Anthropogenic activities such as mining and the metallurgical industry are the main sources of mercury contamination. Mercury is one of the most serious environmental problems in the world. This study aimed to investigate, using experimental kinetic data, the effect of different inorganic mercury (Hg2+) concentrations on the response of microalga Desmodesmus armatus stress. Cell growth, nutrients uptake and mercury ions from the extracellular medium, and oxygen production were determined. A Compartment Structured Model allowed elucidating the phenomena of transmembrane transport, including influx and efflux of nutrients, metal ions and bioadsorption of metal ions on the cell wall, which are difficult to determine experimentally. This model was able to explain two tolerance mechanisms against mercury, the first one was the adsorption of Hg2+ions onto the cell wall and the second was the efflux of mercury ions. The model predicted a competition between internalization and adsorption with a maximum tolerable concentration of 5.29 mg/L of HgCl2. The kinetic data and the model showed that mercury causes physiological changes in the cell, which allow the microalga to adapt to these new conditions to counteract the toxic effects. For this reason, D. armatus can be considered as a Hg-tolerant microalga. This tolerance capacity is associated with the activation of the efflux as a detoxification mechanism that facilitates the maintenance of the osmotic balance for all the modeled chemical species. Furthermore, the accumulation of mercury in the cell membrane suggests the presence of thiol groups associated with its internalization, leading to the conclusion that metabolically active tolerance mechanisms are dominant over passive ones.

In addition to plant-derived, fungal pigments have become an alternative in respect to synthetic ones. Besides Monascus sp., several pigment-producing fungi do not have culture conditions well-established yet. In this research, media composition, light wavelength and co-culture were evaluated, results were reported in Absorbance Units per gram of biomass (AU/Bgr). For Fusarium oxysporum a C:N ratio above 7 was advantageous, using both complex and defined media; blue LED light increased the AU/Bgr value from 18013 to 344; co-culture did not enhance pigment production. In Aspergillus chevalieri a high C:N ratio with glucose as carbon source was ideal. When exposing cultures to light, UV and red light gave the highest pigmentation; moreover, differential UV-VIS spectra in all wavelengths suggested production of additional pigments. Particularly a pigment observed when cultured in green light was also found in co-culture with yeast and there was an improvement of AU/Bgr value of 52549%. This is the first report regarding light effect and co-culture for these fungi, as well as C:N ratio for A. chevalieri.

Brewer's spent grain (BSG) is a promising feedstock for ABE fermentation. Sulfuric acid pretreatment of BSG at pH 1, 121°C and different solid loadings (5-15% w/w) was investigated. Enzymatic hydrolysis and ABE fermentation by Clostridium beijerinckii DSM 6422 of non-washed and washed pretreated BSG were performed to compare monosaccharide release and butanol production. Pretreatment at 15% w/w BSG resulted in higher availability of sugars in both the enzymatic hydrolysates and pretreatment liquid, and overall yields of 75gbutanol/kg BSG and 95gABE/kg BSG were obtained. When the enzymatic hydrolysate from the washed pretreated BSG was fermented, butanol (6.0±0.5g/L) and ABE (7.4±1.0g/L) concentrations were lower compared with 7.5±0.6g/L butanol and 10.0±0.8g/L ABE from a control. The fermentation of the liquid released in the pretreatment at 15% w/w resulted in a butanol production of 6.6±0.8g/L with a total ABE of 8.6±1.3g/L after overliming.
