RESUMEN

Protein O-linked ß-N-acetylglucosamine modification (O-GlcNAcylation) plays a crucial role in regulating essential cellular processes. The disruption of the homeostasis of O-GlcNAcylation has been linked to various human diseases, including cancer, diabetes, and neurodegeneration. However, there are limited chemical tools for protein- and site-specific O-GlcNAc modification, rendering the precise study of the O-GlcNAcylation challenging. To address this, we have developed heterobifunctional small molecules, named O-GlcNAcylation TArgeting Chimeras (OGTACs), which enable protein-specific O-GlcNAcylation in living cells. OGTACs promote O-GlcNAcylation of proteins such as BRD4, CK2α, and EZH2 in cellulo by recruiting FKBP12F36V-fused O-GlcNAc transferase (OGT), with temporal, magnitude, and reversible control. Overall, the OGTACs represent a promising approach for inducing protein-specific O-GlcNAcylation, thus enabling functional dissection and offering new directions for O-GlcNAc-targeting therapeutic development.

This Letter proposes an optical-pulse-based reconfigurable phase control method, enabling a dual-band phased array receiver to operate in two modes: dual-band-independent operation and dual-band fusion. The method utilizes optical pulses and optical delay to compensate for phase differences across frequency bands. An electrical phase shifter is employed to compensate for phase residual in both bands. All phase operations to both bands are processed concurrently in one link, thereby maintaining inter-band phase coherence. Experimental results verify the ability of dual-band-independent beamforming and inter-band phase coherence maintaining. A four-channel dual-band (X- and Ku-band) phased array antenna (PAA) receiver is constructed to measure PAA patterns and demonstrate band fusion. The pulse compression results in all directions reveal a doubled improvement in range resolution, which shows the potential for enhancement of radar performance.

This paper addresses the problem of fault detection in DC microgrids in the presence of denial-of-service (DoS) attacks. To deal with the nonlinear term in DC microgrids, a Takagi-Sugeno (T-S) model is employed. In contrast to the conventional approach of utilizing current sampling data in the traditional event-triggered mechanism (ETM), a novel integrated ETM employs historical information from measured data. This innovative strategy mitigates the generation of additional triggering packets resulting from random perturbations, thus reducing redundant transmission data. Under the assumption of faults occurring within a finite-frequency domain, a resilient event-based H-/H∞ fault detection filter (FDF) is designed to withstand DoS attacks. The exponential stability conditions are derived in the form of linear matrix inequalities to ensure the performance of fault detected systems. Finally, the simulation results are presented, demonstrating that the designed FDF effectively detects finite-frequency faults in time even under DoS attacks. Furthermore, the FDF exhibits superior fault detection sensitivity compared to the conventional H∞ method, thus confirming the efficacy of the proposed approach. Additionally, it is observed that a trade-off exists between fault detection performance and the data releasing rate (DRR).

RESUMEN

High-speed photonic reservoir computing (RC) has garnered significant interest in neuromorphic computing. However, existing reservoir layer (RL) architectures mostly rely on time-delayed feedback loops and use analog-to-digital converters for offline digital processing in the implementation of the readout layer, posing inherent limitations on their speed and capabilities. In this paper, we propose a non-feedback method that utilizes the pulse broadening effect induced by optical dispersion to implement a RL. By combining the multiplication of the modulator with the summation of the pulse temporal integration of the distributed feedback-laser diode, we successfully achieve the linear regression operation of the optoelectronic analog readout layer. Our proposed fully-analog feed-forward photonic RC (FF-PhRC) system is experimentally demonstrated to be effective in chaotic signal prediction, spoken digit recognition, and MNIST classification. Additionally, using wavelength-division multiplexing, our system manages to complete parallel tasks and improve processing capability up to 10 GHz per wavelength. The present work highlights the potential of FF-PhRC as a high-performance, high-speed computing tool for real-time neuromorphic computing.

Stochasticity is an inherent feature of biological neural activities. We propose a noise-injection scheme to implement a GHz-rate stochastic photonic spiking neuron (S-PSN). The firing-probability encoding is experimentally demonstrated and exploited for Bayesian inference with unsupervised learning. In a breast diagnosis task, the stochastic photonic spiking neural network (S-PSNN) can not only achieve a classification accuracy of 96.6%, but can also evaluate the diagnosis uncertainty with prediction entropies. As a result, the misdiagnosis rate is reduced by 80% compared to that of a conventional deterministic photonic spiking neural network (D-PSNN) for the same task. The GHz-rate S-PSN endows the neuromorphic photonics with high-speed Bayesian inference for reliable information processing in error-critical scenarios.

Real-time acquisition of target signals is preferred for mobile communication systems. However, under the requirement of ultra-low latency for next-generation communication, traditional acquisition methods need to temporally locate the target signal from a large amount of raw data with correlation-based computing, introducing extra latency. We propose a real-time signal acquisition method based on an optical excitable response (OER) by pre-designing a single-tone preamble waveform. The preamble waveform is designed to be within the amplitude and bandwidth of the target signal, so no extra transceiver is required. The OER generates a corresponding pulse to the preamble waveform in the analog domain, which simultaneously triggers an analog-to-digital converter (ADC) to acquire target signals. The dependence of OER pulse on the preamble waveform parameter is studied, leading to a pre-design of the preamble waveform for an optimal OER. In the experiment, we demonstrate a millimeter-wave (26.5-GHz) transceiver system with target signals of orthogonal frequency division multiplexing (OFDM) format. Experimental results show that the response time is less than 4 ns, which is far lower than the ms-level response time of traditional all-digital time-synchronous acquisition methods.

We experimentally demonstrate an all-optical nonlinear activation unit based on the injection-locking effect of distributed feedback laser diodes (DFB-LDs). The nonlinear carrier dynamics in the unit generates a low-threshold nonlinear activation function with optimized operating conditions. The unit can operate at a low threshold of -15.86 dBm and a high speed of 1 GHz, making it competitive among existing optical nonlinear activation approaches. We apply the unit to a neural network task of solving the second-order ordinary differential equation. The fitting error is as low as 0.0034, verifying the feasibility of our optical nonlinear activation approach. Given that the large-scale fan-out of optical neural networks (ONNs) will significantly reduce the optical power in one channel, our low-threshold scheme is suitable for the development of high-throughput ONNs.

Temporal alignment between the demultiplexing signal and sampled signal for complex wideband signals greatly increases the difficulty of designing high-speed and high-resolution photonic analog-to-digital converters (PADCs). We present a vector description to decouple the timing skew from the phase frequency response in time-demultiplexing PADC. We demonstrate that the calibration can be optically implemented with true time delay effects and the broadband input can be one-time calibrated through several single-frequency signals. For verification, we configure out a 40 GSa/s PADC with eight-interleaved sub-channels. The timing skew-induced spurs across the whole Nyquist band are effectively suppressed, making the PADC acquire a wideband signal with 16 GHz instantaneous bandwidth. The spurious-free dynamic range (SFDR) is enhanced to ∼55 dB, and the effective number of bits (ENOB) is improved from ∼5.5 bits to ∼8 bits within 20 GHz. It is nearly 1 bit better than the recently reported time-demultiplexing PADC under the comparable input frequencies.

The radio-frequency (RF) signal processing in real time is indispensable for advanced information systems, such as radar and communications. However, the latency performance of conventional processing paradigm is worsened by high-speed analog-to-digital conversion (ADC) generating massive data, and computation-intensive digital processing. Here, we propose to encode and process RF signals harnessing photonic spiking response in fully-analog domain. The dependence of photonic analog-to-spike encoding on threshold level and time constant is theoretically and experimentally investigated. For two classes of waveforms from real RF devices, the photonic spiking neuron exhibits distinct distributions of encoded spike numbers. In a waveform classification task, the photonic-spiking-based scheme achieves an accuracy of 92%, comparable to the K-nearest neighbor (KNN) digital algorithm for 94%, and the processing latency is reduced approximately from 0.7 s (code running time on a CPU platform) to 80 ns (light transmission delay) by more than one million times. It is anticipated that the asynchronous-encoding, and binary-output nature of photonic spiking response could pave the way to real-time RF signal processing.

A novel bacterium, designated NAS39T, was isolated from the interfacial sediment of Taihu Lake in PR China and its taxonomic position was investigated by using a polyphasic approach. Cells of the isolate were Gram-stain-negative, aerobic, non-motile, catalase-positive, yellow and rod-shaped. Phylogenetic analyses based on 16S rRNA gene sequences supported that strain NAS39T formed a cluster within the genus Flavobacterium, and was most closely related to Flavobacterium laiguense LB2P30T (98.4 %), followed by Flavobacterium tiangeerense 0563T (97.4 %). The average nucleotide identity values between strain NAS39T and F. laiguense LB2P30T and F. tiangeerense 0563T were 82.5 and 75.3 %, respectively. The digital DNA-DNA hybridization values between strain NAS39T and F. laiguense LB2P30T and F. tiangeerense 0563T were 40.9 and 18.6 %, respectively. The genomic DNA G+C content was 34.1 mol%. The major respiratory quinone was menaquinone-6. The dominant cellular fatty acids were iso-C15 : 0 and summed feature 3 comprising C16 : 1 ω7c/C16 : 1 ω6c. The polar lipids comprised phosphatidyl ethanolamine, two amino lipids, three amino phospholipids and two unidentified lipids. Based on the phenotypic, chemotaxonomic, genotypic and phylogenetic characteristics, strain NAS39T (=MCCC 1K06094T=KACC 22328T) represents a novel species of the genus Flavobacterium, for which the name Flavobacterium taihuense sp. nov. is proposed.

Spontaneous intracerebral hemorrhage (ICH) accounts for 10-20% of all strokes and contributes to higher mortalities and severe disabilities. The aims of this study were, therefore, to characterize novel biomarkers, metabolic disruptions, and mechanisms involving ICH. A total 30 ICH patients and 30 controls were enrolled in the study, and their clinical characteristics were analyzed. Nontargeted metabolomic analysis was conducted using ultra-performance liquid chromatography quadrupole time-of-flight tandem mass spectrometry (UPLC/Q-TOF). Multivariate statistical analysis and receiver operating characteristic curve analysis were used for screening and evaluating the predictive ability of biomarkers. ICH patients showed significantly higher systolic blood pressure, diastolic blood pressure, blood glucose levels, white blood cell counts, neutrophil count, percentage of neutrophils and globulin and a lower albumin/globin ratio when compared with controls. In sum, 11 important metabolites were identified, which were associated with disruption of fatty acid oxidation and sphingolipid and phospholipid metabolism, as well as increased inflammation, oxidative stress, and vascular pathologies. Further multiple logistic regression analyses of these metabolites showed that l-carnitine and phosphatidylcholine (20:3/22:6) have potential as biomarkers of ICH, and the area under the curve, sensitivity, specificity were 0.974, 90%, and 93%, respectively. These findings provide insights into the pathogenesis, early prevention, and diagnosis of ICH.

Dysfunction of the homeostasis-maintaining systems in specific cell types or tissues renders the organism susceptible to a range of diseases, including cancers. One of the emerging mechanisms for maintaining tissue homeostasis is cellular senescence. Here, we report that the Hippo pathway plays a critical role in controlling the fate of ovarian cells. Hyperactivation of Yes-associated protein 1 (YAP1), the major effector of the Hippo pathway, induces senescence in cultured primary human ovarian surface epithelial cells (hOSEs). Large tumor suppressor 2 (LATS2), the primary upstream negative regulator of YAP1, is elevated in both YAP1-induced and natural replicative-triggered senescence. Deletion of LATS2 in hOSEs prevents these cells from natural replicative and YAP1-induced senescence. Most importantly, loss of LATS2 switches ovarian cells from YAP-induced senescence to malignant transformation. Our results demonstrate that LATS2 and YAP1, two major components of the Hippo/YAP signaling pathway, form a negative feedback loop to control YAP1 activity and prevent ovarian cells from malignant transformation. Human cancer genomic data extracted from TCGA datasets further confirm the clinical relevance of our finding.

BACKGROUND: To compare the accuracy of photogrammetry, intraoral scanning and conventional impression techniques for complete-arch implant rehabilitation. METHODS: A master cast containing 6 implant abutment replicas was fabricated. Group PG: digital impressions were taken 10 times using a photogrammetry system; Group IOS: intraoral scanning was performed to fabricate 10 digital impressions; Group CNV: splinted open-tray impression technique was used to fabricate 10 definitive casts. The master cast and conventional definitive casts were digitized with a laboratory reference scanner. For all STL files obtained, scan bodies were converted to implant abutment replicas using a digital library. The accuracy of a digitizer was defined by 2 main parameters, trueness and precision. "Trueness" was used to describe the deviation between test files and reference file, and "precision" was used to describe the closeness between test files. Then, the trueness and precision of three impression techniques were evaluated and statistically compared (α = 0.05). RESULTS: The median trueness was 24.45, 43.45 and 28.70 µm for group PG, IOS and CNV; Group PG gave more accurate trueness than group IOS (P < 0.001) and group CNV (P = 0.033), group CNV showed more accurate trueness than group IOS (P = 0.033). The median precision was 2.00, 36.00 and 29.40 µm for group PG, IOS and CNV; Group PG gave more accurate precision than group IOS (P < 0.001) and group CNV (P < 0.001), group CNV showed more accurate precision than IOS (P = 0.002). CONCLUSIONS: For complete-arch implant rehabilitation, the photogrammetry system showed the best accuracy of all the impression techniques evaluated, followed by the conventional impression technique, and the intraoral scanner provided the least accuracy.

The relatively high recombination rate of charges remains the most critical limiting factor for solar-driven water splitting for hydrogen generation. Herein, a tandem 0D/2D/2D NbS2 quantum dot/Nb2 O5 nanosheet/g-C3 N4 flake (NSNOCN) system is designed. Owing to the unique spatial-arrangement and elaborate morphology of 0D NbS2 , 2D Nb2 O5 , and 2D g-C3 N4 in the newly designed NSNOCN, plenty of spatial charge-transfer cascades from g-C3 N4 to NbS2 via Nb2 O5 are formed to accelerate separation and transfer of charges significantly, thus contributing to a high photocatalytic H2 generation rate of 13.99 mmol h-1 g-1 (an apparent quantum efficiency of 10.8% at 420 nm), up to 107.6 and 43.7 times by contrast with that of g-C3 N4 and Nb2 O5 , respectively. This work can provide a new platform in the design of artificial photocatalytic systems with high charge-transfer efficiency.

Although the role of the Hippo signaling pathway in development and tumorigenesis has been extensively studied in multiple organs, its role in ovarian follicle development remains largely unknown. Here, we report that Yes-Associated Protein 1 (YAP1), the major effector of Hippo signaling, is spatiotemporally expressed in ovarian granulosa cells and plays a critical role in the regulation of follicle development. We found that the active form of YAP1 (nuclear YAP1) was predominantly expressed in proliferative granulosa cells, whereas the inactive form of YAP1 (cytoplasmic YAP1) was mainly detected in luteal cells (terminally differentiated granulosa cells). Pharmacological inhibition of YAP1 activity disrupted mouse ovarian follicle development in vitro and in vivo. Foxl2 promoter-driven knockout of Yap1 in ovarian granulosa cells resulted in increased apoptosis of granulosa cells, decreased number of corpora lutea, reduced ovarian size, and subfertility in transgenic mice. However, Cyp19a1 promoter-driven knockout of Yap1 in differentiated granulosa cells of preovulatory follicles and luteal cells of corpora lutea had no effect on ovarian morphology and fertility. Mechanistic studies demonstrated that YAP1 interacted with epidermal growth factor receptor and TGF-ß signaling pathways to regulate granulosa cell proliferation, differentiation, and survival. Results from this study identify YAP1 as a critical regulator of granulosa cell proliferation and differentiation. Balanced expression and activation of YAP1 is essential for follicle development and successful reproduction. Exonuclease 1 (EXO1) is required for both DNA repair and meiosis. Inactivation of EXO1 gene in mice leads to infertility. This study aimed to investigate whether variants in the EXO1 gene contribute to human premature ovarian failure (POF). The coding region of EXO1 was sequenced in 186 Han Chinese patients with non-syndromic POF. No plausible mutation was detected. The results suggest that mutations in the coding region of EXO1 may not be responsible for POF in Han Chinese women.

In this retrospective study, the relationship between demographic characteristics, past medical history, general lifestyle habits and susceptibility of premature ovarian failure (POF) in Han Chinese population was investigated. Five hundred and fifty-three patients with POF and 400 women with normal ovarian function were recruited. A questionnaire was designed to gather information from responders. Logistic regression was carried out to calculate odds ratios (OR), 95% confidence intervals (95% CI) and P-values. History of pelvic surgery, mumps, having relatives with menstrual abnormalities and exposure to chemical agents were significantly associated with increased risk of POF (OR 5.53 [2.15 to 14.23]; 3.26 [2.38 to 4.47]; 28.12 [8.84 to 89.46]; 4.47 [2.09 to 9.58]). Vegetarian diet, tea and mineral water consumption reduced the risk of POF (OR 0.27 [0.19 to 0.37]; 0.04 [0.03 to 0.07]; 0.63 [0.47 to 0.85], respectively). Heredity, pelvic surgery, mumps and exposure to chemical agents were identified as risk factors for POF, whereas vegetarian diet, tea consumption and mineral water drinking were protective. Therefore, genetic consultation could help those women whose relatives manifested an early or premature menopause to avoid the consequences of possible premature ovarian function cessation. Avoidance of exposure to endocrine disrupters and flavonoids intake should be considered.

Currently, there is a lack of relevant research on the efficacy difference between SHD combined with IBG and PVIBGT in the treatment of osteonecrosis of the femoral head(ONFH). Firstly, this study intends to compare the effectiveness of surgical hip dislocation combined with impacting bone grafts (SHD-IBG) and pedicled vascularised iliac bone graft transfer (PVIBGT) in treating ONFH. And the study investigates patients who suffered from hip preservation failures from both groups to better comprehend failure reasons. 30 patients (34 hips) with ARCO stage IIIA femoral head necrosis were selected between January 2012 and July 2022. They were divided into group A(SHD-IBG) and group B (PVIBGT) according to different surgical methods. Firstly, compared the 1-year effect between SHD-IBG and PVIBGT at 1 year postoperatively; Secondly, assessed the medium and long-term efficacy of SHD-IBG hip preservation treatment; Lastly, based on study of the femoral head removed from patients with hip preservation failure in the two groups, the reasons for the failure of hip preservation were comprehensively analyzed in the two groups. Group A: 11 males (13 hips), 4 females (4 hips);Group B: 9 males (11 hips), 6 females (6 hips).Firstly, the average Harris scores of the two groups at 1 year after surgery: preoperative: 70.7, 1 year after surgery: 78.9 in group A; preoperative: 69.5, 1 year after surgery: 81.5 in group B. The differences were statistically significant (P < 0.05).Compared to the preoperative period, quantitative analysis by DCE-MRI showed an increase in perfusion in the necroticarea and an improvement in hyperperfusion in the repair-responsive area one year after the surgery. Secondly, in group A, the hip preservation rate was 88.2% at 2.5-11 (average of 77 months) years of follow-up, and the mean Harris score at the last follow-up was 73.2.Semi-quantitative analysis of postoperative DCE-MRI showed that the perfusion curves of necrotic and repaired areas were similar to those of the normal area. This suggests the instability within the femoral head had been effectively improved, and the perfusion had partially recovered. Thirdly, according to Micro-CT and pathologica studies of patients with hip preservation failure in these two groups, all these patients' femoral head was significantly collapsed and deformed. Their trabeculae was thin and partially disorganized, with fractures in the subchondral bone and separation of the cartilage from the subchondral bone. The necrotic areas had sparse trabeculae, disorganized arrangement, loss of continuity, and disappearance of cells in the trabecular traps. The necrotic area was covered with fibrous tissue, and partial restoration was observed in the repair area. Mechanical finite element analysis showed that the maximum equivalent force was observed in the weight- bearing area and the cortical bone surrounding the shaft of femurand. The result of DCE-MRI showed that the repair reaction area exhibited abnormal hyperperfusion. In this study, the efficacy of SHD-IBG and PVIBGT was compared at 1 year after operation, and the long-term follow-up of SHD-IBG was 2.5-11 (mean 77 months) years, combined with DCE-MRI results, we found that the short-term effect of PVIBGT was more significant than that of SHD-IBG. SHD-IBG can achieve satisfactory hip preservation in the medium and long term follow-up.

Silkie chicken, an important chicken breed with high medicinal and nutritional value, has a long history of being used as a dietary supplement in China. However, the compounds with health-promoting effects in Silkie chickens remain unclear. In the present study, we conducted a comprehensive analysis of metabolic and lipidomic profiles to identify the characteristic bioactive compounds in Silkie chickens, using a common chicken breed as control. The results showed that the levels of 13 metabolites including estradiol, four lipid subclasses including cardiolipin (CL), eight lipid molecules, and three fatty acids including docosahexaenoic acid (C22:6) were significantly increased in Silkie chickens, which have physiological activities such as resisting chronic diseases and improving cognition. These characteristic bioactive compounds have effects on meat quality characteristics, including improving its water-holding capacity and umami taste and increasing the content of aromatic compounds and phenols. The differentially expressed genes (DEGs) between the two chicken breeds revealed the regulatory network for these characteristic bioactive compounds. Fifteen DEGs, including HSD17B1, are involved in the synthesis of characteristic metabolites. Eleven DEGs, including ELOVL2, were involved in the synthesis and transport of characteristic lipids and fatty acids. In summary, we identified characteristic bioactive compounds in Silkie chickens, and analyzed their effects on meat quality characteristics. This study provided important insight into Silkie chicken meat as a functional food.

Objective: To explore the short-term effectiveness of hip revision surgery guided by artificial intelligence preoperative planning (AIHIP) system. Methods: The clinical data of 22 patients (23 hips) who were admitted between June 2019 and March 2023 and met the selection criteria were retrospectively analyzed. There were 12 males and 10 females with an average age of 69.7 years (range, 44-90 years). There were 19 hips in the first revision, 3 hips in the second revision, and 1 hip in the third revision. The causes of revision included 12 hips with prosthesis loosening, 4 hips with acetabular cup loosening, 3 hips with osteolysis, 2 hips with acetabular dislocation, 1 hip with postoperative infection, and 1 hip with prosthesis wear. There were 6 hips in stage ⅡA, 9 hips in stage ⅡB, 4 hips in stage ⅡC, 3 hips in stage ⅢA, and 1 hip in stage ⅢB according to Paprosky staging of acetabular bone defect. The replacement of prosthesis type, operation time, hospitalization stay, ground active condition, and postoperative infection, fracture, prosthesis loosening, and other adverse events were recorded. The function of the affected limb was evaluated by Harris score before operation, at 1 week and 6 months after operation, and the range of motion of the hip joint was compared before operation and at 6 months after operation. Results: The operation time was 85-510 minutes, with an average of 241.8 minutes; the hospitalization stay was 7-35 days, with an average of 15.2 days; the time of disassociation from the walker was 2-108 days, with an average of 42.2 days. All the 22 patients were followed up 8-53 months (mean, 21.7 months). No adverse events such as prosthesis loosening or infection occurred in the rest of the patients, except for postoperative hematoma of the thigh in 1 patient and dislocation of the hip in 1 hip. The matching degree of acetabular cup was completely matched in 22 hips and mismatched in 1 hip (+2), the matching rate was 95.65%. The matching degree of femoral stem was completely matched in 22 hips and generally matched in 1 hip (-1), and the matching rate was 100%. The Harris scores were 55.3±9.8 and 89.6±7.2 at 1 week and 6 months after operation, respectively, which significantly improved when compared with before operation (33.0±8.6, P<0.05), and further improved at 6 months after operation than at 1 week after operation ( P<0.05). The function of hip joint was evaluated by Harris score at 6 months after operation, and 21 hips were good and 2 hips were moderate, which could meet the needs of daily life. The range of motion of hip joint was (111.09±10.11)° at 6 months after operation, which was significantly different from (79.13±18.50)° before operation ( t=-7.269, P<0.001). Conclusion: AIHIP system can improve the accuracy of revision surgery, reduce the difficulty of surgery, and achieve good postoperative recovery and satisfactory short-term effectiveness.
