The power and commensurate prior distributions are informative prior distributions that incorporate historical data as prior knowledge in Bayesian analysis to improve inference about a phenomenon under study. Although these distributions have been developed for analyzing non-spatial data, little or no attention has been given to spatial geostatistical data. In this study, we extend these informative prior distributions to a Gaussian spatial process, which enables the elicitation of prior knowledge from historical geostatistical data for Bayesian analysis. Three informative prior distributions were developed for spatial modeling, and an efficient Markov Chain Monte Carlo algorithm was developed for performing Bayesian analysis. Simulation studies were used to assess the adequacy of the informative prior distributions. Hierarchical models combined with the developed informative prior distributions were applied to analyze transcranial magnetic stimulation (TMS) brain mapping data to gain insights into the spatial pattern of a patient's response to motor cortex stimulation. The study quantified the uncertainty in motor response and found that the primary motor cortex of the hand is responsible for most of the movement of the right first dorsal interosseous muscle. The findings provide a deeper understanding of the neural mechanisms underlying motor function and ultimately aid the improvement of treatment options for individuals with health issues.

Sensorized insoles (SIs) have been used as a wearable instrument to study human gait and have the potential to identify and predict pathologies and injuries. However, most of these sensorized insoles are only statically calibrated, relying on a scale and known weights to establish a relationship between electrical signals and the load applied on laboratory benches while ignoring the dynamic interaction between person and instrument. This study proposes and verifies a calibration method complementary to static calibration to compensate for different dynamic interactions between the insole and the individual during gait. In order to perform this comparison, a laboratory test was proposed with 32 volunteers (18 men and 14 women). Each volunteer walked on a double-belt instrumented treadmill (Bertec at 1000 Hz, Bertec Corp, Columbus, OH) while wearing an experimental resistive sensorized insole (SI). The SI data were compared with the instrumented treadmill and adjusted using an optimization algorithm to create a dynamic coefficient to complement and optimize the results. This study also verifies the impact of the method considering three different types of gait: pronated, neutral, and supinated. After using this technique and considering static calibration, the Pearson correlation coefficient between the SI and the instrumented treadmill improved by 12%.

Many challenges lie ahead when dealing with COVID-19, not only related to the acceleration of the pandemic, but also to the prediction of personal protective equipment sets consumption to accommodate the explosive demand. Due to this situation of uncertainty, hospital administration encourages the excess stock of these materials, over-stocking products in some hospitals, and provoking shortages in others. The number of available personal protective equipment sets is one of the three main factors that limit the number of patients at a hospital, as well as the number of available beds and the number of professionals per shift. In this scenario, we developed an easy-to-use expert system to predict the demand for personal protective equipment sets in hospitals during the COVID-19 pandemic, which can be updated in real-time for short term planning. For this system, we propose a naive statistical modeling which combines historical data of the consumption of personal protective equipment sets by hospitals, current protocols for their uses and epidemiological data related to the disease, to build predictive models for the demand for personal protective equipment in Brazilian hospitals during the pandemic. We then embed this modeling in the free Safety-Stock system, which provides useful information for the hospital, especially the safety-stock level and the prediction of consumption/demand for each personal protective equipment set over time. Considering our predictions, a hospital may have its needs related to specific personal protective equipment sets estimated, taking into account its historical stock levels and possible scheduled purchases. The tool allows for adopting strategies to control and keep the stock at safety levels to the demand, mitigating the risk of stock-out. As a direct consequence, it also enables the interchange and cooperation between hospitals, aiming to maximize the availability of equipment during the pandemic.

Introduction: Patients undergoing cancer treatment usually have comorbidities, and psychiatric disorders are commonly seen in these patients. For the treatment of these psychiatric disorders, the use of psychotropic drugs is common, turning these patients susceptible to untoward drug interactions. Therefore, the aim of this study was to estimate the prevalence of clinically relevant drug-drug interactions (DDI) between chemotherapeutic and psychotropic agents in patients with cancer treated at an oncology service in southern Brazil. Methods: An observational epidemiological study with a cross-sectional census-type design was carried out between October and December 2020. The drug-drug interactions were identified through consultation and analysis of the Medscape Drug Interaction Check and Micromedex databases. The interactions were classified as major, when the interaction can be fatal and/or require medical intervention to avoid or minimize serious adverse effects and moderate, when the interaction can exacerbate the patient's condition and/or requires changes in therapy. Results: A total of 74 patients was included in the study among the 194 patients seen in the oncology service during the period studied. A total of 24 (32.4%) DDIs were found, 21 (87.5%) of which were classified as being of major risk and 3 (12.5%) as moderate risk. According to the mechanism of action, 19 (79.1%) were classified as pharmacodynamic interactions and 5 (20.9%) as pharmacokinetic interactions. Conclusion: It was shown that a considerable percentage of patients undergoing intravenous chemotherapy are at risk of pharmacological interaction with psychotropic drugs. Thus, it is essential that the oncologist considers all psychotropic drugs and other drugs used by patients in order to avoid drug-drug interactions.

Cellulose cryogels are promising eco-friendly materials that exhibit low density, high porosity, and renewability. However, the applications of these materials are limited by their lower mechanical and water resistance compared to petrochemical-based lightweight materials. In this work, nanocelluloses were functionalized with cationic and anionic groups, and these nanomaterials were combined to obtain strong and water-resilient cryogels. To prepare the cryogels, anionic and cationic micro- and nanofibrils (CNFs) were produced at three different sizes and combined in various weight ratios, forming electrostatic complexes. The complex phase was concentrated by centrifugation and freeze-dried. Porous and open cellular structures were assembled in all compositions tested (porosity >90 %). Compressive testing revealed that the most resistant cryogels (1.7 MPa) were obtained with equivalent amounts of negatively and positively charged CNFs with lengths between 100 and 1200 nm. The strength at this condition was achieved as CNF electrostatic complexes assembled in thick cells, as observed by synchrotron X-ray tomography. In addition to mechanical strength, electrostatic complexation provided remarkable structural stability in water for the CNF cryogels, without compromising their biodegradability. This route by electrostatic complexation is a practical strategy to combine and concentrate nanocelluloses to tailor biodegradable lightweight materials with high strength and wet stability.

Introduction: Interpersonal neural synchronization (INS) demands a greater understanding of a brain's influence on others. Therefore, brain synchronization is an even more complex system than intrasubject brain connectivity and must be investigated. There is a need to develop novel methods for statistical inference in this context. Methods: In this study, motivated by the analysis of fNIRS hyperscanning data, which measure the activity of multiple brains simultaneously, we propose a two-step network estimation: Tabu search local method and global maximization in the selected subgroup [partial conditional directed acyclic graph (DAG) + multiregression dynamic model]. We illustrate this approach in a dataset of two individuals who are playing the violin together. Results: This study contributes new tools to the social neuroscience field, which may provide new perspectives about intersubject interactions. Our proposed approach estimates the best probabilistic network representation, in addition to providing access to the time-varying parameters, which may be helpful in understanding the brain-to-brain association of these two players. Discussion: The illustration of the violin duo highlights the time-evolving changes in the brain activation of an individual influencing the other one through a data-driven analysis. We confirmed that one player was leading the other given the ROI causal relation toward the other player.

Inulinase (ß-2,1-D- fructan fructanohydrolase), EC 3.2.1.7, targets the ß-2,1 linkage of inulin, a polyfructan consisting of linear ß-2,1 linked fructose, and hydrolyzes it into fructose. This use provides an alternative to produce fructose syrup through the hydrolysis of inulin. The objective of this work was to study the production, characterization and applications of inulinases from the fungal endophyte CCMB 328 isolated from the Brazilian semi-arid region. Response Surface Methodology (RSM) was employed to evaluate the effect of variables (concentration of glucose and yeast extract), on secreted inulinase activities detected in the culture medium and also in the inulin hydrolysis. The results showed that the best conditions for inulinase production by CCMB 328 are 9.89 g / L for glucose and 1.09 g / L for yeast extract. The concentration of 0.20 mol/L of NaCl and KCl increased the activity of inulinase from CCMB 328 by approximately 63% and 37%, respectively. The results also showed that the inulinase has potential for inulin hydrolysis, whose conversion yields roughly 72.48 % for an initial concentration of inulin at 1% (w/v).

OBJECTIVE: To estimate the incidence of potential in-hospital adverse reactions  with the use of alert drugs in a general hospital in southern Brazil. Method: Cross-sectional study, carried out in a hospital in southern Brazil. The  electronic medical records (TASY®) of patients hospitalized between January  and August 2020, who were prescribed one of the drugs earmarked for  tracking adverse drug reactions, were evaluated: the drugs included  flumazenil, fexofenadine hydrochloride, naloxone, promethazine, diphenhydramine and loperamide. RESULTS: A total of 13,476 medical records were reviewed and 204 (1.5%)  were included in the study in which tracker use was indicated in the management of adverse drug reactions. In this study a total of 18 different signs or symptoms were found in medical records, with  pruritus/hyperemia/urticaria being the most reported symptoms (n = 76).  Among the drug classes that caused most adverse drug reactions, opioids were  the most mentioned (n = 44). It should be noted that in 49 medical  records the information on which drug caused the adverse events was not  reported. Regarding the cause of hospitalization of patients who used  creening drugs, cancer was the most frequent (n = 37). CONCLUSIONS: This study indicates that the use of trackers can be a tool to  estimate the occurrence of adverse drug reactions and to establish adverse  events related to the use of medications, which should be reported to the  pharmacovigilance service, with a view to patient safety.


OBJETIVO: Estimar la incidencia de potenciales reacciones adversas intrahospitalarias con el uso de prescripciones alertantes en un  hospital general del sur de Brasil.Método: Estudio transversal, realizado en un hospital del sur de Brasil. Se  evaluaron las historias clínicas electrónicas (TASY®) de los pacientes hospitalizados entre enero y agosto de 2020, a los que se les  prescribió uno de los medicamentos destinados al seguimiento de reacciones  adversas a medicamentos: los medicamentos incluían flumazenil, clorhidrato de fexofenadina, naloxona, prometazina, difenhidramina y  loperamida. RESULTADOS: Se revisaron 13.476 historias clínicas y se incluyeron 204 (1,5%) en el estudio en el que se indicó el uso de prescripciones alertantes en  el manejo de reacciones adversas a medicamentos. En este estudio se  encontró un total de 18 signos o síntomas diferentes en las historias clínicas,  siendo el prurito, la hiperemia y la urticaria los síntomas más reportados (n =  76). Entre las clases de fármacos que causaron la mayoría de las reacciones  adversas a medicamentos, los opioides fueron los más mencionados (n = 44).  Cabe señalar que en 49 historias clínicas no se reportó la información sobre  qué fármaco causó los eventos adversos. En cuanto a la causa de  hospitalización de los pacientes que utilizaron prescripciones alertantes, el  cáncer fue la más frecuente (n = 37). Conclusiones: Este estudio indica que el uso de alertadores puede ser una  herramienta para estimar la incidencia de reacciones adversas a medicamentos y establecer eventos adversos relacionados con el uso de medicamentos, los cuales deben ser reportados al servicio de  armacovigilancia, con miras a la seguridad del paciente.

Learning techniques involve unraveling regression structures, which aim to analyze in a probabilistic frame the associations across variables of interest. Thus, analyzing fraction and/or proportion data may not be adequate with standard regression procedures, since the linear regression models generally assume that the dependent (outcome) variable is normally distributed. In this manner, we propose a statistical model called unit-Lindley regression model, for the purpose of Statistical Process Control (SPC). As a result, a new control chart tool was proposed, which targets the water monitoring dynamic, as well as the monitoring of relative humidity, per minute, of Copiapó city, located in Atacama Desert (one of the driest non-polar places on Earth), north of Chile. Our results show that variables such as wind speed, 24-hour temperature variation, and solar radiation are useful to describe the amount of relative humidity in the air. Additionally, Information Visualization (InfoVis) tools help to understand the time seasonality of the water particle phenomenon of the region in near real-time analysis. The developed methodology also helps to label unusual events, such as Camanchaca, and other water monitoring-related events.

Core-shell (CS) hydrogels show great potential for the controlled release of fertilizers. In this work, we prepared an alginate-coated gelatin-cellulose nanocrystals (CNCs) hydrogel by a simple layer-by-layer process. CNCs were prepared from cotton linter fibers by the sulfuric acid process. They were incorporated into the gelatin hydrogel, and an external alginate membrane was applied to the inner membrane. Compared to neat gelatin hydrogel, the compressive modulus of the nanocomposite with 5.0 wt% CNCs was enhanced by 288 %. In addition, the CS hydrogel showed a slow-release property and better water retention capacity than neat gelatin hydrogel. The main results of this work are listed below: compression test revealed that the addition of the CNC increases the mechanical properties of the hydrogel, and ii) the addition of a second layer of alginate to CNC-reinforced gelatin hydrogel increase the water retention and improve the sustained release of fertilizer. Our study provides easy and green routes to produce CS hydrogels for potential agricultural applications.

This supplementary dataset is supportive of the randomized sham-controlled, double-blind, crossover clinical trial investigating polarity- and intensity-dependent effects of high-definition transcranial electrical stimulation (HD-tDCS) applied over the right temporo-parietal junction on mean middle cerebral artery blood flow velocity (MCA-BFv) bilaterally. Data of eleven healthy right-handed adults (6 women, 5 men; mean age 31 ± 5.6 years old) were analyzed for MCA-BFv, assessed using transcranial doppler ultrasound on the stimulated and the contralateral hemisphere concomitantly, during and after 3 blocks of 2 min HD-tDCS at 1, 2, and 3 mA. Participants received three electrical stimulation conditions (anode center, cathode center, and sham) randomly ordered across different days. The collected data is publicly available at Mendeley Data. This article and the data will inform future related investigations and safety analysis of transcranial non-invasive brain stimulation.

Since neuronal activity is coupled with neurovascular activity, we aimed to analyze the cerebral blood flow hemodynamics during and following high-definition transcranial direct current stimulation (HD-tDCS). We assessed the mean middle cerebral artery blood flow velocity (MCA-BFv) bilaterally using transcranial doppler ultrasound, during and after HD-tDCS, in eleven right-handed healthy adult participants (6 women, 5 men; mean age 31 ± 5.6 years old), with no evidence of brain or cardiovascular dysfunction. The HD-tDCS electrode montage was centered over the right temporo-parietal junction. The stimulation protocol comprised 3 blocks of 2 min at each current intensity (1, 2, and 3 mA) and an inter-stimulus interval of 5 min between blocks. Participants received three electrical stimulation conditions (anode center, cathode center, and sham) on three different days, with an interval of at least 24 h. Stimulation was well tolerated across HD-tDCS conditions tested, and the volunteers reported no significant discomfort related to stimulation. There was no significant difference in the right or the left MCA-BFv during or after the stimulation protocol across all stimulation conditions. We conclude that at a range of intensities, vascular reaction assessed using middle cerebral artery blood flow is not significantly altered during or after HD-tDCS both locally and remotely, which provides further evidence for the safety of HD-tDCS.

Elucidating cellulose-lignin interactions at the molecular and nanometric scales is an important research topic with impacts on several pathways of biomass valorization. Here, the interaction forces between a cellulosic substrate and lignin are investigated. Atomic force microscopy with lignin-coated tips is employed to probe the site-specific adhesion to a cellulose film in liquid water. Over seven thousand force-curves are analyzed by a machine-learning approach to cluster the experimental data into types of cellulose-tip interactions. The molecular mechanisms for distinct types of cellulose-lignin interactions are revealed by molecular dynamics simulations of lignin globules interacting with different cellulose Iß crystal facets. This unique combination of experimental force-curves, data-driven analysis, and molecular simulations opens a new approach of investigation and updates the understanding of cellulose-lignin interactions at the nanoscale.

Using psychiatric drugs to treat drug dependence and its comorbidities is very common. The objective of this study was to analyze the interactions between prescribed drugs for patients treated at a specialized mental health-care center for persons who use drugs, located in the state of Santa Catarina, Brazil. A cross-sectional study was conducted on secondary data collected from 2010 to 2018. We reviewed the medical records of patients aged 18 years or older who took psychotropic drugs and had any type of substance dependence. The analysis of psychotropic drug interactions was conducted in three databases: Medscape, Drug Interactions Checker, and Micromedex. We included 1,022 of the 2,322 patients attending the care center during the study period. Psychotropic drug interactions were found in 779 (76.4%) study participants, and they presented 2,292 (100%) interactions, out of which 136 (6.0%) had minor clinical risk, 537 (23.4%) had moderate risk, and 1,619 (70.6%) had major risk for the patient, totaling 172 incompatible combinations between two psychotropic drugs. Of the total number of interactions, 128 were pharmacokinetic and 44 were pharmacodynamic. The high number of psychotropic drug interactions is a serious public health issue. Psychopharmacological treatment should be carefully addressed to be safe for the patient.

Supramolecular assembly of biobased components in water is a promising strategy to construct advanced materials. Herein, electrostatic complexation was used to prepare wet-resilient foams with improved mechanical property. Small-angle X-ray scattering and cryo-transmission electron microscopy experiments showed that suspensions with oppositely charged cellulose nanofibers are a mixture of clusters and networks of entangled fibers. The balance between these structures governs the colloidal stability and the rheological behavior of CNFs in water. Foams prepared from suspensions exhibited maximum compressive modulus at the mass composition of 1:1 (ca 0.12 MPa), suggesting that meaningful attractive interactions happen at this point and act as stiffening structure in the material. Besides the electrostatic attraction, hydrogen bonds and hydrophobic contacts may also occur within the clustering, improving the water stability of cationic foams. These results may provide a basis for the development of robust all- cellulose materials prepared in water, with nontoxic chemicals.

BACKGROUND: Technological resources, such as smartphones can contribute to the quantitative assessment of posture. PURPOSES: Test the validity and reliability of using a postural assessment application to quantify the frontal plane knee posture in orthostatism and to test the influence of the use of external markers on the precision of this measure. DESIGN: Methodological study. METHODS: The knee frontal plane posture of 30 volunteers were analyzed by two independent examiners. The photographs were taken with different external marker arrangements. The photographs were analyzed at two moments using the Kinovea software and PhysioCode Posture (PCP) application. Reliability was analyzed using the intraclass correlation coefficient (ICC) between measures with each instrument conducted at two moments with a 7-day interval. Concurrent validity of PCP with Kinovea measure was analyzed using Pearson's correlation coefficient. Standard error measurement (SEM), minimum detectable change (MDC) and Bland Altman plots were analyzed. RESULTS: PCP demonstrated excellent intra-rater [ICC = 0.92 (95% confidence interval [CI] 0.90-0.93)] and inter-rater [ICC = 0.88 (95%CI 0.85-0.90)] reliability. Concurrent validity analysis showed excellent agreement between PCP and Kinovea software (r = 0.88). The use of markers, independent of positioning, did not influence the measurement properties of measures with both softwares. The SEM was inferior to 1.2°, and the MDC was below 2.85°. No systematic errors were observed in the Bland Altman graphs. CONCLUSIONS: The use of PCP application to measure knee posture was valid and demonstrated excellent intra- and inter-rater reliability levels. The use of external markers did not influence the measurement.

To systematically identify studies about the assessment of pregnant women's expectations, using measuring instruments. An online search was made of Medline/PubMed, SciELO, Google Scholar, CAPES and LILACS databases in national and international publications from their first indexation until December 2018, using words associated with expectations during pregnancy. This review included observational studies that presented instruments to measure the expectations of pregnant women; observational studies that described the instrument development or tested the psychometric properties of an instrument. The studies were independently assessed for inclusion, data extraction and potential risks of bias. Because all study designs were observational, MOOSE was used to evaluate the quality of data. The Terwee's quality criteria were used for quality assessment of the instruments. Thirty-two studies were included in this review. The aim of the identified instruments was to measure expectations, experiences, satisfaction, quality of attachment, and attitudes at delivery, encompassing several aspects of the pregnancy process and childbirth. Most studies measured expectations only by relating them to fear and pain during childbirth. The results of this systematic review indicated that there are currently no instruments measuring pregnant women's expectations about childbirth other than those focusing on fear and pain. This gap indicates a need to develop a specific instrument for assessing and measuring this phenomenon comprehensively.
