The present study was conducted to evaluate the in vitro antioxidant activity of fermented camel milk with Lactococcus lactis subsp. cremoris (FCM-LLC) alone, Rosmarinus officinalis extract (R) alone and their combination and to investigate their hepatopreventive effects against CCl4 liver damage in mice. The antioxidant activity in vitro of FMC-LLC supplemented with R exhibited the highest free radical scavenging and ferric reducing power activities. The results showed that the pretreatment with a combination of FMC-LLC and R significantly alleviated the increased levels of hepatic markers and the elevated lipid levels induced by CCl4 in mice. Meanwhile, the enzymatic antioxidants activities (superoxide dismutase, glutathione peroxidase, and catalase) and GSH level in liver significantly were increased while the malondialdehyde level was significantly improved by pretreatment with FMLLC plus R. These data suggest that FCM-LLC in combination with R. officinalis extract possesses better antioxidant and hepatoprotective activity than FMC-LLC alone.

BACKGROUND: Mentha piperita L. is a flowering plant belonging to the Lamiaceae family. Mentha plants constitute one of the main valuable sources of essential oil used in foods and for medicinal purposes. METHODS: The present study aimed to investigate the composition and in vitro antioxidant activity of Mentha piperita leaf essential oil (MpEO). A single dose of CCl4 was used to induce oxidative stress in rats, which was demonstrated by a significant rise of serum enzyme markers. MpEO was administrated for 7 consecutive days (5, 15, 40 mg/kg body weight) to Wistar rats prior to CCl4 treatment and the effects on serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), and γ -glutamyl transpeptidase (γ-GT) levels, as well as the liver and kidney superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activity and thiobarbituric acid reactive substances (TBARS) levels were evaluated. In addition, histopathological examinations of livers and kidneys was performed. RESULTS: The in vitro antioxidant activity of MpEO was lower than that of silymarin. Pretreatment of animals with MpEO at a dose of 5 mg/kg did not have a significant effect on ALT, AST, ALP, LDH, γGT, urea or creatinine levels in CCl4-induced stress. Whereas pretreatment with MpEO at doses of 15 and 40 mg/kg prior to CCl4, significantly reduced stress parameters (ALT, AST, ALP, LDH, γGT, urea and creatinine) compared to the CCl4-only group. Moreover, a significant reduction in hepatic and kidney lipid peroxidation (TBARS) and an increase in antioxidant enzymes SOD, CAT and GPx was also observed after treatment with MpEO (40 mg/kg) compared to CCl4-treated rats. Furthermore, pretreatment with MpEO at 40 mg/kg can also markedly ameliorate the histopathological hepatic and kidney lesions induced by administration of CCl4. CONCLUSIONS: We could demonstrate with this study that MpEO protects liver and kidney from CCl4-induced oxidative stress and thus substantiate the beneficial effects attributed traditionally to this plant.

Proline plays an important role in plant response to various environmental stresses. However, its involvement in mitigation of heavy metal stress in plants remains elusive. In this study, we examined the effectiveness of exogenous proline (10 and 20 mM) in alleviating cadmium induced inhibitory effects in young olive plants (Olea europaea L. cv. Chemlali) exposed to two Cd levels (10 and 30 mg CdCl2 kg(-1) soil). The Cd treatment induced substantial accumulation of Cd in both root and leaf tissues and a decrease in gas exchange, photosynthetic pigments contents, uptake of essential elements (Ca, Mg and K) and plant biomass. Furthermore, an elevation of antioxidant enzymes activities (superoxide dismutase, catalase, glutathione peroxydase) and proline content in association with relatively high amounts of hydrogen peroxide, thiobarbituric acid reactive substances and electrolyte leakage were observed. Interestingly, the application of exogenous proline alleviated the oxidative damage induced by Cd accumulation. In fact, Cd-stressed olive plants treated with proline showed an increase of antioxidant enzymes activities, photosynthetic activity, nutritional status, plant growth and oil content of olive fruit. Generally, it seems that proline supplementation alleviated the deleterious effects of young olive plants exposed to Cd stress.

Lavandula aspic L. is a strongly aromatic shrub plant of the Lamiaceae family and traditionally used in herbal medicine for the treatment of several skin disorders, including wounds, burns, and ulcers. The present study aimed to investigate the composition and in vitro antioxidant activity of lavender essential oil. In addition, it aimed to evaluate the excision wound healing activity and antioxidant property of a Lavandula aspic L. essential oil formulated in ointment using a rat model. The rats were divided into five groups of six animals each. The test groups were topically treated with the vehicle, lavender ointment (4%) and a reference drug, while the control group was left untreated. Wound healing efficiency was determined by monitoring morphological and biochemical parameters and skin histological analysis. Wound contraction and protein synthesis were also determined. Antioxidant activity was assessed by the determination of MDA rates and antioxidant enzymes (GPx, catalase and superoxide dismutase). The treatment with lavender ointment was noted to significantly enhance wound contraction rate (98%) and protein synthesis. Overall, the results provided strong support for the effective wound healing activity of lavender ointment, making it a promising candidate for future application as a therapeutic agent in tissue repairing processes associated with skin injuries.

AIMS: The present study was undertaken to investigate the protective effect of lipid extract of Dunaliella sp. (LE) rich in polyunsaturated fatty acids (PUFA), against oxidative stress induced by nickel in experimental rats. METHODS: Our investigation evaluated the antioxidant activity of LE using both DPPH and NBT assays. Twenty female albino Wistar rats, randomly allocated into four experimental groups, namely (C): control, (Nit): nickel-treated rats with 5 mg/kg/d of NiCl2 during 30 days, (LEa): lipid extract-administered rats with 5 mg/kg BW/d during 30 days and (Nit + LEa): rats treated with Ni and LE-administered during 30 days. RESULTS: The in vitro antioxidant activity demonstrated that LE presents an important antioxidant potential. In vivo, the (Nit + LEa) cotreatment decreased the level of malondialdehyde and restored the antioxidant enzyme activities (superoxide dismutase, catalase and glutathione peroxidase) in livers and kidneys in comparison with those treated with Ni only. LE administration to rats treated with Ni also ameliorated biochemical and histological parameters as compared to only Ni-treated group. LE of Dunaliella sp., rich in polyunsaturated fatty acids showed a significant hepato- and reno-protective effect against metal-induced toxicity. CONCLUSION: LE of Dunaliella sp., rich in PUFA has been proven to be effective in protection against Ni-induced toxicity.

CONTEXT: Consumption of Sarpa salpa Linn. (Sparidae) in certain periods of the year is inadvisable because it can cause central nervous system disorders resulting in sea food poisoning. AIMS: The present study assesses the cytotoxic effects of compounds, not-yet identified, present in the organ extracts of S. salpa, collected in autumn, the period corresponding to the peak in human health problems. MATERIALS AND METHODS: The toxicity was assessed by mouse bioassay of aqueous extract of the fish organs. Wistar rats received daily extracts of different organs of S. salpa by gastric gavage for 7 d (0.3 mL of extract/100 g body weight BW). The dose of tissue extracts of viscera, liver, brain, and flesh of S. salpa administered to rats was as follows: 172, 313, 2050, and 2660 mg/kg BW, respectively. No deaths occurred during the period of treatment. RESULTS: The lethal dose (LD50) determined for the crude ciguatoxin (neurotoxins) extracts of viscera, liver, brain, and flesh of S. salpa was as follows: 1.2, 2.2, 14.4, and 18.6 g/kg mouse, respectively. Changes in locomotor activity during the first 2 h and failure in breathing and no evident signs of gastrointestinal problems were recorded. We observed (1) induction of oxidative stress, indicated by an increase in lipid peroxidation (TBARS) in groups that received extracts of liver (+425%) or viscera (+433%), and a significant decrease in antioxidant enzyme activities (SOD, CAT, and GPx) in cerebral cortex tissue by 13%, 25%, and 25% (LT: animals receiving liver extracts) and by 16%, 26%, and 27% (VT: animals receiving viscera extracts), respectively. In contrast, the administration of extracts of flesh and brain induced an increase in antioxidant enzyme activities (SOD, CAT, and GPx) in cerebral cortex tissue by 26%, 23%, and 44% (FT: flesh extract) and 28%, 24%, and 46% (BT: brain extract), respectively; (2) a significant decrease for acetylcholinesterase (AChE) activity in cerebral cortex was recorded in FT, BT, LT, and VT by 27, 34, 58, and 78%, respectively. Moreover, a significant decrease of AChE activity in plasma was recorded in FT, BT, LT, and VT by 16, 21, 38, and 48%, respectively; (3) the histological findings confirmed the biochemical results. CONCLUSIONS: Liver and especially the visceral part of S. salpa presented toxicity, which clearly indicates the danger of using this fish as food.

CONTEXT: Kombucha (KT) is claimed to have various beneficial effects on human health, but there is very little scientific evidence available in the literature. OBJECTIVE: The present study investigates the effects of Camellia sinensis (GT) Linn. (Theaceae) and KT, two natural drinks, on cholesterol and antioxidant status using a hypercholesterolemia rat model. MATERIALS AND METHODS: The present study compared the free-radical scavenging abilities and polyphenol levels of GT and KT. Wistar rats fed cholesterol-rich diets were given KT or GT (5 mL/kg body weight per day, po) for 16 weeks, then fasted overnight and sacrificed. The plasma lipid levels, thiobarbituric acid reactive substances (TBARS) and aspartate aminotransferase (AST), alanine aminotransferase (ALT), and γ-glutamyl transpeptidase (GGT) serum levels, antioxidant activities of superoxide dismutase (SOD) and catalase (CAT), and creatinine and urea rats were examined. RESULTS: KT had a phenolic compound of 955 ± 0.75 mg GAE/g) followed, by GT (788.92 ± 0.02 mg GAE/g). The free radical scavenging activity of KT was higher than GT. Compared with GT, KT induced lowered serum levels of TC, TG, VLDL-C, and LDL-C by 26, 27, 28, and 36%, respectively, and increased the serum level of high-density lipoprotein cholesterol (HDL-C). KT induced a 55% decrease of TBARS level in liver and 44% in kidney, compared with those of rats fed a cholesterol-rich diet alone. Moreover, CAT and SOD activities were reduced by 29 and 33%, respectively, in liver and 31 and 35%, respectively, in kidney, after oral administration of KT, compared with those of HCD-fed rats. CONCLUSION: The findings revealed that KT administration induced attractive curative effects on hypercholesterolemic, particularly in terms of liver-kidney functions in rats. Its effect on humans needs to be studied further.

AIMS: The present study was aimed to assess the cytotoxic effects of not-yet identified compounds present in organ extracts of Sarpa salpa, collected in autumn, the period with a peak in health problems. METHODS: The toxicity was assessed by mouse bioassay of extract of the fish's organs. Wistar rats received daily extracts of different organs of S. salpa by gastric gavage for 7 d (0.3 ml of extract/100 g body weight, BW). The dose of tissue extracts of viscera, liver, brain and flesh of S. salpa administered to rats were as follows: 17.2, 31.3, 205, 266 mg/100g BW, respectively. No deaths occurred during the period of treatment. RESULTS: The lethal dose (LD50%) determined for the crude ciguatoxin (neurotoxins) extracts of viscera, liver, brain and flesh of S. salpa were as follows: 1.2, 2.2, 14.4, 18.6 g/kg mouse, respectively. Changes in locomotor activity during the first 2 h and failure breathing and no evident signs of gastrointestinal problems were recorded. We observed: (1) Induction of oxidative stress, indicated by an increase in lipid peroxidation (TBARS) in groups that received extracts of liver (+490%) or viscera (+592%). Accompanied by a significant decrease in antioxidant enzyme activities (SOD, CAT, GPx) in liver tissue by 15%, 17%, 18% (LT: animals receiving liver extracts) and by 19%, 22%, 22% (VT: animals receiving viscera extracts), respectively. In contrast the administration of extracts of flesh and brain induced an increase in antioxidant enzyme activities (SOD, CAT, GPx) in liver tissue by 15%, 19%, 15% (FT: flesh extract) and 18%, 55%, 55% (BT: brain extract), respectively; (2) A significant increase in total metallothionein levels in liver tissue was recorded in (FT), (BT), (LT) and (VT) by 55%, 88%, 255% and 277%, respectively, (3) The histological findings confirmed the biochemical results. CONCLUSIONS: Liver and especially visceral part of S. salpa presented toxicity, which clearly indicates the danger of using this fish as food.

Basil (Ocimum basilicum L.) is distributed worldwide and used in the food, pharmaceutical, and cosmetic industries. Most applications are for the herb basil, recently the basil seeds have also been used commercially; however, little is known about the nutritional and functional properties of the seeds. The present study aimed to investigate a possible protective effect of the methanol extract of O. basilicum seeds (MEOB), based on its phytochemical content, against kidney toxicity induced by CCl4 in adult rats. A single dose of CCl4 was used to induce oxidative stress in rats, which was demonstrated by a significant rise of serum enzyme markers. MEOB was administrated for 15 consecutive days (200 mg/kg body weight) to Wistar rats before CCl4 treatment and the effects on serum urea, creatinine, and uric acid, as well as the kidney superoxide dismutase, catalase, glutathione peroxidase, and glutathione activity and thiobarbituric acid reactive substances and protein carbonyl (PCO) levels were evaluated. In addition, histopathological examinations of kidneys were performed. In the positive control group, CCl4 induced an increase in serum biochemical parameters and triggered oxidative stress in the kidney. MEOB (200 mg/kg BW) resulted in significant reduction of CCl4-elevated levels of kidney markers, urea and creatinine, and a significant increase of uric acid compared with the CCl4-only group. In addition, MEOB pretreatment resulted in a significant reduction in lipid peroxidation and PCO levels in renal tissue compared with CCl4-exposed group. MEOB definitely could prevent the development of pathological changes in the kidneys. Overall, we conclude that MEOB is effective in protecting renal function from CCl4 toxicity.

The aim of this work was to investigate for Sarpa salpa the seasonal trend in the food sources, heavy metals bioaccumulation and the oxidative stress in the organs. In addition, the toxicity was assessed by mouse bioassay of extract of the fish's organs collected in autumn, the peak of occurrence of hallucinatory syndrome. The toxicity was further studied for compounds present in epiphyte collected from the sea at the end of spring and in summer that are digested by the S. salpa in these seasons. We observed a higher lipid peroxydation in different tissues of S. salpa compared to the control fish Diplodus annularis. Furthermore, heavy metals accumulation in organs of these fish showed a significant variation between the two species (P < 0.05). The lethal dose (LD50%) determined for crude ciguatoxin (neurotoxins) extracts of viscera, liver, brain and muscle of S. salpa were as follows: 1.217, 2.195, 14.395, 18.645 g/kg mouse, respectively. We noticed a significant correlation (P < 0.05) between the total amount of toxic dinoflagellates and the level of TBARS in the liver, the brain and the muscle, this for all seasons and all sizes. Moreover, the cytotoxic effect observed for epiphytes extract confirms the transfer of toxins originating from toxic dinoflagellates, which live as epiphytes on P. oceanica leaves, to the fish by grazing. Our work indicates that, toxic phytoplanktons and heavy metals accumulation are responsible for the increase of oxidative stress in the organs of S. salpa. Hence, the edible part of S. salpa, especially the viscera and liver, can cause a threat to human health, and consumption should, for this reason, be dissuaded.

The present study aimed to investigate the preventive effects of methanol fraction from Cassia angustifolia leaf extract (MECA), associated with its phytochemical content, on CCl4-induced hepatic toxicity in adult rats. In the controls, CCl4 induced an increase of serum biochemical parameters and triggered oxidative stress in the liver. MECA caused significant reductions in CCl4-elevated levels of hepatic markers, total cholesterol, triglycerides, and low-density lipoprotein and increased the level of high-density lipoprotein compared to the CCl4 group. Moreover, pretreatment with the MECA produced significant reductions in lipid peroxidation (thiobarbituric acid reactive substances) and protein carbonyl level in liver tissues as compared with the untreated group. The formation of pathological hepatic lesions was strongly prevented by MECA. Overall, this study suggests that administration of MECA has a high potential to quench free radicals and alleviate CCl4-induced hepatotoxicity in rats.

The hyperlipidemia is a serious health problem that increases the risk of many complications including cardiovascular disease. This study aims to evaluate the possible antihyperlipidemic effects of the feather protein hydrolysate (FPH) in a mice fed with a high-fat diet (HFD)-fed mice during 5 weeks. The FPH administration improved dose-dependent lipid profile, as well as the liver and renal dysfunction indices in hyperlipidemic mice. The FPH also restored the antioxidant status in liver, kidney, and heart by lowering the lipid peroxidation and enhancing the antioxidant enzymes (catalase (CAT), glutathione peroxidase (GPx) and superoxide dismutase [SOD]). Moreover, the histological studies proved that FPH administration prevents hepatic steatosis, glomerular hyperfiltration risk, and cardiac muscle hypertrophy. Accordingly, the FPH is a promising novel medicinal ingredient for possible use in the hyperlipidemic treatment and related complications.

The inner bark of cinnamon (Cinnamomum verum) is widely used as a spice. Cinnamon plants are also a valuable source of essential oil used for medicinal purposes. The present study aimed to investigate the composition and in vitro antioxidant activity of essential oil of C. verum bark (CvEO) and its protective effects in vivo on CCl4-induced hepatic and renal toxicity in rats. Groups of animals were pretreated for 7 days with CvEO (70 or 100 mg/kg body weight) or received no treatment and on day 7 a single dose of CCl4 was used to induce oxidative stress. Twenty-four hours after CCl4 administration, the animals were euthanized. In the untreated group, CCl4 induced an increase in serum biochemical parameters and triggered oxidative stress in both liver and kidneys. CvEO (100 mg/kg) caused significant reductions in CCl4-elevated levels of alanine transaminase, aspartate transaminase, alkaline phosphatase, γ-glutamyl transferase, lactate dehydrogenase, total cholesterol, triglycerides, low-density lipoprotein, urea, and creatinine and increased the level of high-density lipoprotein compared with the untreated group. Moreover, pretreatment with CvEO at doses of 70 and 100 mg/kg before administration of CCl4 produced significant reductions in thiobarbituric acid reactive substances and protein carbonyl levels in liver and kidney tissues compared with the untreated group. The formation of pathological hepatic and kidney lesions induced by the administration of CCl4 was strongly prevented by CvEO at a dose of 100 mg/kg. Overall, this study suggests that administration of CvEO has high potential to quench free radicals and alleviate CCl4-induced hepatorenal toxicity in rats.

The present study was designed to examine the preventive effects of camel milk (CM) against the toxic effects of acute exposure to carbon tetrachloride (CCl4) on the heart tissue of mice. Administration of a single dose of CCl4 caused cardio toxicity as monitored by an increase in lipid peroxidation (thiobarbituric acid reactive substances), protein carbonyl level and antioxidant markers (superoxide dismutase, catalase, glutathione peroxidase, glutathione and vitamin C) in the heart tissue. Moreover, CCl4 caused a distinguished rise of plasma aspartate aminotransferase, lactate dehydrogenase, troponin I, and creatine kinase activities. Furthermore, CM ameliorated biochemical and histological parameters as compared to CCl4-treated group. Overall, this study indicates that CM is efficient in inhibiting oxidative stress induced by CCl4 and suggests that the administration of this milk may be helpful in the prevention of cardio-toxicity complications.

The present study was aimed to assess the cytotoxic effects of not-yet identified compounds present in organ extracts of Sarpa salpa, collected in autumn, the period with a peak in health problems. In addition, we studied the cytotoxicity of extracts of epiphytes found in the stomach content of S. salpa collected in summer and of epiphytes collected from the sea in the Sfax area at the end of spring. We tested these fractions in two human hepatic cell lines: HepG2 and WRL68. We observed a significant loss of viable cells when HepG2 cells were exposed for 72 h to acetone extracts of livers of S. salpa at a concentration of 2.5 mg/ml protein. Proteins extracted from brain or muscle did not significantly induce cell death at the studied concentrations (≤10 mg/ml). There was a significant loss of viable cells when treated with liver extract of S. salpa dissolved in DMSO. Extracts of epiphytes collected in late spring showed a cytotoxic effect in a concentration-dependent manner. Moreover, we observed a significantly decreased cell viability of HepG2 at a dilution (1/40) of epiphyte extracts from stomach contents of two fish we had collected. The cytotoxic effect of the observed epiphyte extracts confirms the transfer of toxins originating from toxic dinoflagellates which live in epiphyte on the Posidonia oceanica leaves to fish organs by grazing. Hence, the liver of this fish can cause a threat to human health and consumption should for this reason be dissuaded.
