RESUMEN

Selenium (Se) is an essential trace element in organism. Se deficiency can cause many diseases, including vascular disease. Studies have shown that inflammation is the main inducement of vascular disease, microRNA (miRNA) can influence inflammation in various ways, and Se deficiency can affect miRNAs expression. To study the mechanism of aorta damage caused by Se deficiency, we constructed a Se deficiency porcine aorta model and found that Se deficiency can significantly inhibit miR-223, which downregulates the expression of nucleotide-binding oligomerization domain-like receptor family 3 (NLRP3). Subsequently, we found that in Se deficiency group, NLRP3, and its downstream (caspase-1, apoptosis-related spot-like protein [ASC], IL-18, IL-1ß) expression was significantly increased. In vitro, we cultured pig iliac endothelium cell lines, and constructed miR-223 knockdown and overexpression models. NLRP3 messenger RNA and protein levels were significant increased in the knockdown group, and decreased in the overexpression group. The results of this study show that Se deficiency in porcine arteries can induce inflammation through miR-223/NLRP3.

Pregnant women living with HIV represent one of the most high-priority groups for HIV treatment and health assessment. Although social support has been shown to be a protective factor for improved health-related quality of life (HRQoL), and depression and anxiety have been identified as two major causes of psychological distress among people living with HIV, it is still unclear how social support, anxiety, and depression interact to influence HRQoL. The objective of our study was to demonstrate the nature of predictors, direct effects and mediator effects among social support, anxiety, depression symptoms and HRQoL in pregnant women living with HIV. We investigated a total of 101 pregnant women living with HIV in Yunnan province in China from April 2016 to June 2016. All participants completed the Social Support Rating Scale (SSRS), the Chinese version of the Hospital Anxiety and Depression Scales (HADS) and Quality of Life instruments (EuroQoL Five Dimensions Questionnaire, EQ-5D). The relationships between the variables were examined by Pearson's or Spearman's correlation analysis. Predictor effects were tested using separate multiple regressions, controlling for demographic variables and HIV diagnosis variables. Direct and mediation effects of social support on HRQoL were tested using a structural equation model (SEM). Anxiety and depression symptoms were negatively correlated with subjective social support, support utilization, social support and HRQoL. Social support significantly predicted better HRQoL, and anxiety and depression symptoms significantly predicted poorer HRQoL. Anxiety and depression symptoms partially mediated the associations between social support and HRQoL. Anxiety and depression symptoms completely mediated the associations of objective support and support utilization with HRQoL. Interventions to improve HRQoL in pregnant women living with HIV must consider the mediation effect of anxiety and depression symptoms on the association between social support and HRQoL. Social support interventions are valid only when anxiety and depression symptoms are managed effectively.

BACKGROUND: Nebulized magnesium sulfate (MgSO4) has been used to treat asthma, but the efficacy remains controversial. We aimed to comprehensively review the efficacy of nebulized MgSO4 in treating adult patients with asthma. METHODS: PubMed, Embase, and Cochrane Library were searched for relevant studies published up to July 18, 2016. Randomized controlled trials (RCTs) were included if adult patients with acute or stable asthma had been treated with nebulized MgSO4 compared with placebo or another bronchodilator. Standardized mean differences (SMDs), relative risks (RRs) and 95% confidence intervals (CIs) were calculated. Outcomes included pulmonary function, hospital admission and adverse events. RESULTS: A total of 1386 patients from sixteen trials (1240 acute asthma patients and 146 stable asthma patients) were subjected to meta-analysis. Compared to placebo as normal saline, whether using in acute or stable adult asthma, nebulized MgSO4 did not significantly improve the respiratory function: SMD 0.39 (95% CI -0.03-0.82, P = 0.07), and 1.48 (95% CI -0.14-3.11, P = 0.07), respectively. Furthermore, nebulized MgSO4 did not reduce hospital admission in adult patients with acute asthma (RR, 0.72; 95% CI, 0.52-1.00; P = 0.05), although it was not associated with increased adverse events (RR 1.15; 95% CI, 0.88-1.52; P = 0.31). CONCLUSIONS: Evidence to date suggests that nebulized MgSO4 has no role in the management of adult patients with acute or stable asthma.

In the experiment, the morphology of heavy metals (Pb, Cr, Cd, and Ni, HMs) was characterized using flame atomic absorption spectroscopy. In addition, Fourier transform infrared spectroscopy (FTIR) and three-dimensional excitation-emission matrix fluorescence spectroscopy (3D-EEM) were used to characterize the correlation between environmental factors and metal morphology in the rotting compost from several angles. The results showed that the humus treated with wood vinegar solution had a high degree of humification and rich aromatic structure. FTIR spectroscopy confirmed that the degree of humus aromatization gradually increased during the composting process, which enhanced the complexation of humus (HS) with HMs but had less effect on Ni. In addition, the optimum concentration of wood vinegar (WV) was determined to be 1.75%. The results of the study showed that in the Pb passivation treatment group, the proportion of soluble (Red) and exchangeable states (Exc) converted to oxidized (Oxi) and residual states (Res) was 8%, 14%, 6%, 1%, and 12% in the CK, T1, T2, T3, and T4 treatment groups, respectively; in the Cr passivation treatment group, the proportion of Cr-Red and Cr-Exc converted to oxidized and residual states was 31%, 33%, 25%, 29%, and 25%; in the Cd passivation treatment group, the proportions of Cd-Red and Cd-Exc converted to oxidized and residual states were 5%, 15%, 4%, 9%, and 11%, respectively; whereas the Ni treatment group did not show any significant passivation effect. The proportion of Pb-Oxi was relatively stable, Cr-Oxi was converted to Cr-Res, whereas Cd showed the conversion of Cd-Oxi to Cd-Exc. SUVA254 and SUVA280 showed significant positive correlations with Pb-Res, Cr-Res and Ni-Res, and significant positive correlations with moisture content (MC); whereas MC was significantly negatively correlated with each form of HMs. Total potassium (TK), total nitrogen (TN), and both carbon (TOC) were negatively correlated with Pb-Res and Pb-Exc. Structural equation modeling verified the relationship between environmental factors and HMs, and the composting results showed that the addition of biochar (BC) and a higher percentage of WV could increase compost decomposition and passivate HMs to improve its agronomic function.

Objective: The level of health literacy is one of the important factors affecting health outcomes. Family is an important place to shape personality traits, and people with different personalities will adopt different lifestyles, which will lead to variations in health outcomes. Therefore, this article aims to explore the relationship between health literacy and personality and its influencing factors in different family structures. Methods: This was a cross-sectional study with 1,406 individuals. A questionnaire was utilized to measure health literacy, personality and demographic variables, including family structure. Canonical correlation analysis (CCA) and hierarchical multiple regression analysis were used to examine the relation between health literacy and personality traits between two types of family structure. Results: CCA showed that the canonical correlation coefficients were 0.309 (p < 0.001) and 0.347 (p < 0.001), in two-parent family and single-parent family, respectively. The openness of personality traits exhibited the highest correlation with health literacy. Compared with the remaining personality traits, openness yielded the strongest effect (ß = 0.485 and ß = 0.830) in two types of family structure, respectively. Education and monthly income were significantly associated with health literacy. Conclusion: Our results support the relation between health literacy and personality traits in two types of family structure.

This study aimed to investigate the expression of autophagy-related proteins in a mouse model of neuromyelitis optica (NMO). Mice were assigned to one of four groups: an animal experimental model group (NMO-EAE group, given with exogenous IL-17A), Interleukin-17 monoclonal antibody intervention group (NMO-EAE_0IL17inb), No exogenous interleukin-17 enhanced immune intervention group (NMO-EAE_0IL17), and a control group. Behavioral scores were assessed in each group, and the protein expressions of sequestosome 1 (P62), Beclin-1, the mammalian target of rapamycin (mTOR), phosphoinositide 3-kinase (PI3K-I), and LC3II/LC3I were detected using Western blotting. In the NMO-EAE_0IL17 group, the expression of Beclin-1 decreased, the LC3II/LC3I ratio was lower, and the expressions of P62, mTOR, and PI3K-I increased; after administration of IL-17A inhibitor into the brain tissue, however, the expression of Beclin-1 increased significantly, along with the LC3II/LC3I ratio, while the expressions of P62, mTOR and PI3K-I protein decreased significantly. In terms of behavioral scores, the scores of optic neuritis and myelitis were more serious, onset occurred earlier and the progress was faster, after the administration of IL-17A. In the mechanism of NMO animal model, IL-17A may regulate autophagy and affect the disease process through the activation of the PI3K-mTOR signaling pathway.

Maintaining high activity during prolonged catalysis is always the pursuit in catalytic degradation of organic pollutants. For indoor formaldehyde (HCHO) degradation, the accumulation of intermediates is the major factor limiting the conversion of HCHO to final product CO2 (HCHO-to-CO2 conversion) and long-lasting catalysis. Herein, a three-dimensional radialized nanostructure catalyst self-assembled by MnOOH/MnO2 nanosheets anchored with Pt single atoms (PtSA-MnOOH/MnO2 with a trace platinum loading amount of 0.09%) is developed by thermally assisted two-step electrochemical method, which achieves enhanced CO2 production in catalytic HCHO degradation at the room temperature by the collaborative action of active hydroxyl (OH*) and active oxygen species (O2*). By boosting intermediates' decomposing, the catalyst implements real-time HCHO-to-CO2 conversion (∼85.7%) and long-term continuous HCHO removal (∼98%) during 100 h in a 15 ppm HCHO atmosphere at 25 °C under a weight hourly space velocity of 30000 mL/gcat∙h. Density functional theory calculation shows that the formation energy of O2* from O2 over PtSA-MnOOH/MnO2 is nearly half lower than that over Pt-MnO2 catalyst. And decomposing accumulated intermediates gives the credit to OH* species sustainably generated by the combined action of MnOOH and O2*. The synergistic action between PtSA and MnOOH contributes to the continuous production of O2* and OH* for enhancing CO2 production in indoor catalytic formaldehyde degradation.

OBJECTIVE: Coronavirus disease 2019 (COVID-19) caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) remains pandemic with considerable morbidity and mortality around the world. The aim of this study was to identify the predictors for clinical deterioration in patients with COVID-19 who did not show clinical deterioration upon hospital admission. METHODS: Two hundred fifty-seven patients with confirmed COVID-19 pneumonia admitted to Guangzhou Eighth People's Hospital between 23 January and 21 March 2020 were retrospectively enrolled. Demographic data, symptoms, laboratory values, comorbidities and treatments were all collected. The study endpoint was clinical deterioration within 20 days from hospital admission. Univariate and multivariable logistic regression methods were used to explore the risk factors associated with clinical deterioration. RESULTS: A total of 49 (19%) patients showed clinical deterioration after admission. Compared with patients that did not experience clinical deterioration, clinically deteriorated patients had more dyspnea, cough and myalgia (65.3% versus 29.3%) symptoms and more had comorbidities (89.8% versus 36.1%). Clinical and laboratory characteristics at admission that were associated with clinical deterioration included senior age, diabetes, hypertension, myalgia, higher temperature, systolic blood pressure, C-reactive protein (CRP), procalcitonin, activated partial thromboplastin time, aspartate aminotransferase, alanine transaminase, direct bilirubin, plasma creatinine, lymphocytopenia, thrombocytopenia, decreased albumin and bicarbonate concentration. Medical history of angiotensin-converting enzyme inhibitors/angiotensin receptor blockers, calcium channel blockers and metformin were also risk factors. CONCLUSION: The four best predictors for clinical deterioration were CRP, procalcitonin, age and albumin. A "best" multivariable prediction model, resulting from using a variable selection procedure, included senior age, presentation with myalgia, and higher level of CRP and serum creatinine (bias-corrected c-statistic = 0.909). Sensitivity and specificity corresponding to a cut point of CRP ≥18.45 mg/L for predicting clinical deterioration were 85% and 74%, respectively.

BACKGROUND: This study proposes and validates a method of measuring 3D strain in myocardium using a 3D Cardiovascular Magnetic Resonance (CMR) tissue-tagging sequence and a 3D optical flow method (OFM). METHODS: Initially, a 3D tag MR sequence was developed and the parameters of the sequence and 3D OFM were optimized using phantom images with simulated deformation. This method then was validated in-vivo and utilized to quantify normal sheep left ventricular functions. RESULTS: Optimizing imaging and OFM parameters in the phantom study produced sub-pixel root-mean square error (RMS) between the estimated and known displacements in the x (RMSx = 0.62 pixels (0.43 mm)), y (RMSy = 0.64 pixels (0.45 mm)) and z (RMSz = 0.68 pixels (1 mm)) direction, respectively. In-vivo validation demonstrated excellent correlation between the displacement measured by manually tracking tag intersections and that generated by 3D OFM (R >or= 0.98). Technique performance was maintained even with 20% Gaussian noise added to the phantom images. Furthermore, 3D tracking of 3D cardiac motions resulted in a 51% decrease in in-plane tracking error as compared to 2D tracking. The in-vivo function studies showed that maximum wall thickening was greatest in the lateral wall, and increased from both apex and base towards the mid-ventricular region. Regional deformation patterns are in agreement with previous studies on LV function. CONCLUSION: A novel method was developed to measure 3D LV wall deformation rapidly with high in-plane and through-plane resolution from one 3D cine acquisition.

Plantaginis Semen (PS) has been used to promote diuresis and clear away dampness. Recent reports have shown that PS can be used to treat gouty nephropathy (GN). However, the action and mechanism of PS have not been well defined in treating GN. The present study aimed to define the molecular mechanisms of PS as a potential therapeutic approach to treat GN. A combination of network pharmacology and validation experiments in GN is used to understand the potential mechanism. Information on pharmaceutically active compounds in PS and gene information related to GN was obtained from public databases. The compound target network and protein-protein interaction network were constructed to study the mechanism of action of PS in the treatment of GN. The mechanism of action of PS in the treatment of GN was analyzed via Gene Ontology (GO) biological process annotation and Kyoto Gene and Genomics Encyclopedia (KEGG) pathway enrichment. Validation experiments were performed to verify the core targets. The GN rat model was prepared by the method of combining yeast and adenine. Hematoxylin-eosin (HE) staining was used to observe the morphology of renal tissue in rats. ELISA was applied to detect TGF-ß1, TNF-α, and IL-1ß levels in renal tissue. The expressions of TGF-ß1, TNF-α, and IL-1ß were determined using immunohistochemistry. Through the results of network pharmacology, we obtained 9 active components, 118 predicted targets, and 149 GN targets from the public database. Based on the protein-protein interaction (PPI), 26 hub genes for interaction with PS treating for GN were screened, including MMP9, TNF, IL1ß, and IL6. The enrichment analysis results showed that the treatment of GN with PS was mainly involved in the TGF-ß1 signaling pathway, MAPK signaling pathway, TNF signaling pathway, NF-κB signaling pathway, and PI3K Akt signaling pathway. Validation experiment results showed that PS could reduce the content of urinary protein and UA and deregulate the expression of TGF-ß1, TNF-α, and IL-1ß in the treatment of GN. The molecular mechanism of PS in the treatment of GN indicated the synergistic features of multicomponent, multitarget, and multipathway of traditional Chinese medicine, which provided an essential scientific basis for further elucidating the mechanism of PS in the treatment of GN.

PURPOSE: Plant metabolites have gained considerable attention as the anticancer agents over the last few decades. Previous studies have indicated the potential of taxifolin as an anticancer agent. However, the information on the anticancer activity of taxifolin against skin scar cell carcinoma as well as several other types of cancers is scantly. Against this background, the present study was designed to investigate the anticancer activity of taxifolin against a panel of skin scar cell carcinoma cell lines. MATERIALS AND METHODS: Proliferation rate of the cells was monitored by MTT assay. DAPI and annexin V/PI assay was used to investigate the induction of apoptosis. Flow cytometery was employed to carry out the cell cycle analysis. Transwell assay was used to check the invasion of the cancerous cells. RESULTS: The results indicated that taxifolin inhibits the growth of the skin scar cell carcinoma cell lines. However the anticancer effects were more profound on the SSCC cancer cells (IC50, 20 µM). In contrast the anticancer effects of taxifolin on the non-cancerous skin cells were minimal. Further investigation revealed that the anticancer effects of taxifolin on the SSCC cells is due to the induction of apoptosis and cell cycle arrest. Moreover, taxifolin could also inhibit the invasion of SSCC cells which was associated with downregulation of the MMP-2 and MMP-9 expression indicative of the potential of taxifolin in the treatment of scar cell carcinoma Conclusion: It is was found that taxifolin inhibited the growth of skin scar cell carcinoma growth by triggering apoptosis and cell cycle arrest and also inhibited cell invasion capacity and therefore this study warrants further investigation on the anticancerous potential of taxifolin.

BACKGROUND Recent studies have shown that Escherichia coli induced digestive tract diseases may be related to outer membrane vesicles (OMVs) induced intestinal double-strand breaks (DSBs) in intestinal epithelial cells. This study aimed to compare the impact of OMVs forces on DSBs in intestinal epithelial Caco-2 cells, and provide a new treatment for digestive diseases caused by E. coli. MATERIAL AND METHODS E.coli OMVs were prepared and co-cultured with Caco-2 cells. The uptake of OMVs by Caco-2 cells was observed by confocal microscopy. The γ-H2AX protein was detected by western-blots. The DSBs caused by OMVs was detected by single cell gel electrophoresis. RESULTS The particle size analyzer showed that the average diameters of OMVs centrifuged at 20 000×g and 50 000×g were 217.5±7.29 nm and 186.3±6.59 nm (P<0.05), respectively. Transmission electron microscopy of the OMVs revealed a lipid bilayer structure with a variety of different sizes. Confocal fluorescence microscopy revealed that OMVs almost completely entered Caco-2 cells after 24 hours. The ratio of γ-H2AX protein band gray value normalized data in the OMVs centrifuged at 20 000×g and 50 000×g, and the control group (without OMVs) were 2.23±0.18, 1.58±0.20, 1±0.30 (P<0.05), respectively, while DNA levels of the comet tail (TailDNA%, TDNA%) were 72.21±14.61%, 23.11±4.98%, and 1.02±1.41% (P<0.05), respectively. The corresponding DNA damage was categorized as high (grade 3), moderate (grade 2), and no damage (grade 0). CONCLUSIONS Different sizes of OMVs induced different degrees of DNA damage in intestinal epithelial Caco-2 cells.

RESUMEN

Cerebral venous sinus thrombosis (CVST) is a rare type of cerebrovascular disease, accounting for about 0.5% of all strokes. About 4% of patients with CVST have supratentorial brain parenchymal lesions and brain edema sufficient to cause brain hernia and neurological deterioration, which is called malignant CVST. Malignant CVST refers to the clinical (loss of consciousness, unilateral or bilateral pupil dilation) and imaging signs of supratentorial cortical lesions (ischemia or hemorrhage) accompanied by tentorial hiatal hernia formation at the onset or after treatment with heparin. For patients with malignant CVST, decompressive craniectomy is not only a life-saving treatment, but also can make most patients achieve good functional outcome.

OBJECTIVE: To evaluate the effectivity and the improvement of elbow arthroscopic techniques of treating obstinate tennis elbow using debridement and repair under elbow arthroscopy combined with small incision. METHODS: From March 2014 to February 2017, 36 patients(total 36 elbows) with obstinate tennis elbow were treated consecutively using debridement and repair under elbow arthroscopy combined with small incision open, the two methods alternate. In key process, the first 18 cases [group A including 8 males and 12 females with an average age of (43.89±9.71) years old, the treatment time was(17.39±10.53) months] used direct-looking operation and arthroscopic verification, the latter 18 cases[group B including 7 males and 11 females with an averave age of (44.28±8.04) years old, the treatment time was(15.50±9.18) months] used arthroscopic operation and direct-looking verification. The arthroscopic and gross pathological findings were observed during the operation. After operation serious neurovascular complications were observed. The operation time was compared between two groups. MEPS (Mayo Elbow Performance Score) and VAS scores were recorded preoperatively and postoperatively. RESULTS: All patients were followed up for an average of (17.22±8.47) months in group A and (17.83±8.83) months in group B. There was 1 case of nerve injury without infection. VAS score was improved from preoperative 4.33±1.24 to postoperative 0.61±0.70 in group A, and from 4.50±1.47 to postoperative 0.67±0.69 in group B. MEPS of group A was improved from preoperative 62.22±7.90 to postoperative 93.06±5.18 in group A, and from preoperative 61.94±8.93 to postoperative 92.22±5.21 in group B. There were no statistical differences between two groups in MEPS and VAS score. The operation time in group B(54.06±8.43) min was less than that in group A(73.39±12.78) min. Thirty-two cases were satisfied greatly with treatment results, 3 cases satisfied, 1 case unsatisfied. The main reasons that results in dissatisfaction were nerve injury. CONCLUSIONS: Treating obstinate tennis elbow using debridement and repair under elbow arthroscopy combined with small incision have open and arthroscopic surgery advantages, with thorough therapeutic effect. The procedure is suitable to try to explore and improve the elbow arthroscopic technique.

RESUMEN

Podocyte infolding glomerulopathy (PIG) is a pathologic type of podocyte glomerulopathy reported recently. The characteristic is that the ultrastructure related to podocytes, such as microspheres and microtubules, are folded into the glomerular basement membrane (GBM) under electron microscope. At present, there are few reports about this disease at home and abroad, and most of them are concentrated in Japan. The clinical characteristics and pathogenesis of PIG are still unclear. In this paper, we report a case of clinical manifestations of nephrotic syndrome, renal biopsy indicated PIG, after the treatment of glucocorticoid, hydroxychloroquine and tacrolimus, the patient's clinical symptoms were relieved and urinary protein decreased.

Systemic lupus erythematosus (SLE) is an autoimmune disease with multiple organ involvement. There are still many limitations and individual differences in the treatment based on glucocorticoids and immunosuppressants. In recent years, more and more studies have shown that the combination of traditional Chinese medicine in the treatment of SLE has the advantages of good efficacy, low adverse reactions, and high safety. However, the exact regulatory mechanism and combined traditional Chinese medicine in the treatment of SLE are still unclear. This paper reviews the research on the mechanism of traditional Chinese medicine in the treatment of SLE from metabonomic, immune cells, lymphocyte factors and apoptosis, etc, provides ideas for exploring the mechanism of traditional Chinese medicine in the treatment of SLE with modern methods.

Epidemic cerebrospinal meningitis shows a high degree of consistency with the law of transmission among wei (卫)-qi-ying (营)-blood, in terms of the onset of the season, contagiousness, symptoms, pathogenesis, as well as characteristics of the transmission. It is proposed to use epidemic cerebrospinal meningitis as an example to explore the underlying disease of wei-qi-ying-blood syndrome differentiation system. Epidemic meningitis invades the brain from the upper respiratory tract along the nervous system, and its overall pathogenesis follows from entering the lung system (prodromal period) to entering the blood (bacteremia period, sepsis period) and then entering the brain (shock period). According to the four-dimensional qualitative principle of epidemic pathogen tropism, it corresponds to disease of both wei and qi syndrome, then blazing of both qi and ying syndrome, and then heat blocking pericardium, exuberant heat stirring wind, and internal block and external collapse syndrome. This article explored the laws of transmission among wei-qi-ying-blood and its underlying diseases described in On Warm Heat (《温热论》), and revealed the original appearance of the disease model under the laws of transmission among wei-qi-ying-blood to guide the clinical practice.

Objective: To analyze the clinical features of children with uridine responsive developmental epileptic encephalopathy 50 (DEE50) caused by CAD gene variants. Methods: A retrospective study was conducted on 6 patients diagnosed with uridine-responsive DEE50 caused by CAD gene variants at Beijing Children's Hospital and Peking University First Hospital from 2018 to 2022. The epileptic seizures, anemia, peripheral blood smear, cranial magnetic resonance imaging (MRI), visual evoked potential (VEP), genotype features and the therapeutic effect of uridine were descriptively analyzed. Results: A total of 6 patients, including 3 boys and 3 girls, aged 3.5(3.2,5.8) years, were enrolled in this study. All patients presented with refractory epilepsy, anemia with anisopoikilocytosis and global developmental delay with regression. The age of epilepsy onset was 8.5 (7.5, 11.0) months, and focal seizures were the most common seizure type (6 cases). Anemia ranged from mild to severe. Four patients had peripheral blood smears prior to uridine administration, showing erythrocytes of variable size and abnormal morphology, and normalized at 6 (2, 8) months after uridine supplementation. Two patients suffered from strabismus, 3 patients had VEP examinations, indicating of suspicious optic nerve involvement, and normal fundus examinations. VEP was re-examined at 1 and 3 months after uridine supplementation, suggesting significant improvement or normalization. Cranial MRI were performed at 5 patients, demonstrating cerebral and cerebellar atrophy. They had cranial MRI re-examined after uridine treatment with a duration of 1.1 (1.0, 1.8) years, indicating significant improvement in brain atrophy. All patients received uridine orally at a dose of 100 mg/(kg·d), the age at initiation of uridine treatment was 1.0 (0.8, 2.5) years, and the duration of treatment was 2.4 (2.2, 3.0) years. Immediate cession of seizures was observed within days to a week after uridine supplementation. Four patients received uridine monotherapy and were seizure free for 7 months, 2.4 years, 2.4 years and 3.0 years respectively. One patient achieved seizure free for 3.0 years after uridine supplementation and had discontinued uridine for 1.5 years. Two patients were supplemented with uridine combined with 1 to 2 anti-seizure medications and had a reduced seizure frequency of 1 to 3 times per year, and they had achieved seizure free for 8 months and 1.4 years respectively. Conclusions: The clinical manifestations of DEE50 caused by CAD gene variants present a triad of refractory epilepsy, anemia with anisopoikilocytosis, and psychomotor retardation with regression, accompanied by suspected optic nerve involvement, all of which respond to uridine treatment. Prompt diagnosis and immediate uridine supplementation could lead to significant clinical improvement.

OBJECTIVE@#To analyze 43 leukemia genes in children with acute lymphoblastic leukemia (ALL) in Yunnan province, and provide the basis for the diagnosis and treatment of children with ALL in this area.@*METHODS@#The clinical data of 428 children with newly diagnosed ALL in Yunnan area from January 2015 to December 2020 were retrospectively analyzed. Multiple nested PCR technology was used to detect 43 common leukemia genes.@*RESULTS@#Among the 428 children with ALL, 159 were positive for leukemia genes, with a positive rate of 37.15% (159/428), and a total of 15 leukemia genes were detected. Among the 159 leukemia gene-positive children, ETV6-RUNX1+ accounted for 25.79% (41/159), followed by E2A-PBX1+ and BCR-ABL+, accounting for 24.53% (39/159) and 23.27% (37/159) respectively. MLL+ accounted for 6.29% (10/159), WT1+ accounted for 4.40% (7/159), IKZF1 gene deletion and CRLF2+ accounted for 3.77% (6/159) respectively. The positive rate of MLL (46.15%) was the highest in <1-year old group, the positive rate of ETV6-RUNX1 (10.56%) was the highest in 1-10-year old group, and BCR-ABL+ rate (23.65%) was the highest in >10-year old group. The distribution of leukemia genes in different age groups was statistically significant (P <0.05).@*CONCLUSION@#The most common fusion gene of children with ALL in Yunnan is ETV6-RUNX1, followed by E2A-PBX1 and BCR-ABL.

The present study investigated the underlying molecular mechanism by which Buthus martensii Karsch chlorotoxin (BmK CT) inhibits the invasion and metastasis of glioma cells and the possibility of 131I­labeled BmK CT (131I­BmK CT) as a novel targeted agent for the treatment of glioma. The impact of BmK CT with and without 131I radiolabeling on the invasion and metastasis of glioma cells in vitro was studied. Cell viability was assessed using Cell Counting Kit­8 and plate colony formation assays in order to confirm the cytotoxicity of BmK CT and 131I­BmK CT at different concentrations. Transwell invasion and wound­healing assays were conducted in order to investigate the inhibitory effects BmK CT and 131I­BmK CT on cell migration and invasion. Furthermore, western blotting, ELISA immunofluorescence and a gelatin zymography assay were performed to evaluate changes in the protein expression levels of glioma cells following treatment with BmK CT or 131I­BmK CT. The results indicated that BmK CT inhibits the invasion and metastasis of glioma cells via regulation of tissue inhibitor of metalloproteinase­2 expression and that 131I­BmK CT has the potential to be a novel targeted therapeutic drug for glioma.
