RESUMO

Elevated interleukin-6 (IL-6) levels are associated with type 2 diabetes, but its role in glucose metabolism is controversial. We investigated the effect of IL-6 on insulin-stimulated glucose metabolism in type 2 diabetes patients and hypothesized that an acute, moderate IL-6 elevation would increase the insulin-mediated glucose uptake. Men with type 2 diabetes not treated with insulin [n = 9, age 54.9 ± 9.7 (mean ± SD) yr, body mass index 34.8 ± 6.1 kg/m(2), HbA1c 7.0 ± 1.0%] received continuous intravenous infusion with either recombinant human IL-6 (rhIL-6) or placebo. After 1 h with placebo or rhIL-6, a 3-h hyperinsulinemic-isoglycemic clamp was initiated. Whole body glucose metabolism was measured using stable isotope-labeled tracers. Signal transducer and activator of transcription 3 (STAT3) phosphorylation and suppressor of cytokine signaling 3 (SOCS3) expression were measured in muscle biopsies. Whole body energy expenditure was measured using indirect calorimetry. In response to the infusion of rhIL-6, circulating levels of IL-6 (P < 0.001), neutrophils (P < 0.001), and cortisol (P < 0.001) increased while lymphocytes decreased (P < 0.01). However, IL-6 infusion did not change glucose infusion rate, rate of appearance, or rate of disappearance during the clamp. While IL-6 enhanced phosphorylation of STAT3 in skeletal muscle (P = 0.041), the expression of SOCS3 remained unchanged. Whole body oxygen uptake (P < 0.01) and expired carbon dioxide (P < 0.01) increased during rhIL-6 infusion. In summary, although IL-6 induced local and systemic responses, the insulin-stimulated glucose uptake was not affected. While different contributing factors may be involved, our results are in contrast to our hypothesis and previous findings in young, healthy men.

RESUMO

Yb-doped double-cladding large mode area rod-type photonic crystal fibers are a key component for power scaling in fiber laser systems. Recently, designs with 19-cell core defect, that is with 19 missing air-holes in the center of the photonic crystal cladding, have been proposed, with reported core diameter up to 100 µm. In this paper an analysis of the cut-off wavelength of the first high-order mode in such low-NA fibers is reported, accounting for different approaches for the definition of the cladding effective index. Results have shown that taking into account the finite fiber cross-section and considering the first cladding mode of the actual fiber is mandatory to obtain a correct estimate of the cut-off wavelength.

Exercise-induced adaptations in skeletal muscle oxidative enzymes are suggested to result from the cumulative effects of transient changes in gene expression after each single exercise session. However, for several oxidative enzymes, no changes in mRNA expression are detected up to 8 h after exercise. To test the hypothesis that mRNA expression of many oxidative enzymes is up-regulated late in recovery (10-24 h) after exercise, male subjects (n=8) performed a 90-min cycling exercise (70% VO(2-max)), with muscle biopsies obtained before exercise (pre), and after 10, 18 and 24 h of recovery. The mRNA expression of carnitine-palmitoyltransferase (CPT)I, CD36, 3-hydroxyacyl-CoA-dehydrogenase (HAD), cytochrome (Cyt)c, aminolevulinate-delta-synthase (ALAS)1 and GLUT4 was 100-200% higher at 10-24 h of recovery from exercise than in a control trial. Exercise induced a 100-300% increase in peroxisome proliferator-activated receptor gamma co-activator (PGC)-1alpha, citrate synthase (CS), CPTI, CD36, HAD and ALAS1 mRNA contents at 10-24 h of recovery relative to before exercise. No protein changes were detected in Cytc, ALAS1 or GLUT4. This shows that mRNA expression of several training-responsive oxidative enzymes is up-regulated in human skeletal muscle at 10-24 h of recovery, supporting that exercise-induced adaptations of these oxidative enzymes can be the result of the cumulative effects of transient changes in mRNA expression.

RESUMO

A 1xN interferometer-based router with single-stage all-pass filters in the arms has low loss, a flat passband, and low cross talk. However, we show that the router has substantial cubic dispersion over the channel passband, which is identical from channel to channel. For a 1x4 router with a free spectral range of 100 GHz, the average dispersion slope over a 60% passband is -8x10(3)ps/nm(2), and thus a cascade of four routers incurs a significant system power penalty. A three-stage all-pass filter placed on the input arm reduces the dispersion of all channels by a factor of 16. The router is quite insensitive to variations in the all-pass filter design parameters.
