RESUMO

Aquaculture fish diseases pose a serious threat to the security of food supplies. Fish species vary widely, and because they resemble one another so much, it is challenging to distinguish between them based solely on appearance. To stop the spread of disease, it is important to identify sick fish as soon as possible. Due to a lack of necessary infrastructure, it is still difficult to identify infected fish in aquaculture at an early stage. It is essential to promptly identify sick fish to stop the spread of disease. The purpose of this work is to suggest a machine learning technique based on the DCNN method for identifying and categorizing fish diseases. To solve problems involving global optimization, this paper suggests a brand-new hybrid algorithm called the Whale Optimization Algorithm with Genetic Algorithm (WOA-GA) and Ant Colony Optimization. In this work, for classification, the hybrid Random Forest algorithm is used. To facilitate the increased quality, distinctions between both the proposed WOA-GA-based DCNN architecture and the presently used methods for machine learning have been made. The effectiveness of the proposed detection technique is done with MATLAB. Performance metrics like sensitivity, specificity, accuracy, precision, recall, F-measure, NPV, FPR, FNR, and MCC are compared to the performance of the proposed technique.

Neurons of the human brain are primarily affected by the Huntington's disease (HD), Amyotrophic Lateral Sclerosis (ALS), Parkinson's disease and so on. Classification of these neurodegenerative diseases (NDD) is clinically important to analyze the destruction of nerve cells. Early diagnosis of NDD'S helps in saving the human life. Based on the report of previous studies, motor impairment or human gait cycle is largely affected by the clinical symptoms of NDD. Accurate diagnosis of various neurodegenerative diseases in correct time is very important for early diagnosis of the disease. Diseases can be diagnosed earlier by means of characterizing the gait cycle. In this work, a gait dynamics classification method is proposed for determining the neurodegenerative diseases from the brain signals using multilevel feature extraction method. From force sensitive resistors, the left and right feet signals recorded in 60 one minute are included in the input database. It is obtained through fixing 16 healthy subjects, 13 ALS, 20 HD, and 15 PD. Using six levels of Discrete Wavelet Transform (DWT), the features are determined by means of decomposing the raw signal. Ultimately, the pathological gait signals are classified through exploiting three multilevel feature extraction techniques named as, (Detrended Fluctuation Analysis (DFA), Positive, Negative Peak Histogram Analysis (PNPHA) (proposed Method) and Statistical Temporal parameter Analysis (STA)). Experimental outcomes proved that the gait dynamics are successively distinguished between NDD and group of healthy controls using the proposed method.

INTRODUCTION: Low back pain (LBP) is a musculoskeletal condition that affects many individuals. While physiotherapy and acupuncture are frequently used treatments, determining their specific contributions when used alongside integrated yoga and naturopathy (IYN) therapy for relieving chronic LBP symptoms and enhancing quality of life is important. METHODOLOGY: In the present prospective randomized comparative trial, a total of 60 participants were divided into the following two groups: physiotherapy with IYN and acupuncture with IYN. The treatment duration was 10 days, and assessments were conducted both before (baseline) and after the intervention. Various assessment tools, such as the visual analog scale (VAS), Oswestry Disability Index (ODI), fingertip-to-floor test (FTF), State-Trait Anxiety Inventory (STAI), and Short Form 36 (SF-36), were utilized. The statistical analysis was performed using SPSS version 21.0 (Armonk, NY: IBM Corp). RESULTS: The results revealed significant differences in VAS score, ODI, FTF score, STAI score, and SF-36 score between the physiotherapy and acupuncture groups (p<0.001). A t-test for the equality of means and a Mann-Whitney U test were used to compare the two groups; these tests revealed a significant difference in disability levels, range of motion, and state of anxiety. The acupuncture group showed a significant difference in the ODI (15.9 {12.6, 19.3}; p <0.001) and state anxiety (23.0 {20.0, 26.0}; p<0.001) compared to the physiotherapy ODI (22.4 {18.5, 26.4}) and state anxiety (27.5 {25.0, 30.0}). The physiotherapy group showed a significant difference in range of motion (7.7 {5.7, 9.7}; p<0.001) compared to the acupuncture group (11.6 {9.8, 13.5}). CONCLUSION: The present study findings revealed that both physiotherapy and acupuncture, as interventions along with integrated yoga and naturopathy may be considered an effective treatment strategy for chronic LBP.

Broadening of the genetic base for identification and transfer of genes for resistance to insect pests and diseases from wild relatives of rice is an important strategy in resistance breeding programs across the world. An accession of Oryza nivara, International Rice Germplasm Collection (IRGC) accession number 105710, was identified to exhibit high level and broad-spectrum resistance to Xanthomonas oryzae pv. oryzae. In order to study the genetics of resistance and to tag and map the resistance gene or genes present in IRGC 105710, it was crossed with the bacterial blight (BB)-susceptible varieties 'TN1' and 'Samba Mahsuri' (SM) and then backcrossed to generate backcross mapping populations. Analysis of these populations and their progeny testing revealed that a single dominant gene controls resistance in IRGC 105710. The BC(1)F(2) population derived from the cross IRGC 105710/TN1//TN1 was screened with a set of 72 polymorphic simple-sequence repeat (SSR) markers distributed across the rice genome and the resistance gene was coarse mapped on chromosome 7 between the SSR markers RM5711 and RM6728 at a genetic distance of 17.0 and 19.3 centimorgans (cM), respectively. After analysis involving 49 SSR markers located between the genomic interval spanned by RM5711 and RM6728, and BC(2)F(2) population consisting of 2,011 individuals derived from the cross IRGC 105710/TN1//TN1, the gene was fine mapped between two SSR markers (RMWR7.1 and RMWR7.6) located at a genetic distance of 0.9 and 1.2 cM, respectively, from the gene and flanking it. The linkage distances were validated in a BC(1)F(2) mapping population derived from the cross IRGC 105710/SM//2 × SM. The BB resistance gene present in the O. nivara accession was identified to be novel based on its unique map location on chromosome 7 and wider spectrum of BB resistance; this gene has been named Xa33. The genomic region between the two closely flanking SSR markers was in silico analyzed for putatively expressed candidate genes. In total, eight genes were identified in the region and a putative gene encoding serinethreonine kinase appears to be a candidate for the Xa33 gene.

Background and Objectives: The triaging of COVID-19 patients is of paramount importance to plan further management. There are several clinical and laboratory parameters that help in categorizing the disease severity, triaging, and prognostication. Little is known about the prognostic significance of eosinopenia in predicting the severity of COVID-19 from large hospital data, especially from low- and middle-income countries. The objective of this study is to evaluate the level of eosinopenia as an early prognostic marker for assessing the outcomes in COVID-19 patients and to assess the superiority of eosinopenia as a prognostic marker for assessing the outcomes in COVID-19 patients compared to lymphopenia and neutrophil-to-lymphocyte ratio (NLR). Methods: The study was carried out in a tertiary care hospital. A retrospective longitudinal approach was adopted wherein the hospital records of COVID-19 patients were analyzed. In our study, two separate groups of patients were included for analysis to describe the association between initial eosinophil counts of the patients and the clinical outcomes. In the first group, the disease severity in terms of clinical and radiological parameters was compared in patients of COVID-19 presenting with and without the presence of initial eosinopenia. Commonly used markers for triage, namely lymphopenia and NLR, were compared with the presence of initial eosinopenia among the patients who progressed to moderate and severe disease. In the second group, an analysis of eosinopenia was made among the patients who succumbed to the illness. Results: It was seen that 29.6% of patients with eosinopenia had moderate and severe disease compared to those without eosinopenia where only 10.8% had moderate disease, none had severe disease. It was seen that 19.7% of patients with eosinopenia but no lymphopenia had more severe disease compared to patients with lymphopenia but no eosinopenia where 10.8% of the patients had moderate disease, none had severe disease. In patients younger than 60 years who died of COVID-19, it was found that initial eosinopenia was found in 86%, whereas a high NLR >17 was seen in only 25.6% of patients who died, thus implying that is eosinopenia is an important marker of disease severity in COVID-19. Conclusions: Eosinopenia is an important parameter in the evaluation of COVID-19 and the presence of it should alert the clinicians regarding the further progression of the disease. It is not only an important marker but also an early marker for severe disease.

The cultivar Ajaya (IET 8585) exhibits durable broad-spectrum resistance to bacterial blight (BB) disease of rice and is widely used as a resistance donor. The present study was carried out to decipher the genetics of BB resistance in Ajaya and map the gene(s) conferring resistance. Genetic analysis in the F2 indicated a quantitative/additive nature of resistance governed by two loci with equal effects. Linked marker analysis and allelic tests revealed that one of the resistance genes is xa5. Sequence analysis of a 244 bp region of the second exon of the gene-encoding Transcription factor IIAγ (the candidate gene for xa5) confirmed the presence of xa5. Bulked-segregant analysis (BSA) revealed the putative location of the two quantitative trait loci (QTLs)/genes associated with resistance on chromosomes 5 and 8. Composite interval mapping located the first locus on Chr. 5S exactly in the genomic region spanned by xa5 and the second locus (qtl BBR 8.1) on Chr. 8L. Owing to its differential disease reaction with a set of seven hyper-virulent isolates of Xanthomonas oryzae, a map location on Chr. 8L, which was distinct from xa13 and data from allelism tests, the second resistance locus in Ajaya was determined to be novel and was designated as xaAj. A contig map spanning xaAj was constructed in silico and the genomic region was delimited to a 13.5 kb physical interval. In silico analysis of the genomic region spanning xaAj identified four putatively expressed candidate genes, one of which could be involved in imparting BB resistance in Ajaya along with xa5.

Hypolipidaemic and antioxidant effects of ethanol extract of mature green fruits of Musa AAA (Chenkadali) was evaluated in alloxan induced diabetic rats. The effect of extract at two doses, 500 mg/kg body weight and 1000 mg/kg body weight was analysed and compared with a standard drug, glibenclamide. Rats administered with alloxan showed significantly increased levels of serum triacylglycerol, total cholesterol and alanine amino transferase (ALT) activity. Lipid peroxides increased significantly while reduced glutathione (GSH) decreased considerably in liver and pancreas. Oral administration of the ethanol extract of fruits of Musa AAA (Chenkadali) significantly decreased the levels of serum triacylglycerol, cholesterol and ALT activity. Significant decrease was also observed in the level of lipid peroxides while GSH content increased substantially in liver and pancreas. The effect was dose independent and rats treated with 500 mg/kg body weight showed comparable levels of serum triacylglycerol, cholesterol, ALT activity and liver lipid peroxides to that of normal control and glibenclamide treated groups. Although, there was no significant difference, treatment with 500 mg/kg body weight of the extract showed a higher content of GSH and lower level of lipid peroxides in pancreas compared with glibenclamide. Histopathological examination of pancreas and liver revealed regeneration of islet cells and hepatocytes respectively, which correlate with the biochemical findings. The present study shows that ethanol extract of mature green fruits of Musa AAA (Chenkadali) has antioxidant and hypolipidaemic properties and may be used for treating diabetes mellitus.

Protective effect of ethanol extract of Piper longum Linn. against monosodium glutamate (MSG) induced toxicity was studied. Rats, orally administered with MSG at a dose of 8 mg/g body weight for 20 consecutive days, showed an increase in liver weight and rate of lipid peroxidation. Glutathione (GSH) in serum, liver and kidney showed decreased concentration. Significant increase was noticed in activities of serum alanine amino transferase (ALT) and aspartate amino transferase (AST), levels of serum triacylglycerol, total cholesterol and urea. Histopathological examination of liver and kidney showed central venous congestion, diffuse degeneration and necrosis of hepatocytes in para cortical and midzonal areas of liver and diffuse cortical tubular degeneration of kidney. Oral administration of ethanol extract of P. longum fruits at 300 mg/kg body weight along with MSG significantly reduced the levels of lipid peroxides in serum, liver and kidney, serum AST activity, serum levels of triacylglycerol and total cholesterol. Though, there was an increase in the level of GSH in tissues it was not significant. However, the treatment failed to reduce the levels of ALT and urea. Examination of tissue sections also exhibited normal histological architecture of both the organs. The present study revealed that administration of P. longum provided significant protection to liver and kidney from the oxidative stress of MSG, though the dose rate was not sufficient to provide a complete protection.

The Cyt toxins are able to lyse a wide range of cell types in vitro, unlike the Cry delta-endotoxins. It exerts its activity by the formation of pores within target cell membranes. The structural information available for Cyt2Aa (PDB id: 1CBY) consists of a single domain in which two outer layers of alpha-helix wrap around a mixed beta-sheet. Beta-barrel was suggested as a possible structure of the pores. Hence, this study seeks to investigate the structural properties of other Cytolytic proteins by predicting the three-dimensional (3D) model using Cyt2Aa as template. The predicted models are expected to be significantly more accurate as all the Cyt proteins showed significant similarity with the template (PDB id: 1CBY). The refined homology models revealed similar secondary structures (alpha-helices and beta-sheets) and tertiary features as Cyt2Aa. The variation in the loop regions of the tertiary structure accounts for the differential toxicity.

In this research work, three kinds of nonwoven wound dressings were developed from chicken feather keratin (CFK-NW), keratin­sodium alginate (CFK-SA-NW) and keratin-chitosan (CFK-CS-NW) and characterized using FTIR and SEM. The physical characteristics such as air permeability, thickness and areal density test results revealed the suitability of fabricated materials for wound dressing applications. CFK-SA-NW and CFK-CS-NW indicated a positive antibacterial effect against Gram's positive Staphylococcus aureus and Gram's negative Klebsiella pneumoniae and Escherichia coli bacteria with the zone of inhibition enhanced over >2.0 cm. Moreover, the biomedical potentials of dressing materials has been investigated by cell viability and cytotoxicity tests. Further, the wound healing ability was demonstrated using in vivo model (Albino Wistar rat). The fabricated materials exhibited good support for cell viability and a strong cytocompatibility. Furthermore, the hundred percent wound healing ability of CFK-CS-NW, CFK-SA-NW, CFK-NW and untreated control rats was observed at 15, 17, 21 and 23 days, respectively, Moreover, the wound healing potential of CFK-CS-NW and CFK-SA-NW was found to be better than that of CFK-NW and control group of rats. The outcome of the present study discloses the prospective applications of the developed materials as wound dressing biomaterial.

BACKGROUND AND AIM: Flaxseeds are known to have varying antihypercholesterolemic and antiatherogenic activity due to its lignan secoisolariciresinol diglucoside, alpha-linolenic acid, and omega-3 fatty acids. The beneficial effect of whole grain dietary flaxseed was evaluated experimentally in high cholesterol diet (HCD)-fed Wistar albino rats. MATERIALS AND METHODS: Male Wistar albino rats (200 g) were divided into four groups of 12 rats each. Group I rats kept as control and given basal rat chew diet, Group II as positive control for induction of hypercholesterolemia and atherosclerosis by addition of 1% cholesterol and 15% saturated edible oil to the 1000 g of standard rat chew diet (HCD), Group III rats fed with whole grain flaxseed powder at 7.5 g/kg of rat/day in the standard rat chew diet and kept as flaxseed control, and Group IV rats supplemented with flaxseed at 7.5 g/kg of rat/day along with HCD and maintained for 90 days. RESULTS: Group II rats revealed significantly (p<0.05) higher total cholesterol (TC), triglycerides (TG), low-density lipoprotein cholesterol (LDL-C), and very LDL-C and significantly (p<0.05) reduced levels of high-density lipoprotein cholesterol (HDL-C), whereas tissue antioxidants such as catalase, superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GR), and glutathione S transferase (GST) were significantly (p<0.05) reduced, and lipid peroxidation products of thiobarbituric acid reactive substances (TBARS) level were nonsignificantly (p<0.05) increased in the heart and liver tissues. Flaxseeds supplementation along with HCD significantly ameliorated the serum levels of TC, TG, LDL-C, and HDL-C along with cellular antioxidant enzymes such as catalase, SOD, GPx, GR, GST, and non-significant amelioration of TBARS in the heart and liver tissues compared to Group II rats. Majority of the histopathologically initiated atherosclerotic changes in the aorta and fatty change in the liver of Group II were not observed in the flaxseed supplemented Group IV; however, interestingly proliferation of endothelial cells with new vascular channel formation in the liver and in between cardiac muscle fibers was observed in Group I and Group IV rats. CONCLUSION: The present study established the hypercholesterolemia with initiated atherosclerotic lesion in the aorta but unable to establish the atheromatous plaque in the aorta. Flaxseed supplementation along with HCD showed significant antihypercholesterolemic effect and ameliorated the changes of initiated atherosclerosis in the aorta. It needs further studies to explore all the possible beneficial effects and angiogenic properties of flaxseeds in the laboratory animals and human trials.

In this report, a novel method for the synthesis of silver nanoparticles on the surface of hydroxyapatite is described. Hydroxyapatite crystals are synthesized from acid mineralized solution, a byproduct of bone glue industries, by a simple chemical precipitation method, which results in the formation of hydroxyapatite nanocrystals. The reduction of silver ions occurs by the electron transfer from the hydroxyl groups on the surface of hydroxyapatite. This results in the formation of silver nanorods and needle shaped nanoparticles that are bound on the surface of hydroxyapatite, and the observed silver nanocrystals show anisotropic structure. Thus, hydroxyapatite crystals can be used as a new class of inorganic scaffolds for the synthesis of nanomaterials with implications in designing inorganic-inorganic hybrid nanocomposites for different applications.

BACKGROUND: Acute pancreatitis is a disease with variable outcome; the course of the disease can be modified by early aggressive management in patients with severe pancreatitis. Easily calculable pancreatic scores and investigations can help to triage these patients. We aimed to determine the role of bedside index for severity in acute pancreatitis (BISAP), harmless acute pancreatitis score (HAPS), and systemic inflammatory response syndrome (SIRS) scores on day of admission and C-reactive protein (CRP) at 48 h for predicting the presence of pancreatic fluid collection (PFC) and necrosis on CT scans done at 72 h. METHODS: Of a total of 114 consecutively seen patients of pancreatitis, 64 with acute pancreatitis were enrolled in the study. All individuals had the pancreatitis predicting scores calculated at the time of admission, CRP at 48 h, and contrast-enhanced computed tomography (CECT) abdomen at 72 h from admission. RESULTS: The study population of 64 (55 male) had a mean (+SD) age of 37.7 ± 13 years. Alcohol was the most common (68.8%) etiology in these patients. Based on CECT, patients were divided into 2 groups; group 1 with 41 patients who had mild pancreatitis and group 2 with 23 patients who had pancreatic fluid collection with or without necrosis (PFCN). PFCN were seen in 19 (29.7%) of patients with 2 or more SIRS criteria, 17 (26.6%) of patients with BISAP score ≥3, and 16 patients (25.0%) with HAPS >0 respectively. All three scores were able to predict PFCN significantly. CRP >150 mg/L was noted in 23 patients and was able to predict the presence of fluid collections (p=0.0002) and pancreatic necrosis (p = 0.0004) on CT. CONCLUSION: BISAP, HAPS, and SIRS scores and CRP of 150 mg/L all correlated significantly with the occurrence of fluid collections and pancreatic necrosis on CT at 72 h. None of the scores was superior to the other in this respect.

RESUMO

We describe a patient with situational restriction of elevation in adduction in his left eye. Clinical examination pointed to instability of the left medial rectus pulley. This was corrected by Faden on the medial rectus. The importance of this relatively new concept in identifying and treating orbital pulley instability is discussed.

Internally Transcribed Spacers (ITS) characterization and distribution of Repetitive Extragenic Palindromic (REP) sequences were studied in the genome of 223 field isolates of Bacillus thuringiensis from Madurai, India. They were characterized by morphological, biochemical and molecular methods. One hundred and twenty four of a total 223 isolates fitted ITS characterization of B. thuringiensis varieties known. Significant genomic variation was observed among seven isolates using REP primers. The ITS PCR product (EMBL accession number AJ639659) exhibited 98% nucleotide sequence homology with B. thuringiensis and placed the origin of indigenous isolate LDC-7 closer to B. thuringiensis on the basis of phylogenetic analysis.

RESUMO

Venous pattern particularly in the lower limbs is of great clinical importance, while ligating the veins to prevent the spread of thrombus. Normally deep femoral vein drains the area supplied by profunda femoris artery, follows the pattern of branches of this artery and ends independently into the femoral vein. However in few cases it establishes communication with popliteal vein of femoral vein at a lower level. In the present study the deep femoral vein replaced most of the popliteal vein by establishing communication with the tibial veins.

Rasamanikya a familiar drug, frequently used by Ayurvedic physicians. It also has a high demand in current pharmaceutical industry. Rasamanikya possesses different pharmaceutical methods with many a proved clinical studies. But it is of utmost importance to understand the safety profile of drug based on assurance which could be done by carrying out animal experimentation. In the present study, Rasamanikya was prepared with three methods. The toxicological study was carried out on acute and sub-acute toxicity of the drug. The three samples when compared together showed that Rasamanikya prepared out of classical Abhraka Patra method and modified Sharava Samputa method showed minimal histopathological changes proving its non-toxicity, whereas Rasamanikya prepared out of electric bulb method showed mild toxicity, but with chances of recovery. Acute toxicity study showed no immediate and evident toxic signs and mortality in histopathology reports and liver function test. However, sub-acute toxicity study showed mild to moderate fatty changes in liver.

This research work includes a combination of Fisher's linear discriminant (FLD) analysis and a radial basis network (RBN) for monitoring the combustion conditions for a coal fired boiler so as to allow control of the air/fuel ratio. For this, two-dimensional flame images are required, which were captured with a CCD camera; the features of the images-average intensity, area, brightness and orientation etc of the flame-are extracted after preprocessing the images. The FLD is applied to reduce the n-dimensional feature size to a two-dimensional feature size for faster learning of the RBN. Also, three classes of images corresponding to different burning conditions of the flames have been extracted from continuous video processing. In this, the corresponding temperatures, and the carbon monoxide (CO) emissions and those of other flue gases have been obtained through measurement. Further, the training and testing of Fisher's linear discriminant radial basis network (FLDRBN), with the data collected, have been carried out and the performance of the algorithms is presented.

Since the range of comfort zone or thermo neutral zone of domestic chickens is narrow, they become easily susceptible to heat and cold environmental stress. We evaluated Brahma Rasayana (BR) supplementation on concentrations of certain oxidative stress markers associated with heat stress. A total of 48 egg type male chickens of local strain were divided into six groups (n = 8) for the study. Three groups were fed with BR orally at the rate of 2 g/kg bw daily for 10 days prior to and during the period of experiment. Two of the four groups that were exposed to heat stress (HST i.e. to a temperature of 40 +/- 1 degrees C and relative humidity of 80 +/- 5% in an environmental chamber) for 4 h daily for 5 or 10 days, received BR orally. The other two groups remained as BR treated and untreated non-heat stressed (NHST) controls. There was a significant (P < 0.05) increase in the activities of antioxidant enzymes in blood such as catalase (CAT) and superoxide dismutase (SOD), as well as liver CAT, glutathione peroxidase (GPX) and glutathione reductase (GR) in NHST-BR treated and HST-BR treated (both 5 and 10 days) chickens when compared with untreated controls. A great deal of significant (P < 0.05) variations were seen in serum and liver reduced glutathione (GSH) concentration in NHST-BR treated and HST-BR treated (both 5 and 10 days) chickens. Serum and liver lipid peroxidation levels were found to be significantly (P < 0.05) higher in HST-untreated (both 5 and 10 days) chickens when compared with other groups. Thus BR supplementation during HST brings about enhanced action of enzymatic and non-enzymatic antioxidants, which nullified the undesired side effects of free radicals that are generated during HST.

AIMS: This study is mainly focused on the heterologous expression and accumulation of polyhydroxyalkanoates (PHA) in Escherichia coli. METHODS AND RESULTS: PHA synthase gene (phaC1) from indigenous Pseudomonas sp. LDC-5 was amplified by PCR and cloned in E. coli (Qiagen EZ competent cells). The recombinant E. coli was analysed and confirmed for its expression of phaC1 gene by phase contrast microscopy, Western blot analysis and spectral studies (Fourier-transform infrared spectroscopy). It was further evaluated for its accumulation in different carbon and nitrogen sources. The accumulation of PHA (3.4 g l(-1)) was enhanced in the medium supplemented with glycerol and fish peptone compared to the other carbon and nitrogen sources used in this study. CONCLUSIONS: This study would enable the reduction of cost of PHA production. SIGNIFICANCE AND IMPACT OF THE STUDY: An important part of this study is that E. coli harbouring partial phaC1 gene could accumulate medium chain length PHA significantly. The results demonstrated that the E. coli strain could be a potential candidate for the large-scale production of polymer. The conditions for the higher yield and productivity will be optimized in the next phase using fermentation studies.
