RESUMEN

The effect of MK-467, a peripheral α2 -adrenoceptor antagonist, on plasma drug concentrations, sedation and cardiopulmonary changes induced by intramuscular (IM) medetomidine was investigated in eight sheep. Additionally, the interactions with atipamezole (ATI) used for reversal were also evaluated. Each animal was treated four times in a randomized prospective crossover design with 2-week washout periods. Medetomidine (MED) 30 µg/kg alone or combined in the same syringe with MK-467 300 µg/kg (MMK) was injected intramuscular, followed by ATI 150 µg/kg (MED + ATI and MMK + ATI) or saline intramuscular 30 min later. Plasma was analysed for drug concentrations, and sedation was subjectively assessed with a visual analogue scale. Systemic haemodynamics and blood gases were measured before treatments and at intervals thereafter. With MK-467, medetomidine plasma concentrations were threefold higher prior to ATI, which was associated with more profound sedation and shorter onset. No significant differences were observed in early cardiopulmonary changes between treatments. Atipamezole reversed the medetomidine-related cardiopulmonary changes after both treatments. Sedation scores decreased more rapidly when MK-467 was included. In this study, MK-467 appeared to have a pronounced effect on the plasma concentration and central effects of medetomidine, with minor cardiopulmonary improvement.

OBJECTIVE: To determine whether the current Finnish screening method using a single flexed mediolateral view as scored by osteophyte is sufficient to diagnose mild elbow dysplasia in Labrador retrievers and to determine if an additional craniocaudal oblique projection would result in improvement in the screening protocol. MATERIALS AND METHODS: Thirteen dogs with one mildly affected elbow joint and one elbow joint without radiological evidence of osteophytes were studied. Radiographic and computed tomography studies were performed and the results compared with each other. RESULTS: Medial compartment disease was observed in 14 of 26 joints based on computed tomography. The sensitivity and specificity of the grading based mainly on osteoarthritis was 79 and 92%, respectively. A strong association existed between elbow dysplasia based on computed tomography and medial humeral epicondylar osteophytes on the craniocaudal projection. CLINICAL SIGNIFICANCE: A single mediolateral flexed radiograph is reliable in diagnosing mild elbow dysplasia in Labrador retrievers. However, the craniocaudal oblique projection increases the specificity of the diagnosis, and it is proposed that it be included in the radiographic protocol in this breed.

OBJECTIVE: The study assessed the use and reliability of bathroom scales as an objective measurement tool, and setting a normal variance of static weight bearing between hindlimbs. METHODS: Two groups of dogs were tested: a healthy control group (n = 21) and a group (n = 43) of dogs with confirmed osteoarthritis in at least one stifle joint, with or without hip joint osteoarthritis. Static weight bearing was evaluated manually and measured with two bathroom scales. An orthopaedic examination was done and dynamic weight bearing was measured using a force platform. Radiographs were taken to confirm the presence of osteoarthritis, and dogs were divided into groups of severe and non-severe osteo- arthritic changes. Reliability by repeatability was tested using analysis of variance, and the congruity between static weight bearing and other evaluation methods with Kappa statistics and proportion of agreement. RESULTS: The difference between the hindlimbs proportional to the body weight in control dogs was 3.3% (± 2.7%). The repeatability of measuring static weight bearing in the hindlimbs of osteoarthritic dogs with bathroom scales was 81% with osteoarthritic limbs, and 70% for unaffected limbs. The sensitivity of static weight bearing measurements using bathroom scales was 39% and specificity 85%. CLINICAL SIGNIFICANCE: Bathroom scales are a reliable, simple, and cost-effective objective method for measuring static weight bearing and can be used as an outcome measure when rehabilitating dogs with osteoarthritic changes in the hindlimbs.

The clinical effects and pharmacokinetics of medetomidine (MED) and its enanti-omers, dexmedetomidine (DEX) and levomedetomidine (LEVO) were compared in a group of six beagle dogs. The dogs received intravenously (i.v.) a bolus of MED (40 microg/kg), DEX (20 and 10 microg/kg), LEVO (20 and 10 microg/kg), and saline placebo in a blinded, randomized block study in six separate sessions. Sedation and analgesia were scored subjectively, and the dogs were monitored for heart rate, ECG lead II, direct blood pressure, respiratory rate, arterial blood gases, and rectal body temperature. Blood samples for drug analysis were taken. Peak sedative and analgesic effects were observed at mean (+/- SD) plasma levels of 18.5 +/- 4.7 ng/mL for MED40, 14.0 +/- 4.5 ng/mL for DEX20, and 5.5 +/- 1.3 ng/mL for DEX10. The overall level of sedation and cardiorespiratory effects did not differ between MED40, DEX20 and DEX10 during the first hour, apparently due to a ceiling effect. However, the analgesic effect of DEX20 lasted longer than the effect of the corresponding dose of racemic medetomidine, suggesting greater potency for dexmedetomidine in dogs. Levomedetomidine had no effect on cardio-vascular parameters and caused no apparent sedation or analgesia. The pharmacokinetics of dexmedetomidine and racemic medetomidine were similar, but clearance of levomedetomidine was more rapid (4.07 +/- 0.69 L/h/kg for LEVO20 and 3.52 +/- 1.03 for LEVO10) than of the other drugs (1.26 +/- 0.44 L/h/kg for MED40, 1.24 +/- 0.48 for DEX20, and 0.97 +/- 0.33 for DEX10).

RESUMEN

The defences of family members were studied in order to observe the reactions of the family when faced with a dysfunction in one of its members. Thirty families with a mentally handicapped child were studied, and the attitudes of parents, grandparents and siblings towards the handicapped child were examined. The data showed that the birth of a mentally handicapped child interrupts the normal life-cycle of the family, leading to a crisis. The first reaction in the parents was most often found to be denial, but ultimately adaptation was demonstrated in nearly 80% of the parents. Many of the siblings' and grandparents' reactions differed significantly from those of the parents, being more positive than those of the latter.
