BACKGROUND: High-grain diets that meet the energy requirements of high-producing ruminants are associated with a high risk of rumen disorders. Mild acid treatment with lactic acid (LA) has been used to modify the degradable characteristics of grains to improve the negative effects of high-grain diets. However, the related studies mainly focused on dairy cows and explored the effects on rumen fermentation, production performance, ruminal pH and so forth. And up to date, no studies have reported the hydrochloric acid (HA) treatment of grains for ruminant animals. Therefore, based on metabolomics analysis, the aim of this study was to evaluate the effects of treatment of corn by steeping in 1% LA or 1% HA for 48 h on the rumen and plasma metabolic profiles in beef steers fed a high corn (48.76%) diet with a 60:40 ratio of concentrate to roughage. The inflammatory responses of beef cattle fed LA- and HA-treated corn were also investigated. RESULTS: Based on ultra-high-performance liquid tandem chromatography-quadrupole time-of-flight mass spectrometry (UHPLC-QTOF/MS) metabolomics and multivariate analyses, this study showed that steeping corn in 1% LA or 1% HA modulated the metabolic profiles of the rumen. Feeding beef steers corn steeped in 1% LA or 1% HA was associated with lower relative abundance of carbohydrate metabolites, amino acid metabolites, xanthine, uracil and DL-lactate in the rumen; with higher ruminal pH; with lower concentrations of acetate, iso-butyrate and iso-valerate; and with a tendency for lower ruminal lipopolysaccharide (LPS) concentrations. Moreover, the data showed lower concentrations of plasma C-reactive protein, serum amyloid A, haptoglobin, interleukin (IL)-1ß and IL-8 in beef steers fed 1% LA- or HA-treated corn. The 1% LA treatment decreased the concentrations of plasma LPS, LPS-binding protein and tumour necrosis factor-alpha and the relative abundance of L-phenylalanine, DL-3-phenyllactic acid and tyramine in plasma. The 1% HA treatment decreased the relative abundance of urea in plasma and increased the relative abundance of all amino acids in the plasma. CONCLUSIONS: These findings indicated that LA or HA treatment of corn modulated the degradation characteristics of starch, which contributed to improving the rumen and plasma metabolic profiles and to decreasing inflammatory responses in beef steers fed a high-concentrate diet.

The objective of this study was to determine the effect of palm and/or linseed oil (LSO) supplementation on carcass quality, sensory evaluation and fatty acid profile of beef from crossbred Wagyu beef steers. Twenty four fattening Wagyu crossbred beef steers (50% Wagyu), averaging 640±18 kg live weight (LW) and approximately 30 mo old, were stratified and randomly assigned in completely randomized design into 3 treatment groups. All steers were fed approximately 7 kg/d of 14% crude protein concentrate with ad libitum rice straw and had free access to clean water and were individually housed in a free-stall unit. The treatments were i) control concentrate plus 200 g/d of palm oil; ii) control concentrate plus 100 g/d of palm oil and 100 g/d of LSO, iii) control concentrate plus 200 g/d of LSO. This present study demonstrated that supplementation of LSO rich in C18:3n-3 did not influence feed intakes, LW changes, carcass and muscle characteristics, sensory and physical properties. LSO increased C18:3n-3, C22:6n-3, and n-3 polyunsaturated fatty acids (PUFA), however, it decreased C18:1t-11, C18:2n-6, cis-9, trans-11, and trans-10,cis-12 conjugated linoleic acids, n-6 PUFA and n-6:n-3 ratio in Longissimus dorsi and Semimembranosus muscles.

The objective of this study was to determine the effects of chromium methionine (Cr-Met) chelate supplementation on blood metabolites and fatty acid profile of beef from Holstein steers during late fattening period. Fifteen Holstein steers were allotted randomly into two groups including the control (non Cr-Met feeding, NCM, ave. body weight [BW] = 483±25.7 kg) and the treatment (Cr-Met feeding for 4 months, 4CM, ave. BW = 486±27.5 kg) group. The feeding amount of Cr-Met to animals was limited to 400 ppb/cow/d and was supplemented to total mixed ration. No difference in blood albumin, alkaline phosphatase, urea-nitrogen, calcium, creatine, glucose, total protein, triglyceride, and cholesterol were observed between the treatment groups (p>0.05). The level of high density lipoprotein was higher in the 4CM group than the NCM group, whereas low density lipoprotein was lower in the 4CM group (p<0.05). The fatty acid composition (caprate, laurate, myristate, pentadecanoate, palmitate, palmitoleate, margarate, cis-11 heptadodecanoate, stearate, oleate, trans-vaccenate, linoleate, cis-11 eicosenoate, docosa hexaenoic acid, and docosa pentaenoic acid) of the beef showed no difference between the two groups (p>0.05). The arachidonic acid level tended to be higher in the 4CM than the NCM group (p = 0.07). Cr-Met had no influence (p>0.05) on the ratio of saturated, unsaturated, unsaturated/saturated, monounsaturated/saturated and polyunsaturated/saturated fatty acids whereas the ratio of polyunsaturated fatty acids (PUFA) in the 4CM group was comparatively higher than the NCM group (p<0.05). This study concluded that feeding Cr-Met supplementation in 400 ppb/d to Holstein steers for 4 months during late fattening period can improve some blood metabolites and beef quality by increasing PUFA and gamma-linoleate compositions of beef.

Flight speed is a predictive indicator of cattle temperament and is associated with feed efficiency phenotypes. Genetic markers associated with both traits may assist with selection of calmer animals with improved economic value. A preliminary genome-wide association study determined chromosomal regions on BTA9, and 17 were associated with flight speed. The genes quaking (QKI), glutamate receptor, ionotropic, AMPA 2 (GRIA2) and glycine receptor ß (GLRB) were identified in these regions as potential functional candidates. Beef steers (n = 1057) were genotyped with SNPs located within and flanking these genes. One SNP located near QKI and one near GRIA2 were nominally associated with flight speed (P ≤ 0.05) although neither was significant after Bonferroni correction. Several studies have shown a correlation between flight speed and feed intake or gain; therefore, we also analyzed SNPs on BTA6:38-39 Mb known to be associated with average daily gain (ADG) and average daily feed intake (ADFI) for association with flight speed. Several SNPs on BTA6 were associated with flight speed (P ≤ 0.005), and three were significant after Bonferroni correction. These results suggest that the genes tested are unlikely to contribute to flight speed variation for our cattle population, but SNPs on BTA6 associated with ADG and ADFI may influence temperament. Use of these markers to select for economically important feed efficiency phenotypes may produce cattle with more desirable temperaments.

The effects of a novel direct-fed microbial (DFM) on feedlot performance, carcass characteristics, digestibility, ruminal morphology, and volatile fatty acid (VFA) profile of finishing steers were evaluated. Single-source Angus-crossbred yearling steers (n = 144; initial body weight (BW) = 371 ±â€…19 kg) were used in a randomized complete block design. Steers were blocked by initial BW and randomly assigned to treatments (12 pens/treatment; 4 steers/pen). Treatments included (A) CONTROL (no DFM, tylosin, or monensin, (B) MONTY (monensin sodium [330 mg/animal-daily] and tylosin phosphate [90 mg/animal-daily]), and (C) MONPRO (monensin sodium [same as previous] and Lactobacillus salivarius L28 [1 × 106 CFU/animal-daily]). Treatments were included in a steam-flaked corn-based finisher diet offered once daily using a clean-bunk management for ~149 d. The digestibility assessment was performed from days 70 to 74. Ruminal fluid and rumen tissue samples were collected at the slaughter for VFA profile and papillae morphology analyses, respectively. Data were analyzed using the GLIMMIX procedure of SAS with pen serving as the experimental unit, treatment as fixed effect, and BW block as random effect. Steers offered MONPRO had on average 5.3% less (P < 0.01) dry matter intake (9.56 kg/d) compared with either CONTROL (10.16 kg/d) or MONTY (9.96 kg/d). The carcass-adjusted final BW (613 kg; P = 0.23), overall average daily gain (1.64 kg/d; P = 0.23), and gain-efficiency (0.165; P = 0.61) were not affected by treatments. Steers offered CONTROL had greater (P < 0.01) marbling score and tended (P = 0.06) to have less carcasses grading Select and tended (P = 0.10) to have more carcasses grading Upper-Choice, while other carcass characteristics and liver-abscesses were not affected (P ≥ 0.23) by treatments. The digestibility of nutrients (P ≥ 0.13) and the ruminal VFA profile (P ≥ 0.12) were not affected by treatments. Steers offered MONPRO tended (P = 0.09) to have 16% greater average papillae number compared to other treatments. Yearlings offered finishing diets containing L. salivarius L28 plus monensin did not affect growth performance, digestibility, or ruminal VFA, but reduced feed intake. Carcass quality was negatively affected by treatments, while animals consuming L. salivarius L28 and monensin tended to improve ruminal morphology. Current findings in ruminal morphology and feed intake may warrant further assessment of diets containing L. salivarius L28 on beef cattle food safety aspects.


Antimicrobial resistance is a growing concern to public health and medically important antibiotics have been listed in the Veterinary Feed Directive. Nutritional technologies, such as direct-fed microbials, are being increasingly studied for the development of an effective use on beef cattle production systems. The newly isolated strain of Lactobacillus salivarius L28 has demonstrated pathogenic inhibition of Escherichia coli, Salmonella, and Listeria monocytogenes on in vitro assessments. The potential benefits have warranted the exploration of L. salivarius L28 in a feedlot setting. Single-source Angus-crossbred yearling steers were offered steam-flaked corn-based finishing diets containing no feed additive, or either a combination of tylosin plus monensin or L. salivarius L28 plus monensin. Steers offered L. salivarius L28 plus monensin consumed 5.3% less feed compared with other treatments, while other growth performance variables and the digestibility of nutrients were not affected. Carcasses from cattle supplemented with monensin had slightly lower carcass quality grades than those not supplemented with monensin. Lactobacillus salivarius L28 plus monensin tended to improve steers ruminal morphology. Current findings may warrant further food safety assessments when cattle are offered diets containing L. salivarius L28.

This study was conducted to investigate the effect of heat stress (HS) on physiological, blood, and behavioral parameters, according to the temperature-humidity index (THI), in beef steers. Twelve Korean native beef steers (342.7 ± 13.81 days old, body weight (BW) of 333.0 ± 18.53 kg) were used in this experiment. Beef steers were randomly distributed into three homogenized groups (four beef steers each) for 14 days, namely, threshold (THI = 64-71), mild-moderate (THI = 72-79), and severe (THI = 80-87). Feed and water intake were recorded daily. Physiological parameters, including heart rate and rectal temperature, and behavioral patterns (standing and lying down) were measured weekly. Blood was sampled every week to analyze hormones, heat shock protein (HSP) levels, metabolites, and hematological parameters. All data were analyzed using repeated-measures analysis. Beef steers exposed to severe THI had significantly increased (p < 0.001) water intake, heart rate, and rectal temperature compared to the threshold and mild-moderate THI beef steers. Additionally, increased blood cortisol (p < 0.001), HSP70 (p < 0.001), blood urea nitrogen (BUN) (p = 0.014), and time spent standing (p < 0.001) were observed in beef steers after exposure to severe THI compared to beef steers in the threshold and mild-moderate THI groups. However, dry matter intake, blood glucose, and non-esterified fatty acids were not different among the THI groups. In conclusion, heart rate, rectal temperature, blood cortisol, HSP70, BUN, and time spent standing were closely associated with severe HS conditions in beef steers. These phenomena indicated that beef steers exposed to HS modulated their behavior and blood parameters, as well as their physiological response, to maintain homeostasis.

Ninety-two Angus-crossbred steers (424 ±â€…28.3 kg initial body weight) were used in a 98-d study to assess the effects of increasing Zn supplementation on cattle performance, liver and plasma trace mineral concentrations, blood metabolites, and carcass characteristics. All steers were implanted with a Component TE-200 (200 mg trenbolone acetate + 20 mg estradiol; Elanco Animal Health, Greenfield, IN) on d 0 and fed 300 mg‧steer-1‧d-1 of ractopamine hydrochloride (Zoetis, Parsippany, NJ) from d 70 to 98. Cattle were fed via GrowSafe bunks (GrowSafe Systems Ltd., Airdrie, AB, Canada), and steer served as the experimental unit (n = 22 or 23 steers/treatment). Supplemental Zn was administered through the diet at 0, 100, 150, or 180 mg Zn/kg on a dry matter basis from ZnSO4 (Zn0, Zn100, Zn150, or Zn180, respectively). Cattle were weighed on d -1, 0, 9/10, 20, 41, 59, 69, 70, 78/79, 97, and 98. Blood was collected on d 0, 9/10, 69, 78/79, and 97, and liver biopsies on d 9/10 and 78/79 (n = 12 steers/treatment). Data were analyzed as a complete randomized design. Contrast statements were formed to test the linear, quadratic, and cubic effects of Zn supplementation and test Zn0 vs. Zn supplementation. Day 10 and 70 body weight (BW) and d 0 to 10 and 0 to 70 average daily gain were linearly increased with Zn supplementation (P ≤ 0.05), and greater for Zn supplemented steers (P ≤ 0.03). No effects of Zn supplementation were observed on final BW, dressing percentage, ribeye area, 12th rib fat, or marbling (P ≥ 0.11). Hot carcass weight tended to be 7 kg greater for Zn supplemented steers than Zn0 (P = 0.07), and yield grade linearly increased with increasing Zn supplementation (P = 0.02). Day 10 liver Mn concentrations tended to quadratically decrease (P = 0.08) with increasing Zn supplementation, though d 79 liver Mn concentrations and arginase activity were not influenced by Zn (P ≥ 0.28). Day 10 liver arginase activity tended to be (r = 0.27; P = 0.07) and d 10 serum urea nitrogen was correlated with d 10 liver Mn (r = 0.55; P < 0.0001). Zinc supplementation linearly increased d 10 liver Zn and d 10, 69, 79, and 97 plasma Zn concentrations (P ≤ 0.05). A cubic effect of Zn was observed on d 79 liver Zn (P = 0.01) with lesser liver Zn in Zn0 and Zn150 steers. These data suggest increasing dietary Zn improves growth directly following the administration of a steroidal implant and that steroidal implants and beta agonists differ in their effects on protein metabolism.

ABSTRACT: This study was conducted to evaluate the association between a therapeutic dose of tulathromycin for bovine respiratory disease in beef steers and the antimicrobial and multidrug resistance profiles of the gastrointestinal tract commensals Escherichia coli and Enterococcus spp. and the foodborne pathogens Salmonella enterica and Campylobacter spp. isolated from fecal samples. Individual fecal samples were collected on days 0, 14, and 28 from 70 beef steers that were housed in a single pen and had been treated or not treated with tulathromycin. Samples were cultured for bacterial isolation, and isolates were tested for antimicrobial susceptibility with the broth microdilution method to determine the MICs of clinically relevant antimicrobials used in both human and veterinary medicine. Generalized linear mixed effects models were fitted to estimate the prevalence of the bacterial species and the prevalence of resistant isolates over time and between treated and nontreated cattle and of multidrug-resistant isolates. Model-adjusted mean prevalences of E. coli, Enterococcus spp., S. enterica, and Campylobacter spp. were 99.5, 85.9, 1.5, and 17.7%, respectively. The prevalence of erythromycin-resistant Enterococcus spp. was significantly higher on day 14 (59.7%) than on day 28 (22.2%). A higher prevalence of erythromycin-resistant Enterococcus spp. was found in samples from treated (59.3%) than in samples from nontreated (27.6%) animals. Multidrug resistance (three or more antimicrobial classes) was observed in 8.4% of E. coli isolates and 62.7% of Enterococcus isolates. The administration of tulathromycin was significantly associated with an increased prevalence of erythromycin-resistant Enterococcus spp. isolates.

To determine the effect of CaO-treated Pensacola bahiagrass (Paspalum notatum) hay on intake, ruminal fermentation parameters, and apparent total-tract digestibility of nutrients, nine ruminally cannulated Angus-crossbred steers were used in a triplicated 3 × 3 Latin square design. Steers had ad libitum access to either 1) untreated dry hay (DH; n = 8); 2) hay at 50% DM treated with 8.9% CaCO3 (dry matter [DM] basis; CC; n = 9); or 3) hay at 50% DM treated with 5% CaO (DM basis; CO; n = 8). Water was added to reach 50% DM in the CC and CO diets. Ruminal fluid and blood samples were collected every 3 h for 24 h. Ruminal fluid was analyzed for pH, volatile fatty acids (VFA), and ammonia-nitrogen (NH3-N). Blood was analyzed for plasma urea nitrogen (PUN). Hay and fecal samples were collected for 4 d, four times daily for hay and twice daily for feces, to determine apparent total-tract digestibility of nutrients. The hay provided to steers during the digestibility period was analyzed for in vitro organic matter digestibility (IVOMD) for 48 h. Data were analyzed as repeated measures for blood and ruminal fermentation parameters. Total DM intake was not affected (P ≥ 0.674) by treatment. A treatment effect (P < 0.001) was observed for average ruminal pH, where steers consuming CO had the greatest pH (P < 0.001). Ruminal concentration of NH3-N tended (P = 0.059) to be reduced in steers consuming CO. There was a treatment × time interaction (P = 0.023) on concentrations of PUN, where at 3 h DH and CO were lesser than CC (P ≤ 0.050) and at 21 h DH was lesser than CC (P = 0.020). Total VFA, acetate, propionate, butyrate, branched-chain VFA, and valerate concentrations were affected by treatment (P ≤ 0.035), where a reduction (P ≤ 0.034) occurred in steers consuming CO. No treatment differences were observed for total-tract digestibility of DM (P = 0.186), organic matter (P = 0.169), or crude protein (P = 0.152); however, steers consuming DH had greater neutral detergent fiber (P = 0.038) than CC and tended to be greater than CO (P = 0.082). The CO hay had greater (P = 0.005) IVOMD compared with DH and tended (P = 0.100) to be greater than CC. Bahiagrass hay treated with CaO may reduce ruminal fermentation, as indicated by decreased total VFA concentration without altering DM intake. The addition of CaO did not improve the digestibility of bahiagrass hay in vivo; however, in vitro results are contradictory and warrant further elucidation.


With the ever-growing desire to increase efficiency in beef cattle production, researchers have developed strategies such as treating poor-quality forages with chemicals to increase the digestibility of fiber fractions, consequently increasing their energy value for cattle feeding. Calcium oxide has been proposed as a replacement to more caustic chemicals used in the past (e.g., NaOH) and data indicate that it can promote similar and effective outcomes. The current study evaluated the effects of bahiagrass hay treated with calcium oxide on ruminal fermentation parameters, apparent total-tract digestibility of nutrients, and intake by beef steers consuming hay ad libitum as the sole ingredient in their diet. Additionally, in vitro organic matter digestibility was evaluated on the hay provided to steers to assess treatment effectiveness. Results indicated that steers consuming bahiagrass hay treated with calcium oxide had 1) increased pH and reduced volatile fatty acids concentrations in the rumen; 2) reduced or tendency for reduction on total-tract digestibility of fiber fractions; and 3) no effect on intake, all when compared with steers consuming untreated hay. In contrast, in vitro results indicated that organic matter digestibility was increased when the forage was treated with calcium oxide.

This experiment evaluated forage quality, total nutrient yield, water footprint, and growth performance of beef steers receiving protein supplements while grazing Teff grass ['Tiffany'Eragrostis tef (Zucc.) Trotter] over two consecutive growing seasons. Each year, four 2.66-ha irrigated paddocks (experimental units) were stocked with crossbred beef steers (n = 5 per paddock, initial BW = 289 ± 30 for yr 1; and n = 6, initial BW = 286 ± 23 for yr 2) in a randomized complete block design and stocked continuously for 63 d. Daily supplements [0.45 kg/d of cottonseed meal (Control) enough to avoid a negative ruminal N balance; and 0.50% mean paddock BW animal-daily (approximately 1.65 kg) of sorghum-dried distillers grains plus solubles, (DDGS)] were randomly assigned to two paddocks each. Supplement did not influence forage neutral detergent fiber (NDF), acid detergent fiber, crude protein, or in vitro true digestibility (P ≥ 0.54), except for a tendency (P = 0.08) for a numerical increase in NDF content of paddocks with steers that received DDGS supplementation. Paddock nutrient-yields were similar (P ≥ 0.43) between supplement treatments. Supplementation with DDGS produced greater (P = 0.01) cattle shrunk average daily gain (ADG). Predicted teff dry matter intake (DMI), net energy for maintenance (NEm), and growth (NEg) (P ≤ 0.03) were greater with cattle offered Control treatment. Predicted total DMI was similar (P = 0.14) although predicted dietary NEm, NEg, gain:feed, and total BW gain were greater (P ≤ 0.02) with DDGS. Predicted forage intake was greater (P ≤ 0.05) for cattle offered Control treatment. Teff nutrients remaining on d 56 were similar (P = 0.33) between treatments. Water footprint for total production of forage nutrient components did not differ (P ≥ 0.12) by treatments. Nutrient yield and water use efficiency of continuously stocked teff grass was not affected by supplemental regimen. Using DDGS as a supplement may increase BW gain through increased nutrient utilization without hindering teff nutrient production on a continuous stocking system.

This experiment evaluated the influence of protein supplementation frequency (SF) and amount offered on intake, nutrient digestibility, and ruminal fermentation by rumen-fistulated beef steers consuming low-quality [2.9% crude protein (CP); dry matter (DM) basis], cool-season forage. Seven Angus × Hereford steers (300 ± 27 kg) fitted with ruminal cannulas were randomly assigned to 1 of 7 treatments in an incomplete 7 × 4 Latin square. Treatments, in a 2 × 3 factorial design plus a non-supplemented control (CON), consisted of 2 levels of supplemental soybean meal, 100% (F) or 50% (H) of the estimated rumen-degradable protein requirement, provided daily (D), once every 5 d (5D), or once every 10 d (10D). Experimental periods were 30 d and dry matter intake (DMI) was measured from days 19 to 28. On days 21 (all supplements provided) and 30 (only daily supplements provided; day immediately prior to supplementation for 5D and 10D treatments) ruminal fluid was collected for ruminal pH, ammonia-N (NH3), volatile fatty acids (VFA), and determination of ruminal fermentation variables. Forage and total DM, organic matter (OM), and nitrogen (N) intake increased with supplementation (P ≤ 0.04). However, a linear effect of SF × amount of supplement interaction was observed for forage and total DM, OM, and N intake (P ≤ 0.04), with each variable decreasing as SF decreased, but the decrease being greater with F vs. H. Apparent total tract DM, OM, and neutral detergent fiber digestibility was not affected by supplementation or amount of supplement provided (P ≥ 0.10). In contrast, N digestibility increased with supplementation and for F vs. H (P < 0.01). Digestibility of DM, OM, and N increased linearly as SF decreased (P ≤ 0.03). When all supplements were provided, ruminal NH3, total VFA, and molar proportions of all individual VFA increased with supplementation (P ≤ 0.04), whereas acetate:propionate ratio decreased (P < 0.01). When only daily supplements were provided, none of the aforementioned fermentation parameters were affected (P ≥ 0.09). In summary, reducing the amount of supplemental CP provided to ruminants consuming low-quality forages, when supplementation intervals are >5 d, can be a management tool to maintain acceptable levels of DMI, nutrient digestibility, and ruminal fermentation while reducing supplementation cost.

We evaluated the plasma amine/phenol- and carbonyl-metabolome and whole-blood immune gene expression profiles in beef steers with divergent average daily gain (ADG). Forty-eight Angus crossbred beef steers (21 days postweaning; 210 ± 8.5 kg of body weight) were fed the same total mixed ration ad libitum for 42 days with free access to water. After 42 days of feeding, the steers were divided into two groups of lowest (LF: n = 8) and highest ADG (HF: n = 8). Blood samples were taken from all steers. The blood samples from LF and HF steers were used for further analysis. A subsample of the whole blood was immediately transferred into RNA-protect tubes for RNA extraction and messenger RNA expressions of 84 genes involved in innate and adaptive immune responses. Another subsample of the whole blood was immediately centrifuged to harvest the plasma for subsequent metabolome analysis. The average daily dry matter intake of the steers in LF and HF was 6.08 kg ± 0.57 and 6.04 kg ± 0.42, respectively, and was similar between the two groups (P = 0.72). The ADG (1.09 kg ± 0.13) of LF was lower (P = 0.01) than that of HF (1.63 kg ± 0.20). The expressions of 10 immune-related genes were upregulated (FC ≥ 1.2; P ≤ 0.05) in HF steers; these genes were involved in viral pathogen recognition and eradication, defense against intracellular and extracellular pathogens and parasites, and immune response homeostasis. A total number of 42 carbonyl-containing metabolites and 229 amine/phenol-containing metabolites were identified in the plasma samples of both groups. No alteration in carbonyl-metabolome was detected. Ten metabolites with immunomodulatory, anti-inflammatory, and reactive oxygen-scavenging properties were greater (FDR ≤ 0.05) in HF steers, whereas eight metabolites including arginine, phenylalanine, guanidoacetic acid, and aspartyl-threonine were greater in LF steers. This study demonstrated that beef steers with divergent ADG had altered plasma amine/phenol metabolome and immune-related gene expressions in the blood. Notably, plasma metabolites and immune-related genes of great health benefits were greater in steers with high ADG.

BACKGROUND: This study investigated changes in rumen protozoal and methanogenic communities, along with the correlations among microbial taxa and methane (CH4) production of six Belmont Red Composite beef steers fed tea seed saponins (TSS). Animals were fed in three consecutive feeding periods, a high-grain basal diet for 14 d (BD period) then a period of progressive addition of TSS to the basal diet up to 30 g/d for 20 d (TSS period), followed by the basal diet for 13 d without TSS (BDP post-control period). RESULTS: The study found that TSS supplementation decreased the amount of the protozoal genus Entodinium and increased Polyplastron and Eudiplodinium genera. During BDP period, the protozoa community of steers did not return to the protozoal profiles observed in BD period, with higher proportions of Metadinium and Eudiplodinium and lower Isotricha. The addition of TSS was found to change the structure of methanogen community at the sub-genus level by decreasing the abundance of methanogens in the SGMT clade and increasing the abundance of methanogens in the RO clade. The correlation analysis indicated that the abundance of SGMT clade methanogens were positively correlated with Isotricha, and Isotricha genus and SGMT clade methanogens were positively correlated with CH4 production. While RO clade were positively correlated with the proportion of Metadinium genus, which was negatively correlated with CH4 emission. CONCLUSIONS: These results suggest that different genera of rumen protozoa ciliates appear to be selectively inhibited by TSS, and the change in methanogen community at the subgenus level may be due to the mutualistic relationships between methanogens and rumen ciliates.

Our previous study suggested that supplementation of high-grain diets with ruminally protected and non-protected active dried yeast (ADY) may potentially reduce manure pathogen excretion by feedlot cattle. We hypothesized that feeding ruminally protected ADY might change the fecal bacterial community of finishing cattle. The objective of this study was to investigate the effects of feeding ruminally protected and non-protected ADY to finishing beef steers on their fecal bacterial community. Fresh fecal samples were collected on day 56 from 50 steers fed one of five treatments: 1) control (no monensin, tylosin, or ADY), 2) antibiotics (ANT, 330 mg monensin + 110 mg tylosin·steer-1d-1), 3) ADY (1.5 g·steer-1d-1), 4) encapsulated ADY (EDY; 3 g·steer-1d-1), and 5) a mixture of ADY and EDY (MDY; 1.5 g ADY + 3 g EDY·steer-1d-1). Bacterial DNA was extracted from fecal samples and sequenced using a MiSeq high-throughput sequencing platform. A total number of 2,128,772 high-quality V4 16S rRNA sequences from 50 fecal samples were analyzed, and 1,424 operational taxonomic units (OTU) were detected based on 97% nucleotide sequence identity among reads, with 769 OTU shared across the five treatments. Alpha diversity indices, including species observed, Chao estimate, abundance-based coverage estimator, Shannon, Simpson, and coverage, did not differ among treatments, and principal coordinate analysis revealed a high similarity among treatments without independent distribution. Bacteroidetes and Firmicutes were dominant phyla in the fecal bacterial community for all treatments, with a tendency (P < 0.10) for greater relative abundance of Bacteroidetes but lesser Firmicutes with ANT, EDY, and MDY compared with control steers. Prevotella was the dominant genus in all treatments and steers supplemented with ANT, EDY, and MDY had greater (P < 0.05) relative abundance of Prevotella than control steers, but lesser (P < 0.03) relative abundance of Oscillospira. No differences between ADY and control were observed for the aforementioned variables. Fecal starch contents were not different among treatments, but the relative abundance of Bacteroidetes, as well as Prevotella at genera level, tended (P < 0.06) to be positively correlated to fecal starch content. We conclude that supplementing ruminally protected or non-protected ADY or ANT had no effect on diversity and richness of fecal bacteria of finishing beef cattle, whereas feeding protected ADY or ANT to finishing beef steers altered the dominant fecal bacteria at phylum and genus levels. Therefore, supplementation of ruminally protected ADY may potentially improve intestinal health by stimulating the relative abundance of Prevotella.

Fescue toxicosis is a multifaceted syndrome common in cattle grazing endophyte-infected tall fescue that affects performance; however, little information is available pertaining to its effects on immunity. Recently, it has been shown that supplemental CP can improve performance in weaned steers postvaccination. Thus, the objective of this study was to evaluate the effect of supplemental CP on innate and adaptive immune responses in stocker steers chronically exposed to ergovaline. Angus steers (n = 12 pens; 3 steers/pen) were stratified by weight and assigned to a 2 × 2 factorial arrangement to examine crude protein levels of supplement (14% or 18%) and ergovaline exposure (0 or 185 µg ergovaline/kg BW/d via ground endophyte-free (EF) or endophyte-infected (EI) tall fescue seed, respectively) on immune response. Consumption of low to moderate concentration of ergovaline from EI tall fescue seed was sufficient to induce mild symptoms associated with fescue toxicosis. Blood samples were collected at day 0, 42, and 56 to evaluate infectious bovine rhinotracheitis (IBR) and bovine viral diarrhea virus (BVDV) type 1b titers following vaccine challenge. Additionally, serum cytokine concentrations were evaluated using Quantibody Bovine Cytokine Arrays on day 0, 28, and 42. Data were analyzed using PROC MIXED of SAS with repeated measures. Regardless of treatment, no differences were observed in IBR and BVDV-1b seroconversion following vaccine challenge (P > 0.05). Regardless of crude protein concentration, EI steers had greater concentrations of proinflammatory cytokines (TNF-α, IFN-γ, IL-1α), chemokines (CCL2, CCL4, MIG), anti-inflammatory cytokines (IL-2, -13, -15, -21), and various growth factors (FGF-1, IGF-1, VEGF-A) when compared to EF steers (P < 0.05). Furthermore, VEGF-A and IGF-1 concentrations were greater in EI-14 steers on day 28 compared to EI-18, EF-14, and EF-18 steers (P < 0.05), however, this difference was not observed on day 0 or 42 (P > 0.05). Based on these data, steers exposed to ergovaline have an increase in pro- and anti-inflammatory cytokines and supplemental CP had minimal impact to mitigate this response. However, in the current study, exposure to ergovaline had little to no effect on adaptive immunity and response to vaccination. Together, chronic exposure to ergovaline results in a hyperactive innate immune response, which may lead to an immuno-compromised animal.

Two experiments were conducted to examine eating preference of beef cattle for diets with or without the investigative enteric methane inhibitor 3-nitrooxypropanol (3-NOP). Nine beef steers were housed in individual stalls, each equipped with two feed bunks. The first experiment (Exp. 1) was conducted with a high forage diet and each animal received a diet without 3-NOP (CON) in one bunk and a diet with 3-NOP (dNOP) in the other bunk. The second study (Exp. 2) was conducted with the same animals about 6 months after Exp. 1 where a high grain diet without (CON) or with 3-NOP (dNOP) was offered. In Exp. 1, animals initially preferred CON compared with dNOP. Feed consumption from 0 to 3, 3 to 6, and 6 to 12 h after feeding was lower for dNOP compared with CON. However, dry matter intake (DMI) and feed consumption of dNOP gradually increased during Exp. 1 such that there was no preference between CON and dNOP on day 7. In Exp. 2, there was no preference for or against dNOP. Average DMI was greater for dNOP vs. CON, but interactions between diet and day for DMI and feed consumption rates indicated that daily preference between CON and dNOP was variable. In conclusion, beef steers initially detected a difference between CON and dNOP and selected in favor of CON rather than dNOP when they had not previously been exposed to 3-NOP. However, the animals rapidly acclimatized to a diet with 3-NOP (Exp. 1) and showed no eating preference between CON and dNOP within 7 days. This lack of preference was maintained throughout Exp. 2 when the same animals were fed a high grain diet.

The objectives of this study were to assess the effects of Saccharomyces cerevisiae fermentation products (SCFP; NaturSafe, SCFPns; and Original XPC, XPC; Diamond V) on growth performance, carcass traits, immune response, and antimicrobial resistance in beef steers fed high-grain diets. Ninety Angus steers (initial body weight [BW], 533 ± 9.8 kg) were assigned to a randomized complete design with 6 treatments (n = 15/treatment): 1) control, 2) low (12 g SCFPns·steer-1·d-1), 3) medium (15 g SCFPns·steer-1·d-1), 4) high SCFP (18 g SCFPns·steer-1·d-1), 5) encapsulated XPC (eXPC; 7 g XPC·steer-1·d-1 encapsulated with 9 g capsule material), and 6) antibiotics (ANT; 330 mg monensin + 110 mg tylosin·steer-1·d-1). Steers were fed ad libitum a diet containing 10% barley silage and 90% barley grain concentrate mix (dry matter basis) for 105 d. Increasing SCFPns tended (P < 0.09) to linearly increase feed efficiency. Average daily gain (ADG) tended (P < 0.10) to be greater in steers supplemented with eXPC than control. The SCFPns also tended (P < 0.10) to linearly increase marbling score. Proportion of severely abscessed livers tended (P < 0.10) to be lower in steers supplemented with medium and high SCFPns, eXPC, or ANT. A treatment × days on feed interaction were noticed (P < 0.01) for blood glucose, blood urea nitrogen (BUN), and acute phase proteins. The concentration of blood glucose responded quadratically (P < 0.05) on days 28 and 56, whereas BUN linearly (P < 0.01) increased on day 105 with increasing SCFPns dose. The SCFPns linearly increased haptoglobin (P < 0.03) and serum amyloid A (SAA;P < 0.05) concentrations on day 105, and lipopolysaccharide binding protein (LBP;P < 0.01) on days 56 and 105. The percentage of erythromycin-resistant and erythromycin + tetracycline-resistant enterococci was greater (P < 0.05) with ANT than control, SCFPns, and eXPC, whereas no difference was observed among control, SCFPns, and eXPC. No treatment effect was detected on the percentage of tetracycline-resistant enterococci. These results indicate that feeding SCFPns and eXPC was beneficial in improving ADG, feed efficiency and decreasing liver abscesses in a manner comparable to ANT. Unlike antibiotics, SCFPns or eXPC did not increase antimicrobial resistance. Both SCFPns and eXPC are potential alternatives to in-feed antibiotics.

Polyphenol oxidase (PPO) activity in red clover (Trifolium pratense) has been reported to reduce both proteolysis and lipolysis, resulting in greater N use efficiency and protection of PUFA across the rumen. Although high levels of PPO have been reported in grasses such as cocksfoot (orchard grass; Dactylis glomerata), no in vivo research has determined whether grass PPO elicits the same response as red clover PPO. To test the hypothesis that silage ensiled from grass with high levels of PPO protects N and PUFA across the rumen, 6 steers with ruminal and duodenal cannulas were offered cocksfoot silage (CO; high-PPO grass), perennial ryegrass silage (PR; Lolium perenne; low-PPO grass), or red clover silage (RC; high-PPO control) at 16 g DM/kg BW daily with the experiment consisting of two 3 × 3 Latin squares with 21-d periods, consisting of 12 d of diet adaptation, 6 d of duodenal marker infusion, 2 d of duodenal sampling, and 1 d of ruminal sampling. All silages were well preserved, with DM of 34.4, 55.3, and 45.4% for CO, PR, and RC. Activity of PPO in silages was low due to deactivation but was greater in CO than either PR or RC (0.15 vs. 0.05 and 0.08 µkatal/g DM). Protein-bound phenol (mg/g DM) as a measure of the degree of oxidation and an indication of PPO protection was greatest for RC (15.9) but comparable for PR (10.1) and CO (12.2). Biohydrogenation of C18 PUFA was significantly lower on RC compared to the 2 grass silages with CO greater than PR. Despite lower levels of total fatty acid intake and subsequent duodenal flow, CO resulted in greater levels of phytanic acid and total branched and odd chain fatty acids in duodenal digesta than RC or PR. Ruminal ammonia concentration was greatest for RC, with no difference between the grasses. Duodenal flow of microbial N and efficiency of microbial protein synthesis were lowest for CO and comparable for RC and PR. The CO (high-grass PPO) did not result in elevated levels of C18 PUFA escaping the rumen or improve efficiency of total N transfer through the rumen compared to PR. The RC resulted in a greater flow of N and nonmicrobial N to the duodenum than the 2 grasses with PR greater than CO.

British × Continental steers (n = 168; 7 pens/treatment; initial BW = 362 kg) were used to evaluate the effect of dose/payout pattern of trenbolone acetate (TBA) and estradiol-17ß (E2) and feeding of zilpaterol hydrochloride (ZH) on serum urea-N (SUN), NEFA, IGF-I, and E2 concentrations and LM mRNA expression of the estrogen (ER), androgen (ANR), IGF-I (IGF-IR), ß1-adrenergic (ß1-AR), and ß2-adrenergic (ß2-AR) receptors and IGF-I. A randomized complete block design was used with a 3 × 2 factorial arrangement of treatments. Main effects were implant (no implant [NI], Revalor-S [REV-S; 120 mg TBA + 24 mg E2], and Revalor-XS [REV-X; 200 mg TBA + 40 mg E2]) and ZH (0 or 8.3 mg/kg of DM for 20 d with a 3-d withdrawal). Steers were fed for 153 or 174 d. Blood was collected (2 steers/pen) at d -1, 2, 6, 13, 27, 55, 83, 111, and 131 relative to implanting; LM biopsies (1 steer/pen) were collected at d -1, 27, 55, and 111. Blood and LM samples were collected at d -1, 11, and 19 relative to ZH feeding. A greater dose of TBA + E2 in combination with ZH increased ADG and HCW in an additive manner, suggesting a different mechanism of action for ZH and steroidal implants. Implanting decreased (P < 0.05) SUN from d 2 through 131. Feeding ZH decreased (P < 0.05) SUN. Serum NEFA concentrations were not affected by implants (P = 0.44). There was a day × ZH interaction (P = 0.06) for NEFA; ZH steers had increased (P < 0.01) NEFA concentrations at d 11 of ZH feeding. Serum E2 was greater (P < 0.05) for implanted steers by d 27. Serum trenbolone-17ß was greater (P < 0.05) for implanted steers by d 2 followed by a typical biphasic release rate, with a secondary peak at d 111 for REV-X (P < 0.05) implanted steers. Implanting did not affect mRNA expression of the ANR or ER, but the IGF-IR and the ß1-AR and ß2-AR were less (P < 0.05) for REV-S than NI at d 55 and ß2-AR mRNA was less (P < 0.05) for REV-S than for REV-X. Expression of the IGF-IR and the ß1-AR at d 111 was greater (P< 0.05) for REV-X than for REV-S and NI at d 111, and the ß2-AR was less (P< 0.05) for REV-S than for REV-X. Feeding ZH did not affect mRNA expression of the ß1-AR and ß2-AR. Both implanting and feeding ZH decreased SUN, but a greater dose of TBA + E2 did not result in further decreases. In addition, feeding ZH increased serum NEFA concentrations. Metabolic changes resulting from implanting and feeding ZH may aid in explaining steer performance and carcass responses to these growth promotants.

Brahman × British crossbred steers (n = 40 and 38 in yr 1 and 2, respectively) were used to evaluate the effects of calf management systems following early weaning (EW) on growth performance, muscle gene expression, and carcass characteristics. On the day of EW (d 0), steers were stratified by BW and age (95 ± 14 kg; 74 ± 14 d) and randomly assigned to a control treatment that was normally weaned (NW) on d 180 (n = 10 steers/yr) or to 1 of 3 EW treatments: 1) EW and limit fed a high-concentrate diet at 3.5% of BW (as-fed basis) in drylot until d 180 (EW180; n = 10 steers/yr), 2) EW and limit fed a high-concentrate diet at 3.5% of BW (as-fed basis) in drylot until d 90 and then grazed on bahiagrass pastures until d 180 (EW90; n = 10 steers/yr), or 3) EW and grazed on annual ryegrass pastures until d 60 (yr 1; n = 10 steers) or 90 (yr 2; n = 8 steers) and then on bahiagrass pastures until d 180 (EWRG). Early-weaned steers on ryegrass and bahiagrass pastures were supplemented with high-concentrate diet at 1.0% of BW (as-fed basis) until d 180. From d 180 to 270 (yr 1), all EW steers remained in their respective treatments, whereas NW steers were provided high-concentrate diet at 1.0% of BW (as-fed basis) on bahiagrass pastures. In yr 1, feedlot finishing period began on d 270. In yr 2, the study was terminated on d 180. In both years, EW180 steers were heaviest (P < 0.0001) on d 180. On d 180 of yr 1, EWRG steers were lightest (P < 0.0001) and EW90 steers were heavier (P = 0.05) than NW steers, whereas EW90, EWRG, and NW steers had similar BW on d 180 of yr 2 (P ≥ 0.14). On d 90, muscle PPARγ mRNA expression tended (P = 0.07) to be greater for EW180 steers and was greater (P = 0.008) for EW90 vs. EWRG steers but similar (P = 0.25) between EW180 and NW steers. On d 180, PPARγ mRNA was greater (P ≤ 0.06) for EW180 vs. NW, EW90, and EWRG steers. From d 274 to 302, EW180 steers had the least ADG (P ≤ 0.09), whereas EW90 steers had similar (P = 0.19) ADG compared with EWRG steers but greater (P = 0.03) ADG than NW steers. At slaughter, carcass characteristics did not differ (P ≥ 0.22) among treatments. In summary, EW steers provided a high-concentrate diet in drylot for at least 90 d were heavier at the time of normal weaning than NW steers and EW steers grazed on ryegrass pastures for 60 to 90 d and supplemented with concentrate at 1.0% of BW. Feeding a high-concentrate diet immediately after EW enhanced the muscle PPARγ expression but did not enhance marbling at slaughter.
