RESUMO

The nuclear receptor gene Ecdysone-induced protein 75 (E75), as the component of ecdysone response genes in the ecdysone signaling pathway, has important regulatory function for insect molting. However, the regulatory function of E75 during the molting process of spider mites is not yet clear. In this study, the expression pattern of E75 in the molting process of the spider mite Tetranychus urticae was analyzed. The results showed that there was a peak at 8 h post-molting, followed by a decline 8 h after entering each respective quiescent stage across various developmental stages. During the deutonymph stage, the expression dynamics of E75, observed at 4-h intervals, indicated that the transcript levels of TuE75 peaked at 24 h, coinciding with the onset of molting in the mites. To investigate the function of TuE75 during the molting process, silencing TuE75 through dsRNA injection into deutonymph mites at the age of 8 h yielded a notable outcome: 78% of the deutonymph mites were unable to progress to the adult stage. Among these phenotypic mites, 37% were incapable of transitioning into the quiescent state and eventually succumbed after a certain period. An additional 41% of the mites successfully entered the quiescent state but encountered difficulties in shedding the old epidermis, leading to eventual mortality. In summary, these results suggested that TuE75 plays a key role in the molting process of T. urticae.

The new genus, Cerogamasus gen. nov., with the type species Cerogamasus tibetensis sp. nov., is established. The new genus is easily distinguished from other genera of Parasitidae because the dorsal idiosoma in both sexes bears more than 40 pairs of setae, of which fewer than 7 pairs of podonotal setae are smooth; the seta z5 of the dorsal hexagon is similar to j5 and j6 in form (pilose or distally pilose) while different in length (z5 longer); the seta al of the palpfemur is pectinate, and al1 and al2 of the palpgenu are entire; the gnathotectum is trispinate; peritrematal shields in females are posteriorly free; and the palptrochanter in males has a pointed ventral protuberance. C. anhuiensis sp. nov., C. guizhouensis sp. nov. and C. multidentatus sp. nov. are described based on adult samples; C. tibetensis sp. nov. is described based on deutonymph and adult samples. Cycetogamasus coreanus Athias-Henriot, 1980, is transferred to Cerogamasus gen. nov. as a new combination.

The greater wax moth, Galleria mellonella (Lepidoptera, Pyralidae), is a major bee pest that inflicts considerable harm on beehives, leading to economic losses. It also serves as a valuable resource insect and a model organism. The clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) system plays a crucial role in improving economic insect breeding and developing efficient agricultural pest management systems in Lepidoptera. However, the CRISPR/Cas9 protocols have not been developed for G. mellonella. Here, the Gmebony knockout (KO) strain was established using the CRISPR/Cas9 genome editing system. We obtained Gmebony KO strain in the G4 generation, which took approximately 10 months. When compared with wild-type, the head, notum, and the terminal abdominal surface of 1st to 4th instar larvae in the KO strain changed from yellow to brown, and these regions of the KO strain gradually transformed into a black color from the 5th instar larvae, and the body color of the adult moth in the KO strain changed to black. The developmental period of the early larval and the following larval instars extended. The embryonic hatchability of the Gmebony KO strain was significantly decreased. The pupal body weight of the Gmebony KO strain was not affected. The feasibility of the CRISPR/Cas9 methodology was validated by single-target editing of Gmebony. Our findings provide the first evidence that the ebony gene can serve as a pigmentation reference gene for genetic modifications of G. mellonella. Meanwhile, it can be utilized in the development of genome editing control strategies and for gene function analyses in G. mellonella.

Ontogenetic development of leg chaetotaxy, stylophore and striae of the dorsal integument, and variations of leg setal counts and length of idiosomal setae in Aponychus corpuzae are discussed. The following changes were observed during the ontogenetic development of A. corpuzae: the depth of the depression in stylophore increases; the dorsal striae gradually reduce in number and prominence; the length of dorsal setae sc1, c12, d12 and e1 reduce in size; adult dorsal setae much thicker than those of immature stages. There are a significant number of setae suppressed in A. corpuzae, often resulting in unique patterns and sexual dimorphism. The adult males and females of Aponychus firmianae and A. taishanicus from mainland China are redecribed, and a key to all Aponychus from mainland China is provided.

The genus Neotrichobia Tuttle & Baker is redescribed with a revised diagnosis. The type species Neotrichobia arizonensis Tuttle & Baker, 1968 is redescribed based on larvae, protonymphs, deutonymphs and adults including the paratype male and deutonymph.
