This study focuses on the potential of Rhodotorula mucilaginosa CCT 7688 in simultaneous production of lipids and carotenoids in agroindustrial byproduct-based media and specially aims at establishing a process condition that guarantees high concentrations of both bioproducts, i.e., a carotenoid-rich microbial oil with potential economic value and health benefits attributed to carotenoids and fatty acids. Four different combinations of cultivation modes (batch and fed-batch) and alternative substrates (crude glycerol, sugarcane molasses and corn steep liquor) were tested. The M2-B assay, which comprises the use of an agroindustrial byproduct-based medium without any supplementation (70 g L-1 sugarcane molasses and 3.4 g L-1 corn steep liquor) and batch mode, was selected as the most promising one to produce both compounds. Total carotenoid production and total lipid content were 1794.2 µg L-1 and 43.2% (w/w), respectively, after 144 h of cultivation. The fatty acid profile showed predominance of oleic acid (69.9%) and palmitic acid (23.2%). Thus, R. mucilaginosa CCT 7688 may be used in simultaneous production of lipids and carotenoids successfully; its fatty acid profile is similar to that found in olive oil. Both compounds are economically interesting and have great possibility of future commercial applications.

OBJECTIVE: The effects of monosaccharide constituents of lignocellulosic materials on exopolysaccharide (EPS) production by Mesorhizobium sp. Semia 816 were studied. RESULTS: According to the results, by using sugars commonly found in lignocellulosic biomass as carbon sources (glucose, arabinose and xylose), no significant differences were observed in the production of EPS, reaching 3.39 g/L, 3.33 g/L and 3.27 g/L, respectively. Differences were observed in monosaccharide composition, mainly in relation to rhamnose and glucuronic acid contents (1.8 times higher when arabinose was compared with xylose). However, the biopolymers showed no differences in relation to rheological properties, with EPS aqueous-based suspensions (1.0% w/v) presenting pseudoplastic behavior, and a slight difference in degradation temperatures. Using soybean hulls hydrolysate as carbon source, slightly higher values were obtained (3.93 g/L). CONCLUSION: The results indicate the potential of the use of lignocellulosic hydrolysates containing these sugars as a source of carbon in the cultivation of Mesorhizobium sp. Semia 816 for the production of EPS with potential industrial applications.

The work herewith investigated the production of yeast biomass as a source of protein, using Yarrowia lipolytica NRRL YB-423 and raw glycerol from biodiesel synthesis as the main carbon source. A significant influence of glycerol concentration, initial pH and yeast extract concentration on biomass and protein content was observed according to the 2v (5-1) fractional design. These factors were further evaluated using a central composite design and response surface methodology, and an empirical model for protein content was established and validated. The biomass of Yarrowia lipolytica NRRL YB-423 reached 19.5 ± 1.0 g/L in shaken flasks cultivation, with a protein content of 20.1 ± 0.6% (w/w).

In the present study, a new extracellular polysaccharide (EPS-M816) was obtained during the growth of Mesorhizobium loti Semia 816 on a crude glycerol-based medium. EPS-M816 precipitate mainly consisted of carbohydrates (82.54%) and proteins (11.31%), and the weight average molecular weight was estimated at 1.646 × 106 Da. The biopolymer was characterized by FT-IR and NMR spectroscopy, and was found to have typical functional groups of other rhizobial polysaccharides. Furthermore, the rheological and emulsifying properties were investigated. The EPS-M816 solution (1.0% w/v) showed typical pseudoplastic non-Newtonian fluid behavior, and the addition of sodium and potassium chloride (1 mol L-1) increased the apparent viscosity. Regarding its emulsification activity, EPS-M816 formed emulsions with different food-grade vegetable oils (soybean, rice, canola, sunflower and corn oils), showing emulsification index values over 65% in 24 h, indicative of strong emulsion-stabilizing capacity. The biopolymer was able to form gels with texture parameters similar to those reported for xanthan gum and low syneresis. Overall, these results suggest that EPS-M816 is a good candidate for application in the food, cosmetics and pharmaceutical industries as a thickening, gelling, stabilizing and emulsifying agent.

Since carotenoids are synthesized inside the cell, it is desirable to find an efficient method to extract carotegenic pigments. This study aimed at comparing the effectiveness of different chemical and mechanical techniques to disrupt the cell wall of Sporidiobolus pararoseus and Rhodotorula mucilaginosa yeasts isolated from environmental samples. Among the techniques under study, the ultrasonic bath and the abrasion with glass beads yielded the most promising results for S. pararoseus (84.8 ± 2.3 and 76.9 ± 2.1 µg/g, respectively). The ultrasonic bath yielded the highest specific concentration of carotenoids for R. mucilaginosa (193.5 ± 25.8 µg/g), while the biomass freezing process improved neither the extractability nor the specific concentration of carotenoids. Lyophilization increased the specific concentrations of carotenoids from S. pararoseus and R. mucilaginosa by 20 and 13.7%, respectively, while the freezing process did not significantly affect (p > 0.05) the recovery of carotenoids from both yeasts; thus, it may be eliminated from the process.

Many food, cosmetic and pharmaceutical industries have increased their interest in short-chain esters due to their flavor properties. From the industrial standpoint, enzyme reactions are the most economical strategy to reach green products with neither toxicity nor damage to human health. Isoamyl butyrate (pear flavor) was synthesized by isoamyl alcohol (a byproduct of alcohol production) and butyric acid with the use of the immobilized lipase Lipozyme TL IM and hexane as solvents. Reaction variables (temperature, butyric acid concentration, isoamyl alcohol:butyric acid molar ratio and enzyme concentration) were investigated in ester conversion (%), concentration (mol L-1) and productivity (mmol ester g-1 mixture . h), by applying a sequential strategy of the Fractional Factorial Design (FFD) and the Central Composite Rotatable Design (CCRD). High isoamyl butyrate conversion of 95.8% was achieved at 24 hours. At 3 hours, the highest isoamyl butyrate concentration (1.64 mol L-1) and productivity (0.19 mmol ester g-1 mixture . h) were obtained under different reaction conditions. Due to high specificity and selectivity of lipases, process parameters of this study and their interaction with the Lipozyme TL IM are fundamental to understand and optimize the system so as to achieve maximum yield to scale up. Results show that fusel oil may be recycled by the green chemistry process proposed by this study.

Biosurfactant are surface active compounds with emulsifying capacity and are produced by microorganisms, and they may be affected by factors related to microbial cultivation, such as pH, salinity, incubation time, carbon and nitrogen sources. The aim of this work was to study the influence of the culture conditions on the production of biosurfactants by Phialemonium sp using agroindustrial wastes. The processing parameters of temperature, humidity and pH produced the most significant effects on the production of biosurfactant and emulsifying activity. The maximum concentration of biosurfactant obtained in this study was equivalent to a surface tension reduction of 8.5 mg L-1 surfactin commercial solution using wheat bran, pH of 4.5, and 0.5% of soybean oil added at 30°C. Under these conditions, 83.4 EU g-1 of emulsifying activity, 16.4 g L-1 of emulsifier index and 18.3 U g-1 lipolytic capacity were obtained.

This paper presents the evaluation of some important parameters for the purification of phycocyanin using ion exchange chromatography. The influences of pH and temperature on the equilibrium partition coefficient were investigated to establish the best conditions for phycocyanin adsorption. The equilibrium isotherm for the phycocyanin-resin system was also determined. The separation of phycocyanin using the Q-Sepharose ion exchange resin was evaluated in terms of the pH and elution volume that improved the increase in purity and recovery. The highest partition coefficients were obtained in the pH range from 7.5 to 8.0 at 25 degrees C. Under these conditions the equilibrium isotherm for phycocyanin adsorption was well described by the Langmuir model, attaining a Q (m) of 22.7 mg/mL and K (d) of 3.1 x 10(-2) mg/mL. The best conditions for phycocyanin purification using the ion exchange column were at pH 7.5 with an elution volume of 36 mL, obtaining 77.3% recovery and a 3.4-fold increase in purity.
