RESUMO

During meiosis I, homologous chromosomes join together to form bivalents. Through trial and error, bivalents achieve stable bipolar orientations (attachments) on the spindle that eventually allow the segregation of homologous chromosomes to opposite poles. Bipolar orientations are stable through tension generated by poleward forces to opposite poles. Unipolar orientations lack tension and are stereotypically not stable. The behavior of sex chromosomes during meiosis I in the male black widow spider Latrodectus mactans (Araneae, Theridiidae) challenges the principles governing such a scenario. We found that male L. mactans has two distinct X chromosomes, X1 and X2. The X chromosomes join together to form a connection that is present in prometaphase I but is lost during metaphase I, before the autosomes disjoin at anaphase I. We found that both X chromosomes form stable unipolar orientations to the same pole that assure their co-segregation at anaphase I. Using micromanipulation, immunofluorescence microscopy, and electron microscopy, we studied this unusual chromosome behavior to explain how it may fit the current dogma of chromosome distribution during cell division.

INTRODUCTION: Envenomation by black widow spiders manifests clinically with signs of neurotoxicity in paediatric patients. OBJECTIVE: Identify typical neurological signs and symptoms in paediatric patients of different ages, and describe treatment and outcomes in a paediatric hospital in northwest Mexico. MATERIAL AND METHODS: We reviewed 70 clinical records of patients hospitalised due to black widow spider bite between 1978 and 2014. We divided the total into 2 groups: Group 1, infants and preschool children; and Group 2, school-age children and adolescents. The demographic variables were age, sex, birthplace, place where envenomation occurred, body part(s) affected, degree of envenomation according to signs and symptoms, treatment, clinical outcome, and statistical differences. RESULTS: Boys accounted for 61.4% of all cases, and infants younger than one year old made up 14.2%. Most patients (70%) were bitten by the spider at home; the anatomical areas most frequently affected were the legs, neck, thorax, and abdomen. The neurological signs and symptoms displayed by Group 1 were irritability, constant crying, sialorrhoea, nausea, tachycardia, arrhythmias, fatigue when walking, agitation, muscle spasms paraesthesia, tetany, seizures, and nystagmus. Signs in Group 2 included localized pain, headache, sialorrhoea, paraesthesia, profuse sweating, anxiety, muscle weakness, muscle spasms, and fine tremor. The predominant autonomic sign in Group 1 was sialorrhoea (P<.0001) and in Group 2, paraesthesia (P<.0001). Patients who received Fab antivenom treatment displayed better outcomes and shorter hospital stays than those who did not. No deaths were reported. CONCLUSIONS: The neurological signs and symptoms caused by black widow spider bite are predominantly autonomic, and identifying them permits early diagnosis and more effective treatment.

Poecilotheria spiders are considered theraphosids of underestimated clinical importance, with bites from these species inducing symptoms such as severe pain and intense muscle cramps. However, there is no specific treatment for the envenomation caused by these species, which, while native to India and Sri Lanka, are widely distributed worldwide. The present study reports the case of a 31-year-old man bitten by a Poecilotheria regalis specimen. The patient's clinical presentation was similar to Latrodectus envenomation, and patient was treated with an L. mactans antivenom. Most of patient's symptoms improved (fasciculations, pain, erythema, and local swelling), except muscle cramps. A toxicological study conducted on mice did not show that L. mactans antivenom has a neutralizing effect on the toxicity of P. regalis. The present report discusses the envenoming process of Poecilotheria species and the possible neutralizing effect exerted by L. mactans antivenom.

Poisoning by widow-spider (genus Latrodectus) bites occurs worldwide. The illness, termed latrodectism, can cause severe and persistent pain and can lead to muscle rigidity, respiratory complications, and cardiac problems. It is a global health challenge especially in developing countries. Equine serum-derived polyclonal anti-sera are commercially available as a medication for patients with latrodectism, but the use of sera imposes potential inherent risks related to its animal origin. The treatment may cause allergic reactions in humans (serum sickness), including anaphylactic shock. Furthermore, equine-derived antivenom is observed to have batch-to-batch variability and poor specificity, as it is always an undefined mix of antibodies. Because latrodectism can be extremely painful but is rarely fatal, the use of antivenom is controversial and only a small fraction of patients is treated. In this work, recombinant human antibodies were selected against alpha-latrotoxin of the European black widow (Latrodectus tredecimguttatus) by phage display from a naïve antibody gene library. Alpha-Latrotoxin (α-LTX) binding scFv were recloned and produced as fully human IgG. A novel alamarBlue assay for venom neutralization was developed and used to select neutralizing IgGs. The human antibodies showed in vitro neutralization efficacy both as single antibodies and antibody combinations. This was also confirmed by electrophysiological measurements of neuronal activity in cell culture. The best neutralizing antibodies showed nanomolar affinities. Antibody MRU44-4-A1 showed outstanding neutralization efficacy and affinity to L. tredecimguttatus α-LTX. Interestingly, only two of the neutralizing antibodies showed cross-neutralization of the venom of the Southern black widow (Latrodectus mactans). This was unexpected, because in the current literature the alpha-latrotoxins are described as highly conserved. The here-engineered antibodies are candidates for future development as potential therapeutics and diagnostic tools, as they for the first time would provide unlimited supply of a chemically completely defined drug of constant quality and efficacy, which is also made without the use of animals.

Widow spiders are widely known for their potent venom toxins that make them among the few spiders of medical concern. The latrotoxins are the most well-studied widow toxins and include both the vertebrate-specific latrotoxins and the insect-specific latroinsectotoxins (LITs). Previous studies have shown that toxins are not limited to expression in the venom glands of adult spiders; however, gaps exist in latrotoxin screening across all life stages for brown widows, Latrodectus geometricus and southern black widows, Latrodectus mactans. In this study, we screened male and female venom gland, cephalothorax, and abdomen tissues, spiderling cephalothorax and abdomen tissues, and eggs of both L. geometricus and L. mactans, for the presence of three latrotoxins: α-latrotoxin (α-LTX), and α- and δ-latroinsectotoxins (α/δ-LITs). Widows were locally collected. Extracted RNA was used to prepare cDNA that was analyzed by PCR for the presence or absence of latrotoxin expression. Results show that expression profiles between the two species are very similar but not identical. Expression of α-LTX was found in all life stages in all tissues examined for both species. For both species, no LIT expression was detected in eggs and variable patterns of α-LIT expression were detected in spiderlings and adults. Notably, δ-LIT could only be detected in females for both species. Our results show that latrotoxin expression profiles differ within and between widow species. Data on their expression distribution provide further insight into the specific latrotoxins that contribute to toxicity profiles for each life stage in each species and their specific role in widow biology.

Arthropods have an open circulatory system with a simple tubular heart, so it has been estimated that the contractile pumping structure of the cardiac muscle will be less efficient than that of vertebrates. Nevertheless, certain arthropods are known to have far superior properties and characteristics than vertebrates, so we investigated the fine structural features of intercalated discs and cardiac junctions of cardiac muscle cells in the black widow spider Latrodectus mactans. Characteristically, the spider cardiac muscle has typical striated features and represents a functional syncytium that supports multiple connections to adjacent cells by intercalated discs. Histologically, the boundary lamina of each sarcolemma connects to the basement membrane to form an elastic sheath, and the extracellular matrix allows the cells to be anchored to other tissues. Since the intercalated disc is also part of sarcolemma, it contains gap junctions for depolarization and desmosomes that keep the fibers together during cardiac muscle contraction. Furthermore, fascia adherens and macula adherens (desmosomes) were also identified as cell junctions in both sarcolemma and intercalated discs. To enable the coordinated heartbeat of the cardiac muscle, the muscle fibers have neuronal innervations by multiple axons from the motor ganglion.

Introducción: El envenenamiento por mordedura de araña viuda negra es poco frecuente en edades pediátricas sin embargo no está exento de complicaciones y si no se trata a tiempo puede causar la muerte. En México se desconoce el número exacto de estos accidentes. Objetivo: Identificar los signos y síntomas característicos, evolución y tratamiento de los niños atendidos por morderua de araña en el Hospital Infantil del Estado de Sonora, con el propósito de establecer el tratamiento oportuno. Material y Métodos: Se revisaron 43 expedientes de niños hospitalizados por mordedura de araña viuda negra entre 1978-2004, las variables consideradas fueron: edad, género, lugar de origen, sitio del accidente, área corporal afectada, tratamiento previo; grado de envenenamiento por signos y síntomas, tratamiento, evolución clínica. Resultados: Predominó el género masculino 69.7%; sin diferencias entre el número de menores de seis años, escolares y adolescentes. El 44 y 37% de los casos se presentaron en el verano y en horas de la tarde, respectivamente. El 74% de los pacientes tuvieron el contacto con el arácnido dentro del domicilio; las áreas anatómicas más afectadas fueron, miembros superiores y tronco; los signos y síntomas más notables en lactantes y preescolares fueron: irritabilidad, llanto constante, sialorrea, agitación, eritema en área de lesión convulsiones. En escolares y adolescentes fueron los signos y sínotmas dolor en sitio de lesión, dolor abdominal, dolor torácico, ansiedad, espasmos musculares y temblor fino. Los pacientes que recibieron tratamiento con faboterapia específica tuvieron mejor evolución y menor tiempo de hospitalización, cabe señalar que no se presentó mortalidad. Conclusiones: La adecuada identificación de los signos y síntomas ocasionados por la morderura de araña en lactantes y preescolares permite el diagnóstico oportuno y ofrece la mejor alternativa de tratamiento.


INTRODUCTION: Poisoning caused by black widow spider bite, is not commonly observed during childhood, it is not exempt from complications, and must be treated in time or the patient can die. In Mexico we do not have accurate data on the exact number of accidents due to this condition. OBJECTIVE: Identify the signs and characteristic symptoms of children treated at the Children's Hospital from the state of Sonora with the aim of starting early treatment. MATERIAL AND METHODS: We reviewed 43 clinical records of hospitalized patients admitted for black widow spider bite between 1978 - 2004. Demographic variables were: age, gender, place of origin, place where accident took place, treatment given prior to hospital admittance, degree of envenomation measured by signs and symptoms, treatment and clinical evolution. RESULTS: 69.7% of cases were male, we did not find differences in the number of patients between infant, pre-school, elementary school children and teenagers. During summer and afternoon hours we registered 44% and 37% cases respectively; 74% patients were poisoned by the spider at home; the anatomical areas most affected were: arms, hands and thorax. Signs and symptoms in infants were: irritability, constanty cry, sialorrea, agitation erythema on wounded areas and seizures. Among elementary school children and teenagers signs included pain in wounded area, abdominal and thoracic, muscle spasms, fine tremor and residual signs. Patients who received fabotherapic treatment had better evolution and less time of hospitalization. No deaths were reported. CONCLUSIONS: The different signs and symptoms between infants, preschool, elementary school children and teenagers allowed us to clearly identify evenommation and differentiate it form confounding diseases. In our study, specific fabotherapy constituted the best treatment choice.

Se reportan 4 casos de picaduras por arañas compatibles con el tipo viuda negra, en pacientes adultos de la provincia de Ciego de Ávila. Los síntomas y signos sobresalientes fueron: parestesias, vómitos, sudación, cólicos abdominales, adinamia e hipotensión. Los pacientes fueron tratados fundamentalmente con hidrocortisona endovenosa y administración de volumen. La evolución fue satisfactoria y rápida, con recuperación al cuarto día.


4 cases of spider bites, which are compatile with the black widow type were reported in adult patients from the province of Ciego de Avila. The most important symptoms and signs were: paresthesia, vomits, sweating, abdominal colics, acratia and hypotension. The evolution was satisfactory and fast. Patients recovered on the fourth day.

A glândula peçonhenta de Latrodectus mactans compõe-se: 1. De um saco formado por uma membrana de tecido conjuntivo peritonial, que se projeta proximalmente no ducto de peçonha. 2. De um manto externo, formado por 40-45 feixes musculares, circundando 3/4 da glândula, helicoidalmente. Os feixes inserem-se nas extremidades apical e basal da membrana sarcoperitonial; sua contração impele a peçonha, através do ducto, para as garras. 3. Do epitélio glandular, composto: a) de células da glândula principal, b) de células da glândula auxiliar, c) de célula da glândula situada à entrada do ducto. As células das glândulas principal e auxiliar, juntas, formam uma unidade glandular de natureza ragiócrina. A secreção das células da glândula auxiliar passa, através das membranas celulares, para o interior das células glandulares principais, deslocando-se, juntamente com a secreção destas, para o pólo apical, a que ambas confluem, formando uma substância viscosa, fortemente condensada em virtude da eliminação de certos líquidos. Após a formação de 8 a 10 destas porções de secreções, os dois tipos de células esgotam-se; não se dá a regeneração ou substituição por outras células. A quantidade de peçonha, uma vez produzida, é armazenada na cavidade da glândula, sendo suficiente para tôda a vida do animal. As células da glândula situada á entrada do ducto, ou glândula lipócrina, produzem uma substãncia lipóide. Esta substância dissolve uma porção de corpúsculos das secreções segregadas pelas células principais e auxiliares. O líquido resultante é inoculado no tecido da prêsa ou do inimigo, por contração do manto muscular. Ao mesmo tempo, novas porções da peçonha armazenada são transportadas no sentido proximal e dissolvidas no líquido lipóide, preparando-se, assim, nova peçonha para outra picada. As formas das células e os pormenores histológicos estão explicados nas figuras que acompanham êste texto. Os músculos da glândula, ao contrário do que afirmam outros autores, verificamos serem de natureza "tetãnica", apresentando uma estriação transversal, típica. Certas zonas, porém, ricas em sarcoplasma, possuem propriedades "tônicas", mantendo-se sempre em certo grau de contratura que provoca a tensão adequada na membrana sarcoperitonial, o que permite um efeito imediato da contração da parte tetânica, no momento da picada.
