RESUMEN

Blowhole swabs are a simple and non-invasive method for collecting samples from cetaceans and can be used for screening large numbers of animals in the field. This study reports a real-time PCR assay for the detection of Brucella spp. using blowhole swab samples from bottlenose dolphins Tursiops truncatus stranded in the coastal region of Virginia, South Carolina and northern Florida, USA, between 2013 and 2015. We used real-time PCR results on lung samples from the same dolphins in order to estimate the relative sensitivity and specificity of real-time PCR of blowhole swabs. Brucella DNA was detected in lung tissue of 22% (18/81) and in blowhole swabs of 21% (17/81) of the sampled dolphins. The relative sensitivity and specificity of real-time PCR on blowhole swabs as compared to the real-time PCR on lung samples was 94% (17/18) and 100% (63/63), respectively. These results indicate that real-time PCR on blowhole swabs may be used as a non-invasive test for rapid detection of Brucella spp. in the respiratory tract of dolphins. To our knowledge, this is the first report on the use of blowhole swabs for detection of bacterial pathogens by real-time PCR in bottlenose dolphins.

RESUMEN

An adult male pygmy sperm whale ( Kogia breviceps) stranded alive at a beach in Florida, US, in 2016. Main postmortem examination findings included bilateral multifocal variably sized renal cysts, focal renal cystadenoma, and mild dilation of the renal pelvises. The role of these renal lesions in the stranding of this whale is unknown.

Knowledge of an animal's home range is a crucial component in making informed management decisions. However, many home range studies are limited by study area size, and therefore may underestimate the size of the home range. In many cases, individuals have been shown to travel outside of the study area and utilize a larger area than estimated by the study design. In this study, data collected by multiple research groups studying bottlenose dolphins on the east coast of Florida were combined to determine how home range estimates increased with increasing study area size. Home range analyses utilized photo-identification data collected from 6 study areas throughout the St Johns River (SJR; Jacksonville, FL, USA) and adjacent waterways, extending a total of 253 km to the southern end of Mosquito Lagoon in the Indian River Lagoon Estuarine System. Univariate kernel density estimates (KDEs) were computed for individuals with 10 or more sightings (n = 20). Kernels were calculated for the primary study area (SJR) first, then additional kernels were calculated by combining the SJR and the next adjacent waterway; this continued in an additive fashion until all study areas were included. The 95% and 50% KDEs calculated for the SJR alone ranged from 21 to 35 km and 4 to 19 km, respectively. The 95% and 50% KDEs calculated for all combined study areas ranged from 116 to 217 km and 9 to 70 km, respectively. This study illustrates the degree to which home range may be underestimated by the use of limited study areas and demonstrates the benefits of conducting collaborative science.

Papillomaviruses (PVs) are small DNA viruses that have been associated with increased epithelial proliferation. Over one hundred PV types have been identified in humans; however, only three have been identified in bottlenose dolphins (Tursiops truncatus) to date. Using rolling circle amplification and degenerate PCR, we identified four novel PV genomes of bottlenose dolphins. TtPV4, TtPV5 and TtPV6 were identified in genital lesions while TtPV7 was identified in normal genital mucosa. Bayesian analysis of the full-length L1 genes found that TtPV4 and TtPV7 group within the Upsilonpapillomavirus genus while TtPV5 and TtPV6 group with Omikronpapillomavirus. However, analysis of the E1 gene did not distinguish these genera, implying that these genes may not share a common history, consistent with recombination. Recombination analyses identified several probable events. Signals of positive selection were found mostly in the E1 and E2 genes. Recombination and diversifying selection pressures constitute important driving forces of cetacean PV evolution.

Antibodies in the sera of patients with lacaziosis recognized an approximately 193-kDa antigen and other Lacazia loboi antigens. Paracoccidioides brasiliensis gp43 antigen was detected by all evaluated sera, but they failed to detect a protein with the same molecular mass in L. loboi extracts. This study is the first to examine the humoral response to L. loboi antigens by using multiple host sera.
