RESUMO

Neurodegenerative diseases involve the progressive dysfunction and loss of neurons in the central nervous system and thus present a significant challenge due to the absence of effective therapies for halting or reversing their progression. Based on the characteristics of neurodegenerative diseases such as Alzheimer's disease (AD) and Parkinson's disease (PD), which have prolonged incubation periods and protracted courses, exploring non-invasive physical therapy methods is essential for alleviating such diseases and ensuring that patients have an improved quality of life. Photobiomodulation (PBM) uses red and infrared light for therapeutic benefits and functions by stimulating, healing, regenerating, and protecting organizations at risk of injury, degradation, or death. Over the last two decades, PBM has gained widespread recognition as a non-invasive physical therapy method, showing efficacy in pain relief, anti-inflammatory responses, and tissue regeneration. Its application has expanded into the fields of neurology and psychiatry, where extensive research has been conducted. This paper presents a review and evaluation of studies investigating PBM in neurodegenerative diseases, with a specific emphasis on recent applications in AD and PD treatment for both animal and human subjects. Molecular mechanisms related to neuron damage and cognitive impairment are scrutinized, offering valuable insights into PBM's potential as a non-invasive therapeutic strategy.

Lipopolysaccharide (LPS)-induced macrophage activation is a prototype of innate immune response. Although key effector proteins in LPS signaling pathway have been revealed, the map of dynamic protein interactions and phosphorylation as well as the stoichiometry of protein complexes are lacking. Here we present a dynamic map of protein interactions and phosphorylation in MyD88, TRAF6 and NEMO complexes obtained by SWATH-MS. The comprehensive MS measurement leads to quantification of up to about 3,000 proteins across about 21-40 IP samples. We detected and quantified almost all known interactors of MyD88, TRAF6 and NEMO. By analyzing these quantitative data, we uncovered differential recruitment of IRAK family proteins to LPS-induced signaling complexes and determined the stoichiometry of the Myddosome complex. In addition, quantitative phosphoproteomics analysis identified a number of unreported high-confidence phosphosites on the key proteins in LPS signaling pathway. Collectively, data of dynamic protein interactions and phosphorylation presented by this study could be a resource for further study of the LPS signaling pathway.

There remains a significant gap in our quantitative understanding of crosstalk between apoptosis and necroptosis pathways. By employing the SWATH-MS technique, we quantified absolute amounts of up to thousands of proteins in dynamic assembling/de-assembling of TNF signaling complexes. Combining SWATH-MS-based network modeling and experimental validation, we found that when RIP1 level is below ~1000 molecules/cell (mpc), the cell solely undergoes TRADD-dependent apoptosis. When RIP1 is above ~1000 mpc, pro-caspase-8 and RIP3 are recruited to necrosome respectively with linear and nonlinear dependence on RIP1 amount, which well explains the co-occurrence of apoptosis and necroptosis and the paradoxical observations that RIP1 is required for necroptosis but its increase down-regulates necroptosis. Higher amount of RIP1 (>~46,000 mpc) suppresses apoptosis, leading to necroptosis alone. The relation between RIP1 level and occurrence of necroptosis or total cell death is biphasic. Our study provides a resource for encoding the complexity of TNF signaling and a quantitative picture how distinct dynamic interplay among proteins function as basis sets in signaling complexes, enabling RIP1 to play diverse roles in governing cell fate decisions.

OBJECTIVE: To evaluate the effect of health education path on advanced schistosomiasis patients with splenomegaly in perioperative period. METHODS: A total of 60 advanced schistosomiasis patients with splenomegaly in perioperative period were randomly divided into an observation group and a control group (30 cases each group). The patients in the observation group were intervened by the nursing and health education from admission to discharge according to the standardized process of health education path, while those in the control group were implemented with the conventional health education. Then the satisfaction of nursing work, hospital stays, hospitalization expenses and mastery of health knowledge of the patients in the two groups were investigated and the results were compared. RESULTS: The satisfaction rates of nursing work of patients in the observation group and the control group were 90% and 60%, respectively, the difference between them was statistically significant (χ2 = 7.57, P < 0.05). The average hospitals stays of the above two groups were 14.0 ± 3.5 d and 19.0 ± 6.8 d (t = 2.85, P < 0.01), respectively, and the average hospitalization expenses were (1.5 ± 0.5) thousand Yuan and (2.2 ± 0.7) thousand Yuan (t = 24.57, P < 0.01), respectively, both of the differences were statistically significant. On the 10th day after surgery, the mastery rates of the health knowledge of the two groups were 96.7% and 50.0%, respectively, the difference was statistically significant (χ2 = 16.86, P < 0.001). CONCLUSIONS: The implementation of the health education path can promote the rehabilitation of advanced schistosomiasis patients with splenomegaly, and increase the satisfaction as well as improve the mastery of the health knowledge of the patients. Meanwhile, it also can cut down the hospital stays and save the hospitalization expenses.

Taking tobacco as test material, a pot experiment was conducted to study the effect of silicon on translocation of lead (Pb) form soil to tobacco in order to explore effective measures for reducing Pb concentration in tobacco leaf. The results showed that silicon application promoted the transformation of exchangeable Pb into Fe-Mn oxide-bound Pb in non-rhizospheric soil, and into Fe-Mn oxide-bound Pb and residual Pb in rhizospheric soil, which decreased the availability and mobility of Pb in the soil. Silicon application significantly reduced the Pb uptake of tobacco, with the content of Pb being decreased by 6.5% to 44.0% in tobacco, and 3.1% to 60.4% in leaf. Silicon application promoted the transformation of ethanol-extractable, H2O-extractable Pb and NaCl-extractable Pb into HCl-extractable Pb and residual Pb in root, stem and leaf of tobacco, which reduced the toxicity and mobility of Pb in tobacco. Silicon restricted the transportation of Pb from soil to tobacco leaf by reducing the mobility index of Pb from soil to root and the mobility index of Pb from root to stem in soil-tobacco system. Meanwhile, the mobility index of Pb from stem to leaf in soil-tobacco system showed a rising-and-falling trend with the increase of Pb application. Silicon inhibited the Pb migration from soil to tobacco leaf by reducing availability of Pb, mitigating toxicity of Pb to tobacco, and changing the distribution of Pb forms in tobacco, consequently reducing Pb concentration of tobacco leaf. These results demonstrated silicon application could be effective in reducing translocation of Pb from soil to tobacco.
