RESUMO

All chronic wounds are colonised by bacteria; for some, colonisation progresses to become infection. Alginate wound dressings are used for highly exuding chronic wounds as they are very absorbent, taking up large quantities of exudate while maintaining a moist wound bed to support healing. Some alginate dressings are doped with antimicrobials, most commonly silver, but evidence regarding the efficacy of these is largely inconclusive. This manuscript describes the development and in vitro assessment of alginate materials doped with chlorhexidine hexametaphosphate (CHX-HMP), a sparingly soluble salt which when exposed to aqueous environments provides sustained release of the common antiseptic chlorhexidine. Comparator materials were a commercial silver alginate dressing material and an alginate doped with chlorhexidine digluconate (CHXdg). CHX-HMP alginates provided a dose-dependent CHX release which was sustained for over 14 days, whereas CHXdg alginates released limited CHX and this ceased within 24 h. CHX-HMP and silver alginates were efficacious against 5 major wound pathogens (MRSA, E. coli, P. aeruginosa, K. pneumoniae, A. baumannii) in a total viable count (TVC) and an agar diffusion zone of inhibition (ZOI) model. At baseline the silver alginate was more effective than the CHX-HMP alginate in the TVC assay but the CHX-HMP alginate was the more effective in the ZOI assay. After 7 days' artificial aging the CHX-HMP alginate was more effective than the silver alginate for four of the five bacteria tested in both assays. These materials may ultimately find application in the development of wound dressings for chronic wounds that provide sustained antimicrobial protection.

Africa is the third-richest continent in the world in terms of bamboo species. Despite these laudable natural resources, most African countries still use asbestos cement board as one of their major building materials. This is chiefly due to the high cost of equipment and technologies associated with non-asbestos-fiber cement board production. The current research seeks to underscore the possibility of utilizing these massive continent resources for non-asbestos-fiber cement board production by employing the existing production process in the asbestos cement industries via an innovatively developed laboratory-simulated Hatschek process. Non-asbestos-fiber cement boards incorporating kraft and bamboo fibers were successfully produced in the laboratory using this innovative method based on Hatschek technology, with natural fibre addition in the range of 2-6 wt.%. Experimental results revealed that the Flexural strength and deflection of the board improved significantly, producing optimum values of 10.41 MPa and 2.0 mm, respectively for composite board reinforced with 10 wt.% and 6 wt.% of kraft pulp and bamboo fibers, respectively. The SEM morphology of the fractured surfaces revealed the mode of composite fracture as well as good interaction at the fiber-matrix interface. Overall, the mechanical properties of the developed composite boards satisfy the minimum requirements of relevant standards based on fiber cement flat sheets and can be employed for internal building applications in low-cost housing estates in developing countries. The outcome of this research indicates that the current industrial production process based on Hatschek technology can be employed for non-asbestos-fiber cement board production using the studied natural fiber.

The consideration of biopolymers with natural products offers promising and effective materials with intrinsic and extrinsic properties that are utilized in several applications. Electrospinning is a method known for its unique and efficient performance in developing polymer-based nanofibers with tunable and diverse properties presented as good surface area, morphology, porosity, and fiber diameters during fabrication. In this work, we have developed an electrospun sodium alginate (SA) incorporated with pulverized Moringa oleifera seed powder (PMO) as a potential natural biosorbent material for water treatment applications. The developed fibers when observed using a scanning electron microscope (SEM), presented pure sodium alginate with smooth fiber (SAF) characteristics of an average diameter of about 515.09 nm (±114.33). Addition of pulverized Moringa oleifera at 0.5%, 2%, 4%, 6%, and 8% (w/w) reduces the fiber diameter to an average of about 240 nm with a few spindle-like pulverized Moringa oleifera particles beads of 300 nm (±77.97) 0.5% particle size and 110 nm (±32.19) with the clear observation of rougher spindle-like pulverized Moringa oleifera particle beads of 680 nm (±131.77) at 8% of alginate/Moringa oleifera fiber (AMF). The results from the rheology presented characteristic shear-thinning or pseudoplastic behaviour with a decline in viscosity, with characteristic behaviour as the shear rate increases, indicative of an ideal polymer solution suitable for the spinning process. Fourier transform infrared spectroscopy (FT-IR) shows the presence of amine and amide functional groups are prevalent on the alginate-impregnated moringa with water stability nanofibers and thermogravimetric analysis (TGA) with change in degradation properties in a clear indication and successful incorporation of the Moringa oleifera in the electrospun fiber. The key findings from this study position nanofibers as sustainable composites fiber for potential applications in water treatment, especifically heavy metal adsorption.

Seaweed-derived alginate shows promise in the textile industry as a sustainable alternative to synthetic and natural materials. However, challenges arise due to its low mechanical strength. We addressed this limitation by sustainably extracting alginates from European brown algae and employing novel manufacturing methods. Using natural cross-linkers, such as chitosan, ferulic acid, and citric acid, we have successfully modulated the mechanical properties of alginate fibers. Mechanical properties of ferulic acid and citric acid-cross-linked alginate solutions were spinnable, producing fibers with a diameter of 73-75 µm. Ferulic acid cross-linked alginate fibers exhibited stiffness, with a tensile strength of 52.97 MPa and a strain percentage of 20.77, mechanical properties comparable to those of wool, polyester, and rayon. In contrast, citric acid-cross-linked fibers showed partial elasticity, with a tensile strength of 14.35 MPa and a strain percentage of 45.53, comparable to those of nylon. This ability to control the mechanical properties of seaweed-derived fibers represents a significant advancement for their application in sustainable textiles and the fashion industry.

Biomaterials that can stimulate stem cell differentiation without growth factor supplementation provide potent and cost-effective scaffolds for regenerative medicine. We hypothesize that a scaffold prepared from cellulose and silk blends can direct stem cell chondrogenic fate. We systematically prepared cellulose blends with silk at different compositions using an environmentally benign processing method based on ionic liquids as a common solvent. We tested the effect of blend compositions on the physical properties of the materials as well as on their ability to support mesenchymal stem cell (MSC) growth and chondrogenic differentiation. The stiffness and tensile strength of cellulose was significantly reduced by blending with silk. The characterized materials were tested using MSCs derived from four different patients. Growing MSCs on a specific blend combination of cellulose and silk in a 75:25 ratio significantly upregulated the chondrogenic marker genes SOX9, aggrecan, and type II collagen in the absence of specific growth factors. This chondrogenic effect was neither found with neat cellulose nor the cellulose/silk 50:50 blend composition. No adipogenic or osteogenic differentiation was detected on the blends, suggesting that the cellulose/silk 75:25 blend induced specific stem cell differentiation into the chondrogenic lineage without addition of the soluble growth factor TGF-ß. The cellulose/silk blend we identified can be used both for in vitro tissue engineering and as an implantable device for stimulating endogenous stem cells to initiate cartilage repair.

In this study, we characterize the shear and extensional rheology of dilute to semidilute solutions of cellulose in the ionic liquid 1-ethyl-3-methylimidazolium acetate (EMIAc). In steady shear flow, the semidilute solutions exhibit shear thinning, and the high-frequency complex modulus measured in small amplitude oscillatory shear flow exhibits the characteristic scaling expected for solutions of semiflexible chains. Flow curves of the steady shear viscosity plotted against shear rate closely follow the frequency dependence of the complex viscosity acquired using oscillatory shear, thus satisfying the empirical Cox-Merz rule. We use capillary thinning rheometry (CaBER) to characterize the relaxation times and apparent extensional viscosities of the semidilute cellulose solutions in a uniaxial extensional flow that mimics the dynamics encountered in the spin-line during fiber spinning processes. The apparent extensional viscosity and characteristic relaxation times of the semidilute cellulose/EMIAc solutions increase dramatically as the solutions enter the entangled concentration regime at which fiber spinning becomes viable.

Alginate is a linear biodegradable polysaccharide polymer, which is bio-renewable and widely used for various biomedical applications. For the next generation of medical textiles, alginate nanofibres are desirable for their use in wound dressings that are biocompatible, sustainable, and abundantly available. This study has developed a unique manufacturing process for producing alginate nanofibres with exceptional antimicrobial properties of oregano essential oil (OEO) as a natural antimicrobial agent. OEO with varying degrees of concentration was incorporated in an aqueous alginate solution. Appropriate materials and electrospinning process parameter selection allowed us to manufacture alginate fibres with a range of diameters between 38 and 105 nm. A unique crosslinking process for alginate nanofibres using extended water soaking was developed. Mechanical characterisation using micro-mechanical testing of nonwoven electrospun alginate/oregano composite nanofibres revealed that it was durable. An extensive antimicrobial study was carried out on alginate/oregano composite nanofibres using a range of Gram-positive (methicillin-resistant Staphylococcus aureus (MRSA) and Listeria monocytogenes) and Gram-negative bacteria (Klebsiella pneumoniae and Salmonella enterica), which are common wound and food pathogens. The results indicated that increasing the concentration of OEO from 2 to 3 wt % showed improved antimicrobial activity against all pathogens, and activity was significantly improved against MRSA compared to a non-alginate-based control disk containing OEO. Therefore, our research suggests that all-natural alginate/oregano nanofibre composite textiles offer a new generation of medical textiles for advanced wound dressing technology as well as for food packaging applications.

We report studies of the orientation state of multiwalled carbon nanotubes (MWNTs) dispersions in steady and transient shear flows. Uncured epoxy was used as a viscous Newtonian suspending medium and samples were prepared from "aligned" MWNTs using methods previously reported [S. S. Rahatekar et al., J. Rheol. 50, 599 (2006)]. Orientation measurements were performed in both the flow-gradient (1-2) and flow-vorticity (1-3) plane of simple shear flow using in situ x-ray scattering techniques. Steady state measurements in the 1-2 plane indicate that the MWNT orientation is shear rate dependent, with the MWNTs orienting closer to the flow direction at higher shear rates. During steady shear, anisotropy was measured to be higher in the 1-2 plane than in the 1-3 plane, demonstrating that the nanotube orientation state is not unaxially symmetric in shear. It is hypothesized that the steady state MWNT orientation is governed primarily by a rate-dependent state of nanotube aggregation/disaggregation, which was separately characterized by optical microscopy of the same samples under shear. High flux synchrotron radiation allowed for time-resolved structural studies in transient flows. A partial relaxation of flow-induced anisotropy was observed following flow cessation, despite the very small rotational diffusivity estimated for these nanotubes. Long transients are observed in step-down experiments, as the orientation state changes in response to the slow tube aggregation process.

The design of biocompatible implants for neuron repair/regeneration ideally requires high cell adhesion as well as good electrical conductivity. Here, we have shown that plasma-treated chitin carbon nanotube composite scaffolds show very good neuron adhesion as well as support of synaptic function of neurons. The addition of carbon nanotubes to a chitin biopolymer improved the electrical conductivity and the assisted oxygen plasma treatment introduced more oxygen species onto the chitin nanotube scaffold surface. Neuron viability experiments showed excellent neuron attachment onto plasma-treated chitin nanotube composite scaffolds. The support of synaptic function was evident on chitin/nanotube composites, as confirmed by PSD-95 staining. The biocompatible and electrically-conducting chitin nanotube composite scaffold prepared in this study can be used for in vitro tissue engineering of neurons and, potentially, as an implantable electrode for stimulation and repair of neurons.

In the present study gentamicin was encapsulated within calcium alginate beads and incorporated into porous chitosan, gelatin, double-hybrid silk fibroin, chitosan/gelatin and double-hybrid silk fibroin/chitosan scaffolds. Physiochemical, morphological and biological properties of fabricated amenable model systems were evaluated, revealing hemocompatible nature of double-hybrid silk fibroin/chitosan and double-hybrid silk fibroin scaffolds of hemolysis %<5 and porosity >85%. Fourier transform infrared results confirmed the blend formation and scanning electron microscope images showed good interconnectivity. Double-hybrid silk fibroin/chitosan-blended scaffold shows higher compressive strength and compressive modulus than other fabricated scaffolds. A comparative drug release profile of fabricated scaffolds revealed that double-hybrid silk fibroin/chitosan scaffold is a pertinent model system because of its prolonged drug release, optimal hemocompatability and high compressive modulus.

The use of single wall carbon nanotubes (SWCNTs) in current and future applications depends on the ability to process SWCNTs in a solvent to yield high-quality dispersions characterized by individual SWCNTs and possessing a minimum of SWCNT bundles. Many approaches for the dispersion of SWCNTs have been reported. However, there is no general assessment which compares the relative quality and dispersion efficiency of the respective methods. Herein we report a quantitative comparison of the relative ability of "wrapping polymers" including oligonucleotides, peptides, lignin, chitosan, and cellulose and surfactants such as cholates, ionic liquids, and organosulfates to disperse SWCNTs in water. Optical absorption and fluorescence spectroscopy provide quantitative characterization (amount of SWCNTs that can be suspended by a given surfactant and its ability to debundle SWCNTs) of these suspensions. Sodium deoxy cholate (SDOCO), oligonucleotides (GT)(15), (GT)(10), (AC)(15), (AC)(10), C(10-30), and carboxymethylcellulose (CBMC-250K) exhibited the highest quality suspensions of the various systems studied in this work. The information presented here provides a good framework for further study of SWCNT purification and applications.

The research described in this paper primarily involves mesoscale simulations: dissipative particle dynamics (DPD) of packed assemblies of oriented fibers suspended in a viscous medium. Computer simulations have been performed in order to explore how the aspect ratio and degree of fiber alignment affect the critical volume fraction (percolation threshold) required to achieve electrical conductivity. The fiber network impedance was assessed using Monte Carlo simulations after establishing the structural arrangement with DPD. The predictions are compared with the predictions of classical percolation theory and found to be in close agreement. The approach is thus validated and can be extended to systems that cannot be tackled analytically; in particular, the work is motivated by long-standing interest in materials which display a complex percolation behavior.
