RESUMEN

Osteoarthritis (OA), a common joint disorder with articular cartilage degradation as the main pathological change, is the major source of pain and disability worldwide. Despite current treatments, the overall treatment outcome is unsatisfactory. Thus, patients with severe OA often require joint replacement surgery. In recent years, mesenchymal stem cells (MSCs) have emerged as a promising therapeutic option for preclinical and clinical palliation of OA. MSC-derived exosomes (MSC-Exos) carrying bioactive molecules of the parental cells, including non-coding RNAs (ncRNAs) and proteins, have demonstrated a significant impact on the modulation of various physiological behaviors of cells in the joint cavity, making them promising candidates for cell-free therapy for OA. This review provides a comprehensive overview of the biosynthesis and composition of MSC-Exos and their mechanisms of action in OA. We also discussed the potential of MSC-Exos as a therapeutic tool for modulating intercellular communication in OA. Additionally, we explored bioengineering approaches to enhance MSC-Exos' therapeutic potential, which may help to overcome challenges and achieve clinically meaningful OA therapies.

Although supertetrahedral Tn sulfide clusters (n=2-6) have been extensively explored, the synthesis of Tn selenide clusters with n>4 has not been achieved thus far. Reported here are ionic-liquid (IL)-assisted precursor route syntheses, characterizations, and the photocatalytic properties of six new M-In-Q (M=Cu or Cd; Q=Se or Se/S) chalcogenide compounds, namely [Bmmim]12 Cu5 In30 Q52 Cl3 (Im) (Q=Se (T5-1), Se48.5 S3.5 (T5-2); Bmmim=1-butyl-2,3-dimethylimidazolium, Im=imidazole), [Bmmim]11 Cd6 In28 Q52 Cl3 (MIm) (Q=Se (T5-3), Se28.5 S23.5 (T5-4), Se16 S36 (T5-5); MIm=1-methylimidazole), and [Bmmim]9 Cd6 In28 Se8 S44 Cl(MIm)3 (T5-6). The cluster compounds T5-1 and T5-3 represent the largest molecular supertetrahedral Tn selenide clusters to date. Under visible-light illumination, the Cu-In-Q compounds showed photocatalytic activity towards the decomposition of crystal violet, whereas the Cd-In-Q compounds exhibited good photocatalytic H2 evolution activity. Interestingly, the experimental results show that the photocatalytic performances of the selenide/sulfide solid solutions were significantly better than those of their selenide analogues, for example, the degradation time of the organic dye with T5-2 was much shorter than that with T5-1, whereas the photocatalytic H2 evolution efficiencies with T5-3-T5-6 improved significantly with increasing sulfur content. This work highlights the significance of IL-assisted precursor route synthesis and the tuning of photocatalytic properties through the formation of solid solutions.

An axially chiral binuclear µ-hydroxo Pd complex (BPHP) first served as an excellent chiral sensor for discriminating a variety of analytes including amino alcohol, amino amide, amino acid, mandelic acid, diol, diamine, and monoamine by 31P{1H} NMR. A detailed recognition mechanism was proposed based on the single crystal and mass spectrum of Pd-complexes. In general, BPHP sensor, through extracting the acidic hydrogen of an analyte by its Pd-OH group, forms stable diastereomeric complexes with two enantiomers of the analyte giving well distinguishable split 31P{1H} NMR signals for chiral discrimination.

RESUMEN

A new boric agent with bridged structure, boric acid D, was first synthesized and used as an excellent chiral derivative agent for highly efficient enantiodiscrimination of various diols. The derivatization reaction is fast and complete, easy to operate and has high accuracy in measurement of ee values. The characteristic split NMR signals are well-distinguishable with a large chemical shift nonequivalence (up to 0.39 ppm).

A simple chiral binuclear titanium complex ((S)-Ti2L3Na4) was first prepared and served as an excellent chiral sensor for discriminating a variety of chiral ammoniums by 1H NMR. In the presence of trifluoroacetic acid, the in-situ-generated ammoniums of varied amines can be effectively resolved by (S)-Ti2L3Na4. The different binding forces between (S)-Ti2L3Na4 and two enantiomers of analytes generate different shielding effects on two enantiomers, resulting in a split in the distinguishable chemical shift.

A method for nucleic acid amplification, loop-mediated isothermal amplification (LAMP) is a novel, sensitive and rapid technique, which can be applied for disease diagnosis in aquaculture. Using the LAMP method, a highly specific and sensitive diagnostic system for infectious hypodermal and hematopoietic necrosis virus (IHHNV) detection was designed. A set of four primers was designed by targeting the IHHNV genome DNA. By the detection system, target DNA was amplified and visualized on agarose gel within 60min under isothermal condition at 64 degrees C. Without gel electrophoresis, the LAMP amplicon was visualized directly in the reaction tube by addition of SYBR Green I for a naked-eye inspection. The LAMP reaction was also assessed by the white turbidity of magnesium pyrophosphate (a by-product of LAMP) in the tube. The assay had a detection limit of 5-500 copies of DNA template with gel electrophoresis, SYBR Green I and white turbidity with naked-eye inspection. The detection sensitivity of LAMP was 100-fold higher than the PCR. A diagnostic procedure which is rapid and highly sensitive was developed for IHHNV detection.

Vanadium compounds have been recognized for their hypoglycemic effects; however, potential short and long-term vanadium toxicity has slowed the acceptance for therapeutic use. In the present work, three batches of vanadium-enriched chickpea sprout (VCS) were prepared by incubating chickpea seeds in presence of 200, 100, and 50 microg/ml of sodium orthovanadate (SOV). The effects of oral administration of chickpea sprout (CS) and VCS food for 8 weeks on streptozotocin-induced (STZ) diabetic rats were investigated. Both CS and VCS food was found to ameliorate some hyperglycemic symptoms of the diabetic rats, i.e. improve lipid metabolism, decrease blood glucose level, prevent body weight loss, and reduce impairment of diabetic related spatial learning and memory. Serum insulin was substantially elevated in treated diabetic rats, which is probably one important reason for the hypoglycemic effect. Compared with CS alone, VCS100 food exhibited remarkably enhanced effectiveness in alleviating diabetes induced hyperglycemia and memory loss. Moreover, vanadium-enriched chickpeas appeared to abolish the vanadium induced toxicity associated with administration of this metal for diabetes during the 8-week study period. This study suggested further work of the vanadium speciation in CS and novel hypoglycemic mechanism for the antidiabetic activity of vanadium agents. Vanadium containing (VCS) food could be a dietary supplement for the diabetic status.
