RESUMO

The Virtual Brain (TVB) is now available as open-source services on the cloud research platform EBRAINS (ebrains.eu). It offers software for constructing, simulating and analysing brain network models including the TVB simulator; magnetic resonance imaging (MRI) processing pipelines to extract structural and functional brain networks; combined simulation of large-scale brain networks with small-scale spiking networks; automatic conversion of user-specified model equations into fast simulation code; simulation-ready brain models of patients and healthy volunteers; Bayesian parameter optimization in epilepsy patient models; data and software for mouse brain simulation; and extensive educational material. TVB cloud services facilitate reproducible online collaboration and discovery of data assets, models, and software embedded in scalable and secure workflows, a precondition for research on large cohort data sets, better generalizability, and clinical translation.

Working together feels easier with some people than with others. We asked participants to perform a visual search task either alone or with a partner while simultaneously measuring each participant's EEG. Local phase synchronization and inter-brain phase synchronization were generally higher when subjects jointly attended to a visual search task than when they attended to the same task individually. Some participants searched the visual display more efficiently and made faster decisions when working as a team, whereas other dyads did not benefit from working together. These inter-team differences in behavioral performance gain in the visual search task were reliably associated with inter-team differences in local and inter-brain phase synchronization. Our results suggest that phase synchronization constitutes a neural correlate of social facilitation, and may help to explain why some teams perform better than others.

MAIN CONCLUSION: Exogenously applied double-stranded RNA (dsRNA) molecules onto tomato leaves, moved rapidly from local to systemic leaves and were uptaken by agricultural pests namely aphids, whiteflies and mites. Four small interfering RNAs, deriving from the applied dsRNA, were molecularly detected in plants, aphids and mites but not in whiteflies. Double-stranded RNA (dsRNA) acts as the elicitor molecule of the RNA silencing (RNA interference, RNAi), the endogenous and evolutionary conserved surveillance system present in all eukaryotes. DsRNAs and their subsequent degradation products, namely the small interfering RNAs (siRNAs), act in a sequence-specific manner to control gene expression. Exogenous application of dsRNAs onto plants elicits resistance against plant viruses. In the present work, exogenously applied dsRNA molecules, derived from Zucchini yellow mosaic virus (ZYMV) HC-Pro region, onto tomato plants were detected in aphids (Myzus persicae), whiteflies (Trialeurodes vaporariorum) and mites (Tetranychus urticae) that were fed on treated as well as systemic tomato leaves. Furthermore, four siRNAs, deriving from the dsRNA applied, were detected in tomato and the agricultural pests fed on treated tomato plants. More specifically, dsRNA was detected in agricultural pests at 3 and 10 dpt (days post treatment) in dsRNA-treated leaves and at 14 dpt in systemic leaves. In addition, using stem-loop RT-PCR, siRNAs were detected in agricultural pests at 3 and 10 dpt in aphids and mites. Surprisingly, in whiteflies carrying the applied dsRNA, siRNAs were not molecularly detected. Our results showed that, upon exogenous application of dsRNAs molecules, these moved rapidly within tomato and were uptaken by agricultural pests fed on treated tomato. As a result, this non-transgenic method has the potential to control important crop pests via RNA silencing of vital genes of the respective pests.

In this study, the effects of paraffin oil, copper hydroxide, copper oxychloride, wettable sulfur, Beauveria bassiana and deltamethrin, as a toxic reference treatment, on the survival and predation rate of M. pygmaeus were investigated. In each treatment, the prey were classified as slightly, partially or fully consumed. The mortality rate after contact exposure was high (66.6%) when nymphs were treated with copper hydroxide but much lower after residual exposure (6.6%). B. bassiana caused 53.3% and 46.6% mortality via contact and residual exposure, respectively. The total prey consumption was significantly lower in the pyrethroid reference treatment control and B. bassiana treatments. The highest percentage of slightly consumed prey was recorded in the toxic reference and B. bassiana treatments, that of partially consumed prey in the copper hydroxide treatment and, finally, that of fully consumed prey in the paraffin oil treatment. Therefore, assessing the sub-lethal effects by separating the prey killed into slightly, partially and fully consumed is a sensitive approach to detect impacts which otherwise may remain unnoticed. The results provide information for the most appropriate use of M. pygmaeus in IPM programs and introduce more sensitive approaches in the detection of side-effects of pesticides on M. pygmaeus and other hemipteran predators.

Apples depend on insect pollination but intensification of agriculture jeopardizes pollination services in agroecosystems. Concerns about the dependency of crop pollination exclusively on honey bees increase the interest in agricultural practices that safeguard wild pollinators in agroecosystems. The purpose of the study was to assess the potential of floral resource provision in apple orchards to enhance the conservation of hymenopterous pollinating insects and potentially the pollination service to the crop. For this reason, flowering plant mixtures sown in patches inside apple orchards were tested against wild plant patches. Pollinator taxa recorded on the sown and wild plant patches were honey bees, wild bees (Andrena, Anthophora, Eucera, Halictus, Lasioglossum, Megachilidae on both; Systropha only on wild plants; Bombus, Hylaeus, Sphecodes, Nomada, Xylocopa only on sown mixture), syrphids, bee flies. The most abundant pollinator of apple was A. mellifera but wild bees were also recorded (Andrena, Anthophora, Bombus, Xylocopa, Lasioglossum, Megachilidae). The sown mixture attracted a more diverse taxa of pollinators and in greater numbers compared to the weed flora, but it did not have an effect on pollinators visiting apple flowers. Groundcover management with patches of suitable flowering mixtures can enhance pollinator conservation in apple orchards.

Humans interact with each other through actions that are implemented by sensory and motor processes. To investigate the role of interbrain synchronization emerging during interpersonal action coordination, electroencephalography data from 13 pairs of pianists were recorded simultaneously while they performed a duet together. The study aimed to investigate whether interbrain phase couplings can be reduced to similar bottom-up driven processes during synchronous play, or rather represent cognitive top-down control required during periods of higher coordination demands. To induce such periods, one of the musicians acted as a confederate who deliberately desynchronized the play. As intended, on the behavioral level, the perturbation caused a breakdown in the synchronization of the musicians' play and in its stability across trials. On the brain level, interbrain synchrony, as measured by the interbrain phase coherence (IPC), increased in the delta and theta frequency bands during perturbation as compared to non-perturbed trials. Interestingly, this increase in IPC in the delta band was accompanied by the shift of the phase difference angle from in-phase toward anti-phase synchrony. In conclusion, the current study demonstrates that interbrain synchronization is based on the interpersonal temporal alignment of different brain mechanisms and is not simply reducible to similar sensory or motor responses.

Ecological dynamics are strongly influenced by the relationship between prey density and predator feeding behavior-that is, the predatory functional response. A useful understanding of this relationship requires us to distinguish between competing models of the functional response, and to robustly estimate the model parameters. Recent advances in this topic have revealed bias in model comparison, as well as in model parameter estimation in functional response studies, mainly attributed to the quality of data. Here, we propose that an adaptive experimental design framework can mitigate these challenges. We then present the first practical demonstration of the improvements it offers over standard experimental design. Our results reveal that adaptive design can efficiently identify the preferred functional response model among the competing models, and can produce much more precise posterior distributions for the estimated functional response parameters. By increasing the efficiency of experimentation, adaptive experimental design will lead to reduced logistical burden.

Traditional approaches to cognitive modelling generally portray cognitive events in terms of 'discrete' states (point attractor dynamics) rather than in terms of processes, thereby neglecting the time structure of cognition. In contrast, more recent approaches explicitly address this temporal dimension, but typically provide no entry points into cognitive categorization of events and experiences. With the aim to incorporate both these aspects, we propose a framework for functional architectures. Our approach is grounded in the notion that arbitrary complex (human) behaviour is decomposable into functional modes (elementary units), which we conceptualize as low-dimensional dynamical objects (structured flows on manifolds). The ensemble of modes at an agent's disposal constitutes his/her functional repertoire. The modes may be subjected to additional dynamics (termed operational signals), in particular, instantaneous inputs, and a mechanism that sequentially selects a mode so that it temporarily dominates the functional dynamics. The inputs and selection mechanisms act on faster and slower time scales then that inherent to the modes, respectively. The dynamics across the three time scales are coupled via feedback, rendering the entire architecture autonomous. We illustrate the functional architecture in the context of serial behaviour, namely cursive handwriting. Subsequently, we investigate the possibility of recovering the contributions of functional modes and operational signals from the output, which appears to be possible only when examining the output phase flow (i.e., not from trajectories in phase space or time).

The phytophagy of the predator Nesidiocoris tenuis (Hemiptera: Miridae) can trigger defense responses in tomato plants against pests, such as two spotted spider mite Tetranychus urticae (Acari: Tetranychidae) and South American leaf miner Tuta absoluta (Lepidoptera: Gelechiidae). The expression of genes governing Jasmonic Acid (JA) biosynthesis pathway and fluctuations in the levels of underlying metabolites have been rarely studied in mirid-infested plants. In the present study, fifteen 3rd instar nymphs of N.tenuis were caged on each top and lower leaf of tomato plants for 4 d to induce plant defense; after this period the predators were removed. With regard to T. absoluta, oviposition preference; larval period; and pupal weight were significantly reduced in N. tenuis-punctured plants. T. urticae adults exhibited a significantly higher escape tendency and reduced survival on punctured plants. Metabolomics confirmed such observations revealing substantial differences between N. tenuis-punctured and unpunctured (control) plants. Metabolites directly associated with the activation of the JA defense pathway, such as the precursor α-linolenic acid, had increased concentrations. The expression of the defense-related genes PI-II, MYC2, VSP2, and HEL was increased in the top leaves and only VSP2 and MBP2 in the lower leaves; interestingly, in the middle (unpunctured) leaves VSP2, HEL, and MBP2 were also upregulated, indicating systemic signaling. Collectively, phytophagy of N. tenuis caused adverse effects on T. absoluta and T. urticae, whereas the multi-omics approach (phenomics, metabolomics, and genomics) offered valuable insights into the nature of the plant defense responses and provided useful evidence for future applications in integrated pest management, plausibly resulting in the reduction in the required pesticide volumes.

Deep brain stimulation (DBS) has been successfully applied in various neurodegenerative diseases as an effective symptomatic treatment. However, its mechanisms of action within the brain network are still poorly understood. Many virtual DBS models analyze a subnetwork around the basal ganglia and its dynamics as a spiking network with their details validated by experimental data. However, connectomic evidence shows widespread effects of DBS affecting many different cortical and subcortical areas. From a clinical perspective, various effects of DBS besides the motoric impact have been demonstrated. The neuroinformatics platform The Virtual Brain (TVB) offers a modeling framework allowing us to virtually perform stimulation, including DBS, and forecast the outcome from a dynamic systems perspective prior to invasive surgery with DBS lead placement. For an accurate prediction of the effects of DBS, we implement a detailed spiking model of the basal ganglia, which we combine with TVB via our previously developed co-simulation environment. This multiscale co-simulation approach builds on the extensive previous literature of spiking models of the basal ganglia while simultaneously offering a whole-brain perspective on widespread effects of the stimulation going beyond the motor circuit. In the first demonstration of our model, we show that virtual DBS can move the firing rates of a Parkinson's disease patient's thalamus - basal ganglia network towards the healthy regime while, at the same time, altering the activity in distributed cortical regions with a pronounced effect in frontal regions. Thus, we provide proof of concept for virtual DBS in a co-simulation environment with TVB. The developed modeling approach has the potential to optimize DBS lead placement and configuration and forecast the success of DBS treatment for individual patients.

Origanum majorana is a medicinal and aromatic plant that belongs to the Lamiaceae family. It is cultivated in several parts of the world and, due to its splendid aroma and taste, is widely used for culinary purposes and in perfumes. The essential oil of the plant, to which is attributed its aroma, contains many secondary metabolites with valuable biological activity. One of them is the pesticide activity, which has attracted much interest. Given the necessity of replacing synthetic pesticides, essential oils are studied in an attempt to find naturally derived products. Thus, the aim of this review paper is to discuss the chemical profile of O. majorana essential oil and to present data regarding its insecticidal, repellent and fumigant activity. Data were collected from 1992 to 2022. Databases, including PubMed, Google Scholar, ScienceDirect and Scopus, were used for the research, and keywords, including O. majorana, sweet marjoram, essential oil, volatiles, pesticide, insecticide and repellent activity, were used. The results of this review paper indicate that O. majorana essential oil can be an alternative agent to manage pests. However, still, much research should be conducted to evaluate its toxicity against beneficial insects and to ensure its safety for human health.

In the past few decades, behavioral and cognitive science have demonstrated that many human behaviors can be captured by low-dimensional observations and models, even though the neuromuscular systems possess orders of magnitude more potential degrees of freedom than are found in a specific behavior. We suggest that this difference, due to a separation in the time scales of the dynamics guiding neural processes and the overall behavioral expression, is a key point in understanding the implementation of cognitive processes in general. In this paper we use Structured Flows on Manifolds (SFM) to understand the organization of behavioral dynamics possessing this property. Next, we discuss how this form of behavioral dynamics can be distributed across a network, such as those recruited in the brain for particular cognitive functions. Finally, we provide an example of an SFM style functional architecture of handwriting, motivated by studies in human movement sciences, that demonstrates hierarchical sequencing of behavioral processes.

Neurophysiological evidence shows that interpersonal action coordination is accompanied by interbrain synchronization (IBS). However, the functional significance of this association remains unclear. Using two experimental designs, we explored whether IBS is amenable to neurofeedback (NFB). Feedback was provided either as two balls approaching each other (so-called ball design), or as two pendula, each reflecting the oscillatory activity of one of the two participants (so-called pendulum design). The NFB was provided at delta (i.e., 2.5 Hz) and theta (i.e., 5 Hz) electroencephalography frequencies, and manipulated by enhanced and inverse feedback. We showed that the participants were able to increase IBS by using NFB, especially when it was fed back at the theta frequency. Apart from intra- and interbrain coupling, other oscillatory activities (e.g., power spectral density, peak amplitude, and peak frequency) also changed during the task compared with the rest. Moreover, all the measures showed specific correlations with the subjective postsurvey item scores, reflecting subjective feeling and appraisal. We conclude that the use of IBS for NFB might help in specifying the contribution of IBS to interpersonal action coordination and in providing important information about the neural mechanisms of social interaction and the causal dimension of IBS.

RNAi-mediated insect pest management has recently shown promising results against the most serious pest of tomato, the tomato leafminer, Tuta absoluta. This study aimed to investigate whether dsRNA (dsTa-αCOP) designed to target the T. absoluta-αCOP gene could cause adverse effects to its biocontrol agent, the mirid predator, Nesidiocoris tenuis. Oral exposure of N. tenuis to dsRNA (dsNt-αCOP) designed to target N. tenuis-αCOP resulted in a 61%, 67% and 55% reduction in its transcript level in comparison to the sucrose, dsGFP and dsTa-αCOP treatments, respectively. In addition, significantly higher mortality of 57% was recorded in dsNt-αCOP-treated N. tenuis when compared to the sucrose (7%), dsGFP (10%) and dsTa-αCOP (10%) treatments. Moreover, the predation rate of ~33-39 Ephestia kuehniella eggs per N. tenuis adult dramatically reduced to almost half in the surviving dsNt-αCOP-treated N. tenuis. This worst-case exposure scenario confirmed for the first time that the RNAi machinery is functional in this species and that the risk of exposure through the oral route is possible. In contrast, dsTa-αCOP did not cause any sub-lethal effects to N. tenuis upon oral exposure. Oral exposure of T. absoluta to dsTa-αCOP resulted in 50% mortality. In the context of a biosafety risk assessment of RNAi-mediated insect management, investigating the effects on non-target organisms is essential in order to include this method as part of an integrated pest management strategy. Based on our laboratory assays, RNAi-mediated control is compatible with the biological control of T. absoluta by its natural enemy N. tenuis, adding the RNAi approach in the armoire of integrated pest management of T. absoluta.

Interference competition occurs when access to an available resource is negatively affected by interactions with other individuals, where mutual interference involves individuals of the same species. The interactive phenomena among individuals may be size-dependent, since body size is a major factor that may alter prey consumption rates and ultimately the dynamics and structure of food webs.A study was initiated in order to evaluate the effect of mutual interference in the prey-specific attack rates and handling times of same size class predators, incorporating variation in consumer size. For this purpose, laboratory functional response experiments were conducted using same age predators, that is, newly hatched (first instar) or mature (fifth instar) nymphs of the polyphagous mirid predator Macrolophus pygmaeus preying on Ephestia kuehniella (Lepidoptera: Pyralidae) eggs.The experiments involved four predator density treatments, that is, one, two, three, or four predators of same age, that is, either first- or fifth-instar nymphs, which were exposed to several prey densities. The Crowley-Martin model, which allows for interference competition between foraging predators, was used to fit the data.The results showed that mutual interference between predator's nymphs may occur that affect their foraging efficiency. The values of the attack rate coefficient were dependent on the predator density and for the first-instar nymphs were significantly lower at the highest predator density than the lower predator densities, whereas for the fifth-instar nymphs in all density treatments were significantly lower to that of the individual foragers' ones.These results indicate that mutual interference is more intense for larger predators and is more obvious at low prey densities where the competition level is higher. The wider use of predator-dependent functional response models will help toward a mechanistic understanding of intraspecific interactions and its consequences on the stability and structure of food webs.

Ochratoxin A (OTA) is the most toxic member of ochratoxins, a group of toxic secondary metabolites produced by fungi. The most relevant species involved in OTA production in grapes is Aspergillus carbonarius. Berry infection by A. carbonarius is enhanced by damage to the skin caused by abiotic and biotic factors. Insect pests play a major role in European vineyards, and Lepidopteran species such as the European grapevine moth Lobesia botrana are undoubtedly crucial. New scenarios are also emerging due to the introduction and spread of allochthonous pests as well as climate change. Such pests may be involved in the dissemination of OTA producing fungi even if confirmation is still lacking and further studies are needed. An OTA predicting model is available, but it should be integrated with models aimed at forecasting L. botrana phenology and demography in order to improve model reliability.

Behavioral and physiological evidence suggests that developmental changes lead to enhanced cortical differentiation and integration through maturation and learning, and that senescent changes during aging result in dedifferentiation and reduced cortical specialization of neural cell assemblies. We used electroencephalographic (EEG) recordings to evaluate network structure and network topology dynamics during rest with eyes closed and open, and during auditory oddball task across the lifespan. For this evaluation, we constructed a hyper-frequency network (HFN) based on within- and cross-frequency coupling (WFC and CFC, respectively) at 10 oscillation frequencies ranging between 2 and 20 Hz. We found that WFC increased monotonously across the lifespan, whereas CFC showed a U-shaped relationship. These changes in WFC and CFC strengths coevolve with changes in network structure and network topology dynamics, namely the magnitude of graph-theoretical topology measures increased linearly with age (except for characteristic path length, which is going shorter), while their standard deviation showed an inverse U-shaped relationship with a peak in young adults. Temporal as well as structural or nodal similarity of network topology (with some exceptions) seems to coincide with variability changes, i.e., stronger variability is related to higher similarity between consecutive time windows or nodes. Furthermore, network complexity measures showed different lifespan-related patterns, which depended on the balance of WFC and CFC strengths. Both variability and complexity of HFNs were strongly related to the perceptual speed scores. Finally, investigation of the modular organization of the networks revealed higher number of modules and stronger similarity of community structures across time in young adults as compared with children and older adults. We conclude that network variability and complexity measures reflect temporal and structural topology changes in the functional organization and reorganization of neuronal cell assemblies across the lifespan.

A green approach for the encapsulation of Mentha pulegium essential oil in commercial baker's yeast and its evaluation as a pesticide against the insect pest Myzus persicae are presented. Upon treating aqueous yeast cell dispersion with the essential oil, the formation of essential-oil-loaded microparticles of about 9 µm is observed, with a loading capacity ranging from 29 to 36%, depending upon the encapsulation conditions. The thermal properties of the microparticles were characterized using differential scanning calorimetry and thermogravimetric analysis, confirming the protection of the essential oil from the cells. Encapsulation prolonged the insecticidal activity of the essential oil by 3 days.

Research on the perception of facial emotional expressions (FEEs) often uses static images that do not capture the dynamic character of social coordination in natural settings. Recent behavioral and neuroimaging studies suggest that dynamic FEEs (videos or morphs) enhance emotion perception. To identify mechanisms associated with the perception of FEEs with natural dynamics, the present EEG (Electroencephalography)study compared (i) ecologically valid stimuli of angry and happy FEEs with natural dynamics to (ii) FEEs with unnatural dynamics, and to (iii) static FEEs. FEEs with unnatural dynamics showed faces moving in a biologically possible but unpredictable and atypical manner, generally resulting in ambivalent emotional content. Participants were asked to explicitly recognize FEEs. Using whole power (WP) and phase synchrony (Phase Locking Index, PLI), we found that brain responses discriminated between natural and unnatural FEEs (both static and dynamic). Differences were primarily observed in the timing and brain topographies of delta and theta PLI and WP, and in alpha and beta WP. Our results support the view that biologically plausible, albeit atypical, FEEs are processed by the brain by different mechanisms than natural FEEs. We conclude that natural movement dynamics are essential for the perception of FEEs and the associated brain processes.

The dynamical systems approach addresses Bernstein's degrees of freedom problem by assuming that the neuro-musculo-skeletal system transiently assembles and dismantles its components into functional units (or synergies) to meet task demands. Strikingly, little is known from a dynamical point of view about the functioning of the muscular sub-system in this process. To investigate the interaction between the dynamical organisation at muscular and behavioural levels, we searched for specific signatures of a phase transition in muscular coordination when a transition is displayed at the behavioural level. Our results provide evidence that, during Fitts' task when behaviour switches to a different dynamical regime, muscular activation displays typical signatures of a phase transition; a reorganisation in muscular coordination patterns accompanied by a peak in the variability of muscle activation. This suggests that consistent changes occur in coordination processes across the different levels of description (i.e., behaviour and muscles). Specifically, in Fitts' task, target size acts as a control parameter that induces a destabilisation and a reorganisation of coordination patterns at different levels of the neuro-musculo-skeletal system.
