Hemorrhagic, oedema-forming activities and histopathological alterations in the mouse footpad induced by Bothrops and Crotalus snake venoms from Argentina. Hemorrhagic and oedema-forming activities of various Bothrops and Crotalus snake venoms from Argentina were studied, together with histological alterations in the mouse footpad. The highest oedema-forming activity was found in the venom of B. jararaca, followed by B. jararacussu, B. neuwiedii diporus, B. alternatus, and Crotalus durissus terrificus. Regarding hemorrhage, the highest activity was found in the venom of B. neuwiedii diporus, followed by B. jararacussu, B. alternatus, and B. jararaca. No hemorrhage was observed after injection of Crotalus durissus terrificus venom, in agreement with histological observations of injected footpads. Histological analysis revealed a conspicuous inflammatory reaction in the injected footpad, characterized by oedema and an inflammatory infiltrate rich in polymorphonuclear leucocytes. Necrotic blood vessels and dilated lymphatic vessels were observed after injection of B. jararaca and B. jararacussu venoms, and myonecrosis was evident in tissue of mice injected with B. alternatus and B. neuwiedii diporus.

Con el objeto de identificar signos precursores de alteraciones del comportamiento en cachorros, se evaluaron respuestas conductuales durante la primera consulta clínica, coincidiendo con la primo vacunación. Se trabajó con caninos de ambos sexos (n = 31), entre 30 y 60 días de edad, tanto mestizos como de razas puras. Siguiendo la secuencia en la exploración clínica de rutina, al ingresar se recolectaron datos sobre exploración ambiental (Exp) sobre el suelo y luego las respuestas a maniobras sobre la camilla (Mc) y de sujeción y sumisión forzadas (Sf) efectuadas por el profesional. Durante la anamnesis se realizaron preguntas referentes a miedo frente a estímulos novedosos, tiempo de descanso y actitudes durante el juego. El 74% de los cachorros regresó junto al humano conocido (propietario) luego de explorar el ambiente en el suelo. En la camilla, el 81% aceptó el contacto físico iniciado por el veterinario y un 84% aceptó la sumisión forzada. Ningún cachorro intentó escapar y sólo uno presentó reacciones neurovegetativas. Frente a estímulos novedosos, el 61% no presentó miedo y, cuando se presentó la respuesta, el orden de susceptibilidad fue a ruidos, objetos móviles y personas desconocidas. Frente al descanso, 71% de los propietarios no los escuchó llorar durante la noche y durante el juego, 61% de los cachorros dejó de morder ante la solicitud del propietario. En la primera consulta la mayoría de los cachorros no presentó signos precursores de alteraciones de la conducta. La aplicación de maniobras semiológicas sencillas -observación del comportamiento exploratorio, posición de sumisión, reacción a la palpación y auscultación- brindó información sobre hiperactividad, miedo, ansiedad o socialización inadecuada. Esta información fue utilizada como base para hacer modificación de la conducta. Con intervenciones tempranas el veterinario puede, no sólo mejorar la relación humano/animal, sino también el bienestar de los perros urbanos.


With the object of identify early precursors of any conduct disorder on puppies' behaviour when they assisted to the primo vaccination. Thirty one (n = 31) puppies with 30 and 60 days of age, belonging to both sexes and several breeds were observed. According to the clinical examination routine sequence, data from the floor environmental exploration (Exp), stretcher manipulation (Sm), subjection and forced submission (Fs) by the professional were registered. In addition, data from oral responses by owners regarding puppy reaction to novel stimuli, resting, and play episodes were evaluated. 74% percent of puppies returned to the well-know human (owner) after the floor environmental exploration. At the stretcher phase, 81% did accept physical contact initiated and the 84% did accept the forced submission initiated by the professional. None puppy attempt to run away and only one showed neurovegetative responses. Regarding to novel stimuli reaction, 61% of puppies did no display any fear signals and for the rest, the susceptibility ranking was: noises, mobile objects and the unfamiliar human items. Regarding to resting episodes, 71% of the owners did not listened their puppies weep at nights, and for the play episodes 61% of puppies stopped biting at the owner request. During the first consultation, most of the dogs did not present any precursor sign of behavior alteration. The appliance of easy semiologic procedures -observation of the exploratory behaviour, submission position, answer to the palpation and auscultation- give information about hyperactivity, fear, anxiety and inadequate socialization. This information was used to made behaviour modification. With early intervention, the veterinary can improve the human/animal relationship and the urban dogs welfare.

The therapeutic effects of Argentine propolis ear drop formulation on canine otitis externa were evaluated. Forty-eight dogs with symptoms of otitis externa were randomly assigned to double-blinded, controlled clinical trial to evaluate the efficacy of topical formulation with propolis versus a topical placebo in the treatment of otitis externa. The propolis preparation and placebo were administrated into both external ear canals, twice daily for 14 days. Throughout the study, clinical examination and microbiological analysis of dogs ear exudates were made. The most frequent microorganisms isolated in culture media were: Malassezia pachydermatis (54.2 percent), Staphylococcus aureus (43.8 percent), coagulase-negative Staphylococcus (25.0 percent), Pseudomonas aeruginosa (20.8 percent), Candida albicans (18.8 percent), Proteus mirabilis (16.7 percent), Streptococcus spp. (16.7 percent), Enteroccocus faecalis (12.5 percent), Escherichia coli (12.5 percent), Staphylococcus intermedius (6.3 percent), Klebsiella spp. (4.2 percent), andCandida glabrata (2.1 percent). Whereas the control group did not recover from the infectious ear disease, the propolis preparation exhibited antimicrobial activity against most of the microorganisms isolated from samples of the treated group. In addition, no propolis-adverse effects were observed. This allowed propolis-treated patients to show a significant improvement of the clinical parameters. Thus, this new Argentine propolis ear drop formulation may be used for topical treatment of otitis externa in dogs.


Os efeitos terapêuticos da formulação em gotas óticas de própolis procedentes da Argentina foram avaliados no tratamento da otite externa canina. Quarenta e oito cães com sintomas de otite externa foram distribuídos aleatoriamente em ensaio clínico duplo-cego controlado para avaliar a eficácia da formulação tópica com a própolis contra um placebo tópico no tratamento da otite externa. A preparação de própolis e placebo foi administrada em ambos os canais da orelha externa, duas vezes por dia, durante 14 dias. Ao longo do estudo, os cães foram submetidos a exame físico e à análise microbiológica de exsudatos auriculares. Os mais frequentes microrganismos isolados em meios de cultura foram: Malassezia pachydermatis (54,2 por cento), Staphylococcus aureus (43,8 por cento), Staphylococcus coagulase-negativo (25,0 por cento), Pseudomonas aeruginosa (20,8 por cento), Candida albicans (18,8 por cento), Proteus mirabilis (16,7 por cento), Streptococcus spp.(16,7 por cento), Enterococcus faecalis (12,5 por cento), Escherichia coli (12,5 por cento), Staphylococcus intermedius (6,3 por cento), Klebsiella spp.(4,2 por cento) e Candida glabrata (2,1 por cento). A preparação de própolis apresentou atividade antimicrobiana contra a maioria dos microrganismos isolados de amostras do grupo de tratamento, sendo que os do grupo-controle não se recuperaram da doença infecciosa auricular, e não foram observados efeitos adversos à própolis. Isso permitiu aos pacientes tratados com própolis melhora significativa dos parâmetros clínicos. Essa nova formulação da própolis argentina para o ouvido apresenta potencial utilidade no tratamento tópico da otite externa em cães.

Authors tried to find out a correlation between chronological age, inorganic phosphorus (P), calcium (Ca) and alkaline phosphatase (ALP) with some radiological signs obtained from a periodical study of different sexes and breeds (small-middle-large) of dogs right foreleg from weaning to the end of osseous growth. They confirmed the period when ossification centres come in sight. The first centre appears at 3 weeks of life (humerus) and the last one at 8-10 weeks (scapular tuberosity). Epiphyseal fusion begins with the accessory carpal bone (16-20 weeks). Osseous development finished around 40 weeks with the fusion of proximal humerus and radius epiphyseal plates. During the work the ALP and P serum decreased values were significant (p less than 0.001), but Ca were not significant (p less than 0.05). There were good correlations with chronological age and both P (r = 0.98) and ALP values (r = 0.90). There was none correlation between age and Ca. When the osseous growth finished, Ca and P reached adult levels, but ALP was still elevated.

Mice of 18 and 20 g were injected intradermally with 0.1 ml of serial dilution of venom in saline solution 0.9 buffered pH 7.2. Groups of 4 animal were formed, they were sacrificed 2 hours after inoculation. Skin of every mouse was put out, and the haemorrhagic area was measured Bothrops alternatus, Bothrops jararaca, Bothrops jararacussu and bothrops neuwiedii venoms were used. Crotalus durissus terrificus did not show any haemorrhagic activity.

RESUMO

Bothrops alternatus venom was inoculated in gastrocnemius muscle of mice between 18 and 20g. The dose was 50 micrograms in 0.1 ml in ClNa 0.85% solution. Groups of 5 animals were sacrified between 3, 6 and 12 hours after inoculation. Hystopathology was performed with muscle and different organs fixed with Bouin. The venom reproduced a local inflammatory reaction. Histopathology observations of muscle revealed myolytic and coagulative necrosis. There and six hours after inoculation, hepatocellular degeneration and tumefaction in area of central veins was observed. In kidneys there were cortical congestion and hydropic tumefaction of proximal and distal tubules. There were no anormality in heart, lung and brain.

Toxic and enzymatic activity of Bothrops moojeni venom from Argentine. Hemorrhagic, edema forming, proteolytic and indirect hemolytic activities, myonecrosis and muscle regeneration using optic microscopy induced by Bothrops moojeni snake venom from Argentina were studied. The described activities were present in this venom. Myonecrosis was intense 60 minutes after venom injection, increasing its activity during the first 3 hours. Myolytic lesions rise above coagulative ones. Hemorrhage was minimum the same as the edema. After 3 hours polymorphonuclear inflammatory infiltrate was observed, with neutrophils. Macrophages were visible after 48 hours. After 3 days fibroblasts were present. At the 7 day muscle fiber regeneration were present, showing striations alternating with calcified areas. Bothrops moojeni venom injuries are intense and the recuperation of them can generate important tissue lose, with functional consequences.

RESUMO

Myonecrotic and oedema-inducing activities of Bothrops jararaca of Argentina were studied. For oedema-inducing activity 0.05 ml of different solutions of venom in 0.9% NaCl were injected in mice. The dose of 0.86 micrograms/20 g mouse induced an oedema of 30% respect the other member in one hour. The myonecrotic effects were studied injecting mice gastrocnemius muscle with 70 micrograms of venom in 0.1 ml of 0.9% NaCl, which were sacrificed in different time. The inoculation area was obtained for hystophatological process. Animal sacrificed 30 minutes after the inoculation showed oedema, hemorrhage and inflammatory infiltrate. Those sacrificed in one and three hours after the inoculation also had got necrosis.

A study of hemorrhagic, edema-forming, proteolytic and myonecrotic activities induced by newborn Bothrops alternatus snake venoms from Argentina was performed. The analyzed venom had these activities with markedly differences when compared with the venom of adult specimens. Hemorrhagic activity was four times higher, while edema forming activity was ten times or more. But proteolytic activity was similar in both groups. Hystological analysis showed intense hemorrhage and muscular fiber myolisis after 60 minutes of venom injection. Myonecrosis and inflammatory exudate were higher in the following hours. Hemorrhage decreased after 24 hours and muscular fiber regeneration started after the first week with granulation tissue formation. On the fourth week regenerating muscular fiber areas and granulation tissue were still observed. As a conclusion, we can assume that the injury induced in the snake accident by newborn Bothrops alternatus snakes would be more intense than those induced by the adult ones.

Descreve-se a sintomatologIa e o tratamento de dois cäes picados por cascavel da America do Sul (Crotalus durissus terrificus) na Argentina. Sinais neurológicos foram evidentes poucos minutos após o acidente, incluindo anestesia local, ataxia do membro afetado e fascia neurotóxica. As enzimas aminotransferase da alanina, aminotransferase do aspartato, quinase da creatinina, desidrogenase de lactato e o cálcio foram determinados para analisar dano muscular. Falha rEnal näo foi observada, mas algumas alteraçöes foram detectadas na urina, incluindo densidade baixa, sedimento com grumos granulares e células pequenas redondas. Biópsias de músculo foram obtidas de ambas as pernas para histopatología, revelando edema e fibras necróticas isoladas. Ambos os cäes receberam tratamento quatro horas após o acidente, pela via intravenosa. A antitoxina foi administrada diluída em 250ml de soluçäo Ringer lactato na dose capaz de neutralizar 8mg de toxina. Dexametasona foi aplicada antes de administrar a antitoxina. A evoluçäo clínica foi boa e ambos os pacientes estavam com boa saúde no segundo dia após a acidente

Bothrops alternatus venom was intramuscularly inoculated (3mg/kg) into 12 dogs, 30 to 65 days old. Spontaneou bleeding commenced twenty minutes later. Blood samples obtained 3 and 20 minutes after venom inoculation presented spontaneuous clotting formation. Plasmatic fibrinogen decreased within 3 minutes. Partial thromboplastin time (PTT) and one-stage prothrombin time (PT) were found. Plasma did not coagulate 40 minutes after inoculation. Platelet counts did not vary but their function was altered. Histopathology pointed out severe muscular necrosis and massive hemorrhage in the inoculation area. Regional ganglia showed intense hemorrhage. The 45 and 65-day-old animals showed alveolar thickening of the septum and generalized congestion, but the 30-day-old animals showed thrombosis of small arteries and arterioles. Renal lesions were different with the age. Cortical tubular necrosis was present in puppies, and intense cortical tubular hydropic degeneration was present in adult dogs. Thymus hemorrhage and necrosis were present.

Se realizó el estudio de las actividades hemorrágica, edematizante, proteolítica y mionecrótica inducida por veneno de viboreznos de Bothrops alternatus de Argentina. Los venenos estudiados poseen las actividades mensionadas, mostrando marcadas diferencias respecto al veneno de ejemplares adultos. Cuadriplica la capacidad hemorrágica, supera en más de diez veces la edematizante. La actividad proteolítica es la que menor diferencia posee. El análisis histológico mostró intensa hemorragia y miólisis de fibras musculares a los 60 minutos de exposición al veneno. La mionecrosis y el exudado inflamatório fueron más altos en las siguientes horas. La hemorragia disminuyó de 24 horas y la regeneración de las fibras musculares se inició en la primer semana con formación de tejido de granulación. A la cuarta semana se observaban áreas de regeneración de fibras musculares y aún se observada tejido de granulación. Se concluye que la injuria producida en los accidentes por viboreznos de Cothrops alternatus es más intensas que la inducida por ejemplares adultos de la misma especie.

Se estudiaron las actividades hemorrágica, edematizante, proteolítica, hemolítica indirecta (in vitro) y por microscopía óptica la mionecrosis y regeneración de fibras musculares inducidas por veneno de Bothrops moojeni de Argentina. El veneno demostró poseer las actividades mensionadas. La actividad mionecrótica fue intensa a partir de los 60 minutos de exposición incrementando su intensidad a las 3 horas con predominio de lesiones de fibras musculares de tipo miolótico, respecto a necrosis coagulativa. En los cortes histológicos, la hemorragia fue mínima al igual que el edema, mientras que a las 3 horas se observó infiltrado inflamatorio polimorfonuclear con predominio de neutrófilos y a partir de las 48 horas se sumaron los macrófagos. Al tercer día se observaron fibroblastos, los que se incrementaron paulatinamente; a los 7 días se observó regeneración de algunas fibras musculares que presentaban estriaciones, a partir de la curta semana, éstas alternaban con áreas calcificadas. La recuperación de las lesiones fue parcial, con fibras musculares de menor diámetro que las normales, alterando con áreas de calcificación y otras de fibrosis. Las lesiones causadas por el veneno de Bothrops moojeni son intensas y por la recuperación parcial de las mismas, puede generar pérdida importante de tejidos, con secuelas funcionales.

Se estudiaron las actividades hemorrágica, edematizante, proteolítica, hemolítica indirecta (in vitro) y por microscopía óptica la mionecrosis y regeneración de fibras musculares inducidas por veneno de Bothrops moojeni de Argentina. El veneno demostró poseer las actividades mensionadas. La actividad mionecrótica fue intensa a partir de los 60 minutos de exposición incrementando su intensidad a las 3 horas con predominio de lesiones de fibras musculares de tipo miolótico, respecto a necrosis coagulativa. En los cortes histológicos, la hemorragia fue mínima al igual que el edema, mientras que a las 3 horas se observó infiltrado inflamatorio polimorfonuclear con predominio de neutrófilos y a partir de las 48 horas se sumaron los macrófagos. Al tercer día se observaron fibroblastos, los que se incrementaron paulatinamente; a los 7 días se observó regeneración de algunas fibras musculares que presentaban estriaciones, a partir de la curta semana, éstas alternaban con áreas calcificadas. La recuperación de las lesiones fue parcial, con fibras musculares de menor diámetro que las normales, alterando con áreas de calcificación y otras de fibrosis. Las lesiones causadas por el veneno de Bothrops moojeni son intensas y por la recuperación parcial de las mismas, puede generar pérdida importante de tejidos, con secuelas funcionales. (AU)

RESUMO

A study of hemorrhagic, edema-forming, proteolytic and myonecrotic activities induced by newborn Bothrops alternatus snake venoms from Argentina was performed. The analyzed venom had these activities with markedly differences when compared with the venom of adult specimens. Hemorrhagic activity was four times higher, while edema forming activity was ten times or more. But proteolytic activity was similar in both groups. Hystological analysis showed intense hemorrhage and muscular fiber myolisis after 60 minutes of venom injection. Myonecrosis and inflammatory exudate were higher in the following hours. Hemorrhage decreased after 24 hours and muscular fiber regeneration started after the first week with granulation tissue formation. On the fourth week regenerating muscular fiber areas and granulation tissue were still observed. As a conclusion, we can assume that the injury induced in the snake accident by newborn Bothrops alternatus snakes would be more intense than those induced by the adult ones.

RESUMO

Toxic and enzymatic activity of Bothrops moojeni venom from Argentine. Hemorrhagic, edema forming, proteolytic and indirect hemolytic activities, myonecrosis and muscle regeneration using optic microscopy induced by Bothrops moojeni snake venom from Argentina were studied. The described activities were present in this venom. Myonecrosis was intense 60 minutes after venom injection, increasing its activity during the first 3 hours. Myolytic lesions rise above coagulative ones. Hemorrhage was minimum the same as the edema. After 3 hours polymorphonuclear inflammatory infiltrate was observed, with neutrophils. Macrophages were visible after 48 hours. After 3 days fibroblasts were present. At the 7 day muscle fiber regeneration were present, showing striations alternating with calcified areas. Bothrops moojeni venom injuries are intense and the recuperation of them can generate important tissue lose, with functional consequences.
