Automated measurements of the ratio of concentrations of methane and carbon dioxide, [CH4]:[CO2], in breath from individual animals (the so-called "sniffer technique") and estimated CO2 production can be used to estimate CH4 production, provided that CO2 production can be reliably calculated. This would allow CH4 production from individual cows to be estimated in large cohorts of cows, whereby ranking of cows according to their CH4 production might become possible and their values could be used for breeding of low CH4-emitting animals. Estimates of CO2 production are typically based on predictions of heat production, which can be calculated from body weight (BW), energy-corrected milk yield, and days of pregnancy. The objectives of the present study were to develop predictions of CO2 production directly from milk production, dietary, and animal variables, and furthermore to develop different models to be used for different scenarios, depending on available data. An international dataset with 2,244 records from individual lactating cows including CO2 production and associated traits, as dry matter intake (DMI), diet composition, BW, milk production and composition, days in milk, and days pregnant, was compiled to constitute the training dataset. Research location and experiment nested within research location were included as random intercepts. The method of CO2 production measurement (respiration chamber [RC] or GreenFeed [GF]) was confounded with research location, and therefore excluded from the model. In total, 3 models were developed based on the current training dataset: model 1 ("best model"), where all significant traits were included; model 2 ("on-farm model"), where DMI was excluded; and model 3 ("reduced on-farm model"), where both DMI and BW were excluded. Evaluation on test dat sets with either RC data (n = 103), GF data without additives (n = 478), or GF data only including observations where nitrate, 3-nitrooxypropanol (3-NOP), or a combination of nitrate and 3-NOP were fed to the cows (GF+: n = 295), showed good precision of the 3 models, illustrated by low slope bias both in absolute values (-0.22 to 0.097) and in percentage (0.049 to 4.89) of mean square error (MSE). However, the mean bias (MB) indicated systematic overprediction and underprediction of CO2 production when the models were evaluated on the GF and the RC test datasets, respectively. To address this bias, the 3 models were evaluated on a modified test dataset, where the CO2 production (g/d) was adjusted by subtracting (where measurements were obtained by RC) or adding absolute MB (where measurements were obtained by GF) from evaluation of the specific model on RC, GF, and GF+ test datasets. With this modification, the absolute values of MB and MB as percentage of MSE became negligible. In conclusion, the 3 models were precise in predicting CO2 production from lactating dairy cows.

Mercury (Hg) concentrations and their associated toxicological effects in terrestrial ecosystems of the Gulf of Mexico are largely unknown. Compounding this uncertainty, a large input of organic matter from the 2010 Deepwater Horizon oil spill may have altered Hg cycling and bioaccumulation dynamics. To test this idea, we quantified blood concentrations of total mercury (THg) in Seaside Sparrows (Ammospiza maritima) and Marsh Rice Rats (Oryzomys palustris) in marshes west and east of the Mississippi River in 2015 and 2016. We also tested for a difference in THg concentrations between oiled and non-oiled sites. To address the potential confounding effect of diet variation on Hg transfer, we used stable nitrogen (δ15N) and carbon (δ13C) isotope values as proxies of trophic position and the source of primary production, respectively. Our results revealed that five to six years after the spill, THg concentrations were not higher in sites oiled by the spill compared to non-oiled sites. In both species, THg was higher at sites east of the Mississippi River compared to control and oiled sites, located west. In Seaside Sparrows but not in Marsh Rice Rats, THg increased with δ15N values, suggesting Hg trophic biomagnification. Overall, even in sites with the most elevated THg, concentrations were generally low. In Seaside Sparrows, THg concentrations were also lower than previously reported in this and other closely related passerines, with only 7% of tested birds exceeding the lowest observed effect concentration associated with toxic effects across bird species (0.2 µg/g ww). The factors associated with geographic heterogeneity in Hg exposure remain uncertain. Clarification could inform risk assessment and future restoration and management actions in a region facing vast anthropogenic changes.

Short-chain fatty acids (SCFA) and lactate in ruminal fluid are products resulting from the microbial fermentation of substrates and can be used to reflect the composition and activity of the ruminal microbiome. Determination of SCFA and D-/L-lactate in ruminal fluid currently requires two separate protocols, which is time-consuming and costly. In this study, we have optimised and validated a simple and unified 3-nitrophenylhydrazine (3-NPH) derivatisation protocol and a 20 min chiral-LC-MS method for the simultaneous quantification of all SCFA and D- and L-lactate in ruminal fluid. This method, which requires no sample pretreatment or purification shows adequate sensitivity (limit of detection (LOD): 0.01 µg/mL), satisfactory accuracy (recovery: 88-103%), and excellent reproducibility (relative standard deviation (RSD) for repeated analyses < 3% for most analytes). The application of this method to a cohort of 24 animals allowed us to reveal a large inter-cow variation in ruminal SCFA and lactate level, the concentration range for each species, the widespread correlation between different SCFA, and the strong correlation between D- and L-lactate.

Quantification of SARS-CoV-2 RNA copies in wastewater can be used to estimate COVID-19 prevalence in communities. While such results are important for mitigating disease spread, SARS-CoV-2 measurements require sophisticated equipment and trained personnel, for which a centralized laboratory is necessary. This significantly impacts the time to result, defeating its purpose as an early warning detection tool. The objective of this study was to evaluate a field portable device (called MINI) for detecting SARS-CoV-2 viral loads in wastewater using real-time reverse transcriptase loop-mediated isothermal amplification (real-time RT-LAMP). The device was tested using wastewater samples collected from buildings (with 430 to 1430 inhabitants) that had known COVID-19-positive cases. Results show comparable performance of RT-LAMP against reverse transcriptase polymerase chain reaction (RT-qPCR) when detecting SARS-CoV-2 copies in wastewater. Both RT-LAMP and RT-qPCR detected SARS-CoV-2 in wastewater from buildings with at least three positive individuals within a 6-day time frame prior to diagnosis. The large 96-well throughput provided by MINI provided scalability to multi-building detection. The portability of the MINI device enabled decentralized on-site detection, significantly reducing the time to result. The overall findings support the use of RT-LAMP within the MINI configuration as an early detection system for COVID-19 infection using wastewater collected at the building scale.

INTRODUCTION: Acute right iliac fossa (RIF) pain is a common presenting symptom in surgical patients, with a wide range of differentials, particularly in premenopausal females. This study explores ultrasound usage in the management of women aged 16-55 years presenting with RIF pain. METHODS: A total of 1,082 patients who presented to a tertiary hospital over 12 months were included. Data were collected from patients' electronic records, including initial clinical impression, imaging, management, operative findings, histology and subsequent hospital attendances within 6 weeks and within 6 months. RESULTS: Following clinical assessment, 607 (56%) of patients underwent an ultrasound. Of these, 280 (25.9%) patients received no radiological imaging on initial presentation, and 252 (42%) had pathology identified on ultrasound. The most common finding was an ovarian cyst, closely followed by unexplained free pelvic fluid. Of the 607 patients scanned, 29 (4.8%) had an ultrasound diagnosis of appendicitis; 254 of 1,082 (23.5%) patients underwent operative management. Of the 254 patients who had surgery, 179 (70.5%) had preoperative imaging. Of the 29 (11.4%) cases where the intraoperative finding was gynaecological, 15 (51.7%) cases had not had any preoperative imaging. The negative appendicectomy rate was 21.3% (45/211). Of the 45 patients who had a histologically normal appendix, 22 (48.9%) had not had any previous imaging. Ultrasound had a specificity of 78% for diagnosing appendicitis. CONCLUSIONS: In patients who underwent operative management, a negative finding or finding not requiring surgical management was associated with no preoperative imaging. This supports the use of ultrasound scans as an adjunct in a multimodal approach to the assessment of women presenting with RIF pain.

Abstract: The present study describes the pharmacological analysis of the effects of carbachol, a cholinergic agonist, on hippocampal theta activity. Knowing that this activity is critically related to cognitive function and altered in patients with neurodegeneration, pharmacological efforts aiming to directly modulate hippocampal theta activity becomes of central importance. In a recently developed complete septo-hippocampal preparation, carbachol elicited significant theta power enhancement with 1 μM. Concentrations under 1 μM and over 2 μM carbachol caused significant reduction in the power of hippocampal theta activity. Carbachol effects were completely blocked with the cholinergic antagonist scopolamine. At the experimental level, it is the first time the direct action of a cholinergic agonist is evaluated in the septo-hippocampal pathway completely isolated. However, carbachol as a cholinergic agonist is a drug with a certain level of nonspecific response. That is why to correct this experimental limitation, we used scopolamine (cholinergic antagonist) which allowed us to corroborate the effects on the cholinergic pathway. In summary, electrophysiological assays demonstrated an effective concentration range of carbachol specifically modulating hippocampal theta activity.


Resumen: El presente estudio describe el análisis farmacológico de los efectos de carbacol, un agonista colinérgico, sobre la actividad theta del hipocampo. Sabiendo que esta actividad está críticamente relacionada con la función cognitiva y alterada en pacientes con neurodegeneración, los esfuerzos farmacológicos destinados a modular directamente la actividad theta del hipocampo se vuelven de gran importancia. En una preparación completa que contiene la región septal media conectada al hipocampo, desarrollada recientemente, 1 μM de carbacol provocó un incremento significativo a nivel de potencia en la actividad theta del hipocampo. Las concentraciones menores de1 μM y mayores a 2 μM causaron una reducción significativa en la potencia de la actividad theta. Los efectos del carbacol fueron completamente bloqueados con la escopolamina, antagonista colinérgico. A nivel experimental, es la primera vez que se evalúa la acción directa de un agonista colinérgico en la vía septo-hipocámpica completamente aislada. Sin embargo, el carbacol como agonista colinérgico es un fármaco que presenta cierto nivel de respuesta inespecífica. Es por eso que para corregir esta limitante experimental, se utilizó escopolamina (antagonista colinérgico) lo que nos permitió corroborar los efectos sobre la vía colinérgica. En resumen, nuestros estudios electrofisiológicos demostraron un intervalo de concentración eficaz del carbacol que modula específicamente la actividad theta del hipocampo.

The case of a 42- year old woman with leiomyosarcoma of the kidney, a very rare renal lesion, is presented. Leiomyosarcomas are the most common of the primary renal sarcomas which account for less than 1% of renal tumours in adults.


Se presenta el caso de una mujer de 43 años de edad con un leiomiosarcoma del riñón - una lesión renal muy rara. Los leiomiosarcomas son los más comunes de los sarcomas renales primarios, y representan menos del 1% de los tumores renales en adultos.

ECMO es un procedimiento de rescate de recién nacidos (RN) con fallo respiratorio hipoxémico severo que no responde al tratamiento convencional. Previo a la instrumentación de esta técnica en pacientes, se realizó el entrenamiento experimental en animales. Existe escasa bibliografía sobre la utilización de porcinos como modelo animal en ECMO. Se reportan, en cambio, la implementación de otros animales, tales como el cordero. Objetivo: determinar la utilidad y el comportamiento modelo animal eligido, su respuesta al procedimiento de ECMO, describir la técnica utilizada y sus resultados. Método: se utilizaron dos porcinos de la raza Landrace, de 30 y 45 días de vida. Se canularon la vena yugular interna y la arteria carótida interna derechas; se realizó el ensamblado y purgado del circuito. El bypass se inició con un flujo de bomba de 20 mL/K/min, aumentandose hasta 100-120 mL/K/min; logrado un flujo de 80 mL/K/min se disminuyeron los parámetros de Asistencia Respiratoria Mecánica (ARM) a nivel de reposo. Durante el curso de ECMO se practicaron diferentes acciones relacionadas con el manejo del sistema de perfusión y evaluación del comportamiento clínico del modelo. Finalmente se suspendió electivamente el procedimiento; luego el animal fue dacanulado y recuperado. Resultados: tiempo promedio de ECMO: 24.5 horas. Signos vitales del animal estables. No hubo complicaciones relacionadas con el circito, ni presencia de sangrado sistémico. Se lograron tasas deseadas de hemofiltración. Ambos animales sobrevivieron. Como complicación se registró hematuria en un caso. Conclusión: El modelo animal elegido, para el entrenamiento en el manejo de ECMO, fue satisfactorio.

Diadema antillarum populations at many Caribbean locations have failed to recover from the pathogen-induced mortality events of the 1980s. It has become clear that the massive decline of this herbivorous urchin and the wide-spread absence of a population recovery lead to numerous long-term ecological consequences and reef degradation. While few quantitative studies on pre-mortality exist, great effort has been put forth to monitor remaining populations and their recovery. However, the patchy distribution of D. antillarum coupled with paucity of long-term studies based on the same methods applied at the same locations undermines the value of local as well as regional comparisons. In Dominica, ongoing quantitative assessments of D. antillarum began in 2001. Surveys of D. antillarum abundance are being carried out in 4-month intervals at six 100 m² sites; spread over 38 km along the west coast. The density of D. antillarum has differed significantly between sites, ranging from 0.81(SD= 0.04) to 3.13 m-2 (SD= 2.10), and increased by 61.11% during the first five years of this study. Seasonal fluctuations, possibly related to spawning aggregations, are also evident. The current abundance of D. antillarum on Dominican reefs contrasts that of Caribbean locations with recorded incidents of mass mortality events, and in some cases resembles pre-mortality densities from the early 1970s. Prior to this study, no systematic quantitative assessments of D. antillarum were carried out in Dominica. It is thus unclear in what way Dominica’s D. antillarum were affected by the mass mortality events observed elsewhere in the 1980s. The increase in D. antillarum density so far observed may thus be the recovery from a pathogen-induced disturbance or from Hurricane Lenny in November 1999. Locally, D. antillarum is important grazer on Dominican’s reefs, where over-fishing has drastically reduced the number of herbivorous fishes. On a regional scale, the island’s D. antillarum may represent a source of larvae for downstream islands, given the duration of the echinoplutei stage and a range of larval dispersal of hundreds of kilometers. Las poblaciones de Diadema antillarum de muchas localidades del Caribe, no se han recuperado después de los eventos de mortandad, inducidos por un patógeno, en la década de 1980. Se ha podido demostrar que la muerte masiva de este erizo herbívoro y la ausencia de recuperación, han tenido consecuencias ecológicas que han producido la degradación de los arrecifes. Existen pocos estudios pre-mortalidad, pero se han realizado grandes esfuerzos para monitorear las poblaciones restantes y su recuperación. Sin embargo, la distribución en parches de D. antillarum combinado con pocos estudios a largo plazo usando la misma metodología en los mismos sitios, mina la posibilidad de comparaciones locales y regionales. En Dominica, el monitoreo cuantitativo de D. antillarum empezó en 2001. Cada cuatro meses se realizan determinaciones de abundancia de D. antillarum en seis sitios de 100 m²; distribuidos a lo largo de 38 km de la costa oeste. La densidad de D. antillarum, fue significativamente diferente entre sitios, con ámbitos de 0.81 (SD= 0.04) a 3.13 m-2 (SD= 2.10), y aumentó 61.11% durante los primeros cinco años de estudio. Fluctuaciones estacionales, posiblemente relacionadas con agregaciones reproductivas, son evidentes. La abundancia actual de D. antillarum en los arrecifes de Dominica contrasta con otros sitios en el Caribe que sufrieron mortandades masivas y en algunos casos se parece a densidades pre-mortalidad de inicios de la década de 1970. Antes de este trabajo, no se había realizado ningún estudio sistemático de Diadema en Dominica, por lo que no es claro como fueron afectadas las poblaciones en este sitio. El aumento en la densidad de D. antillarum observado hasta ahora puede deberse a recuperación tras una perturbación por un patógeno o por el Huracán Lenny de noviembre 1999. Localmente, D. antillarum es un herbívoro importante en los arrecifes de Dominica donde la sobrepesca ha disminuido el número de peces herbívoros. En escala regional, las poblaciones de D. antillarum de la isla de Dominica pueden representar una fuente de larvas corriente abajo, dada la duración de estadio equiinopluteo y el ámbito de dispersión de las larvas, que es de cientos de kilómetros.

Se efectuó vitrectomía a cielo abierto en 23 ojos de 18 niños (entre 4 y 35 meses de edad) que presentaban una Retinopatia del prematuro (R.P.) en etapa cicatrizal grado V - desprendimiento de retina total con configuración en embudo cerrado. Al finalizar el seguimiento, la retina estaba aplicada en el polo posterior en 8 ojos (8/23 = 34,7%). El seguimiento osciló entre 1 y 48 meses (media = 14 meses; mediana = 12 meses). Se analiza la posible influencia de factores clínico-patológicos y quirúrgicos en el resultado anatómico

In a study in western Kenya of malaria-infected adult women who had been treated with chloroquine, we compared the level of chloroquine and its principal metabolite, desethylchloroquine, in urine, measured using a newly developed modified Haskins test, with the level of chloroquine in whole blood, determined by high-performance liquid chromatography. Over a 28-day follow-up period, 277 matched urine and blood samples from 81 women were evaluated. A high correlation was observed between the level of chloroquine in whole blood (in ug/l) and that of chloroquine+desethylchloroquine in urine (in ug/l). The test was easily performed and may be useful for monitoring use of chloroquine in a community and determining a pre-study or post-treatment ingestion or absorption of the drug in in vivo studies of parasite sensitivity
