RESUMO

Two in vitro and in vivo experiments were accomplished to investigate the impacts of integrated feed additives (IFA, combination of protected fat, lysine, and methionine plus yucca extract, eucalyptus essential oil, and direct-fed microbial) on gas production (GP), ruminal fermentation and degradability parameters and lamb performance, digestibility, and nitrogen balance. In the in vitro experiment, responses of graded doses (0, 25, 50, 75, 100, and 125 g IFA/kg DM) were evaluated. In the in vivo experiment, 15 Barki male lambs (30.43 ± 0.74 kg BW ± SE) were individually allotted in complete randomized design into three treatments (five animal/treatment) as control (basal diet without additive), low dose (50 g/kg DM) and high dose (100 g/kg DM) for 120 days. In vitro results showed that both doses (100 and 125 g/kg DM) quadratically decreased (p < 0.001) GP and ammonia nitrogen (NH-3N) concentration. In vivo experiment revealed that dry matter intake was increased (p = 0.016) with low dose compared to high dose. Inclusion of high dose decreased blood serum glucose concentrations (p > 0.05) and ruminal protozoal populations (p = 0.094) compared with low dose and control diet. Both doses of IFA increased (p < 0.05) serum urea, creatinine, and triglyceride concentrations, while decreased (p < 0.001) ruminal NH3-N concentrations. These results suggested that, although IFA was effective to modify in vitro ruminal fermentation process and feed degradability, such aspects did not improve in vivo performance of growing lambs.

In vitro and in vivo experiments were carried out to investigate the effects of the supplementation of different levels of humic and fulvic acids alone or their combination (2:1 ratio) on ruminal fermentation constituents, and nutrients digestibility in goats. The treatments in Exp. 1 were the following: (1) basal substrate (50% concentrate: 50% forage) was incubated humic at 0, 2, 4, and 6 g/kg DM; (2) fulvic at 0, 1, 2, and 3 g/kg DM; and (3) a combination of humic and fulvic (in a 2:1 ratio) at 0, 3, 6, and 9 g/kg DM" of treatments. The results of Exp. 1 revealed that methane (CH4) production was linearly decreased (P < 0.001) upon increasing humic doses. Whereas, the combination of fulvic acid with humic acid resulted in a quadratic decrease (P < 0.001) in net CH4 production. Supplementing humic and fulvic acids, either separately or in combination, resulted in reduced (P < 0.05) ammonia nitrogen (NH3-N) and total volatile fatty acid (VFA) concentrations. In Exp. 2 to further examine the findings obtained in Exp. 1, forty Damascus non-lactating goats (2-3 years of age and body weight 29 ± 1.5 kg) were fed the same basal diet as in Exp. 1, plus one of four treatments. Treatments were the following: (1) control (no supplement); (2) basal diet plus 5 g humic alone; (3) basal diet plus 2.5 g fulvic alone, and (4) basal diet plus 7.5 g their combination. Goats fed diets supplemented with humic acid, fulvic acid, either alone or in combination, increased concentrations of butyrate (P = 0.003), total VFA (P < 0.001), and improved (P < 0.001) digestibility of nutrients, but reduced (P < 0.001) ruminal NH3-N concentrations. In conclusion, applying humic and fulvic acids alone or in combination attenuated in vitro CH4 production, while improved intake and diet digestibility without adverse effect on rumen fermentation profiles in Damascus goats.

BACKGROUND: Montmorillonite clay modified by organosulfur surfactants possesses high cation exchange capacity (CEC) and adsorption capacity than their unmodified form (UM), therefore they may elevate the adverse impact of aflatoxin B1 (AFB1) on ruminal fermentation and methanogenesis. Chemical and mechanical modifications were used to innovate the organically modified nano montmorillonite (MNM). The UM was modified using sodium dodecyl sulfate (SDS) and grounded to obtain the nanoscale particle size form. The dose-response effects of the MNM supplementation to a basal diet contaminated or not with AFB1 (20 ppb) were evaluated in vitro using the gas production (GP) system. The following treatments were tested: control (basal diet without supplementations), UM diet [UM supplemented at 5000 mg /kg dry matter (DM)], and MNM diets at low (500 mg/ kg DM) and high doses (1000 mg/ kg DM). RESULTS: Results of the Fourier Transform Infra-Red Spectroscopy analysis showed shifts of bands of the OH-group occurred from lower frequencies to higher frequencies in MNM, also an extra band at the lower frequency range only appeared in MNM compared to UM. Increasing the dose of the MNM resulted in linear and quadratic decreasing effects (P < 0.05) on GP and pH values. Diets supplemented with the low dose of MNM either with or without AFB1 supplementation resulted in lower (P = 0.015) methane (CH4) production, ruminal pH (P = 0.002), and ammonia concentration (P = 0.002) compared to the control with AFB1. Neither the treatments nor the AFB1 addition affected the organic matter or natural detergent fiber degradability. Contamination of AFB1 reduced (P = 0.032) CH4 production, while increased (P < 0.05) the ruminal pH and ammonia concentrations. Quadratic increases (P = 0.012) in total short-chain fatty acids and propionate by MNM supplementations were observed. CONCLUSION: These results highlighted the positive effects of MNM on reducing the adverse effects of AFB1 contaminated diets with a recommended dose of 500 mg/ kg DM under the conditions of this study.

The current study investigated the effect of adding different doses of galangal rhizome powder (Alpinia galanga) to the diets of lactating buffaloes on milk production, composition and in vitro ruminal fermentation kinetics. Twenty buffaloes were divided into four groups: (T1) a control diet comprising of concentrates, corn silage and rice straw at 50: 35: 15 on a DM basis, (T2) a control diet plus 15 g of galangal/animal/day and (T3) a control diet plus 30 g of galangal/animal/day and (T4) a control diet plus 45 g of galangal/animal/day. Results showed that inclusion of galangal in diets decreased (p < 0.05) the concentrations of butyrate, ammonia-nitrogen, CO2, and total protozoa count as compared with T1. Acetate concentration was lower in T4 as compared with other groups (p < 0.05). While propionate concentration was lower (p < 0.05) in T2 as compared with T1. T3 and T4 lowered (p < 0.05) methane, CO2 productions, and protozoa count. T3 increased (p < 0.05) milk yield, energy corrected milk (ECM), yield of total solids, total protein, and solids not fat (SNF) than recorded for T1. While T4 recorded the lowest values (p < 0.05) of milk yield, ECM, total solids, fat, and total protein. The current results support the hypothesis that the inclusion of galangal in lactating buffaloes' diets at 30 g per animal daily improved feed intake and milk yield.

Effects of cinnamon, thyme, and peppermint essential oils blend (EOB) on milk production, serum constituents, and energy balance of early lactating does and performance of their kids were investigated. Twenty-seven late pregnant Damascus goats were grouped into two treatments: EOB (n = 14) received orally 1.5 ml EOB/d and control (n = 13) received orally 1.5 ml water/d. The trail started 15 days before the expected kidding date and lasted for 30 days postpartum while sample collection continued until the eighth week of lactation. Milk yield and composition, maternal metabolic profile, immunoglobulin (IgG), and weights of kids were monitored throughout 8 consecutive weeks postpartum. Energy corrected milk (ECM), milk energy value (MEV), net energy for lactation (NEL), and energy balance were estimated. Does treated with EOB had higher (p < 0.05) milk yield, ECM, and NEL than control. Treated does had higher (p = 0.02) energy balance than control does. The EOB increased (p < 0.05) concentrations of serum albumin, cholesterol, triglycerides, and IgG. Treatment with EOB improved (p < 0.05) weaning weight and total weight gain of single kids. Administration of EOB for 45 days during the transition period could improve milk production, metabolic status and energy balance of goats and performance of kids.

Eighty multiparous lactating Holstein cows (635 ± 33 kg) were used to study the effect of feeding Saccharomyces cerevisiae and/or Aspergillus oryzae on lactational performance for 14 weeks. Cows were assigned in a completely randomized experimental design, with repeated measures into four treatments, and were fed a basal diet of concentrates and forage at a ratio of 592:408, respectively. The treatments were: (1) the basal diet with no additive (Control treatment); (2) the basal diet supplemented with 3.5 g of live S. cerevisiae/cow daily (SC treatment); (3) the basal diet supplemented with 3.5 g A. oryzae fermentation extract/cow daily (AO treatment); and (4) the basal diet supplemented with 3.5 g of live S. cerevisiae + 3.5 g A. oryzae fermentation extract/cow daily (AOSC treatment). The AO and AOSC treatments increased (p < .05) feed intake and daily milk production, with a low milk fat content for the AO treatment. Feeding SC treatment decreased (p = .002) serum glucose concentration, while the AOSC treatment increased serum glutamate-oxaloacetate transaminase concentration. It is concluded that S. cerevisiae supplementation did not enhance milk production; however, A. oryzae fermentation extract improved feed intake and milk production.

The effects of supplementing diet of growing lambs with microbial feed additive mixture (direct-fed microbial; DFM) based on Saccharomyces cerevisiae, lactic acid bacteria and exogenous enzymes on feed intake, nutrient digestibility, nitrogen (N) utilization, ruminal fermentation, blood chemistry and growth performance were studied. The study was a completely randomized design with 12 growing Barki lambs divided into two groups of six lambs per treatment. Lambs were offered a control diet of peanut hay and concentrates (1:1 dry matter (DM) basis) or the control diet supplemented with DFM at 0.5 g/day (Bactozyme treatment) for 16 weeks. There was no effect on feed efficiency but Bactozyme lambs had increased (p = 0.009) feed intake, average daily gain (p = 0.042) and final body weight (p = 0.047). Bactozyme treatment had greater neutral detergent fiber (p = 0.020) and acid detergent fiber (p = 0.034) digestibility compared with the control treatment. Metabolism experiment showed that the feed additive mixture increased (p<0.05) N intake and N retention. Bactozyme treatment had greater blood total protein (p = 0.027), and globulin (p = 0.025) concentrations compared with the control treatment. It is concluded that supplementing growing Barki lambs with DFM at 0.5 g daily enhanced final body weight gain and fiber digestion without affecting feed efficiency.

BACKGROUND: Berseem clover is the main forage crop grown in Mediterranean regions. There are plenty of cultivars that possess variability in their productivity and quality among the different cuts. Therefore, accurate agronomic and qualitative characterization is crucial for selecting the most promising cultivars for breeding and feeding purposes. In the present study, the agronomic characteristics, ruminal degradability and fermentation measures of five cuts of the five most prominent Egyptian beseem clover cultivars (Helaly, Serw, Giza6, Gemmeza1 and Sakha4) were evaluated. RESULTS: The Giza6 cultivar produced a significantly higher fresh yield and high whole plant dry matter content on the 3rd cut. Giza6 was among the superior cultivars in crude protein content and had the highest values of gas production at the 3rd cut, and ruminal degraded acid detergent fiber and propionate concentrations at the 2nd cut, at the same time as presenting the lowest ammonia concentrations at the 3rd and 5th cuts. Overall, the 3rd cut produced significantly higher fresh yield across all cultivars, except for Sakha4. The 3rd cut also produced a higher leaf to stem ratio compared to the 4th and 5th cuts. The 1st three cuts had higher degraded neutral detergent fiber than the 4th and 5th cuts. The 1st cut was characterized by low propionate concentration in the Helaly and Gemmeza1 cultivars compared to the other cuts. CONCLUSION: Expanded production of the high-yielding, high-quality cultivar Giza6 is recommended. When harvesting, three cuts is optimal for berseem clover cultivation for breeding and feeding programs. © 2020 Society of Chemical Industry.

Twenty Santa Inês ewes used to evaluate effects of oral administration of Brazilian red propolis extract on blood metabolites, milk production, and lamb performance were randomly grouped (n = 10 ewes/group) to control without propolis administration and propolis treated (3 g red propolis extract/ewe/day) 21 days before expected lambing date. Blood samples were collected weekly, and daily milk yield was recorded twice weekly until 7 weeks postpartum. Propolis administration increased (P < 0.05) total leukocyte counts, protein, and globulin and glucose concentrations, decreased (P < 0.05) somatic cell counts, and enhanced (P < 0.05) yields of milk, fat, protein, and lactose. Propolis supplementation increased (P < 0.05) average daily gain and milk conversion ratio but had no effect on lamb birth and weaning weights. The prepartum administration of propolis extract supported positively the transition of ewes from pregnancy to lactation with health benefits achieved for both of ewes and lambs performances.

Brazilian red propolis was administered orally to Santa Inês ewes, and evaluation was made of general health and hematological, biochemical, and parasitic responses during and after flushing. Thirty mature, nonlactating, nonpregnant Santa Inês ewes (40 ± 2 kg and 2.5 years old) were grazing tropical pasture (Brachiaria decumbens and Pennisetum purpureum) and, as flushing after synchronization, were supplemented with a concentrate-roughage mixture at a rate of 4 % body weight (BW). Ewes were divided according to BW and fecal egg count (FEC) into two groups (n = 15 each): control and propolis that received propolis ethanolic extract at rates of, respectively, 0 and 3 g/ewe/day. The treatments lasted 21 days until the end of flushing period. BW and body condition score (BCS) were recorded, and blood and fecal samples were taken weekly for 8 weeks. Mean values of BW and BCS were not (P > 0.05) affected by propolis administration. Propolis increased (P < 0.01) total leukocytes (WBC), but no significant differences were observed for other hematological parameters. Propolis increased (P < 0.01) total protein and globulin concentrations and decreased (P < 0.01) triglycerides, glutamate oxaloacetate transaminase, and glutamate pyruvate transaminase. Propolis decreased (P < 0.05) FEC. Propolis administration had good impact on ewe health and may be a promising feed additive during critical periods such as flushing.

A synbiotic composed of alginate nanoencapsulated prebiotic (pomegranate peel phytogenics) and multi-species probiotics (Lactococcus lactis, Lactobacillus plantarum, Lactobacillus paracasei, and Saccharomyces cerevisiae) has been developed as a potential eco-friendly alternative to antibiotics. The physicochemical properties of the encapsulated synbiotic were evaluated, and its gastric and storage tolerance, as well as its antioxidant and antimicrobial activity, were tested and compared to that of the non-encapsulated synbiotic (free synbiotic). The results showed that the prebiotic pomegranate peel ethanolic extract contained seven phenolic compounds, with cinnamic being the most abundant (13.26 µL/mL). Sodium alginate-CaCl2 nanocapsules were effective in encapsulating 84.06 ± 1.5% of the prebiotic's phenolic compounds and 98.85 ± 0.57% of the probiotics. The particle size of the alginate-CaCl2 nanoencapsulated synbiotic was 544.5 nm, and the polydispersity index and zeta potential values were 0.593 and -12.3 mV, respectively. Thermogravimetric analysis showed that the alginate-CaCl2 nanoencapsulated synbiotic had high thermal stability at high temperatures, with only 2.31% of its weight being lost within the temperature range of 70-100 °C. The count of viable probiotics in the nanoencapsulated synbiotic was significantly higher than that in the free synbiotic after exposure to gastric acidity and storage for six months at room temperature. The percent inhibition values of the nanoencapsulated synbiotic and ascorbic acid (as a standard antioxidant) were comparable and significantly greater than those of the free synbiotic. The half-maximal inhibitory concentrations (IC50) of the nanoencapsulated synbiotic and ascorbic acid were significantly lower than those of the free synbiotic (3.96 ± 0.42 µg/mL and 4.08 ± 0.79 µg/mL for nanoencapsulated synbiotic and ascorbic acid, respectively, vs. 65.75 ± 2.14 µg/mL for free synbiotic). The nanoencapsulated synbiotic showed the highest significant antimicrobial activity against Escherichia coli (ATCC 8739). Both the nanoencapsulated and free synbiotics showed antimicrobial activity against Staphylococcus aureus (ATCC 6538), similar to that of gentamicin, although the nanoencapsulated synbiotic showed significantly higher inhibition activity compared to the free synbiotic. The nanoencapsulated synbiotic showed antimicrobial activity comparable to gentamicin against Pseudomonas aeruginosa (ATCC 90274), whereas the free synbiotic showed the least antimicrobial activity (p < 0.05). Both synbiotics showed significantly higher antimicrobial activity against Salmonella typhi (ATCC 6539) than gentamicin. Both synbiotics showed antifungal activity against Aspergillus niger and Aspergillus flavus, with a stronger effect observed for the nanoencapsulated synbiotic. However, the activity of both synbiotics was significantly lower than that of fluconazole (an antifungal drug).

Forty-five male Barki lambs (30.62 ± 4.54 kg BW) were allocated into three treatments designed to evaluate the effect of replacing soybean meal nitrogen with urea supplemented with natural clinoptilolite on performance and carcass characteristics. Lambs were randomly assigned to nine pens blocked according to age and initial BW into three blocks of three pens each assigned to one of three treatments: Control diet: animals received only a basal diet ad libitum, UR diet: urea replaced 68% of SBM nitrogen in the control diet or UR-Z diet: UR diet supplemented with 20-g zeolite for 141 days. Lambs fed on UR-Z diet increased (p < 0.05) total intakes of DM, crude protein digestibility, ADG, FCR, blood glucose and cholesterol concentration. Lambs fed on UR-Z diet decreased (p < 0.05) ruminal NH3 -N concentration compared to the UR diet. Carcasses from lambs fed on UR diet had greater (p < 0.05) ether extract content, fat thickness, meat yellowness and saturation attributes than those fed on UR-Z and control diets. Sheep fed on UR-Z diet made higher economic profit than UR and control diets. Partial replacement of soybean meal with urea supplemented with zeolite seemed to be economically efficient, with comparable performance and carcass attributes in growing lambs diet.

This study was conducted to investigate the effects of 10 weeks supplementation of Quebracho tannins (QT; 0 [control], 100 [QT100] or 200 g/[cow·d] [QT200]) to 30 multiparous postpartum buffalo cows (10 cows per group) on milk yield and composition, blood metabolites and reproductive performance. Supplementation of QT100 had no significant effect on milk yield, whereas QT200 decreased (P < 0.05) this trait. Compared with the control group, both QT levels decreased (P < 0.05) fat-corrected milk (FCM) yield, but no significant effects were found on percentages of milk fat and protein. Contrariwise, yields of milk fat, lactose and milk protein were decreased (P < 0.05) when QT200 was supplemented. The solids nonfat (SNF) percentage and yield were decreased (P < 0.05) with QT100 supplementation. Moreover, QT tended to numerically reduce total number of ovarian follicles, number of small follicles, peripheral progesterone concentration and conception rate. Supplementation of QT200 numerically increased number of large follicles, mean diameter of large follicle, number and diameters of corpora lutea. The inclusion of QT200 shortened days open (DO) and decreased number of services per conception. Contrariwise, QT did not show significant effects on serum total protein, albumin, globulin, glucose, cholesterol and triglycerides concentrations. Supplementation of QT100 caused an increase (P < 0.05) of serum urea compared with that in control and QT200 groups. Generally, QT decreased (P < 0.05) serum creatinine concentration. Therefore, the supplementation of a commercial QT to early lactating Egyptian buffalo cows displayed negative consequences on their productive and reproductive performances.

Guava leaves (Psidium guajava, GL), a high-phenolic- and flavonoid-containing plant resource capable of substituting the high-quality forage, may help in mitigating ruminal methane (CH4) emission without adverse impact on nutrient degradability if supplemented at an appropriate level. In order to test this hypothesis, rumen fermentation, CH4 production, and nutrient degradability of GL either solely or as a substitute of berseem hay (Trifolium alexandrinum, BH) were evaluated in a diet containing 50:50 concentrate to roughage. Five different levels of GL (0, 12.5, 25, 37.5, and 50%) were tested in vitro after 24 h incubation using a semi-automated gas production (GP) system. The current findings indicated that merely the presence of GL resulted in significantly lower values for cumulative GP (P < 0.001), CH4 emission (P < 0.05), truly degraded dry matter (TDDM; P < 0.001), truly degraded organic matter (TDOM; P < 0.001), and ammonia nitrogen (NH3-N) concentration (P < 0.001); however, pH (P < 0.001) and partitioning factor (P < 0.001) were higher. The total and individual volatile fatty acid (VFA) concentrations were drastically declined with GL as compared to BH (P < 0.05). A negative linear correlation was recorded between the levels of GL and GP including CH4 production (P < 0.05). The addition of GL up to 25% did not pose any negative effect on both TDDM and TDOM values along with NH3-N concentration. In addition, the inclusion of GL up to 25% did not affect the total or individual VFA concentration. Conclusively, in a medium concentrate diet, use of 25% GL and 25% BH in animal diet could be a promising alternative for mitigating the CH4 production without any deleterious effect on nutrient degradability.

Leucaena (Leucaena leucocephala), a leguminous shrub promising to cope with feed scarcity in the tropics, may help in mitigating ruminal methane (CH4) emission in the tropics as well. Two experiments were conducted to evaluate the effect of Leucaena and major secondary compounds of this plant in ruminants. At first, effects of Leucaena tannins and mimosine on ruminal CH4 and nutrient degradability were tested in vitro. Incubations were made with Leucaena without or with polyethylene glycol (PEG) to exclude the tannins effects, as well as with Bermuda grass (Tifton) and lucerne hays, both either untreated or supplemented with mimosine at the same concentration that has been provided by the tested Leucaena (6.52 mg/g DM). Furthermore, in an in vivo experiment a control diet (per kg DM 700 g Tifton hay) and Leucaena diets (per kg DM 350 g Tifton hay and 350 g Leucaena), either with or without 20 g PEG/d per head, were evaluated in six Santa Inês sheep following a double Latin square design. In vitro, Leucaena resulted in the lowest (p < 0.05) gas and CH4 production and the highest (p < 0.05) partitioning factor, a measure for microbial efficiency, whereas the amount of truly degraded organic matter (TDOM) was lowest (p < 0.05) with Tifton among the experimental forage plants. Mimosine addition to lucerne and Tifton as well as PEG addition to Leucaena had no effect on ruminal CH4 and TDOM. In vivo Leucaena, compared to the Tifton diet, enhanced (p < 0.05) intake, faecal nitrogen excretion, body nitrogen retention and the excretion of urinary purine derivatives as an indicator for microbial protein synthesis and availability. This was independent of PEG addition. Leucaena also decreased (p < 0.001) CH4 emission per unit of digested organic matter by 14.1% and 10.8%, without and with PEG, respectively. No significant diet differences were observed in total-tract nutrient digestibility. The study demonstrated efficiency of Leucaena to mitigate in vivo methane emission of sheep, but did not reveal which constituent of Leucaena was primarily responsible for that since no clear efficiency of either tannins or mimosine could be demonstrated.

In view of the disadvantages of human and equine rabies immunoglobulin still there is urgent needs for safe and cost-control anti-rabies immunoglobulins especially for person who have been severely exposed (categories III) to the virus. Our attempt to produce a less immunogenic and cheaper anti-rabies immunoglobulin affordable for those people living in developing countries, has been harnessed the ovine as a bioreactor instead the horse. The animals have been intramuscular immunized, and the plasma processed with 5% caprylic acid to yield IgG with purity of 95%. Moreover, antibody apparently indicated that the titer and neutralizing indexes were harmonized, especially at higher antibody dilution. The results showed that three immunized sheep were produced about 7000 IU of purified anti-rabies antibody. Sheep's IgG has low immunogenic effect than human and horse antibodies when injected into the mouse. Pure concentrated ovine antibody may serve as a possible alternative to currently available anti-rabies human or equine immunoglobulin.
