RESUMO

The longhorn beetle Graphisurus fasciatus (Degeer) ranges from southeastern Canada to Florida and west to Texas, and has frequently been caught during field trials testing attraction of other cerambycid species to their synthesized pheromones. Collections of headspace volatiles from live beetles revealed that males but not females produce a polyketide compound identified as (4R,6S,7E,9E)-4,6,8-trimethylundeca-7,9-dien-3-one ([4R,6S,7E,9E]-graphisurone). Field trials verified that beetles of both sexes were attracted to the synthesized compound, indicating that it is an aggregation-sex pheromone. This structure represents a new structural motif among cerambycid pheromones, and a new natural product. While this study was in progress, the same compound was isolated from males of the South American cerambycid Eutrypanus dorsalis (Germar), in the same subfamily (Lamiinae) and tribe (Acanthocinini) as G. fasciatus. Field trials in Brazil confirmed that (4R,6S,7E,9E)-graphisurone is also an aggregation-sex pheromone for E. dorsalis, and a possible pheromone for two additional sympatric lamiine species, Hylettus seniculus (Germar) (Acanthocinini) and Oreodera quinquetuberculata (Drapiez) (tribe Acrocinini). These results indicate that graphisurone may be shared among a number of related species, as has been found with many components of cerambycid pheromones.

Longhorned beetles (Cerambycidae) are a diverse family of wood-boring insects, many species of which produce volatile pheromones to attract mates over long distances. The composition and structure of the pheromones remain constant across many cerambycid species, and comparative studies of those groups could, therefore, reveal the chemoreceptors responsible for pheromone detection. Here, we use comparative transcriptomics to identify a candidate pheromone receptor in the large and economically important cerambycid genus Monochamus, males of which produce the aggregation-sex pheromone 2-(undecyloxy)-ethanol ("monochamol"). Antennal transcriptomes of the North American species M. maculosus, M. notatus, and M. scutellatus revealed 60-70 odorant receptors (ORs) in each species, including four lineages of simple orthologs that were highly conserved, highly expressed in both sexes, and upregulated in the flagellomeres where olfactory sensilla are localized. Two of these orthologous lineages, OR29 and OR59, remained highly expressed and conserved when we included a re-annotation of an antennal transcriptome of the Eurasian congener M. alternatus. OR29 is also orthologous to a characterized pheromone receptor in the cerambycid Megacyllene caryae, suggesting it as the most likely candidate for a monochamol receptor and highlighting its potential as a conserved lineage of pheromone receptors within one of the largest families of beetles.

A novel trisubstituted tetrahydropyran was isolated and identified from the sex-specific volatiles produced by males of the cerambycid beetle Macropophora accentifer (Olivier), a serious pest of citrus and other fruit crops in South America. The compound was the major component in the headspace volatiles, and it was synthesized in racemic form. However, in field trials, the racemate was only weakly attractive to beetles of both sexes, suggesting that attraction might be inhibited by the presence of the "unnatural" enantiomer in the racemate. Alternatively, the male-produced volatiles contained a number of minor and trace components, including a compound tentatively identified as a homolog of the major component, as well as a number of unsaturated 8-carbon alcohols and aldehydes. Further work is required to conclusively identify and synthesize these minor components, to determine whether one or more of them are crucial components of the active pheromone blend for this species.

We describe the identification and field testing of 3-methylthiopropan-1-ol (methionol) as a male-produced aggregation-sex pheromone for the cerambycid beetle Knulliana cincta cincta (Drury) (subfamily Cerambycinae, tribe Bothriospilini). The corresponding sulfoxide, 3-methylsulfinylpropan-1-ol, was also produced sex-specifically by males, but its function remains unclear because the measured release rates of this compound from five different types of release devices were very low to undetectable. Unexpectedly, adults of the cerambycine Elaphidion mucronatum (Say) (Elaphidiini), primarily females, also were attracted by methionol, despite males of this species producing an aggregation-sex pheromone of entirely different structure, (2E,6Z,9Z)-2,6,9-pentadecatrienal.

Cerambycid beetles of the subfamily Lamiinae use male-produced aggregation-sex pheromones that are attractive to both sexes. Terpenoid pheromones have been identified from species in the tribes Acanthoderini and Acanthocinini native to North and South America, comprised of (E)-6,10-dimethyl-5,9-undecadien-2-one (geranylacetone), the structurally related 6-methylhept-5-en-2-one (sulcatone), and/or specific enantiomers or nonracemic ratios of enantiomers of the related compounds (E)-6,10-dimethyl-5,9-undecadien-2-ol (fuscumol), its acetate ester, (E)-6,10-dimethyl-5,9-undecadien-2-yl acetate (fuscumol acetate), and 6-methylhept-5-en-2-ol (sulcatol). Here, we present new information about the chemical ecology of six acanthoderine and acanthocinine species native to the eastern USA. The pheromone of Astyleiopus variegatus (Haldeman) previously was identified as a blend of (S)-fuscumol and (S)-fuscumol acetate, and we report here that geranylacetone is a synergistic component. Males of Aegomorphus modestus (Gyllenhal), Lepturges angulatus (LeConte), and Lepturges confluens (Haldeman) were found to produce similar blends composed of the enantiomers of fuscumol acetate and geranylacetone, whereas males of Astylidius parvus (LeConte) and Sternidius alpha (Say) produced both enantiomers of fuscumol together with (R)-fuscumol acetate and geranylacetone. Field experiments with synthesized chemicals revealed that species with similar pheromone composition nevertheless differed in their responses to individual components, and to various blends of components, and in how attraction was influenced by chemicals that were pheromone components of other species. Sulcatone and/or sulcatol antagonized attraction of some species to pheromones of the geranylacetone class, suggesting that there is an adaptive advantage in an ability to detect these heterospecific compounds, such as in avoiding cross attraction to other cerambycid species, as yet unknown, that use pheromones composed of both chemical classes.

Nine strains of five species of entomopathogenic hypocrealean fungi were tested against adults of the Asian longhorned beetle, Anoplophora glabripennis (Motschulsky). These strains have been developed as commercial biopesticide products in the United States, Brazil, South Korea, or the European Union (EU). Metarhizium anisopliae (Metschnikoff) (Hypocreales: Clavicipitaceae) ESALQ E-9 and Metarhizium brunneum (Petch) F52 (formerly M. anisopliae F52) (Hypocreales: Clavicipitaceae) killed 100% of treated beetles with the shortest survival times. Virulence differed among the five strains of Beauveria bassiana (Balsamo) (Hypocreales: Cordycipitaceae) tested, ranging from 0 to 77.3% mortality within 28 d. Two Isaria fumosorosea (Wize, 1904) (Hypocreales: Cordycipitaceae) (formerly Paecilomyces fumosoroseus) strains and the Lecanicillium muscarium (Petch) Zare & Gams (Hypocreales: Cordycipitaceae) strain used in Mycotal were not pathogenic to A. glabripennis adults. Within the entomopathogenic fungi tested, the Metarhizium strains may be the most appropriate for further evaluation.

Adult and juvenile herbivores of the same species can use divergent feeding strategies, and thus may inhabit and consume different parts of the plant. Because the expression of chemical defences often differs between host plant tissues, this variation may result in disparate performance outcomes for adult and juvenile conspecifics that feed on distinct dietary substrates. The goal of this study was to evaluate how host range may differ between adults and juveniles in a generalist herbivore. We addressed the impacts of among- and within-plant defence variation using the wood-feeding Asian longhorned beetle (Anoplophora glabripennis) and three host plants having a range of putative resistance. Impacts of host plants on adult and offspring performance were assessed using a series of controlled bioassays. We evaluated adult-feeding and egg-laying behaviours in choice and no-choice experiments using the different hosts, and subsequent offspring establishment. We then evaluated host plant chemical composition related to nutrition and defence. Different plants had strong impacts on adult performance, but these patterns did not extend to effects on offspring. Females were capable of developing eggs when provided Acer rubrum, but not Populus deltoides or Populus tomentosa. Females that produced eggs by feeding on A. rubrum, however, deposited eggs into all three plant species. Larvae hatched and consumed tissues in all three hosts. The differences between adult and juvenile utilization of Populus spp. were reflected in markedly higher salicinoid phenolic concentrations in bark (>2% dw), while wood had trace quantities. Our results demonstrate that plant resistance mechanisms can differentially act upon adult and juvenile life stages of a polyphagous herbivore when there is differential expression of chemical defences among plant tissue types. Anoplophora glabripennis has been a globally successful invader due in part to its broad host range, and our results suggest a mechanism that permits the beetle to exploit marginally resistant plants. This study has implications for how host range differs between insect feeding stages, which is particularly important for invasive, polyphagous species encountering novel food sources.

Microbial symbionts play pivotal roles in the ecology and physiology of insects feeding in woody plants. Both eukaryotic and bacterial members occur in these systems where they facilitate digestive and nutrient provisioning. The larval gut of the Asian longhorned beetle (Anoplophora glabripennis) is associated with a microbial consortium that fulfills these metabolic roles. While members of the community vary in presence and abundance among individuals from different hosts, A. glabripennis is consistently associated with a fungus in the Fusarium solani species complex (FSSC). We used amplicon sequencing, taxon-specific PCR, culturing, and imaging to determine how bacterial and fungal communities differ between life stages and possible modes of symbiont transfer. The bacterial and fungal communities of adult guts were more diverse than those from larvae and eggs. The communities of larvae and eggs were more similar to those from oviposition sites than from adult female guts. FSSC isolates were not detected in the reproductive tissues of adult females, but were consistently detected on egg surfaces after oviposition and in frass. These results demonstrate that frass can serve as a vehicle of transmission of a subset for the beetle gut microbiota. Vertically transmitted symbionts are often beneficial to their host, warranting subsequent functional studies.

Males of several species in the large cerambycid beetle subfamily Lamiine, native to South and North America, are known to produce aggregation-sex pheromones composed of three related structures: the sesquiterpene catabolic product geranylacetone, its corresponding alcohol (known as fuscumol), and the acetate ester of the alcohol (fuscumol acetate). Here, we show that males of two lamiine species native to the eastern USA, Astylopsis macula (Say) and Leptostylus transversus (Gyllenhal) (both tribe Acanthocinini), produce pheromones composed of the structurally related (S)-6-methylhept-5-en-2-ol ([S]-sulcatol). Males of both species also produced 6-methylhept-5-en-2-one (sulcatone), and males of A. macula produced trace amounts of (R)-fuscumol acetate. Field experiments demonstrated that both species were attracted by (S)-sulcatol. Sulcatone was not attractive as a single component, nor did it influence attraction of A. macula to (S)-sulcatol, while its effect on L. transversus was unclear. Data from the field experiments, and a separate experiment using traps fitted with a timer mechanism that rotated trap jars, revealed that the two species overlap only slightly in both seasonal and daily flight period, thus minimizing interspecific attraction. Racemic fuscumol acetate and fuscumol antagonized attraction of both species to sulcatol. The identification of sulcatol as a cerambycid pheromone extends the known range of pheromone components in the subfamily Lamiinae.

Asian longhorned beetles (ALB; Anoplophora glabripennis), are invasive wood borers susceptible to Metarhizium brunneum. This fungus can be prepared as dried microsclerotia which, after rehydration, produce infective conidia within weeks. Wood samples coated with formulated microsclerotia were attached to trees in the Ohio USA ALB-eradication zone and collected after 4-week periods. Adult ALB exposed to these samples had 100% mortality. In an experiment comparing formulations with or without humectant hydrogel, hydrogel did not significantly increase mortality of exposed ALB. In a dose-response experiment with 5 application rates, ALB survival decreased with increasing application rate and conidial density.

During field screening trials conducted in Brazil in 2015, adults of both sexes of the cerambycid beetles Cotyclytus curvatus (Germar) and Megacyllene acuta (Germar) (subfamily Cerambycinae, tribe Clytini) were significantly attracted to racemic 3-hydroxyhexan-2-one and racemic 2-methylbutan-1-ol, chemicals which previously have been identified as male-produced aggregation-sex pheromones of a number of cerambycid species endemic to other continents. Subsequent analyses of samples of beetle-produced volatiles revealed that males of C. curvatus sex-specifically produce only (R)-3-hydroxyhexan-2-one, whereas males of M. acuta produce the same compound along with lesser amounts of (2S,3S)-2,3-hexanediol and (S)-2-methylbutan-1-ol. Follow-up field trials showed that both sexes of both species were attracted to synthetic reconstructions of their respective pheromones, confirming that males produce aggregation-sex pheromones. The minor pheromone components of M. acuta, (S)-2-methylbutan-1-ol and (2S,3S)-2,3-hexanediol, synergized attraction of that species, but antagonized attraction of C. curvatus to (R)-3-hydroxyhexan-2-one. Beetles of other cerambycine species also were attracted in significant numbers, including Chrysoprasis linearis Bates, Cotyclytus dorsalis (Laporte & Gory), and Megacyllene falsa (Chevrolat). Our results provide further evidence that 3-hydroxyhexan-2-one is a major component of attractant pheromones of numerous cerambycine species world-wide. Our results also highlight our increasing understanding of the crucial role of minor pheromone components in imparting species specificity to cerambycid pheromone blends, as is known to occur in numerous species in other insect families.

We describe the identification, synthesis, and field bioassays of a novel aggregation-sex pheromone produced by males of Susuacanga octoguttata (Germar), a South American cerambycid beetle. Analyses of extracts of headspace volatiles produced by adult beetles revealed a sex-specific compound emitted by males which was identified as (Z)-7-hexadecene by microchemical and spectroscopic analyses. The synthesized pheromone was attractive to beetles of both sexes in field trials. This unsaturated hydrocarbon motif is unprecedented among cerambycid pheromones identified to date. During field bioassays, we serendipitously discovered that adults of S. octoguttata trapped in two Brazilian biomes differed considerably in elytral markings, although males from both populations produced (Z)-7-hexadecene as an aggregation-sex pheromone.

We performed single-sensillum recordings from male and female antennae of the Asian longhorned beetle, Anoplophora glabripennis, that included as stimuli the two components of this species' aggregation-sex pheromone in addition to various general odorants. We compared the aggregation-sex-pheromone-component responses of olfactory sensory neurons (OSNs) to those of OSNs that responded to a variety of plant-related odorants. In the smooth-tipped, tapered, trichoid sensilla on the most distal antennal flagellomeres nos. 10 or 11 of both males and females, we found OSNs with high-amplitude action potentials that were tuned to the aldehyde and alcohol pheromone components and that did not respond to various plant-related volatiles. Because this OSN type responded to both the alcohol and aldehyde components it cannot be considered to be specifically tuned to either component. These large-spiking OSNs were co-compartmentalized in these sensilla with a second, smaller-spiking OSN responding to plant-related volatiles such as geraniol, citronellal, limonene, 1-octanol, nerol and citral. The large-spiking OSNs thus appear to be a type that will be involved in aggregation-sex pheromone pathways targeting a specific glomerulus in the antennal lobe and in generating pheromone-related behavioral responses in A. glabripennis. In other sensilla located in these distal antennal flagellomeres as well as those located more proximally, i.e., mid-length along the antenna on flagellomere nos. 4-7, we found OSNs in blunt-tipped basiconic sensilla that were responsive to other plant-related volatiles, especially the terpenoids, (E,E)-alpha farnesene, (E)-ß-farnesene, ß-caryophyllene, and eugenol. Some of these terpenoids have been implicated in improving attraction to pheromone-baited traps. Some of these same OSNs responded additionally to either of the two sex pheromone components, but because these OSNs also responded to some of the above plant volatiles as shown by cross-adaptation experiments, these OSNs will not be the types that convey sex-pheromone-specific information to the antennal lobe.

An increasing body of evidence suggests that the volatile pheromones of cerambycid beetles are much more diverse in structure than previously hypothesized. Here, we describe the identification, synthesis, and field testing of (2E,6Z,9Z)-2,6,9-pentadecatrienal as a male-produced aggregation-sex pheromone of the cerambycid Elaphidion mucronatum (Say) (subfamily Cerambycinae, tribe Elaphidiini). This novel structure is unlike any previously described cerambycid pheromone, and in field bioassays attracted only this species. Males produced about 9 µg of pheromone per 24 h period, and, in field trials, lures loaded with 10, 25, and 100 mg of synthetic pheromone attracted beetles of both sexes, whereas lures loaded with 1 mg of pheromone or less were not significantly attractive. Other typical cerambycine pheromones such as 3-hydroxy-2-hexanone, syn-2,3-hexanediol, and anti-2,3-hexanediol were not attractive to E. mucronatum, and when combined with (2E,6Z,9Z)-2,6,9-pentadecatrienal, the former two compounds appeared to inhibit attraction. Unexpectedly, adults of the cerambycine Xylotrechus colonus (F.) were attracted in significant numbers to a blend of 3-hydroxyhexan-2-one and (2E,6Z,9Z)-2,6,9-pentadecatrienal, even though there is no evidence that this species produces the latter compound. From timed pheromone trap catches, adults of E. mucronatum were determined to be active from dusk until shortly after midnight.

The compound 1-(1H-pyrrol-2-yl)-1,2-propanedione ("pyrrole") is an important pheromone component of several Asian and South American species of longhorned beetles in the subfamily Cerambycinae. Here, we report the first confirmed identification of this compound as a pheromone component of a cerambycine species native to North America, the rare beetle Dryobius sexnotatus Linsley. Headspace volatiles from males contained (R)-3-hydroxyhexan-2-one and pyrrole (ratio 1:0.13), neither of which were detected in samples from a female. A field bioassay confirmed that adults of both sexes were attracted only to the binary blend of racemic 3-hydroxyhexan-2-one plus pyrrole, and not by either compound alone. Adults of another cerambycine, Xylotrechus colonus (F.), were attracted by 3-hydroxyhexan-2-one, consistent with this compound being the primary component of the pheromone of this species; attraction was not influenced by the presence of pyrrole. This study attests to the effectiveness of pheromone-baited traps in capturing rarely encountered species of cerambycids. It also provides further evidence that pyrrole represents another conserved pheromone motif within the Cerambycinae, now having been found in representatives of five cerambycid tribes from three continents.

Assessing risks of uncertain but potentially damaging events, such as environmental disturbances, disease outbreaks and pest invasions, is a key analytical step that informs subsequent decisions about how to respond to these events. We present a continuous risk measure that can be used to assess and prioritize environmental risks from uncertain data in a geographical domain. The metric is influenced by both the expected magnitude of risk and its uncertainty. We demonstrate the approach by assessing risks of human-mediated spread of Asian longhorned beetle (ALB, Anoplophora glabripennis) in Greater Toronto (Ontario, Canada). Information about the human-mediated spread of ALB through this urban environment to individual geographical locations is uncertain, so each location was characterized by a set of probabilistic rates of spread, derived in this case using a network model. We represented the sets of spread rates for the locations by their cumulative distribution functions (CDFs) and then, using the first-order stochastic dominance rule, found ordered non-dominant subsets of these CDFs, which we then used to define different classes of risk across the geographical domain, from high to low. Because each non-dominant subset was estimated with respect to all elements of the distribution, the uncertainty in the underlying data was factored into the delineation of the risk classes; essentially, fewer non-dominant subsets can be defined in portions of the full set where information is sparse. We then depicted each non-dominant subset as a point cloud, where points represented the CDF values of each subset element at specific sampling intervals. For each subset, we then defined a hypervolume bounded by the outermost convex frontier of that point cloud. This resulted in a collection of hypervolumes for every non-dominant subset that together serve as a continuous measure of risk, which may be more practically useful than averaging metrics or ordinal rank measures. Overall, the approach offers a rigorous depiction of risk in a geographical domain when the underlying estimates of risk for individual locations are represented by sets or distributions of uncertain estimates. Our hypervolume-based approach can be used to compare assessments made with different datasets and assumptions.

Research over the last decade has revealed extensive parsimony among pheromones within the large insect family Cerambycidae, with males of many species producing the same, or very similar aggregation pheromones. Among some species in the subfamily Cerambycinae, interspecific attraction is minimized by temporal segregation, and/or by minor pheromone components that synergize attraction of conspecifics or inhibit attraction of heterospecifics. Less is known about pheromone-based mechanisms of reproductive isolation among species in the largest subfamily, the Lamiinae. Here, we present evidence that the pheromone systems of two sympatric lamiine species consist of synergistic blends of enantiomers of (E)-6,10-dimethyl-5,9-undecadien-2-ol (fuscumol) and the structurally related (E)-6,10-dimethyl-5,9-undecadien-2-yl acetate (fuscumol acetate), as a mechanism by which species-specific blends of pheromone components can minimize interspecific attraction. Male Astylidius parvus (LeConte) were found to produce (R)- and (S)-fuscumol + (R)-fuscumol acetate + geranylacetone, whereas males of Lepturges angulatus (LeConte) produced (R)- and (S)-fuscumol acetate + geranylacetone. Field experiments confirmed that adult beetles were attracted only by their species-specific blend of the enantiomers of fuscumol and fuscumol acetate, respectively, and not to the individual enantiomers. Because other lamiine species are known to produce single enantiomers or blends of enantiomers of fuscumol and/or fuscumol acetate, synergism between enantiomers, or inhibition by enantiomers, may be a widespread mechanism for forming species-specific pheromone blends in this subfamily.

Male Dorysthenes granulosus (Thomson, 1860) (Coleoptera: Cerambycidae: Prioninae) were caught in traps baited with racemic 3,5-dimethyldodecanoic acid (prionic acid) during field screening trials in China that tested known cerambycid pheromones. This species is an important pest of sugarcane (Saccharum officinarum L.). In follow-up dose-response trials, plastic sachets loaded with 1 or 0.1 mg of prionic acid were equally attractive to male beetles, whereas lower doses were no better than controls. Two commercial prionic acid lures also were attractive, suggesting that traps baited with prionic acid can be rapidly incorporated into integrated pest management programs targeting this major pest. It is likely that this compound is a major component of the female-produced sex pheromone of D. granulosus because this species is in the same subfamily as Prionus californicus Motschulsky, 1845, the species from which prionic acid was originally identified.

Research during the 1980s showed that male Xylotrechus pyrrhoderus Bates (Coleoptera: Cerambycidae), a vineyard pest, produce (2S,3S)-2,3-octanediol and (S)-2-hydroxyoctan-3-one as possible pheromone components, but to our knowledge, field tests were not carried out. We confirmed that at least female X. pyrrhoderus were attracted by a 1:1 blend of these two compounds in field trials in Japan. Furthermore, more than 200 males and females of the congener Xylotrechus rufilius Bates were attracted by racemic 2-hydroxyoctan-3-one, and inhibited by syn-2,3-octanediol. Adult X. rufilius recently were intercepted in a shipment from China entering Baltimore, Maryland, raising concerns that this polyphagous species could establish in North America. Our results suggest that traps baited with 2-hydroxyoctan-3-one would be a valuable tool to assess whether X. rufilius has indeed become established, and to monitor for future introductions of X. rufilius.

BACKGROUND: The Asian longhorned beetle (ALB), Anoplophora glabripennis, is a serious wood borer of hardwood trees. Populus deltoides 'Shalinyang' (PdS) is attractive to ALB adults for oviposition but highly resistant to their offspring. Investigation of the chemicals regulating ALB oviposition is scarce in previous studies until now. To determine which chemicals emitted by PdS were attractive and induced oviposition behavior by ALB on non-host poplar tree species, we first: collected and identified the bio-active volatiles produced by PdS using coupled gas chromatography-mass spectrometry (GC-MS) and coupled gas chromatography-electroantennographic detector (GC-EAD); then evaluated which chemicals were attractive in a Y-tube olfactometer bioassay; and finally screened key compounds affecting ALB oviposition using a 'chemical-stimulated oviposition on non-host tree' bioassay. RESULTS: (E)-2-Hexenal, hexyl acetate, (Z)-3-hexenol acetate, 1-hexanol, (Z)-3-hexenol, ß-caryophyllene, and salicylaldehyde emitted from PdS were attractive to ALB. When (E)-2-hexenal, 1-hexanol, (Z)-3-hexenol acetate, and (Z)-3-hexenol were applied to the non-host tree Populus tomentosa, oviposition by ALB females was significantly increased. Furthermore, the mean number of oviposition pits increased as the (Z)-3-hexenol concentrations increased. Further tests on synergy between pairs of chemicals showed that (Z)-3-hexenol stimulated production of the most oviposition pits, but that the percentage of effective oviposition pits (those containing an egg and larva and not empty) decreased. CONCLUSION: (Z)-3-Hexenol is the main chemical component inducing ALB oviposition. These results increase understanding about the oviposition behavior of ALB and could help improve management strategies that regulate ALB behavior by planting mixed-species forests resistant to ALB. © 2023 Society of Chemical Industry.
