RESUMO

Soil salinity is a global major abiotic stress threatening crop productivity. In salty conditions, plants may suffer from osmotic, ionic, and oxidative stresses, resulting in inhibition of growth and development. To deal with these stresses, plants have developed a series of tolerance mechanisms, including osmotic adjustment through accumulating compatible solutes in the cytoplasm, reactive oxygen species (ROS) scavenging through enhancing the activity of anti-oxidative enzymes, and Na+/K+ homeostasis regulation through controlling Na+ uptake and transportation. In this review, recent advances in studies of the mechanisms of salt tolerance in plants are described in relation to the ionome, transcriptome, proteome, and metabolome, and the main factor accounting for differences in salt tolerance among plant species or genotypes within a species is presented. We also discuss the application and roles of different breeding methodologies in developing salt-tolerant crop cultivars. In particular, we describe the advantages and perspectives of genome or gene editing in improving the salt tolerance of crops.

Barley is the fourth most produced cereal crop in the world and one of the major dietary sources of cadmium (Cd), which poses serious threats to human health. Here, we identify a gene that encodes a P-type heavy metal ATPase 3 (HvHMA3) responsible for grain Cd accumulation in barley. HvHMA3 from the high Cd barley variety Haruna Nijo in Japan and the low Cd variety BCS318 in Afghanistan shared 97% identity at the amino acid level. In addition, the HvHMA3 from both varieties showed similar transport activity for Cd and the same subcellular localization at the tonoplast. However, the expression of HvHMA3 was double in BCS318 than in Haruna Nijo. A 3.3-kilobase Sukkula-like transposable element was found to be inserted upstream of the gene in the low Cd variety, which functioned as a promoter and enhanced the expression of HvHMA3. Introgression of this insertion to an elite barley cultivar through backcrossing resulted in decreased Cd accumulation in the grain grown in Cd-contaminated soil without yield penalty. The decreased Cd accumulation resulting from the insertion was also found in some other barley landraces in the world. Our results indicate that insertion of the Sukkula-like transposable element plays an important role in upregulating HvHMA3 expression.

OBJECTIVE: To establish the theories and methods to determine solubility parameters of multiple components for the traditional Chinese material medica (TCMM) with HPLC. METHOD: The mathematical expresses to determin the solubility parameters were established according to chromatographic and Hildebrand-Scatchard theories, The HPLC experiments were carried out at 40 degrees C on an Alltech Apollo C18 column (4.6 mm x 250 mm, 5 microm) eluted with acetone and water in gradient mode. The flow rate was 1.0 mL min(-1), and the detection wavelength was 254 nm. The injection volume was 1 0 microL. RESULT: The mathematical expresses between the retention time and the solubility parameters were established and used to determin caffeine solubility parameter as 28.31 J(1/2) cm(-3/2) in accordance with 28.84 J(1/2) cm(-3/2) reported by literature, and those of aloe-emodin, rhein, emodin, physcione as 39.70 J(1/2) cm(-3/2), 39.08 J(1/2) cm(-3/2), 38.37 J(1/2) cm(-3/2), 36.42 J(1/2) cm(-3/2) respectively. CONCLUSION: The retention time of HPLC can be used to determine the solubility parameters of multiple componets. The established method is useful for the compatibility rule study of traditional Chinese medicine.

ß-Amylase activity (BAA) and thermostability (BAT) are important traits for malt quality. In this study, 138 Tibetan annual wild barley accessions and 20 cultivated genotypes differing in BAA were planted and analyzed in 2009 and 2012. Significant differences were detected among genotypes in BAA and BAT. The cultivated genotypes had a mean BAA of 1137.6 U/g and a range of from 602.1 to 1407.5 U/g, while the wild accessions had a mean of 1517.9 U/g and a range of from 829.7 to 2310.0 U/g. The cultivated genotypes had a mean relative residual ß-amylase activity (RRBAA) of 61.6% and a range of from 22.2% to 82.3%, while the wild barleys had a mean of 57.8% and a range of from 21.9% to 96.1%. Moreover, there was a significant difference among genotypes in the response of RRBAA to the temperature and duration of heat treatment. The wild barleys had wider variation in BAA and BAT than cultivated genotypes.

RESUMO

Praziquantel remains the drug of choice for schistosomiasis, but it has the low bioavailability and single dosage form. This paper reviews the properties of praziquantel in vivo and in vitro, the pathogenic characteristics of schistosomiasis, and the progress of research on the formation of praziquantel, so as to enhance the efficiency of praziquantel against schistosomiasis and provide consults for relevant drug researches.
