BACKGROUND: Microbial polysaccharides have been reported to possess remarkable bioactivities. Physarum polycephalum is a species of slime mold for which the microplasmodia are capable of rapid growth and can produce a significant amount of cell wall-less biomass. There has been a limited understanding of the polysaccharides produced by microplasmodia of slime molds, including P. polycephalum. Thus, the primary objectives of this research were first to chemically characterize the exopolysaccharides (EPS) and intracellular polysaccharides (IPS) of P. polycephalum microplasmodia and then to evaluate their cytotoxicity against several cancer cell lines. RESULTS: The yields of the crude EPS (4.43 ± 0.44 g/l) and partially purified (deproteinated) EPS (2.95 ± 0.85 g/l) were comparable (p > 0.05) with the respective crude IPS (3.46 ± 0.36 g/l) and partially purified IPS (2.45 ± 0.36 g/l). The average molecular weight of the EPS and IPS were 14,762 kDa and 1788 kDa. The major monomer of the EPS was galactose (80.22%), while that of the IPS was glucose (84.46%). Both crude and purified IPS samples showed significantly higher cytotoxicity toward Hela cells, especially the purified sample and none of the IPSs inhibited normal cells. Only 38.42 ± 2.84% Hela cells remained viable when treated with the partially purified IPS (1 mg/ml). However, although only 34.76 ± 6.58% MCF-7 cells were viable when exposed to the crude IPS, but the partially purified IPS displayed non-toxicity to MCF-7 cells. This suggested that the cytotoxicity toward MCF-7 would come from some component associated with the crude IPS sample (e.g. proteins, peptides or ion metals) and the purification process would have either completely removed or reduced amount of that component. Cell cycle analysis by flow cytometry suggested that the mechanism of the toxicity of the crude IPS toward MCF-7 and the partially purified IPS toward Hela cells was due to apoptosis. CONCLUSIONS: The EPS and IPS of P. polycephalum microplasmodia had different chemical properties including carbohydrate, protein and total sulfate group contents, monosaccharide composition and molecular weights, which led to different cytotoxicity activities. The crude and partially purified IPSs would be potential materials for further study relating to cancer treatment.

BACKGROUND: Tuberculosis is the leading infectious cause of death. Steep reductions in tuberculosis-related mortality are required to realize the World Health Organization's "End Tuberculosis Strategy." However, accurate mortality estimates are lacking in many countries, particularly following discharge from care. This study aimed to establish the mortality rate among patients with pulmonary tuberculosis in Vietnam and to quantify the excess mortality in this population. METHODS: We conducted a prospective cohort study among adult patients treated for smear-positive pulmonary tuberculosis in 70 clinics across Vietnam. People living in the same households were recruited as controls. Participants were re-interviewed and their survival was established at least 2 years after their treatment with an 8-month standardized regimen. The presence of relapse was established by linking identifying data on patients and controls to clinic registries. Verbal autopsies were performed. The cumulative mortality among patients was compared to that among a control population, adjusting for age and gender. RESULTS: We enrolled 10964 patients and 25707 household controls. Among enrolled tuberculosis patients, 9% of patients died within a median follow-up period of 2.9 years: 342 (3.1%) during treatment and 637 (5.8%) after discharge. The standardized mortality ratio was 4.0 (95% confidence interval 3.7-4.2) among patients with tuberculosis, compared to the control population. Tuberculosis was the likely cause of death for 44.7% of these deceased patients. CONCLUSIONS: Patients treated for tuberculosis had a markedly elevated risk of death, particularly in the post-treatment period. Interventions to reduce tuberculosis mortality must enhance the early detection of drug-resistance, improve treatment effectiveness, and address non-communicable diseases.

SETTING and OBJECTIVE: Exposure to pollutants is related to the type of dwelling inhabited. Besides tobacco smoke, indoor air pollution is a significant risk factor for chronic respiratory disease (CRD). The prevalence of CRD by type of dwelling was studied in Ho Chi Minh City, Viet Nam.DESIGN: A total of 1561 people living in four type of dwellings were enrolled. Information on respiratory health, lung function, dwelling characteristics and sources of indoor pollution was obtained using a symptom and demographics questionnaire and spirometry. The two main respiratory health outcomes were clinical chronic CRD (CCRD) and chronic obstructive respiratory disease (CORD) (forced expiratory volume in 1 sec/forced vital capacity <0.7). We used binary logistic regression adjusted for age, sex, time spent at home, smoking status, certain occupational exposures, previous tuberculosis, presence of pets, rats or cockroaches at home, wall dampness, biofuel use and use of airconditioning.RESULTS: The prevalence of CCRD (24.3%) and CORD (5.3%) in the type of dwellings studied were not similar (χ² P < 0.0001). CCRD and CORD prevalence was similar in tube houses and apartments. Compared to people living in apartments, those living in rental single rooms had a 46% higher risk of developing CCRD. The odds ratio of having CORD in people living in rental single rooms and in rural houses were respectively 4.64 (95%CI 1.97-10.5) and 2.99 (95%CI 1.21-7.37).CONCLUSION: Type of dwelling was associated with CCRD and CORD morbidity.

RESUMO

A modular facile route has been developed to synthesize functionalized 2,5-di(thiophen-2-yl)-1-H-arylpyrroles from readily available starting materials. These units are compatible with various polymerization conditions and are versatile building blocks for conjugated polymers. The polymers show high thermal stability and solubility in a number of solvents. Characterization of the polymers reveals a correlation between molecular packing, controllable by polymer design, and charge carrier mobility.

Employees' working relationships were long determined to be crucial to their overall wellbeing and performance ratings at work. However, a few studies were found to examine the effects of positive workplace relationships on employees' working manners. This study aimed to investigate the effects of healthy workplace relationships on employees' working behaviors, which in turn affect their performance. In doing so, an integrated model was developed to examine the primary performance drivers of nurses in Vietnamese hospitals and focus on the effects of high-quality workplace relationships on the working attitudes of the staff. This study analyzed a questionnaire survey of 303 hospital nurses using a structural equation modeling approach. The findings demonstrated the positive effects of high-quality workplace relationships on working manners including higher commitment, lower level of reported job stress, and increased perception of social impact. Notably, the results also demonstrated that relationships between leaders and their staff nurses make a significant contribution to the quality of workplace relationship and nurses' performance. In addition, the social impact was illustrated to positively moderate the association between healthy workplace interactions and job stress; however, it had no significant effect on job commitment. Unfortunately, job commitment was surprisingly found to not be related to performance ratings. This paper provides some suggestions for the divergence of performance drivers in the hospital context in Vietnam.

This study examined the distribution of the neuropeptides somatostatin (SS) and calcitonin gene-related peptide (CGRP) in forebrain and midbrain song-control nuclei of male versus female brains in adult zebra finches (Poephila guttata) using immunohistochemical techniques. Vocal learning in songbirds is controlled by an interconnected, highly-localized system of brain nuclei. Male zebra finches produce learned vocalizations, and females do not. This behavioral sexual dimorphism is reflected in a substantial difference in the size of these song-control nuclei: males have larger nuclei containing a greater number of neurons than do females. Interestingly, previous studies describing neurochemical aspects of the song-control system in other songbird species have not reported any obvious sex differences. However, the results of this study showed that the level of neuropeptide staining was substantially greater in telencephalic song-control nuclei of male brains compared to female brains in zebra finches. In some brain regions females lacked any apparent staining, whereas male brains were intensely labeled. In other cases the number of neurons and/or intensity of labeling were diminished in females relative to males. The telencephalic pattern of somatostatin labeling revealed a large number of SS-labeled somata in the magnocellular nucleus of the anterior neostriatum species that occupy habitats with different hydrodynamic conditions. (MAN), the high vocal center (HVC) and the robust nucleus of the archistriatum (RA) of males, whereas female song-control nuclei contained many fewer or no labeled cells in these regions. Area X of the striatum appeared to contain a slightly higher level of somatostatin immunoreactivity than surrounding striatum in males, but the region corresponding to Area X in females could not be distinguished from the rest of striatum. The telencephalic pattern of CGRP staining was less extensive than that seen for SS. In male brains the magnocellular core region of lateral MAN contained many darkly labeled neurons, and RA was stained by a dense field of anterograde terminal label. Well labeled somata were also seen in medial MAN of males, but HVC was devoid of immunoreactivity. Area X in male brains contained a light field of terminal immunoreactivity. The pattern of labeling seen in males indicates that CGRP acts selectively as a neuromodulator along the efferent projection from lateral MAN to RA and Area X but not in the HVC-to-RA pathway. No telencephalic song-control nuclei in female brains contained CGRP staining. Although some diencephalic and midbrain nuclei contained well-labeled somata or fibers with either or both neuropeptides, there was little or no evidence of a sex difference in neuropeptide expression in these regions. This latter finding suggests that the greater anatomical specialization seen in cortical regions of songbirds compared to those regions in non-oscine species is accompanied by a greater neurochemical differentiation, whereas thalamic and midbrain regions may be more conserved across sex as well as species. These findings indicate that male neurons produce high levels of somatostatin and calcitonin gene-related peptide in major telencephalic vocal-control regions in zebra finches, whereas female neurons produce less or none at all. These dramatic neurochemical sex differences may be directly related to production of learned vocalizations in males, as well as to other aspects of song behavior and courtship.
