More than a quarter of the world's tropical forests are exploited for timber1. Logging impacts biodiversity in these ecosystems, primarily through the creation of forest roads that facilitate hunting for wildlife over extensive areas. Forest management certification schemes such as the Forest Stewardship Council (FSC) are expected to mitigate impacts on biodiversity, but so far very little is known about the effectiveness of FSC certification because of research design challenges, predominantly limited sample sizes2,3. Here we provide this evidence by using 1.3 million camera-trap photos of 55 mammal species in 14 logging concessions in western equatorial Africa. We observed higher mammal encounter rates in FSC-certified than in non-FSC logging concessions. The effect was most pronounced for species weighing more than 10 kg and for species of high conservation priority such as the critically endangered forest elephant and western lowland gorilla. Across the whole mammal community, non-FSC concessions contained proportionally more rodents and other small species than did FSC-certified concessions. The first priority for species protection should be to maintain unlogged forests with effective law enforcement, but for logged forests our findings provide convincing data that FSC-certified forest management is less damaging to the mammal community than is non-FSC forest management. This study provides strong evidence that FSC-certified forest management or equivalently stringent requirements and controlling mechanisms should become the norm for timber extraction to avoid half-empty forests dominated by rodents and other small species.

CASE HISTORY: Two clusters of mortality among endangered tuturuatu/tchuriwat'/shore plover (Thinornis novaeseelandiae) have occurred at captive breeding facilities around New Zealand in recent years. In the first, four chicks died at Pukaha National Wildlife Centre (Mount Bruce, NZ) in February 2016, and in the second five adult birds at the Cape Sanctuary (Cape Kidnappers, NZ) died in 2022. CLINICAL FINDINGS: In 2016, four chicks were noted to become weak, have increased vocalisations and closed eyes prior to death. The remaining chicks were treated for 5 days with amoxycillin/clavulanate orally twice daily. Water containers and brooders were cleaned and disinfected with chlorhexidine. No further mortality was seen.In the 2022 cluster, three adult breeding birds died acutely and five others showed inappetence, weight loss and diarrhoea approximately 10 days after heavy rains flooded the local river. The five birds were treated with amoxycillin/clavulanate orally twice daily and oral fluids for 5 days. Two birds died and three survived. No breeding occurred in the aviaries in the following season. PATHOLOGICAL FINDINGS: In 2016, the chicks showed pulmonary changes ranging from congestion and oedema to heterophilic inflammation consistent with septicaemia.In 2022, the adult birds showed proliferation of bacteria in the distal small intestine associated with mucosal ulceration and heterophilic infiltration. Acid-fast staining of the caecal contents in one bird showed organisms consistent with Cryptosporidium spp. LABORATORY FINDINGS: Aerobic bacterial cultures of the lung and liver of two affected chicks carried out in 2016 showed heavy growth of Plesiomonas shigelloides. The same organism was cultured from water trays and holding tanks containing water boatmen (Sigara arguta) on which the chicks were fed.In 2022, cultures from the livers of three dead birds each showed a mixed bacterial growth with differing dominant organisms (Aeromonas sobria, Hafnia alvei, Citrobacter freundii and an Enterococcus sp.). PCR and sequencing confirmed Cryptosporidium parvum in the caecum of one bird. Fresh faeces from 24 breeding birds from the captive breeding facilities were negative by PCR for Cryptosporidium spp.The captive breeding facilities obtain water for the aviaries and aquatic invertebrates to feed to the chicks from local freshwater sources. Water quality testing at the Cape Sanctuary revealed concentrations of faecal indicator bacteria in excess of safe drinking water guidelines, with peaks following heavy rainfall. CLINICAL RELEVANCE: Fluctuations in water quality associated with mammalian faecal bacteria can adversely affect bird health and impact on captive rearing of endangered wildlife.

Coronavirus disease-2019 (Covid-19) nonpharmaceutical interventions have proven effective control measures for a range of respiratory illnesses throughout the world. These measures, which include isolation, stringent border controls, physical distancing and improved hygiene also have effects on other human pathogens, including parasitic enteric diseases such as cryptosporidiosis. Cryptosporidium infections in humans are almost entirely caused by two species: C. hominis, which is primarily transmitted from human to human, and Cryptosporidium parvum, which is mainly zoonotic. By monitoring Cryptosporidium species and subtype families in human cases of cryptosporidiosis before and after the introduction of Covid-19 control measures in New Zealand, we found C. hominis was completely absent after the first months of 2020 and has remained so until the beginning of 2021. Nevertheless, C. parvum has followed its typical transmission pattern and continues to be widely reported. We conclude that ~7 weeks of isolation during level 3 and 4 lockdown period interrupted the human to human transmission of C. hominis leaving only the primarily zoonotic transmission pathway used by C. parvum. Secondary anthroponotic transmission of C. parvum remains possible among close contacts of zoonotic cases. Ongoing 14-day quarantine measures for new arrivals to New Zealand have likely suppressed new incursions of C. hominis from overseas. Our findings suggest that C. hominis may be controlled or even eradicated through nonpharmaceutical interventions.

Smallholder farmers might adopt different farming practices to cope with multiple stressors depending on their livelihood assets, and with varying environmental and economic outcomes. Ongoing global change is triggering stronger and different stressors that threaten conventional farming practices; however, this could be resolved if livelihood assets that drive decision making are actionable and thus can be modified. This study assessed the influence of farmers' livelihood assets, risk perception, and shocks on the choice of non-conventional farming practices for smallholder coffee farmers in San Martín, Peru. Using household survey data, we collected data on 162 coffee plantations along an elevation gradient. We operationalized the sustainable livelihoods framework for the adoption of shade and input coffee farming strategies and explored farmers' motives to change them. Despite associated high risks with pest and disease pressure, coffee price volatility and climate change, these risks did not explain the current shade and input farming strategies. While in the past five years, farmers adapted shade and input management in response to pest and disease and climate change pressures, these occurred in diverging directions: we found higher human and social assets associated with higher shade levels, and a trend for higher physical and financial assets associated with higher input use. These findings illustrate that two main factors affect decisions on farming practices related to shade and input management and they relate to different livelihood capitals. This suggests a potential for conflicting decision-making, push-and-pulling decisions in different directions. Further the disconnect between livelihood assets and perceptions suggests that perception of risk and shocks might not be sufficient to motivate decision making under changing conditions. Such insights in decision-making typologies and drivers can inform the development of farming practices that enhance resilience and sustainability of smallholder coffee production in Peru and elsewhere in the tropics.

BACKGROUND: Lurbinectedin (PM01183) has synergistic antitumor activity when combined with doxorubicin in mice with xenografted tumors. This phase I trial determined the recommended dose (RD) of doxorubicin (bolus) and PM01183 (1-h intravenous infusion) on day 1 every 3 weeks (q3wk), and obtained preliminary evidence of antitumor activity for this combination in small-cell lung cancer (SCLC). PATIENTS AND METHODS: Patients with advanced solid tumors received doxorubicin and PM01183 following a standard dose escalation design and expansion at the RD. Twenty-seven patients had relapsed SCLC: 12 with sensitive disease (platinum-free interval ≥90 days) and 15 with resistant disease (platinum-free interval <90 days). RESULTS: Doxorubicin 50 mg/m2 and PM01183 4.0 mg flat dose was the RD. In relapsed SCLC, treatment tolerance at the RD was manageable. Transient and reversible myelosuppression (including neutropenia, thrombocytopenia, and febrile neutropenia) was the main toxicity, managed with dose adjustment and colony-stimulating factors. Fatigue (79%), nausea/vomiting (58%), decreased appetite (53%), mucositis (53%), alopecia (42%), diarrhea/constipation (42%), and asymptomatic creatinine (68%) and transaminase increases (alanine aminotransferase 42%; aspartate aminotransferase 32%) were common, and mostly mild or moderate. Complete (n = 2, 8%) and partial response (n = 13, 50%) occurred in relapsed SCLC, mostly at the RD. Response rates at second line were 91.7% in sensitive disease [median progression-free survival (PFS)=5.8 months] and 33.3% in resistant disease (median PFS = 3.5 months). At third line, response rate was 20.0% (median PFS = 1.2 months), all in resistant disease. CONCLUSION: Doxorubicin 50 mg/m2 and PM01183 4.0 mg flat dose on day 1 q3wk has shown remarkable activity, mainly in second line, with manageable tolerance in relapsed SCLC, leading to further evaluation of this combination within an ongoing phase III trial.

A series of laboratory experiments in a thermally driven, rotating fluid annulus are presented that investigate the onset and characteristics of phase synchronization and frequency entrainment between the intrinsic, chaotic, oscillatory amplitude modulation of travelling baroclinic waves and a periodic modulation of the (axisymmetric) thermal boundary conditions, subject to time-dependent coupling. The time-dependence is in the form of a prescribed duty cycle in which the periodic forcing of the boundary conditions is applied for only a fraction δ of each oscillation. For the rest of the oscillation, the boundary conditions are held fixed. Two profiles of forcing were investigated that capture different parts of the sinusoidal variation and δ was varied over the range 0.1≤δ≤1. Reducing δ was found to act in a similar way to a reduction in a constant coupling coefficient in reducing the width of the interval in forcing frequency or period over which complete synchronization was observed (the "Arnol'd tongue") with respect to the detuning, although for the strongest pulse-like forcing profile some degree of synchronization was discernible even at δ=0.1. Complete phase synchronization was obtained within the Arnol'd tongue itself, although the strength of the amplitude modulation of the baroclinic wave was not significantly affected. These experiments demonstrate a possible mechanism for intraseasonal and/or interannual "teleconnections" within the climate system of the Earth and other planets that does not rely on Rossby wave propagation across the planet along great circles.

BACKGROUND: We investigated the association of BRCA1 and XPG mutations with response rate (RR), progression-free survival (PFS) and overall survival (OS) in a subset of patients from a phase 3 clinical trial comparing the efficacy and safety of trabectedin + pegylated liposomal doxorubicin (PLD) versus PLD alone in patients with recurrent ovarian cancer. PATIENTS AND METHODS: A candidate array was designed based on the Breast Cancer Information Core database for BRCA mutation analyses. An exploratory analysis of BRCA1/XPG mutation status was conducted using a two-sided log-rank test and 0.05 significance in germline DNA samples from 264 women with failed first-line platinum-based chemotherapy, randomized (1 : 1) to trabectedin + PLD or PLD alone. RESULTS: Overall, 41 (16%) of the 264 women had BRCA1(mut) (trabectedin + PLD: n = 24/135, 18%; PLD: n = 17/129; 13%) and 17 (6%) had XPG(mut) (trabectedin + PLD: n = 8/135, 6%; PLD: n = 9/129, 7%). A higher RR was observed in BRCA1(mut) patients (20/41; 49%) versus BRCA1(wt) patients (62/223; 28%). Within the BRCA1(mut) group, trabectedin + PLD-treated patients had longer PFS and longer OS than PLD-treated patients (median PFS 13.5 versus 5.5 months, P = 0.0002; median OS 23.8 versus 12.5 months, P = 0.0086), whereas in BRCA1(wt) patients, OS was not significantly different (median OS: 19.1 versus 19.3 months; P = 0.9377). There were no differences in OS or PFS of patients with XPG(mut) between the two treatment arms. However, trabectedin + PLD-treated patients with XPG(mut) had a trend toward shorter PFS (median PFS: 1.9 versus 7.5 months; P = 0.1666) and OS (median OS: 14.5 versus 20.7 months; P = 0.1774) than those with XPG(wt). CONCLUSIONS: In this exploratory analysis, patients with recurrent ovarian cancer carrying the BRCA1(mut) had improved outcomes with trabectedin + PLD treatment compared with PLD alone. Prospective evaluation of BRCA status is likely an important evaluation for DNA-damaging agents and may significantly impact interpretation of clinical studies. XPG may be a biomarker of poor outcome in these patients.

Mountainous grasslands in South America, characterized by their high diversity, provide a wide range of contributions to people, including water regulation, soil erosion prevention, livestock feed provision, and preservation of cultural heritage. Prior research has highlighted the significant role of grazing in shaping the diversity and productivity of grassland ecosystems, especially in highly productive, eutrophic systems. In such environments, grazing has been demonstrated to restore grassland plant diversity by reducing primary productivity. However, it remains unclear whether these findings are applicable to South American mountainous grasslands, where plants are adapted to different environmental conditions. To address this uncertainty, we conducted a meta-analysis of experiments excluding livestock grazing to assess its impact on plant diversity and productivity across mountainous grasslands in South America. In alignment with studies in temperate grasslands, our findings indicated that herbivore exclusion resulted in increased aboveground biomass but reduced species richness and Shannon diversity. The effects of grazing exclusion became more pronounced with longer durations of exclusion; nevertheless, they remained resilient to various climatic conditions, including mean annual precipitation and mean annual temperature, as well as the evolutionary history of grazing. In contrast to results observed in temperate grasslands, the reduction in species richness due to herbivore exclusion was not associated with increased aboveground biomass. This suggests that the processes governing (sub)tropical grassland plant diversity may differ from those in temperate grasslands. Consequently, further research is necessary to better understand the specific factors influencing plant diversity and productivity in South American montane grasslands and to elucidate the ecological implications of herbivore exclusion in these unique ecosystems.

INTRODUCTION: Lateral thoracotomy is replacing traditional median sternotomy for atrial septal defect (ASD) closure in children in order to improve cosmetic outcomes. Continuous paravertebral block has been described as an effective and safe analgesic technique in children. The aim of this study is to assess pain management by continuous perfusion of local anesthetic through a thoracic paravertebral catheter (PVC) in a pediatric population after thoracotomy closure of ASD, and its effectiveness in a fast-track program. METHODS: Descriptive cross-sectional study. Analgesic effectiveness, perioperative and safety-related data were analyzed in 21 patients who underwent thoracotomy closure of ASD with PVC. In the postoperative period, patients received continuous perfusion of bupivacaine 0.125% and fentanyl (1 mcg.ml-1) at 0.2 ml.kg-1.h-1 through the PVC. RESULTS: The median of mean pain scale score for each patient was 1.5. All patients were extubated in the operating theatre. No patient with PVC required opioid rescue. The median length of stay in the Pediatric Intensive Care Unit was 48 hours. There were 3 adverse events related to PVC: 1 due to malposition and 2 due to accidental removal. No other complications or cases of local anesthetic toxicity were recorded. CONCLUSIONS: PVC provides effective, safe, opioid-saving analgesia in the postoperative period of ASD closure by thoracotomy in the context of a fast-track protocol.

Despite increasing attention for relationships between species richness and ecosystem services, for tropical forests such relationships are still under discussion. Contradicting relationships have been reported concerning carbon stock, while little is known about relationships concerning timber stock and the abundance of non-timber forest product producing plant species (NTFP abundance). Using 151 1-ha plots, we related tree and arborescent palm species richness to carbon stock, timber stock and NTFP abundance across the Guiana Shield, and using 283 1-ha plots, to carbon stock across all of Amazonia. We analysed how environmental heterogeneity influenced these relationships, assessing differences across and within multiple forest types, biogeographic regions and subregions. Species richness showed significant relationships with all three ecosystem services, but relationships differed between forest types and among biogeographical strata. We found that species richness was positively associated to carbon stock in all biogeographical strata. This association became obscured by variation across biogeographical regions at the scale of Amazonia, resembling a Simpson's paradox. By contrast, species richness was weakly or not significantly related to timber stock and NTFP abundance, suggesting that species richness is not a good predictor for these ecosystem services. Our findings illustrate the importance of environmental stratification in analysing biodiversity-ecosystem services relationships.

The occurrence of a gastrointestinal illness among a class of 96 undergraduate veterinary students in New Zealand prompted laboratory and questionnaire-based investigations. Cryptosporidium parvum was the only enteropathogen identified in 4/7 faecal specimens analysed. The C. parvum isolates carried a rare IIa GP60 allele, indicating a point-source outbreak. The infection source could not be microbiologically traced, but the investigation suggested contact with calves during a practical class as the most likely exposure. A total of 25/80 respondents to a questionnaire were defined as cases using a clinical case definition (31% attack rate). The inferred median incubation period was 5 days (range 0-11 days), and the median illness duration was 5-6 days (range 2-23 days), corroborating previous observations in experimental cryptosporidiosis. Disease was self-limiting, characterized by abdominal discomfort, diarrhoea, and in some cases, vomiting. Originating from a rural area and having had previously handled ruminants were associated with a significant risk reduction in males. All the three students who reported chronic use of steroid inhalers for treatment of asthma were cases. This case highlighted, once again, the potential hazard for explosive outbreaks of cryptosporidiosis.

Humid tropical forests are increasingly exposed to devastating wildfires. Major efforts are needed to prevent fire-related tipping points and to enable the effective recovery of fire-affected areas. Here, we provide a synthesis of the most common forest restoration strategies, thereby focusing on post-fire forest dynamics in the humid tropics. A variety of restoration strategies can be adopted in restoring humid tropical forests, including natural regeneration, assisted natural regeneration (i.e. fire breaks, weed control, erosion control, topsoil replacement, peatland rewetting), enrichment planting (i.e. planting nursery-raised seedlings, direct seeding) and commercial restoration (i.e. plantation forests, agroforestry). Our analysis shows that while natural regeneration can be effective under favourable ecological conditions, humid tropical forests are often ill-adapted to fire, and therefore less likely to recover unassisted after a wildfire event. Active restoration practices may be more effective, but can be costly and challenging to implement. We also identify gaps in knowledge needed for effective restoration of humid tropical forests after fire, hereby taking into account the ecosystems and socio-economic conditions in which these fires occur. We suggest to incorporate fire severity in future studies, to better understand and predict post-fire ecosystem responses. In addition, as fire poses a recurring and intensifying threat throughout the recovery process, more emphasis should be placed on post-restoration management and the prevention of fire throughout the different phases of the restoration process. Furthermore, as tropical wildfires are increasing in scale, establishing collaborative capacity and setting priorities for efficient resource allocation should become a major priority for restoration practitioners in the humid tropics. Finally, as global fire regimes are changing and expected to intensify in the context of climate change, land use and land cover change, we suggest to put continuous effort into fire monitoring and modelling to inform the development of effective restoration strategies in the long-run.

RESUMO

Stopping droplets from bouncing or splashing after impacting a surface is fundamental in preventing cross-contamination, and the spreading of germs and harmful substances. Here we demonstrate that dielectrowetting can be applied to actively control the dynamics of droplet impact. Moreover, we demonstrate that dielectrowetting can be used to prevent droplet bouncing and suppress splashing. In our experiments, the dielectrowetting effect is produced on a flat substrate by two thin interdigitated electrodes connected to an alternating current potential. Our findings show that the strength of the electric potential can affect the dynamic contact angle and regulate the spreading, splashing and receding dynamics at the right time-scales.

RESUMO

Synchronization phenomena in a fluid dynamical analogue of atmospheric circulation is studied experimentally by investigating the dynamics of a pair of thermally coupled, rotating baroclinic annulus systems. The coupling between the systems is in the well-known master-slave configuration in both periodic and chaotic regimes. Synchronization tools such as phase dynamics analysis are used to study the dynamics of the coupled system and demonstrate phase synchronization and imperfect phase synchronization, depending upon the coupling strength and parameter mismatch.

Frequency entrainment and nonlinear synchronization are commonly observed between simple oscillatory systems, but their occurrence and behavior in continuum fluid systems are much less well understood. Motivated by possible applications to geophysical fluid systems, such as in atmospheric circulation and climate dynamics, we have carried out an experimental study of the interaction of fully developed baroclinic instability in a differentially heated, rotating fluid annulus with an externally imposed periodic modulation of the thermal boundary conditions. In quasiperiodic and chaotic amplitude-modulated traveling wave regimes, the results demonstrate a strong interaction between the natural periodic modulation of the wave amplitude and the externally imposed forcing. This leads to partial or complete phase synchronization. Synchronization effects were observed even with very weak amplitudes of forcing, and were found with both 1:1 and 1:2 frequency ratios between forcing and natural oscillations.

AIM: The aim of the study was to investigate the role of muscarinic acetylcholine receptor (mAChR) activity in the regulation of endothelial (e), neuronal (n) and inducible (i) nitric oxide synthase (NOS) activity and expression in experimentally induced inflammation of rat dental pulp tissue. METHODOLOGY: Inflammation was induced by application of bacterial lipopolysaccharide (LPS) to the pulp. Extirpated pulp-tissue samples were incubated in saline solution until the various experiments were performed. Saline-treated pulp and healthy pulp tissues were used as controls. NOS activity was measured by the production of [U-(14)C]-citrulline from [U-(14)C]-arginine. Nitrite/nitrate assay was evaluated by the conversion of nitrate to nitrite in the presence of nicotinamide adenine dinucleotide phosphate. i-nos, e-nos and n-nos mRNA levels were measured using reverse-transcriptase polymerase chain reaction by co-amplification of target cDNA with a single set of primers. RESULTS: Application of LPS to the pulp increased NOS activity and nitrate production (P < 0.001), generated by iNOS over-activity and expression. Pilocarpine acting on mAChRs triggered a biphasic action on NOS activity and NO accumulation. At low concentrations, pilocarpine induced a negative effect associated with a decrease in i-nos mRNA level, whilst at high concentration, it produced a positive effect associated with increased e-nos and n-nos mRNA levels. In control pulp tissue, only the positive effect of pilocarpine was observed. CONCLUSIONS: Irreversible pulpitis changes mAChR conformation increasing its efficiency of coupling to transducing molecules that in turn induce activate iNOS. The capacity of pilocarpine to prevent NO accumulation and iNOS activity, by acting on mAChR mutation induced by pulpitis, might be useful therapeutically as a local treatment.

A reliable, simple, and affordable liquid tensiometer is presented in this paper. The instrument consists of 72 ultrasonic transmitters in a tractor beam configuration that levitates small liquid samples (droplets) in air. Under operation, the instrument imparts a pressure instability that causes the droplet to vibrate while still levitating. Droplet oscillations are then detected by a photodiode, and the signal is recorded by an oscilloscope. The frequency of these oscillations is obtained and then used to obtain the effective surface tension of the sample. The instrument operates at the millisecond scale time (t < 12.5 ms), with very small liquid volumes (∼0.5 µl), and the sample is recoverable after testing. The instrument has been experimentally validated with acetone, ethanol, Fluorinert FC-40, water, and whole milk.

A temporal analysis of the evolution of Gaussian wave packets in cylindrical capillary jets is presented through both a linear two-mode formulation and a one-dimensional nonlinear numerical scheme. These analyses are normally applicable to arbitrary initial conditions but our study focuses on pure-impulsive ones. Linear and nonlinear findings give consistent results in the stages for which the linear theory is valid. The inverse Fourier transforms representing the formal linear solution for the jet shape is both numerically evaluated and approximated by closed formulas. After a transient, these formulas predict an almost Gaussian-shape deformation with (i) a progressive drift of the carrier wave number to that given by the maximum of the Rayleigh dispersion relation, (ii) a progressive increase of its bell width, and (iii) a quasiexponential growth of its amplitude. These parameters agree with those extracted from the fittings of Gaussian wave packets to the numerical simulations. Experimental results are also reported on near-Gaussian pulses perturbing the exit velocity of a 2-mm diameter water jet. The possibility of controlling the breakup location along the jet and other features, such as pinch-off simultaneity, are demonstrated.

RESUMO

In a paper published in this journal in 2001 by Dong et al. [W. G. Dong, X. Y. Huang, and Q. L. Wo, J. Acoust. Soc. Am. 110, 120-126 (2001)] it was claimed that acoustic chaos was obtained experimentally by the nonlinear interaction of two acoustic waves in a duct. In this comment a simple experimental setup and an analytical model is used to show that the dynamics of such systems corresponds to a quasiperiodic motion, and not to a chaotic one.
