RESUMO

The previous study has shown that transcriptional factor MEOX1 could promote proliferation and sphere formation ability of non-small cell lung cancer (NSCLC) cells, however, we found that MEOX1 mRNA was lowly expressed in lung cancer tissues compared to that in normal adjacent tissues, and MEOX1 mRNA expression was positively correlated with the survival of lung cancer patients, especially in lung adenocarcinoma patients. Functional experiments using in vitro and in vivo experiments revealed that stable overexpression of MEOX1 significantly suppressed the proliferation ability, promoted cell cycle arrest in G2 phase, and apoptotic ability of NSCLC cells. Additionally, it was identified that MEOX1 and CCNB1 mRNA expression exhibited a negative correlation in different lung cancer tissues. Mechanistically, we indicated that MEOX1 bound to the transcriptional initiation site of CCNB1 and thus suppressed CCNB1 expression. Notably, CCNB1 overexpression rescued the inhibition of MEOX1 overexpression on NSCLC progression. This study deciphers a novel MEOX1/CCNB1 axis suppressing NSCLC progression.

BACKGROUND: The application of multiparametric magnetic resonance imaging (mpMRI) for diagnosis of prostate cancer has been recommended by the European Association of Urology (EAU), National Comprehensive Cancer Network (NCCN), and European Society of Urogenital Radiology (ESUR) guidelines. The purpose of this study is to systematically review the literature on assessing the accuracy of mpMRI in patients with suspicion of prostate cancer. METHOD: We searched Embase, Pubmed and Cochrane online databases from January 12,000 to October 272,018 to extract articles exploring the possibilities that the pre-biopsy mpMRI can enhance the diagnosis accuracy of prostate cancer. The numbers of true- and false-negative results and true- and false-positive ones were extracted to calculate the corresponding sensitivity and specificity of mpMRI. Study quality was assessed using QUADAS-2 tool. Random effects meta-analysis and a hierarchical summary receiver operating characteristic (HSROC) plot were performed for further study. RESULTS: After searching, we acquired 3741 articles for reference, of which 29 studies with 8503 participants were eligible for inclusion. MpMRI maintained impressive diagnostic value, the area under the HSROC curve was 0.87 (95%CI,0.84-0.90). The sensitivity and specificity for mpMRI were 0.87 [95%CI, 0.81-0.91] and 0.68 [95%CI,0.56-0.79] respectively. The positive likelihood ratio was 2.73 [95%CI 1.90-3.90]; negative likelihood ratio was 0.19 [95% CI 0.14,-0.27]. The risk of publication bias was negligible with P = 0.96. CONCLUSION: Results of the meta-analysis suggest that mpMRI is a sensitive tool to diagnose prostate cancer. However, because of the high heterogeneity existing among the included studies, further studies are needed to apply the results of this meta-analysis in clinic.

Two unprecedented examples of cluster organic frameworks (TBA)3Cu[V6O13(L)2]2·4DEF (2) (TBA)Ag[V6O13(L)2] solvent (3) (TBA = tetrabutylammonium, H3L = tris(hydroxymethyl)-4-picoline, DEF = N, N'-diethylformamide) based on Lindqvist-type polyoxometalate (POM) secondary building units (SBUs) have been constructed successfully. Compounds 2 and 3 are the second cases of cluster organic frameworks based on Lindqvist-type POM cluster SBUs. Furthermore, the cluster organic framework of 2 exhibits efficient electrocatalytic activity and strong durability in oxygen reduction reaction.

Autoimmune thyroid disease (AITD) shows the highest incidence among organ-specific autoimmune diseases and is the most common thyroid disease in humans, including Graves' disease (GD) and Hashimoto's thyroiditis (HT). The susceptibility to autoimmune diseases is affected by increased autoantibody levels, susceptibility gene polymorphisms, environmental factors, and psychological factors, but the pathogenesis remains unclear. Various cytokines and related genes encoding them play important roles in the development and progression of AITD. CD152, an expression product of the CTLA-4 gene, downregulates T cell activation. The A/A genotype polymorphism in the CT60 locus may reduce the production of thyroid autoantibodies. The C1858T polymorphism of the PTNP22 gene reduces the expression of its encoded LYP, which increases the risk of GD and HT. GD is an organ-specific autoimmune disease involving increased secretion of thyroid hormone, whereas HT may be associated with the destruction of thyroid gland tissue and hypothyroidism. These two diseases exhibit similar pathogenesis but opposite trends in the clinical manifestations. In this review, we focus on the structure and function of these cytokines and related genes in AITD, as well as the association of polymorphisms with susceptibility to GD and HT, and attempt to describe their differences in pathogenesis and clinical manifestations.

RESUMO

A new approach to prepare heterometallic cluster organic frameworks has been developed. The method was employed to link Anderson-type polyoxometalate (POM) clusters and transition-metal clusters by using a designed rigid tris(alkoxo) ligand containing a pyridyl group to form a three-fold interpenetrated anionic diamondoid structure and a 2D anionic layer, respectively. This technique facilitates the integration of the unique inherent properties of Anderson-type POM clusters and cuprous iodide clusters into one cluster organic framework.

OBJECTIVE: To construct an N-2a cell line stably expressing PcDNA 3.1-platelet derived growth factor-galanin (GAL) and explore the effect of over-expressed GAL on proliferation and apoptosis of N-2a cell in vitro. METHODS: The vector containing the target gene was transfected into N-2a cells by liposome, and cell clones stably over-expressing GAL was obtained via G418 screening. GAL mRNA and protein levels were determined by reverse transcriotion-polymerase chain reaction (RT-PCR) and Western blot. The proliferation of N-2a cells was detected by MTT.The cell cycle and apoptosis were detected by flow cytometry. RESULTS: RT-PCT and Western blot indicated that GAL genes were highly expressed in the transfected N-2a cells (i.e.GAL-N-2a). As shown by MTT, the proliferation of the N-2a cells transfected with PcDNA 3.1-PDGF-GAL was significantly slower than the control group(P<0.05). Compared with the non-transfected cells in the control group, the N-2a cells with endogenously overexpressed GAL were arrested at the G0/G1 phases, and the over-expressed GAL protein significantly induced the N-2a cell apoptosis in a concentration-dependent fashion. CONCLUSION: Eukaryotic expression vector PcDNA 3.1-PDGF-GAL can encode the expression of GAL in N-2a cells. Aslo, it can inhibit cell proliferation and promote the cell apoptosis.

OBJECTIVE: To explore the difference of biological activities between two Chinese Herbs polysaccharides. METHODS: Extracted two kinds of Chinese Herbs Polysaccharides by hot water, and determined the content of Polysaccharides. Determined the antibacterial activity with turbidimetry. Explored antioxidant activity using an in vitro antioxidant analysis, probed into the effects on cell growth with MTT colorimetric analysis. RESULTS: The two polysaccharides both showed activities on the antibacterial, anti-oxidation and promotion of cell growth but different in degree. The Radix Paeoniae Rubra polysaccharide was stronger than Rhizoma Chuanxiong in antioxidant and antimicrobial activities, and weaker in promotion of cell growth activity. CONCLUSION: Activity differences exist between the two polysaccharides and the activities show some complementary effects. The differences provide important value in research on polysaccharide compound.

RESUMO

Quantum dots (QDs) are luminescent nanocrystals with rich surface chemistry and unique optical properties that make them useful as probes or carriers for traceable targeted delivery and therapy applications. QDs can be functionalized to target specific cells or tissues by conjugating them with targeting ligands. Recent advancement in making biocompatible QD formulations has made these nanocrystals suitable for in vivo applications. This review provides an overview of the preparation of QDs and their use as probes or carriers for traceable, targeted therapy of diseases in vitro and in vivo. More specifically, recent advances in the integration of QDs with drug formulations for therapy and their potential toxicity in vitro and in vivo are highlighted. The current findings and challenges for optimizing QD/drug formulations with respect to optimal size and stability, short-term and long-term toxicity, and in vivo applications are described. Lastly, we attempt to predict key trends in QD/drug formulation development over the next few years and highlight areas of therapy where their use may provide breakthrough results in the near future.

RESUMO

In this paper we implement the 7-point checklist, a set of dermoscopic criteria widely used by clinicians for melanoma detection, on smart handheld devices, such as the Apple iPhone and iPad. The application developed is using sophisticated image processing and pattern recognition algorithms, yet it is light enough to run on a handheld device with limited memory and computational speed. When combined with a commercially available handheld dermoscope that provides proper lesion illumination, this application provides a truly self-contained handheld system for melanoma detection. Such a device can be used in a clinical setting for routine skin screening, or as an assistive diagnostic device in underserved areas and in developing countries with limited healthcare infrastructure.

In order to understand growth of plant and its effect on ecological restoration of riparian zone, pilot-scale Scirpus yagara Ohw riparian zone had been studied for one year. Height and density of Scirpus yagara Ohw in different water depth showed that the water depth endurance of Scirpus yagara Ohw was not strong, and it growed and germinated better in shallow water. Contrast between harvest area and without harvest area showed that harvest was good for germination and growth of Scirpus yagara Ohw in next year. Distribution of biomass and N and P fixation between the underground and overground Scirpus yagara Ohw showed that N and P fixation of the underground was 50% and 126% more than that of the overground, respectively. Harvest of the overground Scirpus yagara Ohw can remove 40.5% of the total N and 30.6% of the total P. Dunked in water for 130 days, the overground Scirpus yagara Ohw lose 27.10% of its dry weight, 40.80% of the total N and 76.80% of tha total P. And the released pollutants have no serious cumulation in water.

In the title compound, [Fe(C(5)H(3)N(2)O(4))(2)](n), each Fe atom lies on a centre of symmetry, in an octahedral coordination environment consisting of two chelate rings [Fe-N = 2.154 (3) A and Fe-O = 2.180 (3) A] and two carboxylate O atoms [Fe-O = 2.111 (2) A] from imidazole-4,5-dicarboxylate ligands. Extensive hydrogen-bonding interactions exist between layers constructed of Fe(4) squares, forming tunnels along the a axis with large voids.
