RESUMEN

Listeria monocytogenes can adapt, persist, and form biofilms on food premises surfaces, representing a challenge for food safety, since they led to disease transmission, food contamination and spoilage during production. Physical interventions (scrubbing and wiping) can help controlling formation, nevertheless when biofilms are formed, they are usually very resistant to current control strategies used in the food industry. Biofilm attachment and formation is influenced by environment characteristics, substrate properties and microbial motility. The purpose of this study was to evaluate the ability of L. monocytogenes to attach and form biofilms on different surfaces (wood, nylon, and polycarbonate) representative of the materials used during produce harvesting and storage. Multi-strain L. monocytogenes biofilms were grown in a CDC Biofilm reactor at 20 ± 2 °C up to 96-h and characterized for: a) attachment strength by enumerating cells after rinsing; b) hydrophobicity and interfacial tension by contact angle measurements; c) biofilm architecture by Laser Scanning Confocal Microscopy. All experiments were done in triplicate. Material, incubation, and solvent significantly affected the hydrophobicity and wetting properties of L. monocytogenes biofilms (P < 0.05). The type of material and incubation time significantly influenced hydrophobicity and wetting properties of L. monocytogenes biofilms (P < 0.05). Highest contact angle and lowest interfacial tension were observed on polycarbonate coupons. The data presented contributes to understanding Listeria biofilms grow on different surfaces commonly used in produce harvesting and storage. The data obtained in this study can be used when evaluating intervention strategies to control this pathogen in food premises.

Introduction: This study investigated attachment networks in a sample of Italian young adults. Attachment networks were defined in terms of attachment functions, attachment strength, the presence of a primary figure, and full-blown attachments. Method: Participants were 405 young adults, and we studied the effects of the demographic variables of gender, romantic status (whether single, involved in a romantic relationship for less or more than 24 months) and employment (whether university students or workers) on the structure of attachment networks. Participants were asked to answer the WHO-TO questionnaire, and derived indexes were analyzed using mixed ANOVAs, linear and logistic regression techniques. Results: Results indicated that while friends still had great importance in the network, partners were acquiring increasing relevance; at the same time, parents, and particularly mothers, remained central figures, particularly for the secure base function. Regarding the demographic variables, we observed that women reported stronger bonds with their mothers than men did, while the importance of friends was higher for men than for women. Additionally, our study supports previous findings underlining the importance of romantic partners in this phase of life, with participants involved in romantic relationships for longer than 24 months showing a fully developed attachment bond with their partners. Finally, for workers, the transfer of functions from the family-of-origin to external figures seemed to be fostered. Discussion: In conclusion, young Italian young adults go through a phase of intensive restructuring of attachment bond networks, particularly in relation to the consolidation of romantic relationships and work commitments.

This study explored attachment networks in committed couples who differed in parenting choice and relationship status. Attachment networks were defined in terms of attachment functions, attachment strength, the presence of a primary figure, and full-blown attachments. Participants were 198 couples, married or cohabiting, either expecting their first child or childless-by-choice. Results indicated that participants relied most strongly on partners for all attachment functions except secure base, for which they relied on mothers to a similar extent. Furthermore, expectant women reported more proximity seeking and stronger attachments to mothers, while expectant men relied more on fathers for safe haven. Married participants indicated less proximity seeking to partners than cohabiting couples, and married women reported less reliance on partners for safe haven than married men and cohabiting women. This study supports previous findings underlining the particular importance of partners for members of committed couples. Further, it extends past research by showing the robustness of this finding across parenting choice, and by revealing gender differences in the attachment networks of committed couples.

Attachment of cells to substrate surface is the premise for biofilm formation. To shelter microalgae cells from fluid shear stress and offer larger areas for microalgae attachment, the inerratic microgrooves, which can act as anchor points that offer larger areas for microalgae attachment and induce vortex to protect cells from hydraulic shear stress, were designed and fabricated into substrate surface. The results indicated that the shear stress on the surface with V-grooves was weaker than that on the surface with U-grooves, and 45° V-grooves with the width of 200 µm were benefit for cells attachment. The initial attachment time was shortened to 50 min under the hydraulic shear stress of 0.02 Pa compared to that of 135 min on the surface without microgrooves. Subsequently, the biofilm biomass concentration on the surface with 45° V-grooves increased by 14.29% to 165.84 g m-2 compared with that on flat substrates.

Sargassum horneri is one of the most important contributors to the rafts of floating seaweed in the waters off the coasts of northeastern Asia. These rafts serve as spawning and nursery grounds for many marine organisms, including Japanese saury and yellowtail. Thus, the details of the attachment/detachment mechanisms of S. horneri are of commercial significance for the aquaculture industry. Here, we describe variations in the attachment strength of S. horneri as it relates to its developmental stage and depth along a bottom gradient. We measured the attachment strength/dislodgement force of S. horneri samples with holdfast detachment in Shidagaura Cove, Shimoda, Japan, from December 2014 to May 2015. After we had determined the dislodgement forces required to detach thalli from the substratum (using a spring scale device) in the field, we transferred released individuals to the laboratory and measured selected morphological traits. Attachment strength was linearly related to the holdfast basal area when thalli were immature (prior to mid-March), but not when they were mature (mid-March to May). Thus, attachment strength was maintained through the reproductive phase and declined thereafter, allowing released thalli to join the drifting raft community. Rafting may be a mechanism by which the species expands its distribution range, as floating thalli continuing to shed germlings that are able to recruit when suitable habitat is encountered. Attachment strengths were greater in the shallows than in deeper water, reflecting the differences in wave forces experienced at different depths.

Non-native invasive species are a major threat to biodiversity, especially in freshwater ecosystems. Freshwater ecosystems are naturally rather isolated from one another. Nonetheless, invasive species often spread rapidly across water sheds. This spread is to a large extent realized by human activities that provide vectors. For example, recreational boats can carry invasive species propagules as "aquatic hitch-hikers" within and across water sheds. We used invasive gobies in Switzerland as a case study to test the plausibility that recreational boats can serve as vectors for invasive fish and that fish eggs can serve as propagules. We found that the peak season of boat movements across Switzerland and the goby spawning season overlap temporally. It is thus plausible that goby eggs attached to boats, anchors, or gear may be transported across watersheds. In experimental trials, we found that goby eggs show resistance to physical removal (90 mN attachment strength of individual eggs) and stay attached if exposed to rapid water flow (2.8 m·s(-1)for 1 h). When exposing the eggs to air, we found that hatching success remained high (>95%) even after eggs had been out of water for up to 24 h. It is thus plausible that eggs survive pick up, within-water and overland transport by boats. We complemented the experimental plausibility tests with a survey on how decision makers from inside and outside academia rate the feasibility of managing recreational boats as vectors. We found consensus that an installation of a preventive boat vector management is considered an effective and urgent measure. This study advances our understanding of the potential of recreational boats to serve as vectors for invasive vertebrate species and demonstrates that preventive management of recreational boats is considered feasible by relevant decision makers inside and outside academia.
