The prevalence of multidrug-resistant Campylobacter species in wastewater effluents presents a formidable challenge at the intersection of environmental sustainability and public health. This study examined the presence of multidrug-resistant Campylobacter in wastewater effluents in the Eastern Cape Province, South Africa, and its implications for environmental ecosystems and public health. Forty-five samples from household effluent (HHE) and wastewater treatment plant effluent (WWTPE) were collected at different geographical locations within the province between April and September 2022. The counts of the presumptive Campylobacter genus ranged from 5.2 × 103 to 6.03 × 104 CFU/mL for HHE and 4.93 × 103 to 1.04 × 104 CFU/mL for WWTPE. About 42.55% of the samples were positive for Campylobacter species. Five virulence determinants including the cadF and wlaN were detected in all the isolates; however, flgR (19.23%), ciaB, and ceuE (15.38%) were less prevalent. The antibiogram profiles of confirmed Campylobacter isolates revealed high resistance (>55%) against all tested antibiotics ranging from 55.77% (nalidixic acid) to 92.30% (erythromycin), and resistance against the other antibiotics followed the order ciprofloxacin (51.92%), azithromycin (50%), and levofloxacin (48.08%). On the contrary, gentamicin was sensitive against 61.54% of the isolates, followed by imipenem (57.69%) and streptomycin (51.92%). The WWTPE's antibiotic resistance index (ARI) was 0.19, lower than the permitted Krumperman threshold of 0.2; and HHE's ARIs were higher. The isolates' respective multiple antibiotic resistance indexes (MARI) varied between 0.08 and 1.00. Among the phenotypically resistant Campylobacter isolates examined, 21 resistance determinants encoding resistance against ß-lactam, carbapenems, aminoglycosides, phenicol, quinolones, tetracyclines, and macrolides were detected, which explains the phenotypic resistance observed in the study. This study concludes that the wastewaters in the study areas are important reservoirs of multidrug-resistant and potentially pathogenic Campylobacter species, suggesting the need for proper treatment of the wastewaters to eliminate the organisms in the effluents before discharge the final effluent to the receiving watershed.

BACKGROUND: Virulence determinants are crucial to the risk assessment of pathogens in an environment. This study investigated the presence of eleven key virulence-associated genes in Vibrio cholerae (n = 111) and Vibrio mimicus (n = 22) and eight virulence determinants in Vibrio alginolyticus (n = 65) and Vibrio parahaemolyticus (n = 17) isolated from six important water resources in Eastern Cape, South Africa, using PCR techniques. The multiple virulence gene indexes (MVGI) for sampling sites and isolates as well as hotspots for potential vibriosis outbreaks among sampling sites were determined statistically based on the comparison of MVGI. RESULT: The PCR assay showed that all the V. cholerae isolates belong to non-O1/non-O139 serogroups. Of the isolates, Vibrio Cholera (84%), V. mimicus (73%), V. alginolyticus (91%) and V. parahaemolyticus (100%) isolates harboured at least one of the virulence-associated genes investigated. The virulence gene combinations detected in isolates varied at sampling site and across sites. Typical virulence-associated determinants of V. cholerae were detected in V. mimicus while that of V. parahaemolyticus were detected in V. alginolyticus. The isolates with the highest MVGI were recovered from three estuaries (Sunday river, Swartkopps river, buffalo river) and a freshwater resource (Lashinton river). The cumulative MVGI for V. cholerae, V. mimicus, V. alginolyticus and V. parahaemolyticus isolates were 0.34, 0.20, 0.45, and 0.40 respectively. The targeted Vibrio spp. in increasing order of the public health risk posed in our study areas based on the MVGI is V. alginolyticus > V. parahaemolyticus > V. cholerae > V. mimicus. Five (sites SR, PA5, PA6, EL4 and EL6) out of the seventeen sampling sites were detected as the hotspots for potential cholera-like infection and vibriosis outbreaks. CONCLUSIONS: Our findings suggest that humans having contact with water resources in our study areas are exposed to potential public health risks owing to the detection of virulent determinants in human pathogenic Vibrio spp. recovered from the water resources. The study affirms the relevancy of environmental Vibrio species to the epidemiology of vibriosis, cholera and cholera-like infections. Hence we suggest a monitoring program for human pathogenic Vibrio spp. in the environment most especially surface water that humans have contact with regularly.

Bacterial species responsible for food infections and intoxication are sometimes carried through the food production and processing. Very few published literatures exist on integrons among antibiotic-resistant staphylococcal strains from foods of animal origin in Gauteng Province, South Africa, hence this study. A total of 720 samples (360 meat and 360 dairies) from a community abattoir of a research farm in South Africa, using conventional bacteriological and molecular methods. Nine (9) bacterial strains, including Bacillus subtilis AYO-123, Acinetobacter baumannii AYO-241, Staphylococcus lentus AYO-352, among others were identified and submitted to GenBank. More bacterial strains were recovered from raw meat (90.5%) than dairy products (9.5%). Resistance was shown (0-100%) to Imipenem, Meropenem, Norfloxacin, Clindamycin, and 22 other antibiotics, without any carbapenem-resistant Acinetobacter baumannii and methicillin/vancomycin-resistant Staphylococcus species (MRSS/VRSS). Virulence genes for fibronectin-binding protein A (FnbA) were predominant (56.24%) followed by the circulating nucleic acids (cna) gene (43.75%). Others were staphylococcal enterotoxin A (sea, 41%), staphylococcal enterotoxin B (seb, 23.5%). Co-presence of sea and seb genes occurred in 11.76% of the isolates, but no coa genes was amplified. Antibiotic resistance genes (ARGs), tetK (70.58%), linA (29.4%), and ermA (11.76%) were detected, but none of the mecA and vat genes was amplified. Class 2 integron (50%) was more predominantly detected than integron 1 (25%), but no Class 3 integron was detected. Bacteria with both the detected virulence and antibiotic resistance genes are of potential risks to human health.

Enterococci are regarded as a better indication of faecal pollution in freshwater and marine waters. Their levels in seawater are positively connected with swimming-related gastrointestinal disorders. This study used an Enterococcus-specific polymerase chain reaction (PCR) to characterize the isolates. Classes 1 and 2 integrons were examined for environmental Enterococcus isolates using a standard biological procedure. All strains were assessed against a panel of 12 antibiotics from various classes using disc diffusion methods. The microdilution method was used to work out the minimum inhibitory concentration (MIC) according to the CLSI guiding principles. The combination therapy of the resistant drugs was evaluated using a checkerboard assay and a time-dependent test for assessing their bactericidal or bacteriostatic activity. The gene diversity of the tested organisms was analyzed with the aid of Enterobacterial Repetitive Intergenic Consensus (ERIC) PCR. In total, 57 Enterococcus spp. environmental samples were recovered, in which Enterococcus faecalis (33.33%) and Enterococcus faecium (59.65%) were the dominant species. Resistance to linezolid, ciprofloxacin, erythromycin, gentamicin, vancomycin, rifampicin, and tetracycline was prevalent. Fifty (50) strains tested positive for class 1 integron, more frequent in Enterococcus faecium and Enterococcus faecalis isolates, with no gene cassette array discovered. A combination of gentamicin (MIC 4 µg/mL) with vancomycin (MIC 256 µg/mL) antibiotics against Enterococcus faecalis showed antibacterial activity. In contrast, the combination of ciprofloxacin (1 µg/mL) with Ampicillin (16 µg/mL) antibiotics against Enterococcus faecalis showed a bacteriostatic effect. The ERIC-PCR analysis pointed out that most of the assessed isolates have close genetic similarities.

This study focused on evaluation of the levels, seasonal variations and human health risks associated with polychlorinated naphthalenes (PCNs) in water and sediment samples of Markman Canal using solid phase and soxhlet extraction methods respectively, followed by clean-up and quantification with gas chromatograph coupled with microelectron capture detector. The sum of eight PCNs congener's (∑8PCNs) in water and sediments varied from 0.035 to 0.699 µg/L and 0.260 to 6744.16 µg/kg dw, respectively. Highest PCNs concentrations were recorded in water during winter, while sediment samples collected during spring contained maximum levels. The estimated toxic equivalency (TEQ) for water and sediments was 1.19 × 10-7-1.47 × 10-4 µg/L and 4.43 × 10-5-4.19 × 10-1 µg/kg consecutively. The PCN levels and TEQ values revealed that this waterbody is polluted but constitutes no excess health risk. Efforts should be made to control all the activities contributing to pollution of this canal.

Herbicides are chemicals used globally to kill unwanted plants so as to obtain high agricultural yields and good agricultural products. Herbicides are sometimes transported from the farmlands into water bodies mainly through runoffs. These chemicals are recalcitrant, and their accumulation is hazardous to abiotic and biotic components of the ecosystem. At present, the best alternative technology for elimination of herbicides in water is the usage of advanced oxidation processes (AOPs). The AOPs, which are performed homogeneously or heterogeneously, are capable of breaking down complex pollutants in water into carbon dioxide and mineral compounds. In these processes, ·OH is produced and used for degradation process. It is recommended that the total organic carbon (TOC) produced during degradation reaction be monitored because the ‧OH produced or generated can react to form intermediates before complete mineralisation is achieved. Different kinds of AOPs for degradation of herbicides have their specific advantages as well as limitations. This report shows that AOPs are excellent techniques for degradation of herbicides in aqueous solutions, and the mechanisms showed that herbicides were mineralised. The amount and type of photocatalysts, pH of the medium, surface characteristics of the photocatalysts, doping of the photocatalysts, temperature of the medium, concentration of herbicides, presence of competing ions, intensity and irradiation period, and type of oxidants have great influence on the degradation of herbicides in water. Overall, this report showed that most AOPs could not completely degrade herbicides in water and complete degradation can be achieved by developing novel and robust AOPs that will completely mineralise herbicides in water-this will pave way for water and environmental safety.

Detection of perfluorinated compounds (PFCs) in the environment has been a global concern because of the risk they pose due to their endocrine-disruptive properties. This study analyzed the global trends and research productivity of PFCs from 1990 to 2021. A total number of 3256 articles on PFCs were retrieved from the Web of Science focusing on different environmental and biological matrices. An increase in the productivity of research on PFCs was observed during the survey period which indicates that more research and publications on this class of contaminants are expected in the future. Evaluating the most productive countries and the number of citations per country on PFCs research shows that China and the United States of America were ranked in first and second places. It was also observed that research on PFCs received the most attention from scientists in developed countries, with little research emerging from Africa. Hence, research on PFCs in developing countries, especially low-income countries should be promoted. Consequently, more research programs should be implemented to investigate PFCs in countries and regions where research on these contaminants is low. The study will help researchers, government agencies and policymakers to tailor future research, allocation of funds to PFCs research and countries' collaboration.

Polycyclic aromatic hydrocarbons (PAHs) are a large group of diverse hazardous organic compounds that are relatively stable and widely distributed throughout the world's ecosystems due to various anthropogenic activities. They are generally less soluble in water and have a low vapour pressure, but dissolve easily in adipose tissues; and they bioaccumulate into high concentrations in aquatic animals, thereby exerting a variety of hazardous and lethal effects. Despite the plethora of research studies on these pollutants, only few bibliometric reviews on the subject have been documented in the literature. As a result, the present study aimed to assess the research growth on PAHs-related studies across different ecosystems. Science Citation Index-Expanded of Web of Science was explored to obtain the research studies that were conducted between 1991 and 2020, and RStudio was utilized for the data analysis. Annual productivity increased arithmetically over the years, with a 9.2% annual growth rate and a collaboration index of 2.52. Foremost among the trend topics in this field of study include soil, sediments, biodegradation, bioremediation, bioavailability, and source apportionment. China, USA, Spain, France and Germany were the five top-ranked countries in terms of publications and citations over the three decades investigated; however, Korea, Japan, United Kingdom, Germany, and Canada were ranked as the five leading countries in terms of collaboration per published article (MCP ratio). Therefore, efforts to strengthen international collaboration in this field of study especially among the less participating countries and continents are thus encouraged. The findings of this study are expected to provide future direction for the upcoming researchers in identifying the hot spots in this field of study as well as research leaders whom to seek collaboration in their future research plan.

Somatic antigen agglutinable type-1/139 Vibrio cholerae (SAAT-1/139-Vc) members or O1/O139 V. cholerae have been described by various investigators as pathogenic due to their increasing virulence potential and production of choleragen. Reported cholera outbreak cases around the world have been associated with these choleragenic V. cholerae with high case fatality affecting various human and animals. These virulent Vibrio members have shown genealogical and phylogenetic relationship with the avirulent somatic antigen non-agglutinable strains of 1/139 V. cholerae (SANAS-1/139- Vc) or O1/O139 non-agglutinating V. cholerae (O1/O139-NAG-Vc). Reports on implication of O1/O139-NAGVc members in most sporadic cholera/cholera-like cases of diarrhea, production of cholera toxin and transmission via consumption and/or contact with contaminated water/seafood are currently on the rise. Some reported sporadic cases of cholera outbreaks and observed change in nature has also been tracable to these non-agglutinable Vibrio members (O1/O139-NAGVc) yet there is a sustained paucity of research interest on the non-agglutinable V. cholerae members. The emergence of fulminating extraintestinal and systemic vibriosis is another aspect of SANAS-1/139- Vc implication which has received low attention in terms of research driven interest. This review addresses the need to appraise and continually expand research based studies on the somatic antigen non-serogroup agglutinable type-1/139 V. cholerae members which are currently prevalent in studies of water bodies, fruits/vegetables, foods and terrestrial environment. Our opinion is amassed from interest in integrated surveillance studies, management/control of cholera outbreaks as well as diarrhea and other disease-related cases both in the rural, suburban and urban metropolis.

Acinetobacter baumannii (A. baumannii) plays a significant part in nosocomial infections world over and is re-emerging as a formidable pathogen due to the wide range of antibiotic resistance factors it acquires and environmental resilience. The high attendance of patients (outpatients and inpatients) into the health care facilities formed the basis for the selection of the hospitals. Consequently, this study profiled the antibiogram and antibiotic resistance genes of A. baumannii isolated from selected hospital wastewater effluents. A total of twenty-four (24) wastewater samples from three selected hospital drainages were collected and analysed presumptively by culture-dependent methods for A. baumannii. The identity confirmation of A. baumannii was done by the amplification of recA and blaoxa-51 genes. Virulence and antibiotic resistance markers were assessed using polymerase chain reaction. A total of 53 A. baumannii isolates were confirmed and the highest antibiotic resistance profile was 93% (piperacillin). Multiple antibiotic resistance index (MARI) showed a range of 0.23 and 0.46. FimH virulence gene was detected in 29 (55%) of the isolates. Tetracycline and beta-lactam resistance markers were found; 70% and 92% of the isolates possessed tetA and ampC genes. The isolates showed high level of resistance to antibiotics. The multiple antibiotic resistance index (MARI) of ≥ 0.2 indicates that some of the isolates harbour virulence and resistance traits emerging from high-risk source thereby projecting a threat to public health.

Antibiotic resistance has become a major public health concern around the world. This is exacerbated by the non-discovery of novel drugs, the development of resistance mechanisms in most of the clinical isolates of bacteria, as well as recurring infections, hindering disease treatment efficacy. In vitro data has shown that antibiotic combinations can be effective when microorganisms are resistant to individual drugs. Recently, advances in the direction of combination therapy for the treatment of multidrug-resistant (MDR) bacterial infections have embraced antibiotic combinations and the use of nanoparticles conjugated with antibiotics. Nanoparticles (NPs) can penetrate the cellular membrane of disease-causing organisms and obstruct essential molecular pathways, showing unique antibacterial mechanisms. Combined with the optimal drugs, NPs have established synergy and may assist in regulating the general threat of emergent bacterial resistance. This review comprises a general overview of antibiotic combinations strategies for the treatment of microbial infections. The potential of antibiotic combinations with NPs as new entrants in the antimicrobial therapy domain is discussed.

In recent times, there has been a cumulative apprehension regarding organophosphate flame retardants (OPFRs) owing to their high manufacturing and usage after brominated flame retardants were strictly regulated and banned from being distributed and used in many countries. OPFRs are known as the main organic pollutants in the terrestrial and aquatic environment. They are very dangerous to humans, plants and animals. They are also carcinogenic and some have been implicated in neurodevelopmental and fertility challenges. OPFRs are distributed into the environment through a number of processes, including the usage, improper disposal and production of materials. The solid phase extraction (SPE) method is suggested for the extraction of OPFRs from water samples since it provides high quality recoveries ranging from 67% to 105% and relative standard deviations (RSDs) below 20%. In the same vein, microwave-assisted extraction (MAE) is highly advocated for the extraction of OPFRs from sediment/soil. Recoveries in the range of 78% to 105% and RSDs ranging from 3% to 8% have been reported. Hence, it is a faster method of extraction for solid samples and only demands a reduced amount of solvent, unlike other methods. The extract of OPFRs from various matrices is then followed by a clean-up of the extract using a silica gel packed column followed by the quantification of compounds by gas chromatography coupled with a mass spectrometer (GC-MS) or a flame ionization detector (GC-FID). In this paper, different analytical methods for the evaluation of OPFRs in different environmental samples are reviewed. The effects and toxicities of these contaminants on humans and other organisms are also discussed.

Pesticides are synthetic compounds that may become environmental contaminants through their use and application. The high productivity achieved in the agricultural industry can be credited to the use and application of pesticides by way of pest and insect control. As much as pesticides have a positive impact on the agricultural industry, some disadvantages come with their application in the environment because they are intentionally toxic, and this is more towards non-target organisms. They are grouped into chlorophenols, organochlorines, synthetic pyrethroid, carbamates, and organophosphorus based on their structure. The symptoms of exposure to carbamate (CM) and organophosphates (OP) are similar, although poisoning from CM is of a shorter duration. The analytical evaluation of carbamate and organophosphate pesticides in human and environmental matrices are reviewed using suitable extraction and analytical methods.

The concentrations, potential sources, and compositional profile of PBDEs in the surface water and sediment of Nahoon Estuary, East London, South Africa, were investigated with solid-phase extraction and ultra-sonication, respectively, followed by gas-chromatography-electron capture detection. The seasonal range of the contaminants' concentrations in water and sediment samples in spring season were ∑PBDE 329 ± 48.3 ng/L (25.32-785 ng/L) and ∑PBDE 4.19 ± 0.35 ng/g dw (1.91-6.57 ng/g), but ∑PBDE 62.1 ± 1.50 ng/L (30.1-110 ng/L) and ∑PBDE 65.4 ± 15.9 ng/g dw (1.98-235 ng/g) in summer, respectively. NH1 (first sampling point) was the most contaminated site with PBDE in the Estuary. The potential source of pollution is attributed to the stormwater runoff from a creek emptying directly into the Estuary. This study's dominant PBDE congener is BDE-17, ranging from below detection limit to 247 ng/L and 0.14-32.1 ng/g in water and sediment samples, respectively. Most detected at all the sites were BDE-17, 47, 66, and 100. Most BDE-153 and 183 are found in sediment in agreement with the fact that higher brominated congeners of PBDE adsorb to solid materials. There was no correlation between the congeners and organic carbon and organic matter. However, the human health risk assessment conducted revealed that the PBDE concentration detected in the estuary poses a low eco-toxicological risk. Nevertheless, constant monitoring should be ensured to see that the river remains safe for the users, as it serves as a form of recreation to the public and a catchment to some neighbourhoods.

This study investigates the pollution of Markman stormwater runoff, which is a tributary to Swartkops River Estuary. Solid-phase and ultrasonic extraction methods were utilized in the extraction of water and sediment samples, respectively. The pH of the sampling sites was above the EU guideline. The ranges of concentration of [Formula: see text]PBDE obtained in water and sediment samples for all the seasons were 58.47-1357 ng/L and 175-408 ng/g, respectively. Results also showed that BDE-66 was the dominant congener, specifically in the industrial zone, where its concentrations ranged from 2 to 407 ng/g in sediment. Consequently, the high concentration of BDE- 66 in the sediment of stormwater calls for concern. Penta-BDE suggests potential moderate eco-toxicological risk, as evident in the calculated risk assessment. The result showed possible photodegradation along the contaminant's travel time, as only 7% of the PBDE was detected at the point of entry into the Swartkops River Estuary. Markman stormwater may be contributing heavily to the pollution load of Swartkops River, as evident in the alarming concentrations of PBDEs obtained. The industries at this zone should eliminate the contaminants before discharging their effluents into the canal.

There have been many global studies on the occurrence and distribution of pharmaceuticals and personal care products (PPCPs) in the aquatic resources, but reports on the effects of physicochemical properties of water on their concentrations are very scarce. The amounts and removal of these contaminants in various environmental media are dependent on these physicochemical properties, which include pH, temperature, electrical conductivity, salinity, turbidity, and dissolved oxygen. Here, we reviewed the influence of these properties on determination of PPCPs. Reports showed that increase in turbidity, electrical conductivity, and salinity gives increase in concentrations of PPCPs. Also, neutral pH gives higher PPCP concentrations, while decrease in temperature and dissolved oxygen gives low concentration of PPCPs. Nevertheless, it is quite challenging to ascertain the influence of water quality parameters on the PPCP concentration, as other factors like climate change, type of water, source of pollution, persistence, and dilution factor may have great influence on the concentration of PPCPs. Therefore, routine monitoring is suggested as most water quality parameters vary because of effects of climate change.

BACKGROUND: This study describes the characteristics of pregnant women on antiretroviral therapy (ART) and the rate of peripartum virologic suppression in a large prevention of mother-to-child transmission cohort who delivered in some selected maternity centers in Eastern Cape Province, South Africa. In addition, the study examines the factors associated with virologic suppression in the cohort. METHODS: This multicenter, retrospective cross-sectional analysis included medical data of 1709 women with human immunodeficiency virus between September 2015 and May 2016 in Eastern Cape Province. The main outcome measure was the rate of peripartum virologic suppression, defined as viral load (VL) <1000 copies/mL and undetectable viremia (VL <20 copies/mL). Correlates of peripartum virologic suppression and undetectable viremia were examined by fitting logistic regression model analysis. RESULTS: Of 1463 women with available VL results, the overall rate of peripartum suppression was 82%, and undetectable viremia was 56.9%. Being aged 24 years or younger (adjusted odds ratio [AOR], 0.68 [95% confidence interval {CI}, .48-.94]), smoking during pregnancy (AOR, 0.50 [95% CI, .28-.90]), and starting ART in the first trimester were associated with lower odds of viral suppression (<1000 copies/mL). Women who had never defaulted ART had an increased odds of having an undetectable VL (AOR, 3.09 [95% CI, 2.12-4.49]) and virologic suppression (AOR, 3.88 [95% CI, 2.62-5.74]) compared to those who defaulted. CONCLUSIONS: More than half of the women achieved undetectable VL, and 4 in 5 women achieved viral suppression at delivery in the region. Early antenatal booking, combined with enhanced adherence support for pregnant women on ART, would be crucial toward achieving the goal of elimination of mother-to-child transmission in the region.

OBJECTIVES: Decaying wood samples were collected, and actinomycetes were isolated and screened for laccase production. The identity of the efficient laccase-producing isolate was confirmed by using a molecular approach. Fermentation conditions for laccase production were optimized, and laccase biochemical properties were studied. RESULTS: Based on the 16S rRNA gene sequencing and phylogenetic analysis, the isolate coded as HWP3 was identified as Streptomyces sp. LAO. The time-course study showed that the isolate optimally produced laccase at 84 h with 40.58 ± 2.35 U/mL activity. The optimized physicochemical conditions consisted of pH 5.0, ferulic acid (0.04%; v/v), pine back (0.2 g/L), urea (1.0 g/L), and lactose (1 g/L). Streptomyces sp. LAO laccase was optimally active at pH and temperature of 8.0 and 90 °C, respectively, with remarkable pH and thermal stability. Furthermore, the enzyme had a sufficient tolerance for organic solvents after 16 h of preincubation, with laccase activity > 70%. Additionally, the laccase maintained considerable residual activity after pretreatment with 100 mM of chemical agents, including sodium dodecyl sulphate (69.93 ± 0.89%), ethylenediaminetetraacetic acid (93.1 ± 7.85%), NaN3 (96.28 ± 3.34%) and urea (106.03 ± 10.72%). CONCLUSION: The laccase's pH and thermal stability; and robust catalytic efficiency in the presence of organic solvents suggest its industrial and biotechnological application potentials for the sustainable development of green chemistry.

Chlorine is the commonest and cheapest disinfectant used in drinking water and wastewater treatment at household, municipal and industrial levels. However, the uprising of microbial chlorine resistance (MCR) pose critical public health hazard concerns; because, its potentiate exposure to difficult-to-treat resistant pathogens. Therefore, this study aimed at evaluating the burden of MCR in drinking water/wastewater treatment and distribution systems (DWWTDS) via science mapping of research productivity (authors, countries, institutions), thematic conceptual framework, disciplines, research networks and associated intellectual landscape. MCR data were mined from Scopus and Web of Science based on optimized algorithms with the root key term "chlorine* resistant*'' and analysed for pre-set indicator variables. Results revealed 1127 documents from 442 journals and 1430% average growth rate (AGR) of research articles from 2017 to 2019 on MCR. Country-wise, the USA (n = 299), China (n = 119), and Japan (n = 43) ranked in the 1st, 2nd, and 3rd positions respectively, among the top participating countries in MCR research. MCR research had considerable performance in public health and sustainable concern subjects namely, Environmental Sciences & Ecology, Engineering, Microbiology, Water Resources, Biotechnology & Applied Microbiology, Food Science & Technology, Public, Environ & Occupational Health, Chemistry, Infectious Diseases, and Marine & Freshwater Biology; and with noticeable AGR in Environmental Sciences & Ecology (330%) and Infectious Diseases (130%). The study found biofilm-related thrusts (n = 90, 270% AGR) as main research hotspots on MCR. Overall, the study identified and discussed four important thematic areas of public health challenges in MCR that could promote increasing waterborne diseases due to (re)emerging pathogens, enteric viruses and dissemination in DWWTDS. In conclusion, this study provides comprehensive overview of the growing burden of MCR in DWWTDS and standout as a primer of information for researchers on MCR. It recommends direct, intentional and integrated research priorities on MCR to overcome accompanying public health and environmental threats. In addition, chlorine resistance in waterborne fungi have not received research attention. Research activities related to fungal chlorine resistance will be an invaluable future direction in DWWTDS and guide against exposure to waterborne pathogenic fungi and mycotoxins. It is unknown whether chlorine resistance can be acquired by horizontal gene transfer in microorganisms and future research should elucidate this important thrust.

Drug resistance has been recognized in all available therapeutic class of medications for the management of human immunodeficiency virus-1 (HIV-1) infected patients. This makes the continuous study of HIV drug resistance and new treatment options pertinent to patients and researchers globally. The aim of this study is to analyze the complete HIV-1 integrase gene for the possible occurrence of resistance mutations or polymorphisms. We performed genetic analyses on 48 treatment-naive HIV-1-infected patients using nested polymerase chain reaction. Integrase drug-related resistance mutation (DRMs) analysis was performed on all generated sequences according to Stanford HIV drug interpretation program and the International AIDS Society-USA guidelines while phylogenetic analysis was inferred using MEGA 6. The study revealed no major resistance-associated mutation. However, E157Q (2.1%), L74M/I (4.2%), and P142T (2.1%) were the observed accessory and polymorphic mutations. Naturally occurring polymorphism observed were E11D, K14R, D25E, V31I, M50I, V72I, P90T, F100Y, L101I, T124A, T125A, K136Q, D167E, V201I, L234I, A265V, A269K, D278A, and S283G. Phylogenetic analysis delineated all the sequences as HIV-1 subtype C. The study revealed the absence of major integrase inhibitors associated resistance mutations in a setting where integrase inhibitor is administered as salvage therapy in patients developing resistance to first and second-line antiretroviral treatment. However minor and natural polymorphisms were observed and thus may influence the outcome of each treatment regimen. However, additional studies are required to precisely evaluate the impact of these mutations on integrase inhibitors in the Eastern Cape of South Africa.
