubulin appeared localized to the region of the axoneme, the centriolar adjunct and the equatorial part of the acrosome, in contrast MAP1 and MAP2 were localized to the post-nuclear region and in the fibrous sheath of the principal piece of the flagella.

A number of investigations have suggested a fundamental role for lysosomal enzymes in growth control of normal cells and in the loss of growth control which occurs in neoplasia and in cell cultures transformed by oncogenic viruses. In this investigation total enzyme activities and rates of extracellular accumulation of enzyme were measured in long term cultures of gliomas and normal human astrocytes. Marked differences were found between the cultures with regard to total enzyme activity. Exocytosis rates of the enzymes also showed differences, with one group of tumours demonstrating low or intermediate rates and another smaller group with high rates. There was no positive correlation between the enzyme profile of the cultures and their tumorigenicity in nude mice.
