In this study, we conducted a thorough investigation into the mechanisms by which miR-29 influences lipid metabolism. Thirty-two cows were selected and categorized into distinct groups based on their liver triglyceride (TG) content: healthy, mild fatty liver, and moderate fatty liver groups. Dairy cows with moderate fatty liver showed higher levels of hepatic lipid accumulation, MDA content and serum AST, ALT and ALP contents and lower hepatic catalase CAT and SOD activities. Subsequently, hepatocytes isolated from healthy calves were exposed to sodium oleate (SO) in the presence or absence of pre-incubation with miR-29 inhibitor or inhibitor NC. Pre-transfection with miR-29 inhibitor resulted in reduced hepatocyte lipid accumulation and MDA levels, as well as decreased levels of AST, ALT, and ALP in the supernatant. In the miR-29 inhibitor + SO group, there was an increase in the expression of SREBP-1, FAS, SCD1, and Sirt1. Meanwhile, the expression of PPARα, CPT1, CPT2, PGC-1α, NRF-1, UCP2, and miR-29 were observed to be decreased. In comparison to the miR-29 inhibitor + SO group, some of the measured indicators showed partial reversal in the miR-29 inhibitor + siSirt1 + SO group. Collectively, these findings provide evidence that miR-29 may play a crucial role in the pathogenesis of fatty liver in dairy cows.

Slippery liquid-infused porous surfaces (SLIPSs) have attracted wide interest with regard to their excellent liquid repellency properties and broad applications in various fields associated with anti-adhesion. However, the preparation processes depending on the chemical properties of the substrate and the poor stability of the lubricant layer hinder the practical applications. In this work, a facile method to fabricate SLIPSs based on the mussel-inspired polydopamine (PDA)-mediated nanosilica structures is demonstrated. A variety of substrates can be decorated with SLIPSs by successive treatment of PDA-assisted sol-gel process, fluorination, and lubricant filling. The robust uniform and nanotextured silica coating, mediated by the pre-adhered PDA layer, shows enhanced lubricant-locking ability even when subjected to increased evaporation and high shear from flowing water or spinning compared with hierarchical silica rough structures. The obtained SLIPSs exhibit high transparency and excellent resistance against adhesion of liquid/solid contaminants and biofoulings through this pre-adhesion of PDA strategy. The well-defined nanosilica coating of high decoration covering micron-scaled pore walls enables improved durability of the slippery surfaces for antifouling of the porous membrane under pressure-driven filtration and this may be employed as a potential candidate for fouling resistance of porous materials.

Bordetella bronchiseptica is a significant contributor to respiratory disease in pigs, leading to substantial economic losses in the swine industry worldwide. We isolated 52 B. bronchiseptica strains from 542 samples collected from pigs with atrophic rhinitis and bronchopneumonia in central China. Multi-locus sequence typing identified two prevalent sequence types: ST6 (69.23%) and ST7 (30.77%). PCR-based detection of seven virulence genes (fhaB, prn, cyaA, dnt, bteA, fla, and bfrZ) revealed that six of these genes were present in over 90% of the isolates, with bfrZ being the exception at 59.62%. Antimicrobial susceptibility testing, performed using the K-B method, demonstrated high sensitivity to enrofloxacin, polymyxin, and doxycycline but a notable resistance to tylosin, trimethoprim, tobramycin, ciprofloxacin, and amikacin. Remarkably, 86.54% of the isolates exhibited a multidrug-resistant phenotype. Notably, we successfully screened a strain of B. bronchiseptica with a heteroresistance phenotype to gentamicin using population analysis profiling, which is a rare case. Biofilm-formation assays indicated that 96.15% of the isolates possessed biofilm-forming capabilities. These findings provide crucial insights into the prevalence of B. bronchiseptica in central China, facilitating the development of effective preventive measures to safeguard both animal and human health.

An optical sensor membrane was prepared by electrostatic self-assembled technique for online detection of cadmium ion (II) (Cd(II)). The optical indicator 5,10,15,20-tetrakis(4-N-methylpyridyl) porphyrin p-toluenesulfonate (TMPyP) was adsorbed on a hydrolyzed polyacrylonitrile (PAN) membrane by electrostatic attraction and further immobilized through layer-by-layer deposition of poly(allylamine hydrochloride) (PAH) and poly(sodium 4-styrenesulfonate) (PSS) on the membrane surface. The electrostatic self-assembly of polyelectrolytes on the membrane is influenced by pH and salt concentration of polyelectrolytes. The optical sensor membrane shows distinct color and spectral response to Cd(II) under static and flow-through conditions based on the coordination of TMPyP with Cd(II). A faster detection of Cd(II) is achieved at higher feed concentration of Cd(II) or appropriate lower immobilization capacity of TMPyP on the membrane. The flow-through detection is also influenced by the flow rate; higher flow rate led to faster response to Cd(II) during filtration. Compared with the static process, the flow-through conditions are more conducive to faster analysis of ppb level concentration of Cd(II) (10-3 mg L-1) due to a promoted mass transfer and filtration enrichment. Hence, the development of the optical sensor membrane in this study demonstrated the prospect to make membranes multifunctional with advantages for online chromatic warning in addition to adsorption/rejection of heavy metal ions in the solutions that are treated.
