RESUMO

Floral colour is an important agronomic trait that influences the commercial value of ornamental plants. Anthocyanins are a class of flavonoids and confer diverse colours, and elucidating the molecular mechanisms that regulate their pigmentation could facilitate artificial manipulation of flower colour in ornamental plants. Here, we investigated the regulatory mechanism of light-induced anthocyanin biosynthesis during flower colouration in grape hyacinth (Muscari spp.). We studied the function of two B-box proteins, MaBBX20 and MaBBX51. The qPCR revealed that MaBBX20 and MaBBX51 were associated with light-induced anthocyanin biosynthesis. Both MaBBX20 and MaBBX51 are transcript factors and are specifically localised in the nucleus. Besides, overexpression of MaBBX20 in tobacco slightly increased the anthocyanin content of the petals, but reduced in MaBBX51 overexpression lines. The yeast one-hybrid assays indicated that MaBBX20 and MaBBX51 did not directly bind to the MaMybA or MaDFR promoters, but MaHY5 did. The BiFC assay revealed that MaBBX20 and MaBBX51 physically interact with MaHY5. A dual luciferase assay further confirmed that the MaBBX20-MaHY5 complex can strongly activate the MaMybA and MaDFR transcription in tobacco. Moreover, MaBBX51 hampered MaBBX20-MaHY5 complex formation and repressed MaMybA and MaDFR transcription by physically interacting with MaHY5 and MaBBX20. Overall, the results suggest that MaBBX20 positively regulates light-induced anthocyanin biosynthesis in grape hyacinth, whereas MaBBX51 is a negative regulator.

The northwest China is a climate change-sensitive and ecologically vulnerable area. Under the backdrop of global warming, this region exhibits clear characteristics of warming and wetting. In recent years, the causes of climate change in the northwest China has become a widely-focused topic. Previous research has mainly attributed the increase in precipitation to changes in the westerly belt and enhanced local convective activity caused by global warming. South Asia, beside the Tibetan Plateau from the northwest China, is one of the regions with the fastest growth in global atmospheric pollutant emissions. This study utilizes reanalysis data such as ERA5 and MERRA-2. Statistical methods, including Theil-Sen Median trend analysis and Singular Value Decomposition analysis, are employed to analyze the spatiotemporal characteristics of South Asian aerosols, extreme precipitation in the northwest China, and the correlation between the two. The study reveals the existence of two key aerosol-precipitation response areas. Synthetic analysis of the meteorological elements of the response events in both regions is conducted to explore the possible physical mechanisms behind the correlation between South Asian aerosols and precipitation in the northwest China. The result of this study is to provide a new perspective on the causes of extreme precipitation in the arid region of northwest China.

R2R3 MYBs play vital roles in the regulation of flavonoid biosynthesis. However, the regulatory network of R2R3 MYBs in flavonoid biosynthesis is not fully understood in grape hyacinth (Muscari spp.). Here, we identified two R2R3 MYBs, MaMYBF and MaMYB1, as potential regulators of flavonol and anthocyanin biosynthesis, respectively. MaMYBF and MaMYB1 expression was elevated during flower development and was light-induced, and the expression patterns were related to those of the flavonoid structural genes MaFLS and MaDFR, respectively. The BiFC assay verified that MaMYB1 interacts with MabHLH1, but MaMYBF does not. A dual luciferase assay revealed that MaMYBF alone strongly activated pMaFLS, and its activation was attenuated at reduced doses of MaMYBF in the presence of MabHLH1, MaMybA, and MaMYB1. MaDFR transcription mediated by MaMybA and MabHLH1 was inhibited by MaMYB1. Moreover, overexpression of MaMYBF and MaMYB1 in tobacco reduced flower pigmentation and repressed the expression of flavonoid pathway key structural genes. Therefore, MaMYBF regulates the flavonol pathway independently of cofactors. Whereas MaMYB1 regulates anthocyanin biosynthesis by binding to MabHLH1 and disrupting the MaMybA-bHLH complex in grape hyacinth. Our results offer new insights into the intricate regulatory network of flavonoids in grape hyacinth involving the regulation of both flavonol and anthocyanin.

Landfills have served as the final repository for > 50 % municipal solid wastes in the United States. Because of their widespread uses and persistence in the environment, per- and polyfluoroalkyl substances (PFAS) (>4000 on the global market) are ubiquitously present in everyday consumer, commercial and industrial products, and have been widely detected in both closed (tens ng/L) and active (thousands to ten thousands ng/L) landfills due to disposal of PFAS-containing materials. Along with the decomposition of wastes in-place, PFAS can be transformed and released from the wastes into leachate and landfill gas. Consequently, it is critical to understand the occurrence and transformation of PFAS in landfills and the effectiveness of landfills, as a disposal alternative, for long-term containment of PFAS. This article presents a state-of-the-art review on the occurrence and transformation of PFAS in landfills, and possible effect of PFAS on the integrity of modern liner systems. Based on the data published from 10 countries (250 + landfills), C4-C7 perfluoroalkyl carboxylic acids were found predominant in the untreated landfill leachate and neutral PFAS, primarily fluorotelomer alcohols, in landfill air. The effectiveness and limitations of the conventional leachate treatment technologies and emerging technologies were also evaluated to address PFAS released into the leachate. Among conventional technologies, reverse osmosis (RO) may achieve a high removal efficiency of 90-100 % based on full-scale data, which, however, is vulnerable to the organic fouling and requires additional disposal of the concentrate. Implications of these knowledge on PFAS management at landfills are discussed and major knowledge gaps are identified.

RESUMO

Fracturing flowback fluid (FFF) including various kinds of organic pollutants that do harms to people and new treatments are urgently needed. Advanced oxidation processes (AOPs) are suitable methods in consideration with molecular weight, removal cost and efficiency. Here, we summarize the recent studies about AOP treatments towards organic pollutants and discuss the application prospects in treatment of FFF. Immobilization and loading methods of catalysts, evaluation method of degradation of FFF, and continuous treatment process flow are discussed in this review. In conclusion, further studies are urgently needed in aspects of catalyst loading methods, macromolecule organic evaluation methods, industrial process, and pathways of macromolecule organics' decomposition.

In this study, a series of N-benzyl-2-(5-phenylpyridin-2-yl) acetamide-based derivatives were successfully designed and synthesized as anti-cancer agents. KC-180-2 was screened as a potentially leading compound with dual mechanisms of action: Src signaling and tubulin polymerization inhibition. It efficiently suppressed the proliferation of five cancer cell lines (MDA-MB-231, H446, SKOV-3, HepG2, and HT29), with IC50 values ranging from 5 to 188 nM, especially small-cell lung cancer (SCLC) cells (IC50, 5 nM). Correspondingly, it exerted a significant therapeutic effect on the H446 small-cell lung cancer xenograft model, significantly reducing the volume of tumors without obvious toxicity. Mechanistically, this compound significantly inhibited the polymerization of purified tubulin in vitro, inducing G2/M cell cycle arrest and binding to the kinase catalytic domain of the Src protein, which reduced the phosphorylation of Src. Thus, KC-180-2 is a potential lead compound for the further development of a new anti-tumor drug against SCLC.

As a key mediator of cell death and inflammation, receptor-interacting protein kinase 1 (RIPK1) responds to a broad set of inflammatory and pro-death stimuli in human diseases. Inhibitors targeting RIPK1 are being investigated for the treatment of a wide range of human diseases, including ulcerative colitis. In the present study, we designed, synthesized, and investigated the anti-necroptosis and RIPK1-inhibition effects of SZ-15-a symmetrical high-molecular-weight (>500 Da) compound. SZ-15 effectively inhibited necroptosis in U937 and HT-29 cells at concentrations of 1 nM and 10 nM, respectively, and SZ-15 at a concentration of 10 nM almost completely blocked RIPK1, RIPK3, and mixed-lineage kinase domain-like (MLKL) protein phosphorylation induced by necrosis inducers. SZ-15 suppressed the pro-necroptosis function of RIPK1 by downregulating the mRNA expression of pro-inflammatory cytokines, including tumor necrosis factor (TNF)-α, interleukin (IL)-1ß, and IL-6. The activities of SZ-15 were effectively restricted to the gut: The percent recovery of the parent form of SZ-15 in mouse feces was 85.75%. Nevertheless, SZ-15 was effectively absorbed and detected in colon tissues after 1 h at a concentration of 3335 ± 868 ng/g, indicating that membrane permeability was maintained. SZ-15 alleviated dextran sulfate sodium (DSS)-induced ulcerative colitis in vivo by decreasing TNF-α, IL-1ß, IL-22, and IL-6 mRNA expression in colonic tissues. Our preclinical study describes a novel gut-restricted RIPK1 inhibitor that shows great potential for use in the clinical treatment of ulcerative colitis.

Per- and polyfluoroalkyl substances (PFAS) are ubiquitous in landfill leachate due to their widespread applications in various industrial and consumer products. Yet, there has been no cost-effective technology available for treating PFAS in leachate because of the intrinsic persistency of PFAS and the high matrix strength of landfill leachate. We tested a two-step 'Concentrate-&-Destroy' technology for treating over 14 PFAS from a model landfill leachate through bench- and pilot-scale experiments. The technology was based on an adsorptive photocatalyst (Fe/TNTs@AC), which was able to selectively adsorb PFAS despite the strong matrix effect of the leachate. Moreover, the pre-concentrated PFAS on Fe/TNTs@AC were effectively degraded under UV, which also regenerates the material. The presence of 0.5 M H2O2 during the photocatalytic degradation enhanced the solid-phase destruction of the PFAS. Fresh Fe/TNTs@AC at a dosage of 10 g/L removed >95% of 13 PFAS from the leachate, 86% after first regeneration, and 74% when reused three times. Fe/TNTs@AC was less effective for PFBA and PFPeA partially due to the transformation of precursors and/or longer-chain homologues into these short-chain PFAS. Pilot-scale tests preliminarily confirmed the bench-scale results. Despite the strong interference from additional suspended solids, Fe/TNTs@AC removed >92% of 18 PFAS in 8 h under the field conditions, and when the PFAS-laden solids were subjected to the UV-H2O2 system, ~84% of 16 PFAS in the solid phase were degraded. The 'Concentrate-&-Destroy' strategy appears promising for more cost-effective removal and degradation of PFAS in landfill leachate or PFAS-laden high-strength wastewaters.

Cadmium (Cd) is one of the most commonly detected toxic heavy metals in the environment. Ferrous sulfide (FeS) nanoparticles were prepared using sodium carboxymethyl cellulose (CMC) as a stabilizer and tested for removal of Cd from aqueous solutions. Effects of CMC concentration, initial Cd concentration, pH, humic acid (HA) and dissolved oxygen were examined. Fully stabilized FeS (100 mg/L) nanoparticles were obtained using 0.01 wt% CMC. Batch kinetic tests showed that the nanoparticles at 100 mg/L as FeS rapidly removed 93% of 1 mg/L Cd within 4 h at pH 7.0, and the kinetic data were well interpreted by a pseudo-second-order rate model with a rate constant of 6.68 g·mg-1·hr-1. Sorption isotherm was well simulated by a dual-mode isotherm model with a maximum Langmuir sorption capacity of 497.5 mg/L at pH 7.0. Fourier transform infrared (FTIR) spectroscopy and X-ray powder diffraction (XRD) analyses suggested that chemical precipitation and surface complexation between Cd and FeS were dominant immobilization mechanisms. Increasing pH from 4.0 to 8.0 enhanced Cd removal rate from 73.0% to 98.8%, whereas addition of 3 mg/L HA (as total organic matter) inhibited the removal rate by 2.7% and the presence of molecular oxygen had negligible effect. Increasing NaCl or CaCl2 from 0 to 10 mM suppressed Cd removal by 10.1% and 27.7%, respectively. The immobilized Cd remained insoluble when aged for 717 days under anoxic or oxic conditions. This study demonstrated that CMC-stabilized FeS nanoparticles can facilitate long-term immobilization of cadmium in contaminated water.

R3-MYBs negatively regulate anthocyanin pigmentation in plants. However, how R3-MYB repressors finely modulate anthocyanin biosynthesis in cooperation with R2R3-MYB activators remains unclear in monocots. We previously identified two anthocyanin-related R2R3-MYB activators (MaMybA and MaAN2) in grape hyacinth (Muscari spp.). Here, we isolated a R3-MYB repressor, MaMYBx, and characterized its role in anthocyanin biosynthesis using genetic and biochemical markers. The temporal expression pattern of MaMYBx was similar to that of MaMybA and MaAN2, and it was correlated with anthocyanin accumulation during flower development. MaMYBx could be activated either by MaMybA alone or by MaMybA/MaAN2 and cofactor MabHLH1, and it suppressed its own activation and that of MaMybA promoters mediated by MaMybA/MaAN2 and MabHLH1. Like MaMybA, MaMYBx interacted with MabHLH1. MaDFR and MaANS transcription and anthocyanin accumulation mediated by MaMybA/MaAN2 and MabHLH1 were inhibited by MaMYBx. Overexpression of MaMYBx in tobacco greatly reduced flower pigmentation and repressed the expression of late structural and regulatory anthocyanin pathway genes. Thus, MaMYBx finely regulates anthocyanin biosynthesis by binding to MabHLH1 and disrupting the R2R3 MYB-bHLH complex in grape hyacinth. The regulatory network of transcriptional activators and repressors modulating anthocyanin biosynthesis is conserved within monocots. MaMYBx seems a potentially valuable target for flower color modification in ornamental plants.
