RESUMO

Reducing the levels of dietary protein is an effective nutritional approach in lowering feed cost and nitrogen emissions in ruminants. The purpose of this study was to evaluate the effects of dietary Lys/Met ratio in a low protein diet (10%, dry matter basis) on the growth performance and hepatic function (antioxidant capacity, immune status, and glycolytic activity) in Tibetan lambs. Ninety two-month-old rams with an average weight of 15.37 ± 0.92 kg were randomly assigned to LP-L (dietary Lys/Met = 1:1), LP-M (dietary Lys/Met = 2:1) and LP-H (dietary Lys/Met = 3:1) treatments. The trial was conducted over 100 d, including 10 d of adaption to the diets. Hepatic phenotypes, antioxidant capacity, immune status, glycolytic activity and gene expression profiling was detected after the conclusion of the feeding trials. The results showed that the body weight was higher in the LP-L group when compared to those on the LP-M group (P < 0.05). In addition, the activities of the catalase (CAT) and glutathione peroxidase (GSH-Px) in the LP-L group were significantly increased compared with the LP-M group (P < 0.05), while the malondialdehyde (MDA) levels in LP-H group were significantly decreased (P < 0.05). Compared with LP-H group, both hepatic glycogen (P < 0.01) and lactate dehydrogenase (LDH) (P < 0.05) were significantly elevated in LP-L group. For the LP-L group, the hepatocytes were arranged radially with the central vein in the center, and hepatic plates exhibited tight arrangement. Transcriptome analysis identified 29, 179, and 129 differentially expressed genes (DEGs) between the LP-M vs. LP-L, LP-H vs. LP-M, and LP-H vs. LP-L groups, respectively (Q-values < 0.05 and |log2Fold Change| > 1). Gene Ontology (GO) and correlation analyses showed that in the LP-L group, core genes (C1QA and JUNB) enriched in oxidoreductase activity were positively correlated with antioxidant indicators, while the MYO9A core gene enriched in the immune response was positively associated with immune indicators, and core genes enriched in molecular function (PDK3 and PDP2) were positively correlated with glycolysis indicators. In summary, low-protein diet with a low Lys/Met ratio (1:1) could reduce the hepatic oxidative stress and improve the glycolytic activity by regulating the expression of related genes of Tibetan sheep.

BACKGROUND: Vitamin A and retinoic acid (RA, a metabolite of vitamin A), are inextricably involved to the development of skeletal muscle in animals. However, the mechanisms regulating skeletal muscle development by vitamin A remain poorly reported. The current study designed to investigate the underlying mechanism of vitamin A affecting myogenic differentiation of lamb myoblasts through transcriptome sequencing (RNA-Seq) and gene function validation experiments. It provides a theoretical basis for elucidating the regulation of vitamin A on skeletal muscle development as well as for improving the economic benefits of the mutton sheep industry. RESULTS: Newborn lambs were injected with 7,500 IU vitamin A, and longissimus dorsi (LD) muscle tissue was surgically sampled for RNA-Seq analysis and primary myoblasts isolation at 3 weeks of age. The results showed that a total of 14 down-regulated and 3 up-regulated genes, were identified between control and vitamin A groups. Among them, BHLHE40 expression was upregulated in vitamin A group lambs. Furthermore, BHLHE40 expression is significantly increased after initiation of differentiation in myoblasts, and RA addition during differentiation greatly promoted BHLHE40 mRNA expression. In vitro, RA inhibited myoblasts proliferation and promoted myoblasts myogenic differentiation through BHLHE40. Moreover, BHLHE40 was proved to inhibit the expression of the DNA binding inhibitor 3 (ID3), and meanwhile, ID3 could effectively promote myoblasts proliferation and inhibit myoblasts myogenic differentiation. CONCLUSIONS: Taken together, our results suggested that vitamin A inhibited myoblasts proliferation and promoted myoblasts myogenic differentiation by inhibiting ID3 expression through BHLHE40.

BACKGROUND: Anti-p200 pemphigoid is a subepidermal autoimmune blistering disease (AIBD) characterized by autoantibodies against a 200 kDa protein. Laminin γ1 has been described as target antigen in 70% to 90% of patients. No diagnostic assay is widely available for anti-p200 pemphigoid, which might be due to the unclear pathogenic relevance of anti-laminin γ1 autoantibodies. OBJECTIVE: To identify a target antigen with higher clinical and diagnostic relevance. METHODS: Immunoprecipitation, mass spectrometry, and immunoblotting were employed for analysis of skin extracts and sera of patients with anti-p200 pemphigoid (n = 60), other AIBD (n = 33), and healthy blood donors (n = 29). To localize the new antigen in skin, cultured keratinocytes and fibroblasts, quantitative real-time polymerase chain reaction and immunofluorescence microscopy were performed. RESULTS: Laminin ß4 was identified as target antigen of anti-p200 pemphigoid in all analyzed patients. It was located at the level of the basement membrane zone of the skin with predominant expression in keratinocytes. LIMITATIONS: A higher number of sera needs to be tested to verify that laminin ß4 is the diagnostically relevant antigen of anti-p200 pemphigoid. CONCLUSION: The identification of laminin ß4 as an additional target antigen in anti-p200 pemphigoid will allow its differentiation from other AIBD and as such, improve the management of these rare disorders.

BACKGROUND: Compared with corn, wheat contains higher crude protein, amino acids concentration. However, wheat contains a mass of anti-nutritional factors, resulting in increased of the digesta viscosity and impaired the intestinal function in ruminant. OBJECTIVE: This study aimed to investigate the effects of substitution of different amounts of wheat for corn on hepatic metabolism in the Tibetan lamb. METHODS: Ninety Tibetan lambs (Body weight = 12.37 ± 0.92 kg) were randomly assigned to three groups: 0% wheat diet (Control), 10% wheat diet (Low group), and 15% wheat diet (High group). The feeding trial lasted for 130 d, including a 10 d adaption period. Hepatic gene expression profiling was performed via RNA sequencing after the conclusion of the feeding trials. RESULTS: Results showed that greater level of glutathione peroxidase levels in L group compared with those of the C and H groups (P < 0.05). The immune indexes, including interleukin-1ß (IL-1ß), immunoglobulin A (IgA), and IgM were also elevated in L group compared with the other groups (P < 0.05). Compared with H group, the hepatocytes were arranged radially, and hepatic plates anastomosed with each other to form a labyrinth-like structure in L group. Transcriptomic analysis showed 872 differentially expressed genes (DEG) between H and L group, of which 755 were down-regulated and 117 were up-regulated. Through Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis, 32 pathways were significantly enriched (Q-value < 0.05), such as the cAMP signaling pathway, Th1 and Th2 cell differentiation, leukocyte transendothelial migration, platelet activation and adipocytokine signaling pathway. Additionally, the expression of comment DEGs were verified via quantitative reverse-transcription polymerase chain reaction. CONCLUSION: In summary, our findings suggest that wheat can be supplemented up to 10% in Tibetan sheep, contributing to improve the hepatic oxidative stress, immune response and lipid metabolism through regulating the expression of related genes.

A well-developed heart is essential for embryonic survival. There are constant interactions between cardiac tissue motion and blood flow, which determine the heart shape itself. Hemodynamic forces are a powerful stimulus for cardiac growth and differentiation. Therefore, it is particularly interesting to investigate how the blood flows through the heart and how hemodynamics is linked to a particular species and its development, including human. The appropriate patterns and magnitude of hemodynamic stresses are necessary for the proper formation of cardiac structures, and hemodynamic perturbations have been found to cause malformations via identifiable mechanobiological molecular pathways. There are significant differences in cardiac hemodynamics among vertebrate species, which go hand in hand with the presence of specific anatomical structures. However, strong similarities during development suggest a common pattern for cardiac hemodynamics in human adults. In the human fetal heart, hemodynamic abnormalities during gestation are known to progress to congenital heart malformations by birth. In this chapter, we discuss the current state of the knowledge of the prenatal cardiac hemodynamics, as discovered through small and large animal models, as well as from clinical investigations, with parallels gathered from the poikilotherm vertebrates that emulate some hemodynamically significant human congenital heart diseases.

Colostrum is the initial secretion of the mammary glands following parturition, which offers main food, protection, and biological active substances for the new born. The most threatening episode of neonate's life is the initial two weeks after birth. This period is associated with high neonatal mortality and morbidity. These worthwhile losses lead to a poor prolificacy rate, low profitability, and ultimately poor performance in animal production. Hence, both diseases and mortality cause valuable losses in terms of production and economic losses. The survival of neonate is correlated with their immune status and passive immune transfer (PIT). Colostrum provides the primary source of nutrition and immunity (PIT) that protects neonates against infections. It must be given as soon as possible after birth since its immunoglobulins are absorbed within the first 16-27 hours after birth, ideally within 2-4 hours. As a result, immunoglobulin (PIT) is the most important component of distressing infectious immunity, and a passable concentration of immunoglobulin in the blood of newborn lambs is linked to their health and survival rate. In this review, we summarized the importance of colostrum in early life and its association with neonatal lamb's survival, profitability and productivity of sheep farming.

Microplastics (MPs), recognized as an emerging environmental menace, have been extensively investigated in both marine and terrestrial fauna. This study is comprehensive to investigate how polystyrene (PS) affects ruminant animals. The experimental design comprised 24 individually housed lambs, divided into a CON group (diet without PS) and three PS-exposed (25 µm, 50 µm, 100 µm) groups, each with six lambs, the exposure of PS was 100 mg/day, and the duration of exposure was 60 days. The study yielded noteworthy results: (ⅰ) PS leads to a decrease in average daily gain along with an increase in feed conversion rate. (ⅱ) PS decreases rumen ammonia nitrogen. The rumen microbiota diversity remains consistent. However, the relative abundance of Bacteroidetes and Actinobacteria increased in the PS-exposed groups, while the relative abundance of Coriobacteriales_incertae_Sedis and Prevotellaceae_YAB2003_group decreased. (ⅲ) PS leads to decrease in hemoglobin, thrombocytocrit, and albumin levels in lamb blood, thus triggering oxidative stress accumulation, along with swelling of the kidneys and liver. (ⅳ) PS inflicts severe damage to jejunum, consequently impacting digestion and absorption. (ⅴ) PS reduces meat quality and the nutritional value. In conclusion, PS-exposure inhibited lambs' digestive function, adversely affects blood and organs' health status, reducing average daily gain and negatively influencing meat quality. PS particles of 50-100 µm bring worse damage to lambs. This research aims to fill the knowledge void concerning MPs' influences on ruminant animals, with a specific focus on the meat quality of fattening lambs.

Composite materials, valued in aerospace for their stiffness, strength and lightness, require impact monitoring for structural health, especially against low-velocity impacts. The MUSIC algorithm, known for efficient directional scanning and easy sensor deployment, is gaining prominence in this area. However, in practical engineering applications, the broadband characteristics of impact response signals and the time delay errors in array elements' signal reception lead to inconsistencies between the steering vector and the actual signal subspace, affecting the precision of the MUSIC impact localization method. Furthermore, the anisotropy of composite materials results in time delay differences between array elements in different directions. If the MUSIC algorithm uses a fixed velocity value, this also introduces time delay errors, further reducing the accuracy of localization. Addressing these challenges, this paper proposes an innovative MUSIC algorithm for impact imaging using a guided Lamb wave array, with an emphasis on time delay management. This approach focuses on the extraction of high-energy, single-frequency components from impact response signals, ensuring accurate time delay measurement across array elements and enhancing noise resistance. It also calculates the average velocity of single-frequency components in varying directions for an initial impact angle estimation. This estimated angle then guides the selection of a specific single-frequency velocity, culminating in precise impact position localization. The experimental evaluation, employing equidistantly spaced array elements to capture impact response signals, assessed the effectiveness of the proposed method in accurately determining array time delays. Furthermore, impact localization tests on reinforced composite structures were conducted, with the results indicating high precision in pinpointing impact locations.

The early detection of small cracks in large metal structures is a crucial requirement for the implementation of a structural health monitoring (SHM) system with a low transducers density. This work tackles the challenging problem of the early detection of submillimeter notch-type defects with a semielliptical shape and a groove at a constant width of 100 µm and 3 mm depth in a 4.1 mm thick aluminum plate. This defect is investigated with an ultrasonic guided wave (UGW) A0 mode at 550 kHz to investigate the long range in thick metal plates. The mode selection is obtained by interdigital transducers (IDTs) designed to operate with a 5 mm central wavelength. The novel contribution is the validation of the detection by pulse-echo and pitch and catch with UGW transducers to cover a distance up to 70 cm to reduce the transducers density. The analysis of scattering from this submillimeter defect at different orientations is carried out using simulations with a Finite Element Model (FEM). The detection of the defect is obtained by comparing the scattered signals from the defect with baseline signals of the pristine laminate. Finally, the paper shows that the simulated results are in good agreement with the experimental ones, demonstrating the possible implementation in an SHM system based on the efficient propagation of an antisymmetric mode by IDTs.

Thin copper plate is widely used in architecture, transportation, heavy equipment, and integrated circuit substrates due to its unique properties. However, it is challenging to identify surface defects in copper strips arising from various manufacturing stages without direct contact. A laser ultrasonic inspection system was developed based on the Lamb wave (LW) produced by a laser pulse. An all-fiber laser heterodyne interferometer is applied for measuring the ultrasonic signal in combination with an automatic scanning system, which makes the system flexible and compact. A 3-D model simulation of an H62 brass specimen was carried out to determine the LW spatial-temporal wavefield by using the COMSOL Multiphysics software. The characteristics of the ultrasonic wavefield were extracted through continuous wavelet transform analysis. This demonstrates that the A0 mode could be used in defect detection due to its slow speed and vibrational direction. Furthermore, an ultrasonic wave at the center frequency of 370 kHz with maximum energy is suitable for defect detection. In the experiment, the size and location of the defect are determined by the time difference of the transmitted wave and reflected wave, respectively. The relative error of the defect position is 0.14% by averaging six different receiving spots. The width of the defect is linear to the time difference of the transmitted wave. The goodness of fit can reach 0.989, and it is in good agreement with the simulated one. The experimental error is less than 0.395 mm for a 5 mm width of defect. Therefore, this validates that the technique can be potentially utilized in the remote defect detection of thin copper plates.

Delamination represents one of the most significant and dangerous damages in composite plates. Recently, many papers have presented the capability of structural health monitoring (SHM) techniques for the investigation of structural delamination with various shapes and thickness depths. However, few studies have been conducted regarding the utilization of convolutional neural network (CNN) methods for automating the non-destructive testing (NDT) techniques database to identify the delamination size and depth. In this paper, an automated system qualified for distinguishing between pristine and damaged structures and classifying three classes of delamination with various depths is presented. This system includes a proposed CNN model and the Lamb wave technique. In this work, a unidirectional composite plate with three samples of delamination inserted at different depths was prepared for numerical and experimental investigations. In the numerical part, the guided wave propagation and interaction with three samples of delamination were studied to observe how the delamination depth can affect the scattered and trapped waves over the delamination region. This numerical study was validated experimentally using an efficient ultrasonic guided waves technique. This technique involved piezoelectric wafer active sensors (PWASs) and a scanning laser Doppler vibrometer (SLDV). Both numerical and experimental studies demonstrate that the delamination depth has a direct effect on the trapped waves' energy and distribution. Three different datasets were collected from the numerical and experimental studies, involving the numerical wavefield image dataset, experimental wavefield image dataset, and experimental wavenumber spectrum image dataset. These three datasets were used independently with the proposed CNN model to develop a system that can automatically classify four classes (pristine class and three different delamination classes). The results of all three datasets show the capability of the proposed CNN model for predicting the delamination depth with high accuracy. The proposed CNN model results of the three different datasets were validated using the GoogLeNet CNN. The results of both methods show an excellent agreement. The results proved the capability of the wavefield image and wavenumber spectrum datasets to be used as input data to the CNN for the detection of delamination depth.

The carbon-fiber-reinforced polymer (CFRP) bending structure is widely used in aviation. The emergence and spread of delamination damage will decrease the safety of in-service bending structures. Lamb waves can effectively identify delamination damage as a high-damage-sensitivity detection tool. For this present study, the signal difference coefficient (SDC) was introduced to quantify delamination damage and evaluate the sensitivity of A0-mode and S0-mode Lamb waves to delamination damage. The simulation results show that compared with the S0-mode Lamb wave, the A0-mode Lamb wave exhibits higher delamination damage sensitivity. The delamination damage can be quantified based on the strong correlation between the SDC and the delamination damage size. The control effect of the linear array PZT phase time-delay method on the Lamb wave mode was investigated by simulation. The phase time-delay method realizes the generation of a single-mode Lamb wave, which can separately excite the A0-mode and S0-mode Lamb wave to identify delamination damage of different sizes. The A0-mode Lamb wave was excited by the developed one-dimensional miniaturized linear comb transducer (LCT), which was used to conduct the detection experiment on the CFRP bending plate with delamination damage sizes of Φ6.0 mm, Φ10.0 mm, and Φ15.0 mm. The experimental results verify the correctness of the simulation. According to the Hermite interpolation results of the finite-element simulation data, the relationship between the delamination damage size and the SDC was fitted by the Gaussian function and Rational function, which can accurately quantify the delamination damage. The absolute error of the delamination damage quantification with Gaussian and Rational fitting expression does not exceed 0.8 mm and 0.7 mm, and the percentage error is not more than 8% and 7%. The detection and signal processing methods employed in the present research are easy to operate and implement, and accurate delamination damage quantification results have been obtained.

Nearly all cervical cancer cases are caused by infection with high-risk human papillomavirus (HR-HPV) types. The mechanism of cervical cell transformation is related to the powerful action of viral oncoproteins and cellular gene alterations. Transcriptomic data from cervical cancer and normal cervical cells were utilized to identify upregulated genes and their associated pathways. The laminin subunit beta-3 (LAMB3) mRNAwas overexpressed in cervical cancer and was chosen for functional analysis. The LAMB3 was predominantly expressed in the extracellular region and the plasma membrane, which play a role in protein binding and cell adhesion molecule binding, leading to cell migration and tissue development. LAMB3 was found to be implicated in the pathway in cancer and the PI3K-AKT signaling pathway. LAMB3 knockdown decreased cell migration, invasion, anchorage-dependent and anchorage-independent cell growth and increased the number of apoptotic cells. These effects were linked to a decrease in protein levels involved in the PI3K-AKT signaling pathway and an increase in p53 protein. This study demonstrated that LAMB3 could promote cervical cancer cell migration, invasion and survival.

As one of the largest and most diverse classes of specialized metabolites in plants, terpenoids (oprenoid compounds, a type of bio-based material) are widely used in the fields of medicine and light chemical products. They are the most important secondary metabolites in coniferous species and play an important role in the defense system of conifers. Terpene synthesis can be promoted by regulating the expressions of terpene synthase genes, and the terpene biosynthesis pathway has basically been clarified in Pinus massoniana, in which there are multiple rate-limiting enzymes and the rate-limiting steps are difficult to determine, so the terpene synthase gene regulation mechanism has become a hot spot in research. Herein, we amplified a PmDXR gene (GenBank accession no. MK969119.1) of the MEP pathway (methyl-erythritol 4-phosphate) from Pinus massoniana. The DXR enzyme activity and chlorophyll a, chlorophyll b and carotenoid contents of overexpressed Arabidopsis showed positive regulation. The PmDXR gene promoter was a tissue-specific promoter and can respond to ABA, MeJA and GA stresses to drive the expression of the GUS reporter gene in N. benthamiana. The DXR enzyme was identified as a key rate-limiting enzyme in the MEP pathway and an effective target for terpene synthesis regulation in coniferous species, which can further lay the theoretical foundation for the molecularly assisted selection of high-yielding lipid germplasm of P. massoniana, as well as provide help in the pathogenesis of pine wood nematode disease.

Plantago species, which is known to adapt to different climatic conditions, drought, temperature and different soil types. The aim of this study was to compare the effects of P. lanceolata herbage on fattening performance, rumen variables, digestibility, meat quality, meat nutrients and meat textural variables in lambs with the values of Italian ryegrass and sainfoin herbages. The lambs were fed total mix ration (TMR) with concentrated feed (about 30%) plus forage (about 70%) (P. lanceolata, PLA, Italian ryegrass (Lolium multiflorum); IRY or sainfoin (Onobrychis viciifolia) herbages, SAI). Twenty-four male lambs (Akkaraman breed) were assigned to three treatments with 8 lambs in each group. Fattening performance parameters, digestibility and carcass variables were determined. Meat quality and shelf life variables (oxidation, fatty acid profile and textural variables) was detected in the loin (Musculus longissimus), shoulder (M. deltoideus) and leg (M. semitendinosus) muscle samples, which were rested for 24 h +4°C and stored at -20°C for one week, one month, three months and six months. The dry matter (DM) intake, body weight gain, feed efficiency and carcass yield values of lambs, the DM and organic matter (OM) digestions (DMD and OMD) of TMRs, and the pH value, ammonia-nitrogen and short chain fatty acid (SCFA) concentrations of rumen fluid in lambs consuming PLA were similar to those of IRY and SAI (p > 0.05). PLA increased water holding capacity (WHC) and decreased cooking loss of meat compared to feeding with those of IRY and SAI (p < 0.05). The peroxide value (PV) and malondialdehyde (MDA) concentration increased with the stocking time in the meats (p < 0.05). Therefore, PV and MDA concentrations of shoulder, leg and loin meats up to 6th months for PLA were lower than those of IRY and SAI (p < 0.05). The linoleic acid concentration of loin and shoulder meats in PLA were higher than those of IRY and SAI (p < 0.05). The average oleic acid and ∑ω6 fatty acids concentration of meat in PLA was lower than those in IRY and SAI (p < 0.05). Atherogenic index and thrombogenic index values and palmitic acid levels of meat in PLA were similar to those in IRY, but lower than those in SAI (p < 0.05). Springiness, cohesiveness, resilience, hardness and chewiness values of the textural analysis parameters in meat of PLA were lower than those of IRY and SAI. As a result, effect of P. lanceolata on the fattening performance and rumen fermentation of lambs were similar with those of Italian ryegrass (Gramineae family) and sainfoin (Leguminosae family) forages and, it had a positive effect on meat quality (water holding capacity, cooking loss) and meat shelf life criteria (MDA, PV, fatty acid profile and textural variables). For functional lamb meat production, P. lanceolata can be included in the diet of lamb fattening.

Under specific agroclimatic conditions, growth is an adaptability parameter resulting in reduced performance with decreasing animal comfort of a sheep breed. In this study, a total of 1011 lambs' live weights (LW) and average daily weight gains (ADG) in 16 different farms between 2018 and 2020 (birth to 18 months) were determined in Akkaraman herds raised in Çankiri Province of Turkey. Comprehensive Climate Index (CCI) was examined to determine the effect of climate. Thermal comfort limits in CCI values were classified as CCI-1.cold (≤ 5 ℃), CCI-2.comfort (5-25 ℃), and CCI-3.warm (≥ 25 ℃). The climatic values that all lambs examined were exposed to according to index value groups in periods from birth to 18 months of age were prepared daily. The lambs were exposed to comfort environment in the majority of the days in the MP0-3 (77.7%) and MP12-18 (50.1%) periods. The lambs mostly in the MP3-6 period (72.2%) were above the warm stress threshold, but in the MP6-12 period, 54.7% were above the cold stress threshold. Overall mean LW of lambs at birth, at 3, 6, 12, and 18 months were 4.29 kg, 35.1 kg, 39.1 kg, 45.4 kg, and 52.6 kg, while the ADG values between birth-3 months, 3-6 months, 6-12 months, and 12-18 months were 341.8 g, 54.5 g, 42.9 g, and 56.8 g respectively. Periodically, CCI values were realized as 10-15, 25-30, and 18-22 in the growth periods of 0-3, 3-6, and 12-18 months, respectively, and ADG values of the lambs were found to be similar within their own periods. However, when the CCI values of the lambs increase to 5 and above in the 06-12-month period, there is a decrease in their live weight. As a result, climatic index values did not have a negative effect on the growth and development values of lambs in cold conditions in general, but warm conditions affected the animals negatively.

The aim of this study was to compare the performance, intake, digestibility, ruminal parameters, carcass traits, and the yield of commercial cuts of Santa Ines (SI) and Rabo Largo (RL) breeds fed diets with high or low roughage-to-concentrate ratio (R:C) under a tropical climate. Twenty lambs from each breed were individually housed in covered pens and fed the experimental diets for 58 days. The diets were formulated to meet the growth requirements of lambs with a roughage-to-concentrate ratio of 70:30 and 30:70. Significant interactions of breed × diet for nutrient intake were observed (P < 0.05), with SI lambs fed low R:C diet showing higher intake of dry matter, organic matter, crude protein, and total carbohydrates compared to RL lambs fed the same diet. SI lambs fed high R:C diet had higher intake of neutral detergent fiber than RL lambs (P < 0.05). SI lambs displayed better average daily gain and feed efficiency, regardless of diet (P < 0.05). Carcass traits and gastrointestinal components were influenced by breed and diet (P < 0.05). SI lambs fed low R:C diet showed higher subcutaneous fat thickness and better carcass finishing compared to RL lambs (P < 0.05). SI breed lambs exhibited better growth performance, carcass traits, and gastrointestinal characteristics, even when fed diets with a high roughage-to-concentrate ratio.

BACKGROUND: The liver is a principal metabolic organ and has a major role in regulating lipid metabolism. With the development of rapidly fattening livestock in the modern breeding industry, the incidence of hepatic steatosis and accumulation in animals was significantly increased. However, the molecular mechanisms responsible for hepatic lipid metabolic disturbances in a high concentrate diet remain unclear. The objective of this study was to evaluate the effects of increasing concentrate level in a fattening lamb diet on biochemical indices, hepatic triglycerides (TG) concentration, and hepatic transcriptomic profiles. In the present study, 42 weaned lambs (about 3 ± 0.3 months old) were randomly assigned to the GN60 group (60% concentrate of dry matter, GN60, n = 21) or GN70 group (70% concentrate of dry matter, n = 21) for a 3-months feeding trial. RESULTS: No difference was observed in the growth performance or plasma biochemical parameters between the GN60 group and the GN70 group. The hepatic TG concentration was higher in the GN70 group than GN60 group (P < 0.05). Hepatic transcriptomic analysis showed that there were 290 differentially expressed genes identified between GN60 and GN70 groups, with 125 genes up-regulated and 165 genes down-regulated in the GN70 group. The enriched Gene Ontology (GO) items and KEGG pathways and protein-protein interaction (PPI) network of differentially expressed genes (DEGs) revealed that the majority of enriched pathways were related to lipid metabolism. Further analysis revealed that the fatty acid synthesis was up-regulated, while fatty acid transport, oxidation, and TG degradation were down-regulated in the GN70 group when compared with the GN60 group. CONCLUSIONS: These results indicated that GN70 induced excess lipid deposition in the liver of lambs during the fattening period, with high synthesis rates and low degradation rates of TG. The identified mechanisms may help understand hepatic metabolism in lambs with a high concentrate diet and provide insight into decreasing the risk of liver metabolism disorder in animals.

Lamb-Shaffer Syndrome (LSS; OMIM #616803; ORPHA #313892; ORPHA #313884) is an infrequent genetic disorder that affects multiple aspects of human development especially those related to the development of the nervous system. LSS is caused by variants in the SOX5 gene. At the molecular level, SOX5 gene encodes for a transcription factor containing a High Mobility Group (HMG) DNA-Binding domain with relevant functions in brain development in different vertebrate species. Clinical features of Lamb-Shaffer syndrome may include intellectual disability, delayed speech and language development, attention deficits, hyperactivity, autism spectrum disorder, visual problems and seizures. Additionally, patients with the syndrome may present distinct facial dimorphism such as a wide mouth with full lips, small chin, broad nasal bridge, and deep-set eyes. Other physical features that have been reported in some patients include short stature, scoliosis, and joint hypermobility. Here, we report the clinical and molecular characterization of a Spanish LSS cohort of new 20 patients and review all the patients published so far which amount for 111 patients. The most frequent features included developmental delay, intellectual disability, visual problems, poor speech development and facial dysmorphic features. Strikingly, pain insensitivity and hypermetropia seems to be more frequent than previously reported, based on the frequency seen in the Spanish cohort. Eighty-three variants have been reported so far, single nucleotide variants (SNV) and copy number variants represent 47% and 53%, respectively, from the total of variants reported. Similarly to previous reports, the majority of the SNVs variants of the novel patients reported herein fall in the HMG domain of the protein. However, new variants, affecting other functional domains, were also detected. In conclusion, LLS is a rare genetic disorder mostly characterized by a wide range of developmental and neurological symptoms. Early diagnosis would allow to start of care programs, clinical follow up, prospective studies and appropriate genetic counseling, to promote clinical and social improvement to have profound lifelong benefits for patients and their families. Further research is needed to better understand the underlying mechanisms of the syndrome related to SOX5 haploinsufficiency.

To delineate further the clinical phenotype of Lamb-Shaffer Syndrome (LSS) 16 unpublished patients with heterozygous variation in SOX5 were identified either through the UK Decipher database or the study team was contacted by clinicians directly. Clinical phenotyping tables were completed for each patient by their responsible clinical geneticist. Photos and clinical features were compared to assess key phenotypes and genotype-phenotype correlation. We report 16 SOX5 variants all of which meet American College of Medical Genetics/Association for Clinical Genomic Science ACMG/ACGS criteria class IV or V. 7/16 have intragenic deletions of SOX5 and 9/16 have single nucleotide variants (including both truncating and missense variants). The cohort includes two sets of monozygotic twins and parental gonadal mosaicism is noted in one family. This cohort of 16 patients is compared with the 71 previously reported cases and corroborates previous phenotypic findings. As expected, the most common findings include global developmental delay with prominent speech delay, mild to moderate intellectual disability, behavioral abnormalities and sometimes subtle characteristic facial features. We expand in more detail on the behavioral phenotype and observe that there is a greater tendency toward lower growth parameters and microcephaly in patients with single nucleotide variants. This cohort provides further evidence of gonadal mosaicism in SOX5 variants; this should be considered when providing genetic counseling for couples with one affected child and an apparently de novo variant.
