RESUMO

Focused laser differential interferometry (FLDI) is used to measure a well-characterized, 17 kHz screech tone emitted from an underexpanded Mach 1.5 jet. Measurements are made at numerous spatial locations in and around the jet flow-field, where intrusive diagnostics would otherwise influence the flow-field. Results from FLDI measurements are shown to agree with measurements from microphones and analyses of high-speed schlieren. The agreement is used to demonstrate FLDI is a valid and accurate technique for measuring screech tones in jet flow-fields, and furthermore that FLDI can be used to measure jet screech at various spatial locations around the jet, and notably inside of the jet, where microphones and other intrusive diagnostics cannot be used effectively.

This study determined the diversity and seasonality of parasites species of the African clawed frog, Xenopus laevis (Daudin, 1802), from three localities, namely Modjadjikloof, Mokopane and University of Limpopo, Limpopo Province, South Africa. A total of seven parasite species were collected and identified. They included two nematodes, Camallanus kaapstaadi Southwell & Kirshner, 1937 and Batrachocamallanus slomei (Southwell & Kirshner, 1937), a monogenean, Protopolystoma xenopodis (Price, 1943), a cestode, Cephalochlamys namaquensis (Cohn, 1906), a protozoan, Trichodina xenopodos Fantham, 1924, two digeneans, Progonimodiscus doyeri Ortlepp, 1926 and Dollfuschella rodhaini Vercammen-Grandjean, 1960. The most common and abundant parasite species by far were Cm. kaapstaadi, B. slomei and Cp. namaquensis, with Cm. kaapstaadi, B. slomei present in all localities. Trichodina xenopodos was a rare species, only present in host populations from Modjadjiskloof. Modjadjiskloof had the highest species richness (all seven parasite species) followed by Mokopane (five parasite species) and University of Limpopo (3 parasite species). There were also higher infection levels (prevalence and mean intensity) of Cm. kaapstaadi, B. slomei, Cp. namaquensis and Pt. xenopodis in hosts from Modjadjiskloof while Pd. doyeri and D. rodhaini infection levels were greater in Mokopane. The variability between localities shows that parasites with heteroxenous life cycles are more strongly associated with more pristine habitats. The variability in calculated indices (prevalence and mean intensity) also suggests that the occurrence of some of the parasites is affected by season, favouring higher infection rates during summer. This suggests that temperature has a direct role in the reproductive and developmental processes of these parasites. Neither length nor sex had an influence on the prevalence or intensity of parasites.

Avocado sunblotch viroid (ASBVd) is the type species of the family Avsunviroidae and the causal agent of avocado sunblotch disease. The disease is characterised by the presence of chlorotic lesions on avocado fruit, leaves and/or stems. Infected trees may remain without chlorosis for extended periods of time, though distorted growth and reduced yield has been observed in these cases. The molecular effects of ASBVd on avocado, and members of the Avsunviroidae on their respective hosts in general, remain poorly understood. Host global transcriptomic studies within the family Pospiviroidae have identified several host pathways that are affected during these plant-pathogen interactions. In this study, we used RNA sequencing to investigate host gene expression in asymptomatic avocado nursery trees infected with ASBVd. Transcriptome data showed that 631 genes were differentially expressed, 63 % of which were upregulated during infection. Plant defence responses, phytohormone networks, gene expression pathways, secondary metabolism, cellular transport as well as protein modification and degradation were all significantly affected by ASBVd infection. This work represents the first global gene expression study of ASBVd-infected avocado, and the transcriptional reprogramming observed during this asymptomatic infection improves our understanding of the molecular interactions underlying broader avsunviroid-host interactions.

Foot-and-mouth disease virus (FMDV) outer capsid proteins 1B, 1C and 1D contribute to the virus serotype distribution and antigenic variants that exist within each of the seven serotypes. This study presents phylogenetic, genetic and antigenic analyses of South African Territories (SAT) serotypes prevalent in sub-Saharan Africa. Here, we show that the high levels of genetic diversity in the P1-coding region within the SAT serotypes are reflected in the antigenic properties of these viruses and therefore have implications for the selection of vaccine strains that would provide the best vaccine match against emerging viruses. Interestingly, although SAT1 and SAT2 viruses displayed similar genetic variation within each serotype (32 % variable amino acids), antigenic disparity, as measured by r(1)-values, was less pronounced for SAT1 viruses compared with SAT2 viruses within our dataset, emphasizing the high antigenic variation within the SAT2 serotype. Furthermore, we combined amino acid variation and the r(1)-values with crystallographic structural data and were able to predict areas on the surface of the FMD virion as antigenically relevant. These sites were mostly consistent with antigenic sites previously determined for types A, O and C using mAbs and escape mutant studies. Our methodology offers a quick alternative to determine antigenic relevant sites for FMDV field strains.

The present study was designed to assess and compare three different formulations of the new Onderstepoort infectious coryza (IC) quadrivalent vaccine, which contain an NAD-independent strain of Avibacterium paragallinarum (previously known as Haemophilus paragallinarum), and a commercial IC vaccine, not containing an NAD-independent strain, for their safety and ability to protect chickens of varying ages against virulent challenges with four different serovars of A. paragallinarum, including the NAD-independent strain of the C-3 serovar. Four groups of 140 chickens each were vaccinated at the age of 17 weeks and revaccinated at the age of 19 weeks with each of the four vaccine formulations. A similar sized group of non-vaccinated chickens was used as control. Two rounds of challenge were conducted: a group of chicken in each vaccination group was challenged between 31 and 35 weeks of age, while another group was challenged between 51 and 55 weeks of age. The "in-contact" challenge model was used in this experiment. For each vaccination group, the four challenge strains representing four local serovars were used in each challenge round. The efficacy of the vaccines was compared based on overall protection levels obtained and the duration of protection. The safety of the different vaccines was determined by the severity of post-vaccination reactions. The need for the incorporation of the NAD-independent strain in the vaccine was evidenced by the low protection level against NAD-independent challenge recorded in the group of birds vaccinated with the commercial vaccine. The results obtained confirmed not only the variation in virulence of different South African serovars, with serovar C-3 being the most virulent and serovar B having almost no virulence but also the age related increase in susceptibility. The importance of a suitable formulation of the vaccine is discussed.

RESUMO

We report a new preparative method for providing contrast through reduction in electron density that is uniquely suited for propagation-based differential x-ray phase contrast imaging. The method, which results in an air or fluid filled vasculature, makes possible visualization of the smallest microvessels, roughly down to 15 microm, in an excised murine liver, while preserving the tissue for subsequent histological workup. We show the utility of spatial frequency filtering for increasing the visibility of minute features characteristic of phase contrast imaging, and the capability of tomographic reconstruction to reveal microvessel structure and three-dimensional visualization of the sample. The effect of water evaporation from livers during x-ray imaging on the visibility of blood vessels is delineated. The deformed vascular tree in a cancerous murine liver is imaged.

The amino acid sequence motif Arg-Gly-Asp (RGD), located in the surface-exposed betaG-betaH loop of the 1D protein of different serotypes and subtypes of foot-and-mouth disease virus (FMDV), is highly conserved and participates in binding of FMDV to susceptible cells. Previous sequence analyses of the 1D-encoding region of a FMDV serotype SAT1 field isolate from Namibia (NAM/307/98) indicated the presence of a second RGD motif upstream of the conserved betaG-betaH loop RGD. The role of these RGD sequences in virus infection was investigated by mutating the betaG-betaH loop RGD to a KGE tripeptide, using a genome-length infectious chimeric cDNA clone. Although the infectivity of the derived mutant viruses for baby hamster kidney cells (BHK-21) was lost, subsequent replacement of the KGE sequence with RGD in the mutant cDNA clone led to recovery of infectious viruses. Furthermore, viral RNA replication could be demonstrated with the genetically engineered mutant and non-mutant viruses. The presence of virus particles in the transfected cells could be also demonstrated by electron microscopy. These results demonstrate that, in contrast to the betaG-betaH loop RGD motif, the second RGD sequence in the capsid protein 1D of NAM/307/98 does not function as a ligand for receptor binding in BHK-21 cells.

RESUMO

Diffusion-weighted MR imaging is a technique in which image contrast is determined by the motion of water molecules within tissues. This motion is characterized by the apparent diffusion coefficient (ADC). This technique is particularly useful for the early detection of cerebral infarction but many other diseases of the central nervous system are associated with a change in water diffusion and may be assessed by diffusion-weighted MR imaging. This is an easy and fast pulse sequence providing useful data for early diagnosis and prognosis as well as information about underlying pathophysiology. After an overview of the basic concepts of diffusion imaging and the knowledge required for image interpretation, we will assess the potential value of this technique for the diagnosis of the main diseases of the central nervous system.

BACKGROUND AND PURPOSE: Carotid endarterectomy (CEA) is not necessarily beneficial in all patients with symptomatic high-grade (>/=70%) internal carotid artery (ICA) stenosis. Independent risk factors modulate both the individual stroke risk under medical treatment and the combined stroke and death risk after CEA. Endovascular stenting of symptomatic ICA stenosis may be an alternative to CEA in patients with a balanced surgical risk/benefit ratio. METHODS: We included 43 patients (71% men; median age, 67 years) with a recently symptomatic ICA stenosis with >/=70% luminal narrowing in whom the individual sum of medical and surgical risk factors suggested a balanced surgical risk/benefit ratio (risk-modeling appraisal derived from the European Carotid Surgery Trial). After stenting of the stenosed ICA with distal balloon protection, the mean+/-SD follow-up, including clinical and ultrasonographic examinations, was 20+/-11.8 months, with a median number of examinations of 5 per patient. RESULTS: Recanalization of ICA stenoses was technically successful in 40 of 43 procedures (93%). Within the 30-day postinterventional period 1 death occurred (2.5%), and the combined stroke and death rate within follow-up was 5%. Except for 1 asymptomatic ICA occlusion, no restenosis >/=70% occurred during follow-up. CONCLUSIONS: ICA stenting in symptomatic patients with a balanced surgical risk/benefit ratio is technically feasible, with a low periprocedural risk of stroke or death. Furthermore, the risk of future stroke and rate of significant restenosis during long-term follow-up appears to be low, suggesting that ICA stenting may be useful in carotid revascularization and stroke prevention.

BACKGROUND AND PURPOSE: We sought to quantitatively and qualitatively evaluate the release of atheromatous plaque debris induced by carotid stenting procedures. METHODS: Eight patients with severe carotid atheromatous stenoses were treated by stent implantation under distal balloon protection. Blood samplings were obtained after stent deployment in the blood pooled below the inflated protection balloon. The samples were centrifuged and evaluated for plaque debris with the use of light microscopy. The debris release was quantitatively estimated by dividing the total volume of debris obtained by the mean debris size. Five patients without endovascular procedure were used as a control group. RESULTS: The 2 main debris types found were nonrefringent cholesterol crystals (4 to 389 microm; 115 to 8697 in number) and lipoid masses (7 to 600 microm; 341 to 34 000 in number). There was a statistically significant difference compared with the samples obtained in the control group (P:=0.017). CONCLUSIONS: Blood samples collected during stent implantation procedures contain a large quantity of atheromatous plaque debris. This emphasizes the role of distal protection techniques in avoiding migration of this plaque material into the cerebral circulation.

The pharmacokinetics of i.v. and superselective intra-arterial carbon-11 1,3-bis-(2-chloroethyl)-1-nitrosourea ([11C]BCNU) were directly compared for the first time in ten patients with recurrent gliomas using positron emission tomography (PET). Intra-arterial administration of [11C]BCNU achieved concentrations of the drug in the tumor that averaged 50 times higher than with a comparable i.v. dose. These preliminary results suggest that the degree of early metabolic trapping of BCNU in tumor correlates with the clinical response to this chemotherapy.

Computational secondary structure prediction of all 10 Bluetongue virus (BTV-10) RNA transcripts and mutant inverted repeat transcripts were performed. Transcripts with intact 5' and 3' inverted repeat sequences, all indicated base-pairing between the 5' and 3' ends when optimal folding parameters were applied. Secondary structure analysis of the mutant transcripts lacking the inverted repeat sequences indicated alterations in the secondary structures resulting from altered base-pairing. The importance of the inverted repeat sequences in RNA--protein binding was subsequently investigated. Deletion mutant clones lacking the 5' and/or 3' inverted repeat sequences have been constructed. A baculovirus recombinant expressing the BTV NS2 protein and radioactively synthesized RNA transcripts were subjected to nitrocellulose RNA--protein binding assays. The cumulative results suggested that the inverted repeat deletion mutants display weaker binding compared to BTV-10 segment 8 with intact 5' and 3' inverted repeat sequences. Inverted repeats may influence RNA--protein binding by altering the secondary structure of the RNA and consequently the specific NS2 protein-binding sites may no longer be available.

Melatonin is present in human semen, and may affect sperm motility. The presence of melatonin receptors on spermatozoa has not yet been reported. Detection of melatonin-binding sites may be limited because of the masking of such sites by sialic acid. Spermatozoa were obtained from eligible human donors, incubated with neuraminidase to remove sialic acid residues, and saturation binding assays were carried out using 2-125I-melatonin as a receptor probe. Consistent 125I-melatonin binding could only be obtained after spermatozoa were treated with neuraminidase. Scatchard analysis revealed a low-affinity binding site (ML-2) with a Kd value of 127 +/- 6 nM and a Bmax of 25 +/- 4.5 fmol/mg protein. These results present evidence of melatonin-binding sites in spermatozoa. Sialic acid possibly regulates the binding of melatonin to these sites.

Seven patients with intracavernous aneurysms were treated by proximal balloon occlusion of the carotid artery, with no attempt to preserve carotid blood flow. After a 15-minute occlusion trial, two balloons were detached proximal to the neck of these aneurysms. Major clinical improvement occurred in six patients and no procedure-related complications were encountered (the follow-up period was 4-14 months). Because these aneurysms arise proximal to major collateral pathways, proximal carotid occlusions via endovascular techniques are simple and safe and carry with them little risk of thromboembolism.

A case is presented in which three arterial stenoses and one arterial occlusion due to Takayasu's disease were treated by an endovascular approach. The endovascular technique is discussed, and the clinical and angiographic findings after treatment are correlated with cerebral hemodynamic and metabolic parameters as measured by positron emission tomography.

RESUMO

A technique for safe angioplasty of atherosclerotic stenosis of internal and common carotid arteries is described in which temporary balloon occlusion of the internal carotid artery is used to prevent cerebral embolization during manifestation of ulcerated plaques. Its successful use in six atherosclerotic stenosis is described. Five angioplasties of postsurgical stenoses are also reported in which the technique of temporary occlusion was not used. The future of carotid artery angioplasty is discussed.

Four cases in which the diagnosis of carotid-cavernous fistula was made by using CT angiography are illustrated. The diagnosis was confirmed by digital subtraction angiography in all four instances. To our knowledge, this is the first report of the CT angiographic appearance of carotid-cavernous fistulas.

A series comprising 12 patients who had intraarterial local fibrinolysis in the carotid territory is reported. A classification is proposed that divides the different types of occlusions into three groups on the basis of angiographic location. Group 1 (two cases) comprises occlusion of the extra- and/or intracranial carotid artery with patency of the circle of Willis and the lenticulostriate arteries. In this group, there is no brain infarction, the CT findings are normal, and the clinical signs are mainly hemodynamic and intermittent. Fibrinolysis may be performed late and rather safely and completed by surgery or angioplasty of the neck vessel stenosis responsible for the occlusion. Group 2 (five cases) comprises occlusions of the cortical arteries without involvement of the lenticulostriate arteries. The mechanism of the occlusion can be hemodynamic or embolic. Group 3 (five cases) comprises occlusions of intracerebral arteries involving the lenticulostriate arteries. In groups 2 and 3 with brain infarction, fibrinolysis will only be able to restore viability of the area of cerebral tissue surrounding the infarction (penumbra). The time factor is particularly critical in group 3 because lenticulostriate arteries are terminal vessels whose revascularization may induce hemorrhages with increasing frequency as the occlusion time is prolonged. The time factor is less critical in group 2 because collaterals make the ischemia less severe in the infarcted area and the vital and functional consequences of hemorrhage are not as serious as in group 3 because of the location. In this series, all the symptomatic complications of hemorrhage (two cases) occurred in group 3, in patients treated later than 6 hr after clinical onset. Given the time delay inherent in performing CT and angiography and in making the medical decision, it is considered dangerous to undertake fibrinolytic therapy in group 3, unless it can be started before 4 or 5 hr after clinical onset.

Digital subtraction angiography (DSA) with stereoscopic imaging was performed in 40 patients for evaluation of a variety of cerebrospinal disorders. It was facilitated by a C-arm mounted x-ray tube and imaging chain with 7 degrees angulation between image pairs. Stereoscopic digital imaging proved particularly useful in the preoperative assessment of aneurysms, arteriovenous malformations, and primary and metastatic tumors. The technique was also found to be useful as a real-time adjunct to therapeutic radiographic procedures, as an aid in stereotaxic procedures, and in follow-up of postsurgical patients. Although the intravenous route was occasionally used, especially in postoperative follow-up of aneurysms, the procedure was most often carried out via an intraarterial approach. Stereoscopy was useful in supplying depth information regarding the relations between lesions and surrounding normal and abnormal vasculature. This technique combines the demonstrated advantages of intraarterial DSA with the unique advantage of stereoscopic imaging to demonstrate three-dimensional detail, thus contributing significantly to diagnostic confidence. Disadvantages are discussed. Further refinements in the equipment are expected: generation of stereo images with one injection, thus increasing procedure efficiency and patient safety; a video stereoscopic viewing unit; and the ability to obtain precise measurements via computer of depth, position, distance between, and true size of objects.
