RESUMEN

1. A study was conducted to assess the effects of supplemental protease (0 or 15,000 units/kg) and diet type (maize- or wheat-based) on apparent jejunal and ileal digestibility and apparent total tract metabolisability of nutrients in Cobb 500 mixed-sex broilers from 6 to 31 d of age.2. Birds were randomly distributed into 56 metabolism cages (6 birds/cage; 14 replicates/treatment). At 22 d of age, jejunal and ileal digesta contents were collected and pooled from 4 birds/cage to determine apparent digestibility coefficients (DC) and digestible energy (DE). Feed intake was measured, and total excreta were collected from 8 to 11 and 18 to 21 d of age to determine apparent metabolisable energy (AME) and total tract nitrogen and starch metabolisability coefficients.3. Broilers offered the maize-based diet with protease had greater (P < 0.05) jejunal nitrogen DC, starch DC, and DE (8.2, 6.5, and 14.9%, respectively) and ileal nitrogen DC and DE (5.1 and 6.8%, respectively) than those offered the maize-based diet without protease. Ileal starch DC was increased (P < 0.05) by 1.1% with protease supplementation.4. Broilers offered maize-based diets had greater (P < 0.05) nitrogen (7.3%) and starch (0.6%) metabolisability coefficients and AME (4.7%) from 8 to 11 d of age, and nitrogen (4.3%) metabolisability coefficients and AME (2.0%) from 18 to 21 d of age compared with those offered wheat-based diets.5. Thus, protease supplementation and diet type can affect digestive dynamics and nutrient utilisation in broilers.

1. The effect of increasing the dose level of a novel consensus bacterial 6-phytase variant on apparent ileal digestibility (AID) of phosphorus (P), phytic acid (inositol hexa-phosphate, IP6) and ileal IP6 degradation profile was studied in diets containing varying phytate-P (PP) levels.2. Ross 308, one-day-old males (n = 1,800) were allocated to cages (20 birds/cage, six cages/treatment) in a completely randomised design employing a 3 × 5 factorial arrangement (three PP levels: 2.45 (low) 2.95 (medium) and 3.45 g/kg (high); five dose levels of phytase (PhyG): 0, 500, 1,000, 2,000 and 4,000 FTU/kg). Phased diets were based on wheat, corn, soybean meal, rapeseed meal and rice bran (d 0 to 10; 2.60 g/kg digestible P, 7.6 g/kg calcium (Ca); d 11 to 21; 2.10 g/kg digestible P, 6.4 g/kg Ca). Ileal digesta was collected on d 21 for determination of P, IP6 and IP-esters content. Data were analysed by factorial ANOVA; means separation was achieved using Tukey's HSD test.3. Increasing PP reduced AID of IP6 and sum of IP3-6 (%) (P < 0.05) but absolute P-release (g/kg diet) above NC was increased (P < 0.05) at high vs. low PP. Increasing phytase dose exponentially increased (P < 0.001) AID IP6, sum of IP3-6 (%) and digestible IP3-6-P g/kg diet (P < 0.001). AID P was increased but there was an interaction with PP level (P < 0.001). Ileal accumulation of IP5-3-P was universally low with PhyG at ≥1,000 FTU/kg (<0.06 g/100 g DM). At 2,000 and 4,000 FTU/kg, AID IP6 was 97.2, 92.7, 92.6% and 100, 97.2, 97.1%, respectively, at low, medium and high PP. At 2,000 FTU/kg, phytate-P release estimated as the increase (above NC) in ileal digestible sum of IP3-6-P in the diet was 2.26, 2.59 and 3.10 g/kg in low, medium and high PP, respectively.4. The data demonstrated that the novel phytase was effective in breaking down phytate to low IP-esters in diets with varied PP content but the optimal dose level for maximising P-release may differ in diets with varying PP content.

This trial was conducted to investigate the effect of arginine (Arg), threonine (Thr) and Arg + Thr administered in ovo on growth performance, digestive organs and intestinal morphology in broiler chickens. On day 14 of incubation, 400 fertile eggs were randomly allotted to five experimental treatments through injection in the amniotic fluid including: (i) control (none injected), (ii) sham (0.5 ml of 0.5% saline), (iii) Arg (35 mg/egg), (iv) Thr (25 mg/egg) and (v) Arg + Thr (35 + 25 mg/egg). After hatching, chicks were given a commercial corn-soya bean diet up to 42 days of age. Daily feed intake (FI) and body weight (BW) of chicks were measured during different periods of the trial. Digestive organs were measured for their relative weight and intestinal length on days 11 and 42 of age. Intestinal morphometric traits were evaluated on day 11 of the experiment. Supplementing amino acids affected the performance of broiler chicks as Thr significantly increased FI and BW across starter, grower and finisher periods compared with sham and control (p < 0.05). Furthermore, Arg + Thr injection increased jejunal weight compared with control on day 42 (p < 0.05). Moreover, Arg inclusion led to the greatest villus height and crypt depth among treatments in duodenum (p > 0.05); however, amino acid supplemented groups had lower villus height than control in jejunum (p < 0.05). Negative correlations found between digestive organs related to day 11 of age containing pancreas (r = -0.484; p = 0.030), duodenal (r = -0.577; p = 0.007) as well as ileal lengths (r = -0.471; p = 0.035) and FI of entire period. Otherwise, positive relationships were observed between duodenum (r = 0.580; p = 0.007) and ileum (r = 0.582; p = 0.007) weights on day 42 and FI of chickens across the entire phase. In conclusion, Arg and particularly Thr injection into amnion can improve FI and post-hatch growth performance of chickens which may be mediated by the development of digestive organs.

Six expeller-extracted canola meal (ECM) samples produced under different seed conditioning temperatures (90, 95, or 100°C) and screw torques in the second press (low or high) were evaluated in a 3 × 2 factorial arrangement of treatments to determine the effect of processing on standardized ileal amino acid digestibility (SID) in 10 and 24-day-old broilers. A nitrogen-free diet was fed to determine ileal endogenous amino acid flow. Each diet was fed for 5 d to 6 replicate cages of 10 or 7 chicks, and ileal samples were collected at 10 and 24 d, respectively. The endogenous flow (mg/kg DM intake) of CP and all amino acids except Cys decreased (P < 0.01) with age. Conditioning temperature by screw torque interactions were detected (P < 0.05) for apparent ileal digestibility (AID) of CP, Arg, Ile, Lys, Phe, Thr, Ala, Asp, Glu, Pro ,and Ser at d 10, and the same trend was observed at d 24 except for Ile, Phe, Ala, and Pro (P > 0.05). Meals processed under medium conditioning temperature (95°C) at either low or high screw torque had the greatest (P < 0.05) AID of CP and total amino acids. The SID values followed a similar pattern as AID at both ages. When corrected for endogenous losses, the average AID of total amino acids improved approximately 3.5 and 2.0 units, at 10 and 24 d, respectively. The AID and SID values increased (P < 0.05) with age for most AA, but the effect of age was not consistent between ECMs. A negative correlation was detected between NDF and neutral detergent-insoluble nitrogen (NDIN) content of the meals and SID values of Lys at d 24 (r = -0.79, r = -0.76; P = 0.001, respectively). Processing conditions affected CP and amino acid digestibility, likely because of alterations to the chemical composition of ECM and formation of indigestible complexes of amino acids with fiber. The AID and SID values increased with age independent of meal processing conditions.

The objective of this study was to investigate the effect of processing conditions and chemical composition on ileal digestible energy (IDE), AME, and AMEn of 6 expeller-extracted canola meal (ECM) samples subjected to conditioning temperature at 90, 95, or 100°C and high or low screw torque over the second presses in a 3 × 2 factorial arrangement. The ECM samples were incorporated into a corn-soybean meal reference diet at 30% by replacing energy-yielding ingredients. A total of 210 one-day-old male broiler chicks (Ross 308) were fed common starter and grower diets until d 18, and then assigned to 7 experimental diets replicated 6 times, with 5 chicks per cage. After a 5-d diet acclimation period from d 18 to 22, excreta was collected for 72 h. The difference method was used to determine AME, which was corrected to zero N balance to obtain AMEn. Medium seed conditioning temperature resulted in the highest IDE, AME, and AMEn compared with low or high temperature, and high screw torque resulted in higher energy utilization compared with low torque (P < 0.001). There was also an interaction (P < 0.001) between conditioning temperature and screw torque. For ECM subjected to low or medium conditioning temperature at low screw torque, IDE, AME, and AMEn values ranging from 2,137 to 2,705, 2,089 to 2,655, and 1,977 to 2,482 kcal/kg of DM, respectively, were obtained. The mean AMEn values were 2,260 kcal/kg of DM, indicating a 7% reduction compared with AME values. The AMEn values were negatively correlated with neutral detergent fiber (NDF; r = -0.93; P = 0.001) and NDIN (r = -0.87; P = 0.001). Stepwise regression to predict AMEn value resulted in the following equation: AMEn (kcal/kg of DM) = 3,397.8 + (-100.1 × NDF %) + (279.5 × ash %) + (-33.8 × ADF %) (R² = 0.91; SE = 61.9; P = 0.001). These results indicate that AMEn values vary markedly among ECM samples, and chemical constituents, especially the fiber components, may have a considerable effect on AMEn value.

Data from 13 datasets from 4 trials on the effect of a novel consensus bacterial 6-phytase variant (PhyG) on the apparent ileal digestibility (AID) of amino acids (AA) in broilers were used to model AID AA responses. The datasets were obtained from 3 trial locations (New Zealand, Australia and United States) and collectively incorporated variations in diet composition (feedstuff composition, phytate-P (PP) level, limestone solubility), feed form (mash or pellet), bird genetics (strain), and age at sampling (11-35 d of age). In total, 384 observations were analyzed. First, the relationships between AID of AA (as coefficients) and increasing phytase dose level from 0 to 4,000 FTU/kg were evaluated across all datasets using exponential curve fitting. Second, the percentage unit change in AID of AA at each phytase dose level from baseline (basal diet [BD] without phytase) was calculated separately for each dataset and the data then modeled together using exponential curve fitting. The model-predicted mean coefficient of AID of total AA in basal diets was 0.76 (range 0.56 [Cys] to 0.83 [Glu]), which was increased by PhyG to 0.80 and 0.81 at 2,000 and 4,000 FTU/kg, respectively. Exponential increases in the percentage unit improvement in AID of 18 individual and of total AA with increasing phytase dose level were evident (P < 0.05). Improvements (vs. BD) at 2,000 FTU/kg and 4,000 FTU/kg, respectively, were greatest for Cys (+9.2 and +11.0% units), Met (after deduction of synthetic Met, +8.4 and +9.0% units), and Thr (after deduction of synthetic Thr, +6.2 and +7.3% units). The data demonstrated consistent improvements in the AID of AA by the phytase. The modeling results generated from data gathered from birds sampled at different ages and from different dietary settings with correction of synthetic AA for Lys, Met, Thr, and Trp, enabled a more accurate prediction of the digestible AA contribution from the diet by this novel phytase. This will allow diet-specific AA matrix recommendations to be made in commercial feed formulations.

This study was conducted to evaluate the effects of different dietary inclusion of eucalyptus essential oil (EEO) on growth performance, relative organ weight, cecal microflora, nutrient digestibility, serum biochemical parameters, and thigh muscle fatty acid profile in broiler chickens. A total of six hundred 1-day-old male broiler chickens were randomly allocated into 5 treatment groups with 8 replicate pens, and each pen contained 15 birds. The experiment lasted for 42 d. Dietary treatments included corn-soybean meal-based diet supplemented with 0, 250, 500, 750, and 1,000 mg/kg EEO. The results indicated that dietary treatments had no effect on growth performance parameters in the 1 to 10 d period. From day 11 to 24, dietary supplementation of EEO showed a linear decrease in feed conversion ratio (FCR, P < 0.05). From day 25 to 42 and the overall period (1-42 d), broilers fed with different levels of EEO showed a linear increase in body weight gain (BWG) and reduction in feed conversion ratio (linear, P < 0.05). The relative organ weight were unaffected by any of the dietary treatments. With increasing levels of EEO, the cecal Escherichia coli (linear, P = 0.085) count showed a trend in reduction, and the cecal lactic acid bacteria population tended to increase (linear, P = 0.063). The apparent ileal digestibility of ether extract and organic matter were linearly and quadratically increased in response to increasing dietary EEO supplementation (P < 0.05). A trend of linear decrease in total cholesterol in the serum of birds fed with different levels of EEO was recorded (P = 0.074). Eucalyptus essential oil's inclusion increased serum superoxide dismutase linearly but reduced serum malondialdehyde linearly (P < 0.05). Dietary supplementation of EEO affected the fatty acid profile of thigh muscle so that increased the concentrations of total polyunsaturated fatty acids (linear, P < 0.05) and reduced total saturated fatty acid contents (linear, P < 0.05). Taken together, the inclusion of EEO increased BWG and decreased FCR during day 25 to 42 and day 1 to 42, and partially improved cecal microflora balance, nutrient digestibility, antioxidant activity, and thigh muscle fatty acid profile in broiler chickens.

This study was conducted to investigate the effects of zinc (Zn), as a combination of oxide (ZnO) and sulfate (ZnSO4), compared with incremental levels of zinc hydroxychloride (ZH) on tibia traits, intestinal integrity, expression of selected jejunal genes, cecal short chain fatty acids and microbial composition in broilers. Day-old male Ross 308 chicks (n = 784) were randomly allocated to seven dietary treatments, each replicated seven times with 16 chicks per replication. The dietary treatments included a negative control diet (NC) with no supplemental Zn, a positive control (PC) with 100 mg/kg supplemental Zn from an ionic bound source combination (50 mg/kg ZnO + 50 mg/kg ZnSO4), and the NC diet supplemented with one of 20, 40, 60, 80, or 100 mg/kg Zn as ZH. The diets were fed over starter (1-14 d) and grower (14-35 d) phases, with tissue and digesta samples collected from 3 birds per replicate on days 14 and 35. The results showed that dietary Zn level had a significant effect on tibia breaking strength on d 35 (P < 0.05), and tibia Zn concentration both on d 14 and d 35 (P < 0.01). Dietary Zn levels linearly (P < 0.01) increased cecal lactic acid production, increased Lactobacillus, and decreased Bacillus and total bacteria counts (P < 0.05). Inclusion of 80 and 100 mg/kg Zn as ZH tended to upregulate the expression of claudin-1 (P = 0.088) and tight junction protein-1 (P = 0.086). The results obtained in this study suggest that a non-Zn supplemented diet can negatively influence tibia development and gut microbiota composition in broiler chickens. Higher supplemental Zn in the diet alters cecal microbiota population in favor of Lactobacillus and can decrease the total bacterial load. Supplemental Zn level in the feed have the potential to manipulate the jejunal gut integrity at a molecular level.

A total of 945 male Ross 308 broiler chicks were used in a growth study to explore the interaction between dietary crude protein concentration and available phosphorus. Nine experimental treatments were constructed factorially by offering low, medium, or standard protein concentrations without or with low, standard, or high available phosphorus. Diets were based on corn, wheat, and soybean meal and all nutrients other than protein/amino acids and available phosphorus were maintained at or above breeder guidelines. Additional synthetic amino acids were used in the diets with low protein concentration in attempt to maintain digestible amino acid supply. Diets were offered to 7 replicate pens of 15 chicks per pen from day 8 to 35. Growth performance was measured during the grower (day 8-24) and finisher (day 25-35) periods. On day 35 carcass composition was determined, blood was drawn for various biochemical measurements and the tibia was excised for mechanical and compositional analyses. Birds that received the low-protein diet had lower terminal body weight and higher feed conversion ratio compared with those that received diets with adequate crude protein content. However, addition of available phosphorus to the low-protein diet resulted in significant reductions in weight-corrected feed conversion that were not evident in the diet with adequate protein content. Bone architecture was only moderately influenced by dietary treatment but birds that ingested the diets containing low and medium protein concentrations had relatively heavier abdominal fat pad weight. Blood biochemistry, especially ammonia, uric acid, and phosphorus, was influenced by both dietary protein and available phosphorus and trends suggested that both axes are involved in protein accretion and catabolism. It can be concluded that performance losses associated with feeding low protein diets to broiler chickens may be partially restored by additional available phosphorus. The implications for use of exogenous enzymes such as protease and phytase and protein nutrition per se warrants further examination.

This study was designed to compare the effects of nutritional and growth-promoting levels of copper hydroxychloride (CH) with copper sulfate (CuSO4) on growth, carcass characteristics, tibia traits and mineral concentration in broilers fed a conventional wheat-soybean meal-based diet. Day-old Ross 308 male chicks (n = 864) were randomly assigned into 8 dietary treatments with 6 replicates of 18 chicks per treatment. The dietary treatments included a basal diet containing no supplemental copper (Cu) serving as the negative control (NC); basal diet supplemented with 15 or 200 mg/kg Cu as CuSO4; basal diet supplemented with either 15, 50, 100, 150, or 200 mg/kg Cu from CH. Diets were fed over the starter (day 1-14) and grower (day 14-35) phases. Birds in the NC group gained the same body weight and had similar feed conversion ratio (FCR) to birds receiving 15 mg/kg Cu as CuSO4, but birds receiving 15 mg/kg Cu as CH had a lower FCR than the NC birds (day 0-35; P < 0.05). Birds fed 200 mg/kg Cu as CH gained more weight (77 g/bird) and had a lower FCR (3.2 point) compared with those fed 200 mg/kg Cu as CuSO4 (P < 0.01). Based on broken-line regression models, the optimum inclusion level of Cu as CH in the diet for optimal body weight gain and FCR were estimated to be 109.5 and 72.3 mg/kg, respectively (P < 0.001). Carcass characteristics were not affected by dietary Cu sources or levels (P > 0.05). The highest and lowest tibia ash content were observed in birds fed diet with 150 mg/kg Cu as CH and 200 mg/kg Cu as CuSO4, respectively (P < 0.05). Supplementation with 200 mg/kg Cu as CH resulted in higher duodenal mucosa Cu content compared with the diet containing 200 mg/kg Cu as CuSO4 (P < 0.001). In conclusion, supplementation of Cu from CH was more efficacious than CuSO4 in promoting growth performance, both at nutritional and pharmacological levels.

A study evaluating apparent digestibilities of protein and amino acids and their corresponding digestion rates in four small intestinal sites in broiler chickens was completed to further investigate dietary optimisation via synchronised nutrient digestion and absorption. A total of 288 male Ross 308 broiler chickens were offered semi-purified diets with eight protein-rich feedstuffs, including; blood meal (BM), plasma protein meal (PPM), cold pressed (CCM) and expeller-pressed (ECM) canola meal, high (SBM HCP) and low (SBM LCP) crude protein soybean meals, lupins and peas. Diets were iso-caloric, iso-nitrogenous and the test ingredient was the sole source of dietary nitrogen. Each diet was offered to 6 bioassay cages with 6 birds per cage from day 21 to 28 post hatch. On day 28, all birds were euthanized and digesta samples were collected from the proximal jejunum, distal jejunum, proximal ileum and distal ileum to determine apparent protein and amino acids digestibility coefficients, digestion rates and potential digestible protein and amino acids. Dietary protein source significantly influenced energy utilisation, nitrogen retention, apparent protein (N) digestibilities, digestion rates and potential digestible protein along the small intestine. Diets containing BM and SBM LCP exhibited the highest protein digestion rate and potential digestible protein, respectively. Digestibility coefficients and disappearance rates of the majority of amino acids in four sections of the small intestine were influenced by dietary protein source (P < 0.01) and blood meal had the fastest protein digestion rate. In general, jejunal amino acid and protein digestibilities were more variable in comparison to ileal digestibilities, and the differences in protein and amino acid disappearance rates were more pronounced between types of feedstuffs than sources of similar feedstuffs.

An experiment was conducted to investigate the effect of supplemental L-glutamine (L-Gln) and a higher concentration of zinc (Zn) on excreta moisture under nutritionally induced wet droppings via decreased intestinal water reabsorption. A 2 × 2 × 2 factorial arrangement of treatments was used to investigate 3 dietary factors of L-Gln supplementation (0 or 10 g/kg), and added Zn concentration (80 and 160 mg/kg) with or without magnesium chloride (MgCl) (2 g/kg-only in grower diets). A total of 576 male day-old Ross 308 broiler chickens were assigned to the experimental diets. Each diet was replicated 6 times with 12 birds per replicate. Wheat-based diets were formulated to be isocaloric and isonitrogenous. Starter diets were given from day 0 to 9 followed by grower (day 10 to 23) and finisher diets (day 24 to 35). Excreta moisture was measured for all the growth phases. The moisture content of different segments of intestine was assessed for starter and grower phases of feeding. There was no significant effect of any of the 3 main treatments on body weight gain or feed intake of birds during the experiment. Birds fed higher Zn (160 mg/kg) tended (P = 0.09) to have higher weight gain only in the first 9 days of age. Feeding 10 g/kg L-Gln increased the feed conversion ratio of the birds only from hatch until day 9 after which there was no significant effect. No effect of experimental treatments was found on digesta or excreta moisture, except a reduction in ileal moisture at the starter phase resulting from higher Zn concentration in the diets. MgCl at 2 g/kg was not effective in inducing wet droppings in birds fed grower diets. Under the conditions of this study, no positive response was observed in terms of performance or reduction in excreta moisture when birds were fed diets containing 10 g/kg L-Gln or higher concentration of Zn.

This study investigated the effects of fermented soybean meal (FSBM) with or without mannan-oligosaccharide (MOS) prebiotic on growth performance, digestive functions, and hepatic IGF-1 gene expression of broiler chicken. A total of 480 day-old male broiler chickens were fed with 4 experimental diets for 6 wk. Experimental diets included corn-soybean meal diet (CON); corn-soybean meal diet + MOS prebiotic [0.2%, ActiveMOS; Biorigin, Brazile]; corn-FSBM diet [soybean meal (SBM) was totally replaced by FSBM]; and corn-FSBM + MOS prebiotic (MIX). Replacing dietary SBM with FSBM with or without MOS improved body weight gain and feed efficiency for the total grow-out period. However, the addition of MOS to the FSBM diet exhibited a greater body weight gain than other experimental diets. Villus height and villus height to crypt depth of the duodenum and jejunum were increased by feeding FSBM, MOS, and MIX diets. The ileal crude protein and energy digestibilities, as well as the activities of intestinal amylase and protease, and pancreatic protease, were improved by replacing SBM with FSBM, with or without MOS. The concentration of plasma 3-methylhistidine was reduced by FSBM and MOS, and synergistically by their combination. The MOS and FSBM diets upregulated the hepatic IGF-1 gene expression. However, there was an evident synergistic effect of FSBM supplemented with MOS in the upregulation of the hepatic IGF-1 gene expression. The outcomes of the current study indicate the FSBM and MOS had the potential to improve growth performance, hepatic IGF-1 expression, and intestinal morphology of broilers. Overall, the fermented products could be considered as functional feed that exhibits probiotic effects and the synergistic effects of prebiotics added to the fermented feeds may further improve the growth performance and gut health and functionality in broiler chicken.

The search constantly continues to identify potential alternatives to the use of antimicrobial growth promoters (AGP) in broiler production. This trial was conducted with broiler chicks to investigate the effect of different levels of Pulicaria gnaphalodes powder (PGP) in comparison with AGP, and probiotic (PRO) on growth performance, gut microflora, intestinal morphology, antioxidant enzyme activity, and fatty acid profile of meat. Ross 308 male broiler chicks (n = 576) were randomly assigned into 6 dietary treatments with 8 replicate pens per treatment and 12 birds per pen. The dietary treatments consisted of a basal diet as control (CON, with no additive), CON + 0.1% PGP, CON + 0.2% PGP, CON + 0.3% PGP, CON + 0.1% probiotic mixture (PRO), and CON + 0.05% bacitracin methylene disalicylate (AGP). Higher body weight gain and lower feed conversion ratio were obtained in birds fed AGP and 0.3% PGP compared with those fed CON and 0.1% PGP during grower, finisher, and the entire study (P < 0.05). On day 42, birds on PRO, 0.2 and 0.3% PGP treatments had lower counts of Escherichia coli and higher lactobacillus spp. in ileum and cecal contents compared to the CON and 0.1% PGP (P < 0.05). Villus height and villus height to crypt depth ratio of the duodenum were increased (P < 0.05) in response to dietary AGP, PRO, and 0.3% PGP. The diets containing PRO and different levels of PGP increased superoxide dismutase and glutathione peroxidase activities and decreased malondialdehyde level in serum, liver, and thigh muscle (P < 0.05). Total polyunsaturated fatty acid and n-3 fatty acid of birds fed PRO and PGP diets were higher than birds in CON and AGP groups (P < 0.05). In summary, supplementation of PGP could be a potential alternative to AGP in broiler diets due to its combined positive impacts on performance, serum cholesterol, intestinal health, antioxidant activity, and fatty acid profile in meat. Such effects, however, need to be further verified under compromised health or a disease challenge condition.

A total of 90 male Ross 308 broiler chicks were used in a digestibility and performance bioassay to explore the effect of reduction in dietary protein and digestible amino acids and inclusion of an exogenous mono-component protease on amino acid digestibility, net energy, jejunal gene expression, and bird performance. Four dietary treatments were created by the supplementation, or not, of 2 control diets with a mono-component exogenous protease. The control diets were corn/wheat/soybean meal-based and were formulated to be either nutritionally adequate or reduced in protein and amino acids (around 3%). The 2 control diets were supplemented with xylanase and phytase (2000 FYT). Treatments were therefore arranged as a 2 × 2 factorial design. The reduction in diet nutrient density had no significant effect on various experimental outcomes (including bird performance, amino acid digestibility, and net energy [NE]) that were measured with the exception of a reduction in the expression of aminopeptidase N and glucose transporter 2. However, the addition of exogenous protease resulted in an increase in weight gain and a reduction in feed conversion ratio (around 4%; P < 0.05) and an increase in the digestibility of several amino acids (P < 0.05) and starch (P = 0.06). Protease addition also resulted in an increase in both apparent metabolizable energy (AME) (+73 kcal/kg; P < 0.05) and NE (+107 kcal/kg; P < 0.05). The addition of exogenous protease to the diet also increased the jejunal expression of genes responsible for peptide transport (PepT2; P < 0.01) and starch digestion (sucrase isomaltase; P = 0.06). These results confirm the efficacy of exogenous protease in broiler diets that contain both xylanase and phytase and suggest substantial beneficial effects that extend beyond protein and amino acid nutrition. The effect of exogenous protease on energy partitioning, starch digestibility and the efficiency of nitrogen cycling is an area for further study.

This study was conducted to investigate and compare the effect of dietary probiotic mixture (PM) and organic acid (OA) mixture with fermented soybean meal (FSBM) on performance, crop, and ceca microbiota, small intestine morphology, and serum lipid profile in Japanese quails. A total of 800 day-old Japanese quails was randomly allotted to 5 treatments with 8 replicate pens of 20 birds each, for 35 days. The experimental diets consisted of a control corn-soybean meal diet and 4 test diets: 1) control diet + 0.1% PM; 2) control diet + 0.2% OA mixture; 3) control diet + the combination of both PM and OA; and 4) an additives-free diet in which the soybean meal in the control diet was replaced with FSBM. The results indicated that in starter and the entire rearing periods, FSBM, PM, and PM+OA diets had significantly lower FCR compared to control or OA diets (P < 0.05). Birds in the FSBM group gained higher weight than control and OA birds (P < 0.05; 1 to 35 d). At d 21 and 35, birds fed the control diet showed significantly lower numbers of lactic acid bacteria in the crop, while coliforms were higher in the cecal content compared to the other diets (P < 0.05). At d 21, the villus height and villus height to crypt depth ratio in the duodenum and jejunum of birds fed PM, PM+OA, and FSBM diets were greater than in other treatments (P < 0.05). The serum concentrations of cholesterol and low-density lipoprotein cholesterol of birds fed PM, PM+OA, and FSBM diets were significantly lower than birds in control and AO groups (P < 0.05). The results obtained herein suggest that FSBM exhibits probiotic properties and, when used in substitution of SBM in Japanese quail diet, can improve growth performance, balance of desirable gastrointestinal microbiota in crop and ceca, small intestinal morphology, and serum lipid profile-likewise, a probiotic supplement.

This study compared the efficacy of Pediococcus acidilactici, mannan-oligosaccharide, butyric acid, and their combination on growth performance and intestinal health in broiler chickens challenged with S. Typhimurium. Ross 308 male broilers (n = 420) were randomly assigned to one of the 6 treatments, resulting in 5 replicate pens of 14 chicks per treatment. The treatments included a negative control [(NC), no additive, not challenged]; positive control [(PC), no additive, but challenged with S. Typhimurium at d 3 posthatch], and 4 groups whereby birds were challenged with S. Typhimurium at d 3 posthatch and fed diets supplemented with either probiotic [0.1 g/kg Pediococcus acidilactici (PA)], prebiotic [2 g/kg mannan-oligosaccharides (MOS)], organic acid [0.5 g/kg butyric acid (BA)], or a combination of the 3 additives (MA). The S. Typhimurium challenge decreased feed intake, body weight gain and increased feed conversion ratio and reduced jejunum villus height (VH) and VH to crypt depth (CD) ratio (P < 0.05). Birds on the MA treatment exhibited similar performance to birds on the NC treatment (P > 0.05) and had a lower population of Salmonella in the ceca compared with birds on the PC treatment, at d 14 and 21 post-challenge (P < 0.05). The lowest heterophil to lymphocyte ratio was observed in birds on the MA and NC treatments (P < 0.05). Birds fed diets supplemented with MA or PA had greater VH and VH: CD ratio than birds on the PC treatment at d 7, 14 and 21 d post-challenge (P < 0.05). Suppressed amylase and protease activity was observed as a result of the S. Typhimurium challenge; the enzyme levels were restored in birds fed the additive-supplemented diets, when compared to the birds on the PC treatment, particularly at d 21 post-challenge (P < 0.05). These results indicate that dietary supplementation with a combination of PA, BA, and MOS in broiler chickens could be used as an effective tool for controlling S. Typhimurium and promoting growth performance.

Eleven canola seed (CS) samples were collected from different commercial feedmills and crushing plants in Australia and analyzed for nutrient profile. Six of these samples were selected to determine the effect of seed chemical composition and pellet temperature (PT) during steam pelleting on apparent metabolizable energy corrected for nitrogen (AMEn) values of CS for broiler chickens using a 6 × 2 factorial arrangement of treatments. The CS samples were incorporated into a corn-soybean meal diet at 15% by replacing energy-yielding ingredients, and diets were steam pelleted at either 75 or 90°C. A total of 420 18-day-old male broiler chicks (Ross 308) was assigned to 14 experimental diets replicated 6 times, with 5 chicks per cage. After a 5-day diet acclimation period from d 18 to 22, excreta were collected for 72 h using the substitution method to determine AME and AMEn. There was no interaction of seed source and PT for ileal digestible energy (IDE), AME, or AMEn values of CS (P > 0.05). PT did not affect energy availability of CS (P > 0.05) but increasing the PT improved the pellet durability index of the diets by approximately 5.0 percentage points. A significant effect of seed source was detected for all the energy utilization values of CS (P < 0.05). The IDE, AME, and AMEn values of seed samples ranged from 5,239 to 5,645, 4,728 to 5,071, and 4,501 to 4,791 kcal/kg of DM, respectively. The mean AMEn values were 4,664 kcal/kg of DM, indicating a 5.7% reduction compared with AME values. There was a negative correlation between protein and fat content of the seeds (r = -0.93, P = 0.001), and, consequently, AMEn (r = -0.32, P = 0.009). AMEn values were positively correlated with fat content of CS (r = 0.649, P = 0.001). These results indicate that fat and protein content and fiber components may have a considerable effect on energy availability of CS for broiler chickens.

The effectiveness of the addition of oat hulls (OH) as an insoluble fiber for improving nutrient digestibility and performance of birds fed diets containing full-fat canola seed (CS) was studied. A 2 × 2 × 2 factorial arrangement of treatments was used to assess the main effects of canola source (CS vs canola meal plus oil as control), OH (0 or 3%), pellet temperature (75 and 90°C) and their interactions. A total of 576 male day-old Ross 308 chickens were assigned to 8 experimental treatments, each replicated 6 times (12 birds per replicate). All birds were fed a same commercial starter diet for the first 10 d of age. Canola meal and canola oil in the control diets were replaced with CS at 11.6% and 13.5% in the grower (d 10 to 24) and finisher (d 24 to 35) diets, respectively. An interaction was observed between canola source and OH led to improved body weight gain (P < 0.01) and FCR (P < 0.05) in birds fed the combination of CS and OH in grower phase. Pelleting temperature at 75 vs 90°C did not affect performance of broilers. Birds fed diets containing OH had heavier gizzards at 24 and 35 d of age. Inclusion of CS in the diets depressed fat digestibility at d 24 (P < 0.001) and AME of the grower diets. At d 35, there was a significant interaction (P < 0.05) between CS and pellet temperature where birds fed CS diets pelleted at 75°C had higher fat digestibility than birds fed CS pelleted at 90°C. Regardless of canola source or pellet temperature, OH increased fat utilization at d 35 (P < 0.001) but had no effect on AME of the grower diets. In conclusion, CS can replace supplemental oil in broiler diets when an adequate source of insoluble fiber is included in the diet, which may help to maintain feed intake of broilers fed CS in steam-pelleted diets.

Two broiler chicken experiments were conducted to investigate the effects of canola meal (CM) replacing soybean meal (SBM) in diets supplemented with carbohydrase and protease on performance and partitioning of energy. First, a 2 × 2 × 2 factorial arrangement of treatments was employed to evaluate: protein meals (CM vs. SBM), carbohydrase (none or 300 mg/kg), protease (none or 200 mg/kg), and their interactions. Each treatment was fed to 6 replicated pens of 16 male broilers (Ross 308) from d 10 to 35. In the second experiment, 32 broiler chicks were used in a 2 × 2 factorial arrangement to investigate CM and carbohydrase effects on energy partitioning. Birds were transferred into 16 closed-circuit calorimeter chambers (4 chambers/diet; 2 birds/chamber) to measure heat production (HP), metabolizable and net energy (NE) by gaseous exchange, and total excreta collection from d 25 to 28. There were no 3-way interactions among experimental factors for any of the performance parameters measured. Birds given CM diets consumed less feed, had lower BW, and exhibited higher FCR compared to the control birds (P < 0.01). Both enzymes, alone or in combination, improved final BW and FCR (P < 0.05). There was an interaction between carbohydrase and protease for FCR over the grower period (P < 0.01), in which the combination of the enzymes resulted in further improvement of FCR. Energy, DM, and crude protein digestibility values were higher in control birds (P < 0.05). There was an interaction of protein meal and carbohydrase for HP, respiratory quotient (P < 0.05), and NE:ME ratio of the diets (P = 0.06). Inclusion of CM without carbohydrase increased HP and decreased NE and NE:ME ratio of the diets (P < 0.05). Carbohydrase decreased HP and increased retained energy (P = 0.06) and NE and NE:ME ratio (P < 0.05). In conclusion, high CM in the diet negatively affects growth performance through reduction in feed consumption, nutrient digestibility, and NE of the diet, which could partly be restored by enzyme supplementation.
