Characteristics of Mel Ibr Melanoma Line Subclone after Treatment with Chicken Embryo Extract.

We continue analysis of the phenotype of human melanoma cell Mel Ibr subclone obtained previously by treatment of the parental cell line by chicken embryo extract. The present study is focused on detection of markers of epithelial-mesenchymal transition that determine enhanced metastatic and invasive potential of malignant tumors of various locations. Analysis of the expression of E-cadherin and vimentin genes in the subclone and parental cells detected activation of epithelial-mesenchymal transition in the subclone. Immunological markers CD90, CD271, and CD95 were present in the parental population, but were not detected on the subclone cells. In contrast to the parental line, cells of the analyzed subclone retain viability in serum-free medium and formed vessel-like structures characteristic of vasculogenic mimicry.

Changes in the Morphological and Immunological Characteristics of Mel Ibr Melanoma Cells in Response to Chicken Embryo Extract.

Exposure of Mel Ibr human melanoma cells to chicken embryo extract resulted in the appearance of a subclone with morphological and growth characteristics similar to those of embryonic stem cells. The subclone differed from the parental line cells by a sharply reduced percentage of HLA-DR(+) and CD54(+) cells, a significantly elevated percentage of CD63(+) cells, and appearance of CD133(+) and Oct-4A(+) cells. Hence, the subclone cells were characterized by the same features as stem tumor cells and could be responsible for further progress of tumor growth.

[Development of transgenic chicken with a gene of human granulocyte colony-stimulating factor using sperm-mediated gene transfer].

Transgenic poultry with a gene of human granulocyte colony-stimulating factor (gcsf) was developed by arti- ficial insemination with the transfected sperm using the "Lipofectamin 2000" reagent. The ratio of trans- genic chickens carrying the human gcsf genome was 33.3% of the total number of birds obtained. It was shown that the concentration-of the human G-csfprotein in blood serum oftransgenic specimens varied from 50 to 220 pg/ml. The first generation of poultry was obtained as a result of insemination oftransgenic hens by sperm of anti-transgenic cocks, thereby inheritance of the foreign gene was found in 37.5% of the obtained chickens.

The response of ejaculated bovine spermatozoa to gold nanoparticles was studied by the standard method of nuclear chromatin decondensation in vitro. After the treatment of semen samples with a hydrosol containing gold nanoparticles with an average diameter of 3.0 nm and a concentration of 1 x 10(15) particles/mL, the ability of sperm nuclei to decondense in the presence of sodium dodecyl sulfate (SDS) and dithiothreitol (DTT) dramatically changed compared to the control. The frequencies of gametes with nondecondensed ("intact"), partially decondensed, and completely decondensed nuclei correlated as 40 : 32 : 28% and 0 : 36 : 64% in the experiment and the control, respectively. Moreover, the appearance of a sufficiently large number of gametes with destructed and almost completely destroyed nuclei was noticed in the spermatozoa treated with gold nanoparticles. This article suggests the putative mechanisms of action of ultrasmall gold nanoparticles on the structural and functional integrity of the deoxyribonucleoprotein (DNP) complex of mature male gametes.

[The effect of solubilized methandrostenolone on muscle hypertrophy and on the blood biochemical indices in rats]. / Vliianie soliubilizirovannogo metandrostenolola na myshechnuiu gipertrofiiu i nekotorye biokhimicheskie pokazateli krovi u krys.

The paper outlines a new mode of intravascular anabolic steroidal transport. The single intravenous administration of metandrostenolone aqueous solution to rats increases hypertrophy rate of MM. plantaris. It was shown that the action of a single dose (0.05 mg/kg) exceeded that of metandrostenolone oil solution in a dose of 5 mg/kg. The multiple reduction of doses of the anabolic drug and the high level of bioavailability were achieved. This effect was explained by the application of the specific transport agent.

In rabbits with experimental hypocoagulation induced by phenylin, the use of a new dosage form of vitamin K1 for intravenous injections in does of 1 and 5 mg/kg led, in contrast to vicasol in a dose of 0.4 mg/kg, to an increase of the prothrombin index after 2 hours and to its complete normalization after 4 hours. Intravenous injection of vitamin K1 into intact animals did not entail any changes in the activated partial thromboplastin time, thrombin and prothrombin time, in the content of fibrinogen and products of its biotransformation, antithrombin III activity, and fibrinolytic activity or in the count of platelets and their aggregation capacity.

A single intravenous injection of solubilized alpha-tocopherol acetate to Wistar male rats in doses of 25, 50 and 100 mg/kg noticeably increased the animals' working capacity. The stimulation effect appeared 18 hours after the injection and was observable for two weeks. Utilization of glucose reserves in bioenergy supply of muscle activity has been shown. The maintenance of the action for such a long time may be accounted for by high metabolic stability of solubilized alpha-tocopherol acetate.

[The radioprotective and antioxidant properties of solubilized alpha-tocopherol acetate]. / Radiozashchitnye i antioksidantnye svoistva soliubilizirovannogo al'fa-tokoferola atsetata.

A single intravenous administration of solubilized alpha-tocopherol acetate in a dose of 1 mg/kg to CBA x C57Bl x F1 male mice 3 and 24 hours before radiation was demonstrated to increase 30-day survival rates in the animals. Oily alpha-tocopherol acetate had the identical effects when the agent was intramuscularly given in a dose of 10 mg/kg. There was a rise in peripheral leukocyte counts 0.5 hour after solubilized alpha-tocopherol acetate administration. The antioxidative activity of the solubilized agent was 3.3 times as high as that the routine agent when attacking Tween 80. The higher radioprotective activity of solubilized alpha-tocopherol acetate is accounted for by its enhanced bioavailability, antioxidative activity and effects on the factors of nonspecific body's resistance.

The rat experiments were made to compare the wound-healing action of solubilized (aqueous) alpha-tocopherol acetate (SATA) prepared by the original techniques and its official oil-based preparation. A course intravenous SATA (2 mg four times at an interval of 4 days) administration was found to stimulate epithelial regeneration and suppress the formation of connective tissue of uncomplicated skin wounds. By healing the complicated wounds, SATA potentiated the occurrence of an inflammatory reaction and slowed down regeneration, whereas its oily solution failed to produce this effect.

The hand joint is a multi-joint formation, including the wrist, carpal, intercarpal, carpometacarpal, intermetacarpal, distal radioulnar joints. Features of hand joint and periarticular tissue disease were studied in 536 workers exposed to local vibration and functional overstrain. The most prevalent diseases are arthroses with deformation of the wrist, radiocarpal, carpometacarpal joints, often with involvement of the triangular fibrous disk and carpals, periarthroses of the hand joint with compression neuropathy of the median and ulnar nerves.

Features of forming and course of ulnar nerve compression neuropathy in the cubital and Guyin's canals were studied in 86 subjects, suffering from occupational diseases of the arms due to vibration and functional overstrain. Compression neuropathy of the ulnar nerve aggravates the vibration disease and arm disorders caused by functional overstrain--increases frequency, intensity and duration of paroxysmal paleness of the IV-V fingers, aggravates paresthesia, hypalgesia and hypothermia of the ulnar antebrachium, markedly decrease power of the hand, which must be taken into account when solving exert problems and carrying out treatment and rehabilitation.

Defect-unbinding and the Bose-glass transition in layered superconductors.

The low-field Bose-glass transition temperature in heavy-ion irradiated Bi(2)Sr(2)CaCu(2)O(8+delta) increases progressively with increasing density n(d) of irradiation-induced columnar defects, but saturates for n(d) greater or = 1.5 x 10(9) cm(-2). The maximum Bose-glass temperature corresponds to that above which diffusion of two-dimensional pancake vortices between vortex lines becomes possible, and the "linelike" character of vortices is lost. We develop a description of the Bose-glass line that quantitatively describes experiments on crystals with widely different track densities and material parameters.
