BACKGROUND: Tilapia is one of the most essential farmed fishes in the world. It is a tropical and subtropical freshwater fish well adapted to warm water but sensitive to cold weather. Extreme cold weather could cause severe stress and mass mortalities in tilapia. The present study was carried out to investigate the effects of cold stress on the up-regulation of antifreeze protein (AFP) genes in Nile tilapia (Oreochromis niloticus). Two treatment groups of fish were investigated (5 replicates of 15 fish for each group in fibreglass tanks/70 L each): 1) a control group; the fish were acclimated to lab conditions for two weeks and the water temperature was maintained at 25 °C during the whole experimental period with feeding on a commercial diet (30% crude protein). 2) Cold stress group; the same conditions as the control group except for the temperature. Initially, the temperature was decreased by one degree every 12 h. The fish started showing death symptoms when the water temperature reached 6-8 °C. In this stage the tissue (muscle) samples were taken from both groups. The immune response of fish exposed to cold stress was detected and characterized using Differential Display-PCR (DD-PCR). RESULTS: The results indicated that nine different up-regulation genes were detected in the cold-stressed fish compared to the control group. These genes are Integrin-alpha-2 (ITGA-2), Gap junction gamma-1 protein-like (GJC1), WD repeat-containing protein 59 isoform X2 (WDRP59), NUAK family SNF1-like kinase, G-protein coupled receptor-176 (GPR-176), Actin cytoskeleton-regulatory complex protein pan1-like (PAN-1), Whirlin protein (WHRN), Suppressor of tumorigenicity 7 protein isoform X2 (ST7P) and ATP-binding cassette sub-family A member 1-like isoform X2 (ABCA1). The antifreeze gene type-II amplification using a specific PCR product of 600 bp, followed by cloning and sequencing analysis revealed that the identified gene is antifreeze type-II, with similarity ranging from 70 to 95%. The in-vitro transcribed gene induced an antifreeze protein with a molecular size of 22 kDa. The antifreeze gene, ITGA-2 and the WD repeat protein belong to the lectin family (sugar-protein). CONCLUSIONS: In conclusion, under cold stress, Nile tilapia express many defence genes, an antifreeze gene consisting of one open reading frame of approximately 0.6 kbp.

Sediments from Lake Mariut, Egypt, after its rehabilitation, and its anoxic diverted polluted drains were subjected to five sequential steps to define different geochemical fractions of eight studied metals. Results cleared out that 30-50% of its total Cd and total Co contents are easily bioavailable with a high-risk assessment code (RAC) to enter the food chain in the lake basin. Whereas Cu and Fe are safe and the remaining studied metals, i.e., Mn, Zn, Pb, and Cr are of medium risk for the environment. Individual contamination factor (ICF) is high (> 6) for all the studied metals except for Fe and Cu which are tightly held in sediments confirming their safeness to biota. Cadmium accounted for > 94% of the total risk in the study area. Metal pollution loading (MPL) from the sediments was found in the order: Fe > Mn > Zn > Pb > Cu > Cr > Co > Cd.

Marine environment represents a promising source of new, unconventional bioactive compounds with health-promoting abilities, which can be used as food supplements. The present study was carried out to evaluate the efficacy of marine Psychrobacter namhaensis SO89 on growth performance and immune response of Nile tilapia (Oreochromis niloticus). P. namhaensis were isolated from marine environments and phylogenetically identified by 16S rRNA gene sequences. The bacterial isolate was incorporated in Nile tilapia diets (30% crude protein) at three concentrations (0.0, 0.5 and 1.0%; w/w) (designated as T0, T0.5 and T1, respectively), which were equivalent to 0.0, 2.8 × 107 and 5.6 × 107 CFU g-1 diet, respectively. The diets were fed to Nile tilapia fingerlings (4.58 ± 0.14 g average weight) at a daily rate of 3% of their live body weights (BW), 3 times a day for 50 days. The best growth rates and feed utilization efficiency were obtained at 0.5% P. namhaensis SO89 concentration. Hematocrit (Ht%), hemoglobin (Hb%), erythrocytes (RBC) and total leukocyte (WBCs) values were significantly higher in P. namhaensis SO89- fed groups than in the control group. Similarly, immunoglobulin M (IgM), alternative complement hemolysis (ACH50), phagocytic and lysozyme activities significantly increased following dietary P. namhaensis SO89 supplementation at 0.5% concentration compared to the control group. The expression of IL-4 and IL-12 genes was also significantly up-regulated in P. namhaensis SO89-treated groups up to 0.5% concentration. Increasing bacterial concentration to 1% resulted in a significant decrease in fish performance and immune response. The present results suggest that marine psychrotolerant (Psychrobacter namhaensis) can be considered as a novel feed additive in Nile tilapia feeds.

ETHNOPHARMACOLOGICAL RELEVANCE: Oxalis corniculata (O. corniculata) is a member of Oxalidaceae family, widely distributed in Asia, Europe, America, and Africa, used extensively as food and its traditional folkloric uses include management of epilepsy, gastric disorders, and neurodegenerative diseases, together with its use in enhancing health. Numerous pharmacological benefits of O. corniculata are linked to its anti-inflammatory and antioxidant abilities. One of the most prevalent neurodegenerative disorders is Alzheimer's disease (AD) in which neuroinflammation and oxidative stress are its main pathogenic processes. AIM OF THE STUDY: Our research aimed to study the neuroprotective effect of the methanolic extract of Oxalis corniculata Linn. (O. corniculata ME), compared to selenium (Se) against AlCl3-induced AD. MATERIALS AND METHODS: Forty male albino rats were allocated into four groups (Gps). Gp I a control group, the rest of the animals received AlCl3 (Gp II-Gp IV). Rats in Gp III and IV were treated with Se and O. corniculata ME, respectively. RESULTS: The chemical profile of O. corniculata ME was studied using ultraperformance liquid chromatography-electrospray ionization-quadrupole time-of-flight mass spectrometry, allowing the tentative identification of sixty-six compounds, including organic acids, phenolics and others, cinnamic acid and its derivatives, fatty acids, and flavonoids. AlCl3 showed deterioration in short-term memory and brain histological pictures. Our findings showed that O. corniculata ME and selenium helped to combat oxidative stress produced by accumulation of AlCl3 in the brain and in prophylaxis against AD. Thus, Selenium (Se) and O. corniculata ME restored antioxidant defense, via enhancing Nrf2/HO-1 hub, hampered neuroinflammation, via TLR4/NF-κß/NLRP3, along with dampening apoptosis, Aß generation, tau hyperphosphorylation, BACE1, ApoE4 and LRP1 levels. Treatments also promoted autophagy and modulated Wnt 3/ß-catenin/GSK3ß cue. CONCLUSIONS: It was noted that O. corniculata ME showed a notable ameliorative effect compared to Se on Nrf2/HO-1, TLR4/NF-κß/NLRP3, APOE4/LRP1, Wnt 3/ß-catenin/GSK-3ß and PERK axes.

Attention deficit hyperactivity disorder (ADHD) has high incidence rate among children which may be due to excessive monosodium glutamate (MSG) consumption and social isolation (SI). AIM: We aimed to explore the relationships between MSG, SI, and ADHD development and to evaluate the neuroprotective potential of Punicalagin (PUN). METHODS: Eighty male rat pups randomly distributed into eight groups. Group I is the control, and Group II is socially engaged rats treated with PUN. Groups III to VII were exposed to ADHD-inducing factors: Group III to SI, Group IV to MSG, and Group V to both SI and MSG. Furthermore, Groups VI to VIII were the same Groups III to V but additionally received PUN treatment. KEY FINDINGS: Exposure to MSG and/or SI led to pronounced behavioral anomalies, histological changes and indicative of ADHD-like symptoms in rat pups which is accompanied by inhibition of the nuclear factor erythroid 2-related factor 2 (Nrf2)/Heme-oxygenase 1 (HO-1)/Glutathione (GSH) pathway, decline of the brain-derived neurotrophic factor (BDNF) expression and activation of the Toll-like receptor 4 (TLR4)/Nuclear factor kappa B (NF-kB)/NLR Family Pyrin Domain Containing 3 (NLRP3) pathway. This resulted in elevated inflammatory biomarker levels, neuronal apoptosis, and disrupted neurotransmitter equilibrium. Meanwhile, pretreatment with PUN protected against all the previous alterations. SIGNIFICANCE: We established compelling associations between MSG consumption, SI, and ADHD progression. Moreover, we proved that PUN is a promising neuroprotective agent against all risk factors of ADHD.

Microbial bioremediation of heavy metals-polluted industrial effluents has been adopted as one of the most effective eco-friendly tool to cope up with the harmful effects of metals. This study was designed to investigate the biosorption potential of marine actinomycetes isolated from the Alexandrian Mediterranean Seacoast, Egypt, with their potential use in metal remediation of industrial effluents. Among the nine marine actinomycetes isolates, Streptomyces rochei ANH showed the highest versatile metal resistance capability with MIC values of 125 mg/l for Cr6+ and 60 mg/l for both Cd2+ and Pb2+. Additionally, scanning electron micrographs showed complete disintegration of Cr6+-treated biomass compared with the control ones where spores remained intact and connected in long chains. The study also aimed to improve the percentage of Cr6+ biosorption by S. rochei ANH biomass using the statistical designs of Plackett-Burman and Box-Behnken where up to 85% of Cr6+ removal was recorded under the following conditions: pH (5), incubation temperature (30 °C), contact time (3 h), agitation speed (90 rpm), initial Cr6+ concentration (50 mg/l) and living biomass concentration (10 mg/ml). The results also showed that the percentage of Cr6+ biosorption by S. rochei ANH decreased gradually beyond these values. Moreover, the results revealed that the use of the biomass of S. rochei ANH is an effective biotechnological agent for the biological treatment of heavy metal-contaminated tannery effluent where the percentages of metal removal were in the following order: Ni2+ (100%) ≥ Cu2+ ≥ Mn2+ ≥ Fe2+ > Pb2+ (95%) ≥ Cd2+ > Cr6+ (86%). Furthermore, the treated effluent exhibited a stimulating effect on the germination process of Lepidium sativum seeds. Therefore, the present study implies that S. rochei ANH can be considered a powerful candidate to mitigate hazardous heavy metals pollution from industrial effluents and improve the water quality for agricultural purposes.

Heavy metal pollution is one of the most serious environmental and human health risk problem associated with industrial progress. The present study was conducted with the goal of isolation and characterization of metal-resistant lactic acid bacteria (LAB) from the Alexandrian Mediterranean Seacoast, Egypt, with their possible exploitation in metal remediation. Lactobacillus plantarum MF042018 exhibited high degree of resistance, up to 500 and 100 ppm, to both nickel and chromium, respectively, with multiple antibiotic resistance (MAR) index above 0.5. In an attempt to improve chromium removal by L. plantarum MF042018, Plackett-Burman followed by Box-Behnken statistical designs were applied. An initial Cr2+ concentration of 100 ppm and inoculum size of 3% presented the best conditions for the accumulation of chromium by L. plantarum MF042018. The study was also navigated to assess the biosorption capacity of L. plantarum MF042018, the maximum uptake capacity (q) of both Cd2+ and Pb2+ was recorded at pH 2.0 and a temperature of 22 °C after 1 hr. The biosorption process of Cd2+ and Pb2+ was well explained by the Langmuir isotherm model better than the Freundlich isotherm. Furthermore, the results revealed that the use of L. plantarum MF042018 is an effective tool for the treatment of hazardous metal-polluted battery-manufacturing effluent. Therefore, the present study implies that L. plantarum MF042018 can be applied as a promising biosorbent for the removal of heavy metals from industrial wasterwaters.

The water quality of Lake Mariut has been deteriorated for about 5 decades due to continuous discharge of agricultural, municipal, and industrial wastes from Alexandria City and the adjacent land. During the past two decades, some steps were taken for rehabilitation of the lake through primary treatment of the discharged wastes and insulation of the polluted wastewater of QD from the water body in the main basin of the lake. Several parameters of water quality at the surface and near bottom were measured at twelve locations during winter (January) and summer (August) of 2013 and 2014. The present study revealed that the lake water appeared to be well aerated but still containing high concentrations of N and P and suffering hyper-eutrophic conditions. The water quality index (WQI) reflected medium condition in the lake and bad condition in the diverted drains. The P-budget calculation displayed that TP input into the LMMB from UDus and resuspension process from sediments exceeded the output by out flowing at UDds and settling.

A 50-day feeding trial was conducted to evaluate the effects of dietary supplementation of a novel marine psychrotrophic bacterium, Psychrobacter maritimus S, on growth performance and immune responses of Nile tilapia (Oreochromis niloticus) fingerlings. Triplicate groups of Nile tilapia (10 fish each) were fed for 50 days with three different diets: T0 (without probiotic, negative control), T0.5 (supplemented with 3.3 × 108 CFU g-1P. maritimus S), and T1 (supplemented with 6.6 × 108 CFU g-1P. maritimus S). The test diets were fed to fish at a daily rate of 3% of their live weights, three times per day. On the 50th day, fish were weighed and blood samples were collected to determine the main nonspecific humoral and cellular immune responses and digestive enzymes (protease, amylase, and lipase). Fish performance (weight gain, specific growth rate, feed conversion ratio, protein efficiency ratio, and protein productive value) was also calculated. Growth rates and digestive enzyme activity were significantly increased (P < 0.05) with increasing dietary P. maritimus S up to 0.5%, and leveled off with further increase in bacterial concentrations. Phagocytic activity, lysozyme activity, alternative complement hemolysis, and hematological parameters were also significantly increased (P < 0.05) with increasing P. maritimus S concentration to 0.5%, followed by a slight decrease (P > 0.05) at 1% level. The expression of interleukin-4 and interleukin-12 genes was significantly upregulated, while heat shock protein gene was downregulated, with dietary supplementation of P. maritimus S up to 0.5% level. These results suggest that 0.5% of dietary Psychrobacter maritimus S supplementation could be considered as a novel probiont for optimum growth performance and immune response of Nile tilapia fingerlings.
