RESUMO

The objective of this study was to compare two methods for the extraction of high motility sperm; washed semen and swim-up techniques, on 172 ejaculates from 53 patients with suspected infertility. High quality motile sperm had been essential for the success of the assisted reproductive procedures, in which these patients were enrolled. Semen analyses were performed according to the criteria recommended by the WHO. In both methods Ham's F-10 medium plus 7.5% human cord serum were used. The incubations were carried out at 37 degrees C +/- 1. The percentage of motile spermatozoa with forward progression was measured in the fresh sample and after incubation with both methods. The results were analyzed by the two-way analysis of variance. Motility rates were significantly higher in swim-up than fresh samples (mean 41.158%, SD 20.1 vs. 27.572%, SD 16.6; p < 0.0001) and washed semen compared with fresh sample (mean 36.966%, SD 16.3 vs. 27.572% SD 16.6; p < 0.0001).

RESUMO

The oral contraception most employed, consists in a combination of estrogen and progestin, the main action in the prevention of ovulation through the inhibition of the secretion of luteinizing hormone (LH) and follicle stimulating hormone (FSH). The progestin in combination with estrogen produces an endometrium not receptive to the oocyte implantation and changes tubal, this affects capacitation and acrosomal reaction. Since 1960s up to date the reports made had been established that the OC had been developed adverse effects such as increase concentration of HDL, increase of apolipoprotein B-100, insulin resistance, increase in SHBG, atherogenicity, alteration in the coagulation factors and increase in the risk of venous thromboembolic disease. This work was elaborated with literature search on MEDLINE between 1985 to 1998, with the purpose to search the collateral effects of Oral Contraceptives evaluated by other authors.

From January 1, 1987 January 30, 1990, 374 laparoscopic sterilizations using silastic rings (Falope), were carried out. Morbidity and main risk factors, were analyzed. More frequent surgical complications were: mesosalpinx rupture, and fallopian tube rupture in 22 cases (5.8%); abdominal wall emphysema, three cases (0.8%), uterine perforation, two cases (0.5%); other pelvic structures lesions, three cases (0.8%). Morbidity risk factors were: previous abdominal surgery, obesity, salpingitis, use of an uterine mobilizer in a puerperal uterus, and practice of tubal occlusion during the luteal phase of cycle, due to possibility of luteal phase pregnancy; therefore, if a risk factor is present, it is advisable to use other contraceptive technique.

RESUMO

Techniques of "In Vitro" fertilization (IFV) and gamete intrafallopian transfer (GIFT) are proving to be successful in the treatment of infertility. We report here in detail the laboratory procedures of sperm processing for these two techniques also in the present work we pointing out the media quality control methods and the incubation time for the maintenance of spermatozoa fertilizing ability.

The effect of different Inosiplex doses in tissue culture was studied utilizing a cellular line derived from the Hamster Kidney (BHK). Inosiplex doses used were 0.1, 1, 10 and 100 mg/ml of Eagle basal medium adding 10 per cent of heat inactivated fetal calf serum. Increase in cellular growth that was statistically significant was observed in 10 and 100 microgram/ml Inosiplex doses. Also this substance was studied at same concentrations in other two cellular lines, one derived from loose connective tissue in mice (L-929) and the other from cervical carcinoma (HeLa). In the first, 10 and 100 microgram/ml doses produced growth inhibition while in the second no effect was observed.

Se sometieron a experimentacion antitumoral tres mostazas nitrogenadas sinteticas 25 a las que se les ha denominado mostaza I, II y III, cuyos nombres quimicos son clorhidrato de 7 [bis(2'-cloroetil)-amino] 5-heptino, clorhidrato del 3[bis(2'cloroetil)-amino] 1-fenil-1-propino, y clorhidrato del 4[bis(2'cloroetil)-amino]-1-amino-1-ciclohexil-3-butino, respectivamente (cuadro I). Para el analisis de citotoxicidad se utilizaron cultivos de celulas L-929 y Hep-2. Se analizaron la inhibicion del crecimiento y el incremento en celulas multinucleadas. Las dosis efectivas 50 (DE50) con celulas L-929 fueron 50 microgramo/ml para la mostaza nitrogenada I, entre 5 y 10 microgramo/ml para la mostaza nitrogenada II, y menos de 1 microgramo/ml para la mostaza nitrogenada III. Con las celulas Hep-2 las mostazas I y III presentaron menor actividad citotoxica, y fue mayor la DE50 para estas dos mostazas. La mostaza II no mostro actividad inhibitoria sobre el crecimiento; sin embargo, su citotoxicidad se acepto por el efecto sobre las mitosis y la presencia de celulas multinucleadas
