RESUMO

Leaf trichomes protect against various biotic and abiotic stresses in plants. However, there is little knowledge about this trait in oilseed rape (Brassica napus). Here, we demonstrated that hairy leaves were less attractive to Plutella xylostella larvae than glabrous leaves. We established a core germplasm collection with 290 accessions for a genome-wide association study (GWAS) of the leaf trichome trait in oilseed rape. We compared the transcriptomes of the shoot apical meristem (SAM) between hairy- and glabrous-leaf genotypes to narrow down the candidate genes identified by GWAS. The single nucleotide polymorphisms and the different transcript levels of BnaA.GL1.a, BnaC.SWEET4.a, BnaC.WAT1.a and BnaC.WAT1.b corresponded to the divergence of the hairy- and glabrous-leaf phenotypes, indicating the role of sugar and/or auxin signalling in leaf trichome initiation. The hairy-leaf SAMs had lower glucose and sucrose contents but higher expression of putative auxin responsive factors than the glabrous-leaf SAMs. Spraying of exogenous auxin (8 µm) increased leaf trichome number in certain genotypes, whereas spraying of sucrose (1%) plus glucose (6%) slightly repressed leaf trichome initiation. These data contribute to the existing knowledge about the genetic control of leaf trichomes and would assist breeding towards the desired leaf surface type in oilseed rape.

KEY MESSAGE: We provided a gene pool of moderate size for selecting or manipulating the candidate genes that favour the acquisition of seed dormancy, shedding light on the elevation of seed oil content in oilseed rape by blocking lipid degradation in developing seeds. In oilseed rape, the association between the germination potential of premature seeds and the final level of seed lipids, and the underlying mechanism, is elusive. Here, we investigated phenotypic differences in the germination percentage of premature seeds in a collection of oilseed rape cultivars. We compared the dynamic lipid accumulation between the deep-, moderate- and non-dormant genotypes and compared the transcriptomes of the seeds at 40 days after pollination between multiple pairs of deep- and non-dormant genotypes. We identified a wide range of differences in germination percentage of premature seeds and the association between the germination potential and the change of fatty acid content at late stage of seed maturation. The comparisons of transcriptomes between deep- and non-dormant seeds revealed the genetic basis for the dormant difference, e.g. the different expression levels of the genes involved in gibberellic and abscisic acid biosynthesis and/or signalling, fatty acid metabolic pathways, and the structure of seed cell wall. The outbreak of coronavirus disease 2019 (COVID-19) inspires various conspiracy theories, which could divert public attention, alter human behaviors, and consequently affect the spread of the pandemic. Here we estimate the relation of the online attention on COVID-19-related conspiracy theories to human mobility, as well as to the numbers of confirmed COVID-19 cases, during 14 March 2020 to 28 August 2020. We observe that the online attention to COVID-19 conspiracy theories is significantly and negatively related to human mobility, but its negative impact is noticeably less than those of the attention to official information and personal protection measures. Since human mobility significantly promotes the spread of COVID-19, the attention to official information and personal protection measures lowers COVID-19 cases by 16.16% and 9.41%, respectively, while attention to conspiracy theories only reduces the COVID-19 cases by 6.65%. In addition, we find that in the states with higher online attention to COVID-19 conspiracy theories, the negative relation of the attention to conspiracy theories is much weaker than that in states where there is less concern about conspiracies. This study stresses the necessity of restricting the online transmission of unfounded conspiracy theories during a pandemic.

Hongqu is a famous fermented food produced by Monascus and has been used as food coloring, wine starters and food additives for thousands of years in China. Excellent Monascus strain is an important prerequisite for producing high-quality Hongqu. However, the isolation of Monascus pure culture from Hongqu samples is time-consuming and laborious because it is easily interfered by other microorganisms (especially filamentous fungi). Therefore, the development of restrictive medium for Monascus enrichment from Hongqu is of great significance for the preparation and screening of excellent Monascus strains. Results of this study showed that Monascus has good tolerance to lactic acid and ethanol. Under the conditions of tolerance limits [7.5% lactic acid (v/v) and 12.0% ethanol (v/v)], Monascus could not grow but it still retained the vitality of spore germination, and the spore activity gradually decreased with the increasing concentrations of lactic acid and ethanol. More interestingly, the addition of lactic acid and ethanol significantly changed the microbial community structure in rice milk inoculated with Hongqu. After response surface optimization, Monascus could be successfully enriched without the interference of other microorganisms when 3.98% (v/v) lactic acid and 6.24% (v/v) ethanol were added to rice milk simultaneously. The optimal enrichment duration of Monascus by the restrictive medium based on the synergistic stress of lactic acid and ethanol is 8∼24 h. The synergistic stress of lactic acid and ethanol had no obvious effects on the accumulation of major metabolites in the progeny of Monascus, and was suitable for the enrichment of Monascus from different types of Hongqu. Finally, the possible mechanisms on the tolerance of Monascus to the synergistic stress of lactic acid and ethanol were preliminarily studied. Under the synergistic stress of lactic acid and ethanol, the cell membrane of Monascus defends against lactic acid and ethanol into cells to some extent, and the superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities of Monascus were higher than those of other fungi, which significantly reduced the degree of lipid peroxidation of cell membrane, while secreting more amylase to make reducing sugars to provide the cells with enough energy to resist environmental stress. This work has great application value for the construction of Monascus strain library and the better development of its germplasm resources.

To investigate the superhydrophobic properties of different surface textures, nine designs of textures with micro-nanostructures were produced successfully using the laser engraving technique on the surfaces of composite insulator umbrella skirt samples made of silicon rubber. The optimal parameters of the texture designs to give rise to the best hydrophobicity were determined. The surface morphology, abrasion resistance, corrosion resistance, self-cleaning and antifouling property of the different textured surfaces as well as water droplets rolling on the textured surfaces were studied experimentally using a contact angle meter, scanning electron microscope, three-dimensional topography meter and high-speed camera system. It was found that the diamond column design with optimal parameters has the best superhydrophobicity and overall performance. The most remarkable advantage of the optimal diamond column design is its robustness and long-term superhydrophobicity after repeated de-icing in harsh conditions. The reported work is an important step towards achieving superhydrophobic surface without coating for outdoor composite insulator in practical applications.
