The two most common animal-based indicators used to evaluate leg health in broiler chickens are footpad dermatitis (FPD) and gait scoring, but these indicators are less explored in broiler breeders. This study is the first to investigate FPD and gait scoring in broiler breeders during their lifespan from rearing to end of life. In total, eight flocks were monitored (four Ross and four Hubbard) at five different timepoints, in rearing (5 and 15 weeks of age), during the production period (25 and 45 weeks of age) and at the end of the production period (approximately 60 weeks of age). At each visit, 50 hens and 25 roosters were gait scored (six-point scale) and footpads from another 50 hens and 25 roosters were evaluated (five-point scale) (total n = 3000 breeders, 2000 hens and 1000 roosters). Litter quality and air quality were measured at each visit. The results showed that the overall prevalence of FPD in rearing was low and that it increased towards the end of the production, with a mean FPD score of 2 out of a maximum 4 in the hens, indicating moderate lesions and 1.5 in the roosters. In all houses, the litter was dry and loose. FPD was not related to the litter quality, but to air quality, especially the ammonia concentration (P < 0.001). Overall, the gait score were good, and increased with age in both hens (P < 0.001) and roosters of both hybrids (P < 0.001).

In pheasants, which are generally raised in large flocks, stocking density is important for their growth and development, normal behavior and welfare. But there is still a lack of information on the optimum stocking density for game birds. This research was conducted on a total of 200 pheasants to examine the effects of low (5 pheasant/m2) and high (10 pheasant/m2) stocking density rearing practices on body weight, morphological development, behavioral characteristics and welfare parameters such as foot-pad dermatitis and plumage quality. When we look at the study results, it was determined that the density of stocking did not make a difference in the first 13 weeks of age, but pheasants reared at low density at the ages of 14-16 weeks reached higher body weight. In terms of body weight at 14, 15, and 16 weeks, the low stocking density group had an average of approximately 41, 71, and 94 g higher than the high stocking density group, respectively (p<0.05). It had been found that low density provides better morphological development. The low group was found to be significantly higher than the high group in terms of ornithological size, wingspan, back length, tail length, head length and head width, 5.9 cm, 2 cm, 1.3 cm, 7.6 cm, 1.7 mm and 1 mm, respectively (p<0.05). Since the animals in the low group had more individual space, they had the opportunity to perch and therefore exhibit more passive behavior. On the other hand, pheasants in the low group exhibited higher levels of aggressive behavior (p<0.05). More foraging behavior was observed in the higher group and the low-density group consumed more feed in the 14th week (p<0.05). But no significant differences were detected between groups in terms of feed consumption in other weeks (p>0.05). A 1.12 point higher score was calculated in the low group compared to the high group, especially in terms of tail feather quality, and as a result, pheasants reared in low stocking density had better plumage quality (p<0.05). It had been determined that as the stocking density decreases in the rearing of pheasants, they exhibit easier flight and roosting behaviors and therefore they were less affected by feather pecking behavior and had better feather quality. In addition, it had been determined that pheasants grow and develop better in this group, which had more rest opportunities. In conclusion, rearing at low stocking density is recommended for better fattening performance and better welfare conditions.

1. Footpad dermatitis (FPD) can be used as an important indicator of animal welfare and for economic evaluation; however, human scoring is subjective, biased and labour intensive. This paper proposes a novel deep learning approach that can automatically determine the severity of FPD based on images of chicken's feet.2. This approach first determined the areas of the FPD lesion, normal parts of each foot and the background, using a deep segmentation model. The proportion of the FPD for the chicken's two feet was calculated by dividing the number of FPD pixels by the number of feet pixels. The proportion was then categorised using a five-point score for FPD. The approach was evaluated from 244 images of the left and right footpads using five-fold cross-validation. These images were collected at a commercial slaughter plant and scored by trained observers.3. The result showed that this approach achieved an overall accuracy and a macro F1-score of 0.82. The per-class F1-scores from all FPD scores (scores 0 to 4) were similar (0.85, 0.80, 0,80, 0,80, and 0.87, respectively), which demonstrated that this approach performed equally well for all classes of scores.4. The results suggested that image segmentation and a deep learning approach can be used to automate the process of scoring FPD based on chicken foot images, which can help to minimise the subjective bias inherent in manual scoring.

This study was performed to evaluate the effects of different amounts and particle size of zinc oxide (ZnO) on villus height (VH), villus width (VW), crypt depth (CD) and VH to CD ratio (VH: CD), and expression of zonula occludens-1 (ZO-1), occludin (OC) and tumour necrosis factor-α (TNF-α) in broiler breeders. A total of 350 (Ross 308) broiler breeder hens of 54 weeks randomly assigned to seven treatments, included control basal diet (C) without added Zn, C+ 100, and 130 mg Zn per kg of diet from Large (L) (100-1000 nm) and Small (S) (<100 nm) particle size ZnO (LZnO100 and 130; SZnO100 and 130), C and SZnO100 challenged with lipopolysaccharide (C+LPS and SZnO100+LPS). Each diet was fed to five replicates consisting of ten birds each. The middle part of the duodenum, jejunum and ileum was used for morphological assessments. To assess the gene expression of ZO-1, OC and TNF-α in the jejunum samples were excised. Results showed that the supplementing 130 ppm SZnO increased VH:CD in the duodenum (p < 0.05). VW in the duodenum and all the evaluated morphometric indices in jejunum and ileum were not affected by the dietary treatment (p > 0.05). ZO-1 mRNA abundance in C+LPS group compared to SZnO100+LPS group was significantly decreased and increased by LPS and SZnO100 respectively. The SZnO-100 increased OC gene expression in compare to C+LPS group. The expression of TNF-α in C+LPS treatment was higher than other groups (p < 0.05). The lowest and the highest litter moisture and foot-pad dermatitis (FPD) were observed in LZnO-130 and C treatments respectively (p < 0.05). Improving the physical properties of ZnO affect on VH:CD. Broiler breeder diet with ZnO enhance ZO-1, OC and mitigate TNF-α gene expression in jejunum maintenance of gut health in broiler breeders.

In a collection of 6 young binturongs (Arctictis binturong), 3 presented with anorexia, lethargy, and footpad swelling diagnosed by surgical biopsy as calcinosis circumscripta. Despite supportive care over the next 4 years, affected binturongs had progressive weight loss, hyperphosphatemia, and hyperkalemia and developed large radiodense deposits in tissues adjacent to appendicular joints, thoracolumbar vertebrae, and the sternum. Two binturongs died and necropsies showed severe periarticular mineral deposition with fibrosis and granulomatous inflammation. Additionally, binturongs had mineralization of distal renal tubules, mineralization of the renal interstitium, and marked mineralization of the pulmonary interstitium and peribronchiolar smooth muscle. Foot biopsies from the surviving binturong were evaluated by spectroscopic microanalysis and were positive for hydroxyapatite, Ca10(PO4)6(OH)2. Periarticular and footpad mineralization in these binturongs is consistent with hydroxyapatite deposition disease, which is rarely described in humans and animals as a primary familial condition or a secondary condition often associated with renal injury.

1. The aim of this study was to evaluate the effect of graded levels of the microbially derived feed lysozyme, muramidase (MUR) on feed intake (FI), weight gain (WG), feed conversion ratio (FCR), European Performance Index (EPI), dietary N-corrected apparent metabolisable energy (AMEn), footpad dermatitis score (FPD) and other welfare variables, when fed to broilers from 0 to 42d age. 2. A four-phase dietary programme and four experimental pelleted diets were used; a control diet (following breeder recommendations without MUR supplementation), and three diets based on the control diet supplemented with 25,000, 35,000 and 45,000 LSU (F)/kg of MUR, respectively. In addition, all experimental diets contained exogenous xylanase, phytase and a coccidiostat. Each diet was fed to birds in 24 pens (20 male Ross 308 chicks in each pen) following randomisation. Dietary AMEn was determined at 21 d of age, and FPD was evaluated at the end of the study. Data were analysed by ANOVA, using orthogonal polynomials for assessing linear and quadratic responses to MUR activity. 3. The inclusion of MUR did not change FI (P > 0.05), but increased WG in a linear manner (P < 0.05) and reduced FCR in a quadratic manner, with optimum WG and FCR observed in birds fed approximately 35 000 LSU (F)/kg. In accordance with the improvement in FCR, 35 000 LSU (F)/kg MUR supplementation produced the highest EPI (P < 0.05). FPD score was linearly decreased with increased addition of MUR (P < 0.05). Dietary AMEn responded in a quadratic fashion to the MUR inclusion, as the highest values were obtained with the highest inclusion rate (P < 0.05). 4. In conclusion, the results showed that inclusion of MUR improved feed efficiency and the foot health of birds.

We investigated the effect of in-feed and/or in-litter supplemental humate against footpad dermatitis (FPD) in broilers fed diets based on barley. Three hundred and sixty 1-day-old Ross 308 broiler chickens were randomly distributed to 24 floor pens (4 treatments, each consisting of 6 replicate pens; 15 chickens per pen) as a completely randomized design with 2 × 2 factorial arrangement of two levels of supplemental humate in feed (0 and 1 g/kg feed) and litter (0 and 5 g/kg litter). Growth performance, intestinal viscosity, litter quality, and incidence and severity of FPD in broilers were measured. In addition, malondialdehyde (MDA) and superoxide dismutase (SOD) levels were determined in blood and footpad tissues of broilers with different FPD scores. The results revealed that there was no interaction between humate supplementation to feed and litter. Neither dietary nor litter supplementation of humate had a significant effect on growth performance, intestinal viscosity, litter quality, and occurrence of FPD. And also, MDA and SOD levels in serum and footpad tissue did not affect by either dietary or litter supplementation of humate. The presence of FPD (score 1) had no effect on MDA and SOD levels in serum, however, increased the MDA and SOD levels (P < 0.001, P = 0.001, respectively) in footpad tissue of broilers. The intestinal viscosity did not differ between FPD scores 0 and 1. In conclusion, findings of this experiment suggest that humate supplementation to feed and litter did not alleviate FPD development in broilers fed diets based on barley. In addition, the presence of FPD lesions increases the MDA and SOD levels in the footpad tissues.

Buruli ulcer is treatable with antibiotics. An 8-week course of rifampin (RIF) and either streptomycin (STR) or clarithromycin (CLR) cures over 90% of patients. However, STR requires injections and may be toxic, and CLR shares an adverse drug-drug interaction with RIF and may be poorly tolerated. Studies in a mouse footpad infection model showed that increasing the dose of RIF or using the long-acting rifamycin rifapentine (RPT), in combination with clofazimine (CFZ), a relatively well-tolerated antibiotic, can shorten treatment to 4 weeks. CFZ is reduced by a component of the electron transport chain (ETC) to produce reactive oxygen species toxic to bacteria. Synergistic activity of CFZ with other ETC-targeting drugs, the ATP synthase inhibitor bedaquiline (BDQ) and the bc1:aa3 oxidase inhibitor Q203 (now named telacebec), was recently described against Mycobacterium tuberculosis Recognizing that M. tuberculosis mutants lacking the alternative bd oxidase are hypersusceptible to Q203 and that Mycobacterium ulcerans is a natural bd oxidase-deficient mutant, we tested the in vitro susceptibility of M. ulcerans to Q203 and evaluated the treatment-shortening potential of novel 3- and 4-drug regimens combining RPT, CFZ, Q203, and/or BDQ in a mouse footpad model. The MIC of Q203 was extremely low (0.000075 to 0.00015 µg/ml). Footpad swelling decreased more rapidly in mice treated with Q203-containing regimens than in mice treated with RIF and STR (RIF+STR) and RPT and CFZ (RPT+CFZ). Nearly all footpads were culture negative after only 2 weeks of treatment with regimens containing RPT, CFZ, and Q203. No relapse was detected after only 2 weeks of treatment in mice treated with any of the Q203-containing regimens. In contrast, 15% of mice receiving RIF+STR for 4 weeks relapsed. We conclude that it may be possible to cure patients with Buruli ulcer in 14 days or less using Q203-containing regimens rather than currently recommended 56-day regimens.

Background Diabetic neuropathy originating in distal lower extremities is associated with pain early in the disease course, overwhelming in the feet. However, the pathogenesis of diabetic neuropathy remains unclear. Macrophage migration inhibitory factor has been implicated in the onset of neuropathic pain and the development of diabetes. Objective of this study was to observe pain syndromes elicited in the footpad of diabetic neuropathy rat model and to assess the contributory role of migration inhibitory factor in the pathogenesis of diabetic neuropathy. Methods Diabetic neuropathy was made in Sprague Dawley rats by streptozotocin. Pain threshold was evaluated using von Frey monofilaments for 24 weeks. On comparable experiment time after streptozotocin injection, all footpads were prepared for following procedures; glutathione assay, terminal deoxynucleotidyl transferase-mediated biotinylated UTP nick end labeling staining, immunohistochemistry staining, real-time reverse transcription polymerase chain reaction, and Western blot. Additionally, human HaCaT skin keratinocytes were treated with methylglyoxal, transfected with migration inhibitory factor/control small interfering RNA, and prepared for real-time reverse transcription polymerase chain reaction and Western blot. Results As compared to sham group, pain threshold was significantly reduced in diabetic neuropathy group, and glutathione was decreased in footpad skin, simultaneously, cell death was increased. Over-expression of migration inhibitory factor, accompanied by low expression of glyoxalase-I and intraepidermal nerve fibers, was shown on the footpad skin lesions of diabetic neuropathy. But, there was no significance in expression of neurotransmitters and inflammatory mediators such as transient receptor potential vanilloid 1, mas-related G protein coupled receptor D, nuclear factor kappa B, tumor necrosis factor-alpha, and interleukin-6 between diabetic neuropathy group and sham group. Intriguingly, small interfering RNA-transfected knockdown of the migration inhibitory factor gene in methylglyoxal-treated skin keratinocytes increased expression of glyoxalase-I and intraepidermal nerve fibers in comparison with control small interfering RNA-transfected cells, which was decreased by induction of methylglyoxal. Conclusions Our findings suggest that migration inhibitory factor can aggravate diabetic neuropathy by suppressing glyoxalase-I and intraepidermal nerve fibers on the footpad skin lesions and provoke pain. Taken together, migration inhibitory factor might offer a pharmacological approach to alleviate pain syndromes in diabetic neuropathy.

BACKGROUND: Objective of this study is a feasibility-test comparing hock- and footpad-injection in rats with inoculated MatLyLu - adenocarcinoma tumor model. This study compares the development of an adenocarcinoma model (MatLyLu) in 12 Copenhagen rats. Two groups (n = 6) of animals were inoculated with 1 × 106 MatLyLu tumor cells solved in 0.1 ml NaCl either by footpad or hock injection. All animals were examined before tumor inoculation and before euthanasia using a 3.0 Tesla MRI. Histological evaluation of all organs was performed post mortem. RESULTS: Both types of injection were able to induce the adenocarcinoma model using MatLyLu tumor cells. The primary tumor could be visualized in MRI and confirmed histologically. Comparing the risk of reflux and the maximum injection volume during injection, the hock injection was superior to the footpad injection (less reflux, less anatomical restrictions for larger volumes). The hock injection induces a faster tumor growth compared to the footpad injection. As consequence the maximum level of long term discomfort after hock injection was reached earlier, even if it grew on a not weight bearing structure. Early lymph node tumor metastasis could not be observed macroscopically nor detected histologically. Therefore the reproducibility of the MatLyLu tumor model is questionable. CONCLUSION: Hock injection is a feasible alternative technique compared with footpad-injection in rats. It provides a save and easy injection method for various early-terminated applications with the potential to increase animal welfare during tumor models in rats.

1. The performance of the scoring in the Danish footpad dermatitis (FPD) surveillance system was evaluated by determining inter-rater agreement in visual inspection of FPD in broilers between two independent raters (R1 and R2) and the official scoring at a Danish slaughterhouse. 2. FPD scores were evaluated in 1599 chicken feet. The two raters and the slaughterhouse scored equal proportions of score 0. So did R1 and R2 when assessing score 1 and the more severe lesion score 2, whereas the slaughterhouse scored a markedly higher proportion of score 1 and a lower proportion of score 2. Aggregated FPD flock scores ranged from 5 to 163 (R1 and R2) and from 8 to 107 (slaughterhouse). 3. The level of agreement between the two raters was high for scores 0, 1 and 2 and for flock scores. Agreement between raters and the slaughterhouse was lower when R1 and R2 recorded score 2 than when they recorded scores 0 and 1. 4. This study indicates that the occurrence and severity of lesions are underestimated in the official Danish FPD scoring system.

Responses to stocking density (SD), dietary biotin concentration and litter condition were evaluated on 2016 Ross 308 male broilers in the fattening period (day 22-day 42). The birds were placed in 48 pens with either dry or wet litter to simulate the final stocking density of 30 kg (12 broilers/m2 ; normal stocking density, NSD) and 40 kg (16 broilers/m2 ; high stocking density, HSD) of body weight (BW)/m2 floor space. A corn-soybean meal-based diet was supplemented with biotin to provide a normal (NB; 155 µg/kg) or high (HB, 1521 µg/kg) level of dietary biotin. There were six repetitions per treatment. The inappropriate moisture content of litter associated with HSD was avoided (p < 0.05) by good management (SD difference: dry litter, 6.65% vs. wet litter, 13.23%; 42 days), which made it advantageous (p < 0.01) for footpad (SD difference: dry litter, 0.118 vs. wet litter, 0.312; weekly average value) and hock health (SD difference: dry litter, 0.090 vs. wet litter, 0.303; weekly average value) of HSD birds, but not (p > 0.05) for growth and processing yield. In HSD, the biotin effect (gains, FCR) was significantly higher (p < 0.01) than in NSD. The similar response of HSD birds to supplemental biotin was observed (p < 0.05) for lesion scores of footpad and hock in particularly finishing chickens, and a significant interaction (p < 0.01) among stocking density, biotin supplementation and litter condition existed from 35 to 42 days of age. Taken together, increasing dietary biotin improves the performance and well-being of broiler chickens stocked at high densities in litter-independent and litter-dependent manners respectively.

OBJECTIVE: An experiment was conducted to investigate the effect of dietary Ca concentrations in low non-phytate phosphorus (NPP) diets containing phytase on growth performance, bone mineralization, litter quality, and footpad dermatitis (FPD) incidence in growing broiler chickens. METHODS: A total of 1,800 21-day-old Ross 308 growing broiler chickens were allotted to 1 of 6 dietary treatments with 6 replicated cages. Six diets were formulated to provide increasing Ca concentrations of 4.0, 5.0, 6.0, 7.0, 8.0, or 9.0 g/kg in diets. The concentrations of NPP in all diets were maintained at 3.0 g/kg, and phytase was supplemented to all diets at the level of 1,000 fytase units (FTU)/kg. At the end of the 14-d feeding trial, birds were euthanized for tibia sampling, and litter samples were collected from 3 areas in the cage. The FPD incidence was measured based on a 6-point scoring system. RESULTS: Dietary Ca concentrations had no effect on growth performance of growing broiler chickens. However, a tendency (linear, p = 0.05) for decreased feed efficiency was observed as dietary Ca concentrations were increased. The concentrations of Ca and P in the tibia of broiler chickens increased (linear and quadratic, p<0.01) with increasing Ca concentrations in low NPP diets containing phytase. Litter pH, moisture, and N contents were not affected by increasing Ca concentrations in low NPP diets containing phytase. However, a tendency (quadratic, p = 0.10) for increased FPD incidence with increasing dietary Ca concentrations was observed. CONCLUSION: Dietary Ca concentrations from 4.0 to 9.0 g/kg in low NPP diets containing phytase have little effects on growth performance of growing broiler chickens. However, Ca and P concentrations in the tibia are decreased if dietary Ca concentrations are less than 5.0 g/kg. The FPD incidence for growing broiler chickens may be decreased if less than 9.0 g/kg of Ca is included in diets.

One experiment was conducted to evaluate the effects of two incubation temperature profiles (TEM) and two trace mineral (TM) sources with their inclusion levels on performance and footpad skin development of Ross 708 chickens. A total of 1,000 eggs from 29-wk-old breeders were incubated following two TEM profiles: a standard (S) eggshell temperature (37.8±0.2°C) for 21 d of incubation and an early-low late-high (LH) TEM. The second profile had low (37.2±0.2°C) temperature for the first 3 d, and S until the last 3 d when eggshell temperature was 39.2±0.2°C. At hatch, 15 male and 15 female chicks from each TEM were selected, and footpads sampled. Additionally, 168 males per TEM were placed in 24 battery cages with 7 chickens each. The 48 cages were assigned to two TM dietary treatments: one with inorganic (ITM) sources of Zn (120 ppm), Cu (10 ppm), and Mn (120 ppm) and the other with chelated (CTM) mineral sources using lower inclusion levels of Zn (32 ppm), Cu (8 ppm), and Mn (32 ppm). At 7 and 21 d, BW gain and feed conversion ratio (FCR) were obtained and chicks sampled for footpads. Histological analysis assessed thickness and area ofstratus corneum(SC), epidermis, and dermis. Data were analyzed as a completely randomized block design in a 2×2 factorial arrangement of treatments with TEM and sex as main factors for hatch data and TEM and TM diets for 7 and 21 d data. The LH chicks were heavier than S chicks at hatch, but had more residual yolk. However, S TEM male chickens were heavier at 7 and 21 d. The S TEM had better FCR than LH TEM.Papillaedermis parameters at hatch were higher in the S TEM. At 7 d, SC height and area were increased by the S TEM. At 21 d CTM increased dermis height and area. In conclusion, TEM affected footpad skin development and broiler performance. Replacing ITM with reduced levels of CTM increased dermis development without affecting live performance or other skin layers.

Two experiments were conducted in which effects of floor eggs, washed floor eggs, and clean nest eggs were investigated on incubation characteristics and performance in later life of broiler chickens. In both experiments, a young and an older breeder flock were used in a 3×2 factorial design during incubation. In the second experiment, male and female chickens were reared separately until d 35 of age in floor pens. During this grow out trial, an extra group was created in which chickens obtained from clean nest eggs were mixed with chickens obtained from floor eggs, meaning that grow out period was set up as a 4×2×2 factorial design with 4 egg types, 2 breeder ages, and 2 sexes. In both experiments, fertility and hatchability of fertile eggs were lower in floor and washed eggs than in clean nest eggs (hatchability: experiment 1: 74.4 vs. 70.6 vs. 92.6% for floor eggs, washed floor eggs and clean nest eggs, respectively, P<0.001; experiment 2: 78.3 vs. 81.7 vs. 90.2%, respectively, P<0.001). In experiment 2, BW at d 0 of chickens obtained from clean nest eggs was higher than that of chickens from floor eggs and washed floor eggs (41.5 vs. 40.4 and 40.3 g, respectively; P<0.001). This difference disappeared during the grow out period and was absent at slaughter age at d 35 of age. Feed intake (FI), feed conversion ratio (FCR), and mortality during the grow out period were not affected by egg type. Incidence and severity of hock burns and footpad dermatitis were not affected by egg type or breeder age. Litter friability at d 35 of age tended to be lower in pens with chickens obtained from washed floor eggs compared to clean nest eggs. We conclude that incubation of floor eggs or washed floor eggs resulted in lower fertility and hatchability compared to clean nest eggs, but that performance during the grow out period was not affected.

Three rock hyraxes (Procavia capensis) maintained in a zoological collection had chronic hypercalcemia and azotemia. In addition, all animals displayed signs of lameness due to footpad lesions that were histologically characterized as marked metastatic mineralization with granulomatous inflammation, reminiscent of calcinosis circumscripta. Although the animals were managed with aggressive fluid therapy, calciuresis, and dietary modification, all were eventually humanely euthanized due to the severity of their footpad lesions and/or progression of renal disease. Metastatic mineralization was also noted in other soft tissues among the three cases, including the stomach, colon, lung, vascular wall, ovary, and kidney. Varying degrees of interstitial nephritis were confirmed on postmortem examination, and in the absence of other causes for hypercalcemia, metastatic mineralization was presumably the consequence renal dysfunction. The renal pathway is the primary mode of calcium excretion in the rock hyrax. In renal dysfunction, hypercalcemia may develop secondary to decreased calcium excretion. Footpad mineralization is an uncommon sequel to renal dysfunction in domestic animals but has not been reported in rock hyraxes. A retrospective review of mortality data in this collection revealed a notable prevalence of renal lesions, including two additional animals with metastatic mineralization and renal dysfunction. Expanding knowledge of renal diseases will further guide preventative and clinical measures, including screening for metastatic mineralization and therapeutic trials for management of hypercalcemia and calcium mineral deposition in the footpads and other soft tissues.

Epidermal Langerhans cells (LCs) and dermal dendritic cells (dDCs) capture cutaneous antigens and present them to T-cells in lymph nodes (LNs). The function of LCs and Langerin+ dDCs was extensively studied in the mouse, but their anatomical repartition is unknown. Here, we found LCs in back skin, footpads and tail skin of C57BL/6, BALB/c, 129/Sv and CBA/J mice. Langerin+ dDCs were readily observed in back skin of all strains, but only in footpads and tail of BALB/c and CBA/J mice. Similarly, while LCs were equally present in all LNs and strains, Langerin+ dDCs were found in popliteal LNs (draining footpads) only in BALB/c and CBA/J mice. The sciatic LNs, which we identified as the major tail-draining lymphoid organ, were devoid of Langerin+ dDCs in all strains. Thus, functionally different DCs reside in different skin areas, with variations among mouse strains, implying a potential impact on the cutaneous immune reaction.

Footpad dermatitis begins early in life, and there is evidence of individual susceptibility. An experiment was conducted to evaluate the carryover effects of breeder feed restriction programs and incubation temperatures (TEM) on progeny footpad development at hatch, and 7 and 22 d. Cobb 500 fast feathering breeders were subjected to 2 dietary feed restriction programs during rearing: skip-a-day (SAD) and every-day feeding (EDF). At 60 wk of age, eggs from each group were collected and incubated according to 2 TEM, standard (S) eggshell temperature (38.1°C) and early-low late-high (LH). This second profile had low (36.9°C) eggshell temperature for the first 3 d, and standard temperature until the last 3 d when eggs were subjected to elevated (38.9°C) eggshell temperature. At hatch, 15 chicks from each treatment combination were sampled to obtain footpads for histological analysis. Seventy-two chicks per treatment were placed in 48 cages (6/cage), and raised to 22 d. At 7 and 22 d, 1 and 2 chickens, respectively, were sampled for footpads. The BW and group feed intake were recorded to obtain BW gain and feed conversion ratio at 7 and 21 d. Histological analysis assessed thickness and total area of stratus corneum (SC), epidermis, and dermis, and total papillae height. Data were analyzed as randomized complete block design in a 2 × 2 factorial arrangement of treatments. There was a negative effect of LH TEM on performance at both ages. An interaction effect on SC area and papillae height was observed at hatch. Additionally, SAD treatment increased thickness and area of footpad dermis. At 7 d, the SC parameters of the SAD progeny were increased. Epidermis thickness was affected by treatment interaction. Furthermore, LH TEM decreased epidermis thickness and dermis area. At 22 d, interaction effects were observed in thickness and area of SC and epidermis. Incubation S TEM increased thickness and area of dermis. It was concluded that breeder feed restriction programs and incubation TEM profiles may have carryover effects on histomorphological traits of footpads.

Supplementation of a combination of lysolecithin, a synthetic emulsifier, and monoglycerides (LEX) in liquid and dry form to broiler diets with different energy levels was investigated to determine their effect on performance, litter quality and subsequent occurrence of footpad lesions. One thousand two hundred and forty-eight-day-old Ross 308 broilers were assigned to 1 of 6 treatments for a 42-day study: a basal diet with a normal energy content (NE); NE + 300 g/t LEX in liquid form (LEL); NE + 500 g/t LEX in dry form (LED); a basal diet with low energy (LE, -90 kcal/kg starter, -100 kcal/kg grower, finisher), LE + 300 g/t LEL and a LE + 500 g/t LED. Each treatment consisted of 13 pens of 16 birds each. Diets were fed in 3 phases (starter d 0-10, grower d 11-21, finisher d 22-42). Feed intake and weight were measured on d 0, 10, 21, and 42. On d 42 a litter sample was collected from each pen and 2 birds per pen were assessed for footpad lesions and breast scald. Data were analyzed using JMP 16, with means separation achieved using Tukey's HSD; significance was assumed at P < 0.05. Results showed a higher (P < 0.05) cumulative bodyweight gain with LEX supplementation (NE CON = 2,718 g, NE+LED = 2,829, NE+LEL = 2,895, LE CON = 2,722, LE+LED = 2,787, LE+LEL = 2,893; P = 0.0027). An increased feed intake was observed for the LE diets, however cumulative FCR of LE+LED and LE+LEL remained equal to the NE control (1.657 NE CON, 1.657 LE+LED, 1.623 LE+LEL; P > 0.05), suggesting LEX enabled the birds to compensate for the energy gap. Litter dry matter was significantly improved with both LED and LEL supplementation compared to the control groups, and resulted in lower (P < 0.05) occurrence and severity of footpad lesions and breast scalds. Considering the income over feed cost (IOFC) of the NE treatment as the reference point for comparison, all other treatments improved profitability, with NE+LEL and LE+LEL achieving the greatest IOFC with 154.58 and 175.96 €/1,000 birds respectively. In conclusion, feeding broilers a combination of lysophospholipids, a synthetic emulsifier and monoglycerides resulted in improved bird performance. The use of the LEX also improved litter quality and footpad health, therefore improving animal welfare indicators such as breast scald and footpad measurements.

Light flicker is a commonly overlooked factor of artificial light sources. This study aimed to determine the impacts of light-flicker frequency on performance, general health, and mortality of 11-wk Nicholas Select turkey hens. The experiment consisted of 2 trials (block) in a randomized complete block design, with 3 light-flicker frequency treatments (30, 90, or 195 Hz). Turkeys (n = 364 per replicate) were randomly placed into environmentally controlled rooms (3 room replicates per treatment per trial). Group body weight (BW) and feed consumption were measured at 0, 4, 8, and 11 wk, and feed efficiency (mortality corrected feed-to-gain; F:Gm) was calculated for each period. Mortality and culls were collected twice daily. Flock uniformity, feather condition and cleanliness, footpad score, and mobility were evaluated at 10 wk (30 birds per room). Litter quality and ocular weight and dimensions were evaluated (11 wk; 4 birds per room). Data were analyzed using Proc Mixed (SAS 9.4) and significance was declared when P < 0.05. At 8 wk, BW was lower under 30 Hz compared to 195 Hz (P = 0.03). Feed consumption was lowest under 30 Hz (0-4 wk and 4-8 wk; P < 0.01). Mortality-corrected F:G was improved under 30 Hz for 8 to 11 wk and 0 to 11 wk (P = 0.05 and P = 0.04, respectively). Total mortality was lower under 195 Hz compared to 30 Hz (P = 0.02). Uniformity, gait score, feather condition, and litter quality were unaffected by flicker. Footpad scores were improved under 90 Hz (P = 0.01), leading to an improved average footpad score (P = 0.02). Feather cleanliness was improved under 90 Hz compared to both 30 Hz and 195 Hz (P<0.01). Right eyeball dimensions differed across lighting treatments, with the dorso-ventral diameter being larger in birds under 30 Hz compared to 195 Hz (P = 0.05). The anterior-posterior size also increased in birds under 30 Hz compared to 90 Hz (P = 0.03). Light flicker impacted turkey hens, with the results demonstrating negative impacts on early growth and changes to ocular characteristics.
