RESUMO

The vast majority of the energy consumed for urban water services is used to heat tap water. Heat recovery from wastewater is consequently an area of rapidly growing concern, both in research and by commercial interest, promoting the path towards a circular economy. To facilitate a system-wide evaluation of heat recovery from wastewater, this paper compares two one-dimensional models (mechanistic and conceptual) that can describe wastewater temperature dynamics in sewer pipe systems. The models are applied to successfully predict downstream wastewater temperature for sewer stretches in two Swedish cities (Linköping and Malmö). The root mean squared errors for the mechanistic model (Linköping Dataset1 - 0.33 °C; Linköping Dataset2 - 0.28 °C; Malmö - 0.40 °C) and the conceptual model (Linköping Dataset1 - 0.32 °C; Linköping Dataset2 - 0.20 °C; Malmö - 0.44 °C) indicate that both models have similar predictive capabilities, encouraging the use of conceptual models to reduce data requirements and model calibration efforts. Both models are freely distributed and can be easily integrated with wastewater generation and treatment models to facilitate system-wide wastewater temperature dynamics analysis.

OBJECTIVES: Smoking is a major contributor to mortality and morbidity in HIV-positive individuals. Our primary objective was to evaluate the association between smoking status determined by plasma cotinine (P-cotinine) concentration and inflammatory and endothelial biomarkers in HIV-positive versus HIV-negative individuals. METHODS: We studied eight inflammatory/endothelial biomarkers [high-sensitivity C-reactive protein (hsCRP), E-selectin, soluble vascular cell adhesion molecule 1 (sVCAM-1), soluble intercellular adhesion molecule 1 (sICAM-1), matrix metallopeptidase 9 (MMP-9), myeloperoxidase (MPO), tissue type plasminogen activator inhibitor 1 (tPAI) and endothelin] in 105 HIV-positive individuals and 105 HIV-negative individuals matched on age, sex and self-reported smoking status. Smoking status was determined using P-cotinine (a concentration > 14 ng/mL was defined as demonstrating exposure to smoke). We used linear regression models to (1) examine the association between smoking status and inflammatory/endothelial biomarkers in HIV-positive compared with HIV-negative individuals, and (2) to determine whether there was evidence to suggest that the impact of smoking status on the biomarkers differed between HIV-positive and HIV-negative individuals. RESULTS: Of the eight biomarkers, smokers had increased hsCRP, sICAM-1 and MMP-9 concentrations irrespective of HIV status and increasing P-cotinine concentration was associated with increasing hsCRP concentration. We found no interaction between smoking and HIV status. HIV infection was associated with increased hsCRP, E-selectin, sVCAM-1, sICAM-1 and MMP-9 concentrations. Self-reported smoking status differed substantially from smoking status assessed with P-cotinine. CONCLUSIONS: Several biomarkers were associated with smoking status and HIV status. However, our data do not indicate that the effects of smoking on the biomarkers differ between HIV-positive and HIV-negative individuals.

BACKGROUND: Knowledge about the required duration of exposure for elicitation of allergic nickel dermatitis in nickel-allergic individuals is limited. However, it often has been proposed that short skin contact is safe. OBJECTIVES: To examine whether repeated skin contact with nickel over short time periods (3 × 10 min) can elicit allergic nickel dermatitis. METHODS: Sixteen nickel-allergic adults and 10 controls were exposed to, respectively, nickel- and aluminium-containing discs on each volar forearm and on each earlobe for 3 × 10 min. One arm was pretreated for 24 h with sodium lauryl sulfate (SLS) 0·5% under occlusion before exposure. One aluminium and one nickel exposure site were clinically evaluated, and blood flow was measured with laser Doppler flowmetry at day 2 and day 4. RESULTS: Ten of 16 (63%) nickel-allergic participants developed allergic nickel dermatitis on SLS-pretreated arm skin and three of 16 (19%) developed it on normal skin on the earlobe. On the SLS-pretreated arms of nickel-allergic participants, blood flow increased significantly more on the nickel-exposed skin than on the aluminium-exposed skin on days 2 and 4. No change in clinical reactivity or blood flow was found on normal forearm skin in nickel-allergic participants or on any skin in controls. CONCLUSIONS: This experimental study showed that relatively short repeated skin contact (3 × 10 min) with metallic nickel elicits allergic nickel dermatitis in irritated skin and at sites with previous dermatitis. The results support the restrictions in current nickel regulation.

BACKGROUND/PURPOSE: Our objective was to assess epithelialization of suction blister lesions by optical coherence tomography (OCT) and benchmark it to histology using epidermal thickness (ET) as the primary outcome. METHODS: Thirty-two healthy volunteers were recruited to Study 1 and 2. One 10-mm suction blister was raised on each buttock, and the blister roof was excised. Lesions were covered with moisture-retaining dressing. In Study 1, the lesions were OCT-scanned on day 0 (D0), D2 and D4 and excised for histological examination. In Study 2, the progress of epithelialization and skin barrier function were monitored to D14. RESULTS: ET increased from D0 to D2 by 46 µm (P<.001) and from D2 to D4 by 19 µm (P=.004). Compared with histology, OCT overestimated the presence of the epithelium (P<.0001) and ET on D4. Reliable measurements were obtained when the ET of the lesions reached the ET of the normal epidermis from D5-D7 and onwards. The ET development was reflected in decreased transepidermal water loss. CONCLUSION: We found that the OCT technique was poorly discriminative with respect to the neoepithelium and the moist lesion surface material in the early postinjury period. In the later stages, OCT seemed valuable for estimating the advancement of epithelialization.

RATIONALE: There is conflicting evidence on whether patients with asthma experience an accelerated decline in lung function with age. We examined the association between postbronchodilator lung function, asthma, chronic rhinosinusitis (CRS), and atopy with age using a large European sample. METHODS: In 17 centers in 11 European countries, case-control studies were nested within representative cross-sectional surveys of adults aged less than 75 years. Representative samples of participants with asthma, CRS or both and controls were assessed for postbronchodilator ventilatory function, smoking history, atopy, and treatment. Multiple regression was used to assess the interactive effects of age and diagnostic group on decline in postbronchodilator ventilatory function. RESULTS: A total of 3337 participants provided adequate data (778 with asthma, 399 with CRS, 244 with both asthma and CRS and 1916 controls who had neither asthma nor CRS). Participants with asthma had lower FEV1 /FVC (-4.09% (95% CI: -5.02, -3.15, P < 0.001) and a steeper slope of FEV1 /FVC against age (-0.14%/annum [95%CI: -0.19, -0.08]) equivalent to smoking 1-2 packs of cigarettes per day. Those with atopy had a slope equivalent to controls. CONCLUSIONS: People with asthma have a steeper decline in postbronchodilator lung function with age, but neither CRS nor atopy alone were associated with such decline.

Around 90% of the energy requirement for urban water systems management is for heating domestic tap water. In addition, the energy content of wastewater is mainly in the form of heat (85%). Hence, there is an obvious interest in recovering a large portion of this heat. However, city-wide scenario analyses that evaluate heat recovery at various locations while considering impacts on wastewater treatment plant (WWTP) performance are currently very limited. This study presents a comprehensive model-based city-wide evaluation considering four different heat recovery locations (appliance, household, precinct and WWTP effluent) for a Swedish city with varying degrees of implementation using an uncertainty-based approach. Results show that heat recovery at the appliance level, with heat exchangers installed at 77% of the showers at domestic households, leads to a mean energy recovery of 127 MWh/day with a 0.25 °C reduction in mean WWTP inlet temperature compared to the default case without heat recovery. The highest mean temperature reduction compared to the default case is 1.5 °C when heat is recovered at the precinct level for 77% of the domestic wastewater flow rate. Finally, the impact on WWTP nitrification capacity is negligible in this case due to its large existing capacity and design.

BACKGROUND: Multinational time-trend analyses of atopic disease have shown that the East-West gradients in prevalence are shrinking. We set out to clarify whether the disparities in the occurrence of atopy and atopic diseases in Finnish and Russian Karelia during the past 10 years have diminished and how the prevalence of atopy has evolved with successive years of birth. METHODS: Two surveys with identical methodology were performed in 1997/1998 and 2007. The study population comprised randomly selected adults, aged 25-54 years, from Finnish and Russian Karelia. Serum samples were collected for total and specific IgE measurements. Clinical data were obtained by questionnaires. RESULTS: Sensitization rates to birch pollen increased from 7.8% to 14.8% (P < 0.001) and to cat from 6.1% to 10.8% (P < 0.001) in Finland. In Russia, no significant increase was found. Contrary to this, total IgE remained stable in Finland but decreased significantly (P < 0.001) in Russia. Analyses based on years of birth revealed that the prevalence of sensitization to allergens increased with successive birth years in Finland, but remained stable in Russia. Over the 10 years, self-reported physician-diagnosed asthma increased from 5.5% to 8.1% (P = 0.05) and hay fever from 8.1% to 13.2% (P < 0.001) in Finland. CONCLUSIONS: Disparities in the prevalence of atopy and atopic disease between Finnish and Russian Karelia have further grown. The 'allergy epidemic' continues in Finland and is mainly attributable to the years of birth effect shown in atopy prevalence. In Russia, no signs of the epidemic are discernible, although the decrease in total IgE may indicate a change in environmental exposure.

Parathyroid hormone (PTH) plays an essential role in osteoblast proliferation and differentiation. The effects of PTH are known to be mediated by cyclic adenosine monophosphate (cAMP) and calcium and by the activation of protein kinase C (PKC). cAMP is hydrolyzed to the inactive form 5' AMP by cyclic nucleotide phosphodiesterases (PDEs). We have investigated the role of PTH on PDE regulation in UMR-106 osteoblast-like cells. Treatment with 10 nM PTH caused a 3-fold increase in the PDE activity. The activation of PDE could be seen within 2 minutes and reached maximal levels after 20 minutes. The PTH effect was dose dependent with a half-maximal dose of 2 nM. The effect of PTH could be mimicked by the cAMP analogs Bt2 cAMP and forskolin, but not by PTH fragment 3-34, calcium ionophore A23187, or by the PKC activator phorbol 12-myristate 13-acetate. The PDE activity stimulated by PTH could be abolished by the PKA inhibitor H-8. The PDE activated by PTH was inhibitable by low concentrations of the cAMP-PDE-specific inhibitor RO 20-1724 (IC50 = 0.2 microM), but not by low concentrations of the inhibitors of cGMP-stimulated and cGMP-inhibited PDEs MEP-1 and milrinone (IC50 for both compounds > 30 microM). The PTH-stimulated cAMP accumulation was potentiated about 7-fold in the presence of RO 20-1724. H-8 potentiated the PTH-stimulated cAMP accumulation about 4-fold. Our results show that PTH rapidly stimulates the activity of cAMP-PDE in UMR-106 cells. The PDE activation involves cAMP and PKA. Inhibition of PKA can abolish the PTH-stimulated PDE activation and leads to increased accumulation of intracellular cAMP.

Sphingosine 1-phosphate (SPP) potently mobilizes sequestered calcium and is a mitogen in several cell types. In the present investigation, we have evaluated the effect of SPP on intracellular free calcium concentration ([Ca2+]i) and synthesis of DNA in thyroid FRTL-5 cells. SPP rapidly and transiently mobilized sequestered calcium and stimulated entry of extracellular calcium. The entry of calcium, but not the mobilization, was in part inhibited by pretreatment with pertussis toxin (Ptx), and by activation of protein kinase C. SPP did not stimulate the production of inositol 1,4,5-trisphosphate. SPP stimulated the incorporation of 3H-thymidine in a time- and dose-dependent manner. The effect was not inhibited by Ptx. Furthermore, SPP stimulated the activation of the proto-oncogene c-fos. SPP rapidly tyrosine-phosphorylated an approximately 66 kDa protein. This phosphorylation persisted for at least 1 h. Pretreatment of the cells with genistein abolished the SPP-evoked tyrosine phosphorylation, and attenuated the SPP-evoked increase in [Ca2+]i. Furthermore, the SPP-evoked activation of Na+-H+ exchange was inhibited by genistein. The phosphorylation was not attenuated by pretreatment of the cells with Ptx. SPP per se did not affect cellular cAMP levels but attenuated the TSH-evoked increase in cAMP. As the effect of SPP might be due to activation of phospholipase D, we tested whether phosphatidic acid (PA) mobilized calcium or stimulated the incorporation of 3H-thymidine. PA mobilized sequestered calcium but did not stimulate calcium entry. PA very modestly enhanced the incorporation of 3H-thymidine. Our results suggest, that SPP stimulates DNA synthesis and activates entry of calcium in FRTL-5 cells. The effect on calcium entry appears to be dependent, at least in part, on one or several tyrosine kinases.

Previous studies have suggested a role of cyclic guanosine 3', 5'-monophosphate (cGMP) in the differentiation and proliferation of osteoblasts. We studied the effect of ANF (atrial natriuretic factor) on intracellular cGMP accumulation, cGMP efflux, and cGMP-phosphodiesterase (PDE) activity in UMR-106 osteoblast-like cells. ANF rapidly increased both intracellular cGMP and cGMP efflux. ANF-stimulated intracellular cGMP peaked at 2 min in the absence and at 10 min in the presence of 0.25 mM 3-isobutyl-1-methylxanthine. Probenecid, an antagonist of anion transport, blocked the efflux of cGMP (IC(50) = 0.1 mM), ruling out simple diffusion as a mechanism of the efflux. cGMP-PDE activity was increased threefold in crude homogenates from ANF-treated cells (IC(50) = 23 nM). ANF-evoked stimulation of cGMP-PDE activity was reached simultaneously with the peak in intracellular cGMP. Separation of the PDEs by Q-Sepharose chromatography revealed three cGMP-hydrolyzing peaks. The first peak was sensitive to the PDE5 (cGMP-specific PDE) isoenzyme-selective inhibitor zaprinast (IC(50) = 0.45 microM). The second peak was stimulated fourfold by the addition of calcium/calmodulin, indicating the presence of PDE1. The third peak was sensitive to the PDE2 (cGMP-stimulated PDE) isoenzyme-selective inhibitor 9-[2-hydroxy-3-nonyl]adenine (EHNA) (IC(50) = 3 microM), and was activated by over 300% in the presence of 4 microM cGMP. Our results show that ANF-stimulated cGMP is released from UMR-106 cells by a probenecid-sensitive mechanism. ANF also stimulates cGMP hydrolysis by activating cGMP-PDE activity. Three distinct cGMP-hydrolyzing PDEs, namely PDE5, PDE1, and PDE2, are present in the studied cells.

The present study aimed to define the role of adenosine 3',5'-cyclic monophosphate (cAMP)-phosphodiesterase (PDE) activity and the possible involvement of cAMP efflux on parathyroid hormone (PTH)-stimulated intracellular cAMP accumulation in cultured osteoblast-like UMR-106 cells. Treatment of the cells with 10 nM PTH (1-84) rapidly increased the level of intracellular cAMP. PTH stimulation also increased the cAMP efflux rate. The efflux of cAMP could only account for a minor part of the decrease in intracellular cAMP. Six peaks of cAMP-hydrolyzing PDE activity were separated by Q-Sepharose chromatography. The first peak to elute was stimulated by Ca2+/calmodulin and provided less than 2% of the total eluted cAMP-PDE activity. The second peak, providing less than 4% of the cAMP-PDE activity, was stimulated 3-fold by 4 microM cyclic GMP (cGMP) and was sensitive to the PDE2 isoenzyme-selective inhibitor erythro-9-(2-hydroxy-3-nonyl) adenine (EHNA). The third peak, providing less than 10% of the cAMP-PDE activity, was insensitive to rolipram, EHNA, Ca2+/calmodulin, and cGMP. Peaks 4, 5 and 6 were sensitive to rolipram (IC50 < 0.1 microM) and provided approximately 85% of the total cAMP-hydrolyzing activity. It is concluded that cAMP-PDE activity in UMR-106 cells plays a major role in the control of intracellular cAMP accumulation, whereas only moderate amounts of cAMP are extruded from the cells through cAMP efflux. The main cAMP-hydrolyzing PDE isozyme is cAMP-specific/rolipram-sensitive. Ca2+/calmodulin-stimulated PDE, cGMP-stimulated PDE, and presently unidentified cAMP-specific/rolipram-insensitive PDE are also present in UMR-106 cells.

Noradrenaline and ATP evokes a transient increase in the intracellular calcium concentration ([Ca2+]i) in FRTL-5 cells. In a previous study, we showed that 1,25-dihydroxyvitamin-D3 (1,25(OH)2-D3) increases the ATP evoked changes in [Ca2+]i. In the present paper, we found that pre-incubating the cells with 10 nM 1,25(OH)2-D3 for 48 h did not affect the noradrenaline-evoked increase in [Ca2+]i. We subsequently examined if this could be due to an effect of 1,25(OH)2-D3 on alpha 1-adrenergic receptor number, or receptor affinity. Pretreatment with 10 nM 1,25(OH)2-D3 for 48 h decreased the binding of the alpha 1-adrenergic specific antagonist [3H]prazosin by 55% (Bmax for 1,25(OH)2-D3 treated = 27.6 +/- 5.0 fmol/mg protein, untreated = 61.7 +/- 5.4 fmol/mg protein). No effect of 1,25(OH)2-D3 on the affinity for [3H]prazosin was observed. The effect of 1,25(OH)2-D3 on the [3H]prazosin binding was both time- and dose-dependent and could first be seen after 8-12 h of 1,25(OH)2-D3 treatment, indicating a genomic effect. The effect of 1,25(OH)2-D3 could be abolished with the protein synthesis inhibitor cycloheximide. No effect on the [3H]prazosin binding could be seen after a 48 h preincubation with 100 nM of either 24,25-dihydroxyvitamin D3 and 25-dihydroxyvitamin D3, indicating that the effect of 1,25(OH)2-D3 was specific. The cellular cAMP concentration was decreased after 48 h treatment with 10 nM 1,25(OH)2-D3. When TSH was replaced with dibutyryl cAMP or forskolin the [3H]prazosin binding increased. 1,25(OH)2-D3 also reduced the dibutyryl cAMP and forskolin stimulated [3H]prazosin binding.(ABSTRACT TRUNCATED AT 250 WORDS)

The study is an ongoing longitudinal and qualitative psycho-social study of the communicative development in 19 preschool children with cochlear implants, using sign language. The children are video-recorded in natural interactional settings. Analysis of patterns of communication show that 16 of the children use sign language in communication with adults and peers. With regard to oral communication, 13 children were observed to utter single words or speech-like sounds upon an adult's request, but seldom used spoken words spontaneously. Six children used single spoken words in dialogues with adults if the content of the dialogue was about the here and now, and if the topic of reference was clear. None of the children in the study were able to take part in age-adequate play activities with peers when speech was used in communication. The results are discussed in reference to early mother-infant interaction, the development of communication and language, and the significance of early close relationships for children's social and emotional development.

BACKGROUND/OBJECTIVES: Comparable international data on food and nutrient intake is often hindered by the lack of a common instrument to assess food intake. The objective of this study was within the Global Allergy and Asthma European Network of Excellence (GA(2)LEN), we developed and piloted a food frequency questionnaire (FFQ) to assess its validity in Europe. SUBJECTS/METHODS: Five countries participating in GA(2)LEN took part in the pilot study. A total of 200 adults aged 31-75 years were invited to complete a FFQ in two occasions and to give a blood sample. The intra-class correlation coefficient (ICC) was used to assess repeatability of the FFQ. Plasma phospholipid fatty acids (FAs) were analysed by gas chromatography. Pearson correlation was used to analyse the correlation between estimated dietary FA intake and plasma phospholipid FA levels. RESULTS: A total of 177 participants (89%) had complete data on FFQ(1) and plasma phospholipid FAs. In all, 152 participants (76%) completed both FFQs. ICCs between macronutrients ranged from 0.70 (saturated FAs) to 0.78 (proteins) and between 0.70 (retinol) and 0.81 (vitamin D) for micronutrients. Dietary n-3 FAs showed a good correlation with total plasma phospholipid n-3 FAs and with docosahexaenoic acid in the whole sample (0.40) and in individual countries. Poor correlations were observed for other FAs. CONCLUSIONS: The GA(2)LEN FFQ is an appropriate tool to estimate dietary intake for a range of nutrients across Europe regardless of cultural and linguistic differences. The FFQ seems to be useful to estimate the intake of n-3 FAs but not other FAs.

The aim of the present study was to investigate the effect of cAMP on calcium fluxes in Fura 2 loaded thyroid FRTL-5 cells. Preincubating the cells with the phosphodiesterase inhibitor Ro-201724 decreased the ATP-stimulated entry of calcium, while having no effect on the release of sequestered calcium. Pretreatment with forskolin decreased both the release of sequestered calcium and the entry of calcium in response to ATP. We then incubated the cells with phenylisopropyl adenosine (PIA), a P2i-receptor agonist earlier shown to decrease cAMP in FRTL-5 cells. Although we did not observe a decrease in cellular cAMP after PIA, the ATP-evoked calcium response was enhanced. Forskolin decreased calcium entry induced by thapsigargin, a Ca(2+)-ATPase inhibitor, but forskolin had no effect on the thapsigargin-evoked release of sequestered calcium. Addition of calcium to cells stimulated with ATP in a calcium-free buffered resulted in a rapid influx of calcium. This response in [Ca2+]i was decreased in cells pretreated with forskolin. In cells stimulated with thapsigargin, the increase in [Ca2+]i after addition of calcium was inhibited in part by forskolin and enhanced by PIA. The results suggest that cAMP may regulate calcium fluxes in FRTL-5 cells. Furthermore, PIA increased agonist-induced calcium entry through a presently unknown mechanism.

The aim of the study was to explore patterns of communication between 22 children with cochlear implants (CI) and their parents, teachers and peers in natural interactions over a 2-year period. The children, between 2 and 5 years old when implanted, had used the implant between 1 and 3.5 years at the end of the study. Analyses of videorecorded interactions showed that meaningful oral communication was more easily obtained in the home setting than in the preschool setting. Patterns of communication between parent-child, content and complexity of dialogues, quality of peer interactions, communicative styles of adults, and the use of sign language in communication turned out to be important factors when explaining the result of the CI on the individual child's development. The children with the best oral skills were also good signers.
