RESUMO

In this contribution, we described a new fossil of a Pleistocene Tayassuidae from northern Brazil. The specimen is a left dentary with molars assigned to cf. Pecari tajacu recovered from an outcrop of the Rio Madeira Formation, State of Rondônia, Brazil. It represents the first Pleistocene fossil of this clade with stratigraphic provenance in the Amazon region of Brazil. This record contributes to the knowledge on the paleofauna of Rio Madeira Formation as well as extend the past geographic distribution of peccaries in South America.

AIM: To evaluate the clinical outcomes of intentionally replanted maxillary single-rooted teeth with vertical root fractures (VRFs) after being repaired extraorally using 4-methacryloxyethyl trimellitate anhydride/methacrylate-tri-n-butyl borane (4-META/MMA-TBB) resin cement. METHODOLOGY: Twenty-one root filled maxillary single-rooted teeth with VRFs were evaluated. After atraumatic extraction, fractured fragments were adhesively cemented. The teeth were then replanted and splinted to the neighbouring teeth for 2 weeks. Plaque index (PI), gingival index (GI), probing depth (PD) and clinical attachment level (CAL) were assessed at baseline, 6 and 12 months, and radiographic evaluations were made using PAI scores at baseline and 12 months. Mobility was evaluated using periotest values (PTV) at baseline, 1, 3, 6 and 12 months. Replanted teeth, contralateral teeth (control teeth) and adjacent teeth were analysed statistically using repeated measures one-way anova, unpaired t-tests and Wilcoxon matched-pairs signed-rank tests. RESULTS: Two teeth were extracted in the first month after surgery. PI, GI, CAL and PD scores of the replanted teeth were significantly lower at 6 month (P < 0.0001 for all) and 12 month (P < 0.0001 for all) postoperatively when compared to baseline, but the values were not significantly different from those of the control and adjacent teeth. PTV of the test teeth increased significantly (P < 0.0001) after the intervention and decreased to baseline levels by month 12. PTVs were significantly higher (P < 0.05) at baseline, 1, 3 and 6 months in the test teeth when compared with the control teeth, but were not significantly different at month 12. PAI scores of teeth with VRF were significantly lower (P < 0.05) at 12 months compared with baseline. CONCLUSIONS: Adhesive cementation and intentional replantation were an effective treatment modality for this group of vertically fractured maxillary single-rooted teeth. The clinical periodontal parameters decrease by month 6, and the mobility returned to the physiological limits of natural teeth 12 months after replantation.

RESUMO

Cynodonts represent the transition from reptiles to mammals. They are classified as synapsids, or tetrapod animals with mammalian characteristics. We present here the finding of helminth eggs in a coprolite identified as of cynodont origin dated of nearly 240 million years. Microscopy revealed the presence of very well preserved intestinal parasite eggs. Up to now we identified an ascarid egg by morphological characteristics. Based on a previous description of the new genus Ascarites Poinar Jr and Boucot 2006 in coprolites of iguanodons from Belgium, we propose a new species, Ascarites rufferi n.sp. in cynodonts, a host that inhabited the Southern Region of Brazil in the Triassic period.

The parareptiles diversified widely during the Permian and persisted in Pangean ecosystems until the end of the Triassic. However, most parareptiles succumbed to the Permian-Triassic extinction, leading to the exclusive survival of procolophonoids. Procolophonoidea stands out as one of the most diverse parareptile clades, with about 40 species across Pangean land masses. The Early Triassic species Procolophon trigoniceps is known from South Africa, Antarctica, and Brazil. The majority of cranial materials of this procolophonoid are described as anapsid in temporal morphology, however, some skulls discovered in South Africa were found to exhibit temporal fenestration. Once thought to have systematic significance for the genus Procolophon, temporal fenestration was lately proposed to be an anomalous or pathological feature in P. trigoniceps. In this study, we describe new cranial material of P. trigoniceps from the Sanga do Cabral Formation of Brazil that clearly displays temporal fenestration. Aside from the fenestra, the specimen closely resembles more complete Brazilian P. trigoniceps skulls. The recurrent presence of the feature and the varying morphologies exhibited by the temporal fenestrae of P. trigoniceps may substantiate its characterization as an anomalous trait within the species. Furthermore, the occurrence of temporal fenestration in P. trigoniceps specimens from both South America and South Africa underscores parallels between these two Early Triassic ecosystems.

The lineage of sauropodomorph dinosaurs raised some of the most impressive animals that ever walked on Earth. However, the massive titans of the Mesozoic Era originated from far smaller dinosaurs. The Triassic beds from Brazil yielded the earliest part of this evolutionary history. Despite the diverse fossil record of early sauropodomorphs, juvenile specimens, as well as certain species are poorly sampled. This is the case for Unaysaurus tolentinoi, an unaysaurid sauropodomorph from Caturrita Formation (ca. 225 Ma; early Norian, Late Triassic). The holotype and only specimen of U. tolentinoi was excavated from the Água Negra Locality (São Martinho da Serra, Rio Grande do Sul, Brazil) in 1998. More than two decades later, no other fossil vertebrates have been reported from the same fossiliferous site. Here we describe a skeletally immature specimen which was found in association with the holotype of U. tolentinoi. The specimen was discovered after a first-hand examination of the holotype and comprises some isolated vertebrae and elements from the posterior autopodium. According to linear regressions, its metatarsal I is approximately 41.7 mm in length, compared to approximately 75.9 mm in the holotype. The repeated elements and reduced size indicates that it does not belong to the elements originally used to erect U. tolentinoi. Rather, the specimen is assigned to U. tolentinoi by topotypy and shared morphology. In addition to the reduced size, distinct lines of evidence (e.g., neurocentral sutures; bone texture) support its assignment to a skeletally immature individual. In sum, the new material expands the record of U. tolentinoi, and represents an additional juvenile dinosaur from the Caturrita Formation.

SARS-CoV-2 infection triggers distinct patterns of disease development characterized by significant alterations in host regulatory responses. Severe cases exhibit profound lung inflammation and systemic repercussions. Remarkably, critically ill patients display a "lipid storm", influencing the inflammatory process and tissue damage. Sphingolipids (SLs) play pivotal roles in various cellular and tissue processes, including inflammation, metabolic disorders, and cancer. In this study, we employed high-resolution mass spectrometry to investigate SL metabolism in plasma samples obtained from control subjects (n = 55), COVID-19 patients (n = 204), and convalescent individuals (n = 77). These data were correlated with inflammatory parameters associated with the clinical severity of COVID-19. Additionally, we utilized RNAseq analysis to examine the gene expression of enzymes involved in the SL pathway. Our analysis revealed the presence of thirty-eight SL species from seven families in the plasma of study participants. The most profound alterations in the SL species profile were observed in patients with severe disease. Notably, a predominant sphingomyelin (SM d18:1) species emerged as a potential biomarker for COVID-19 severity, showing decreased levels in the plasma of convalescent individuals. Elevated SM levels were positively correlated with age, hospitalization duration, clinical score, and neutrophil count, as well as the production of IL-6 and IL-8. Intriguingly, we identified a putative protective effect against disease severity mediated by SM (d18:1/24:0), while ceramide (Cer) species (d18:1/24:1) and (d18:1/24:0)were associated with increased risk. Moreover, we observed the enhanced expression of key enzymes involved in the SL pathway in blood cells from severe COVID-19 patients, suggesting a primary flow towards Cer generation in tandem with SM synthesis. These findings underscore the potential of SM as a prognostic biomarker for COVID-19 and highlight promising pharmacological targets. By targeting sphingolipid pathways, novel therapeutic strategies may emerge to mitigate the severity of COVID-19 and improve patient outcomes.

This coronavirus pandemic has placed unprecedented restrictions on people's physical activity and routines. Prolonged home stays may lead to fear, panic, anxiety, and depression states, which in turn, can drive to a reduction of active lifestyles. Hence, determining the psychological response in the general population, and the influence level of home-based physical activity development could be relevant during this exceptional Covid-19 disease quarantine period. A multicenter, cross-sectional, and observational study design will be conducted in 12 Iberoamerican countries expecting to enroll 3,096 participants, through a snowball sampling technique. The study started on March 15th, 2020, and it is expected to be completed in August 2020 through online survey that will include demographic data, health status, psychological impact of the Covid-19 outbreak, mental health status, and level of physical activity. This study will be conducted following the principles established by the protocol, the Declaration of Helsinki, and the Ethical Guidelines for Clinical Research. Data from the study will be disseminated in manuscripts for submission to peer-reviewed journals as well as in abstracts for submission to relevant conferences. La pandemia del coronavirus ha dado lugar a unas restricciones sin precedentes en las rutinas y la actividad física de la población. Prolongadas estancias en casa podrían generar miedo, pánico, ansiedad y estados de depresión, los cuales pueden a su vez generar estilos de vida sedentarios. Por ello, ante esta situación excepcional de cuarentena generada por la enfermedad del Covid-19, puede ser relevante determinar cuál es el estado psicológico de la población, así como la influencia que el nivel de ejercicio físico desarrollado en casa puede tener sobre estos parámetros. Para ello, se está elaborando un estudio multicéntrico, transversal y observacional, desarrollado en 12 países iberoamericanos e involucrando a 3.096 personas, a través de una técnica de muestreo de bola de nieve. Los datos serán recogidos entre marzo y agosto de 2020 por medio de cuestionarios electrónicos que incluirán: datos demográficos, estado de salud, impacto psicológico del brote de Covid-19, estado mental, y nivel de actividad física. El estudio será desarrollado siguiendo los principios establecidos en la Declaración de Helsinki y siguiendo las consideraciones éticas de la investigación clínica. Los datos recogidos serán divididos en diferentes publicaciones científicas, las cuales serán sometidas a procesos de revisión por pares de revistas de alto impacto, así como a congresos internacionales de relevancia científica. Present knowledge of Late Triassic tetrapod evolution, including the rise of dinosaurs, relies heavily on the fossil-rich continental deposits of South America, their precise depositional histories and correlations. We report on an extended succession of the Ischigualasto Formation exposed in the Hoyada del Cerro Las Lajas (La Rioja, Argentina), where more than 100 tetrapod fossils were newly collected, augmented by historical finds such as the ornithosuchid Venaticosuchus rusconii and the putative ornithischian Pisanosaurus mertii. Detailed lithostratigraphy combined with high-precision U-Pb geochronology from three intercalated tuffs are used to construct a robust Bayesian age model for the formation, constraining its deposition between 230.2 ± 1.9 Ma and 221.4 ± 1.2 Ma, and its fossil-bearing interval to 229.20 + 0.11/- 0.15-226.85 + 1.45/- 2.01 Ma. The latter is divided into a lower Hyperodapedon and an upper Teyumbaita biozones, based on the ranges of the eponymous rhynchosaurs, allowing biostratigraphic correlations to elsewhere in the Ischigualasto-Villa Unión Basin, as well as to the Paraná Basin in Brazil. The temporally calibrated Ischigualasto biostratigraphy suggests the persistence of rhynchosaur-dominated faunas into the earliest Norian. Our ca. 229 Ma age assignment to Pi. mertii partially fills the ghost lineage between younger ornithischian records and the oldest known saurischians at ca. 233 Ma.

The upper Triassic deposits of the Selous Basin in south Tanzania have not been prospected for fossil tetrapods since the middle of last century, when Gordon M. Stockley collected two rhynchosaur bone fragments from the so called "Tunduru beds". Here we present the results of a field trip conducted in July 2015 to the vicinities of Tunduru and Msamara, Ruvuma Region, Tanzania, in search for similar remains. Even if unsuccessful in terms of fossil discoveries, the geological mapping conducted during the trip improved our knowledge of the deposition systems of the southern margin of the Selous Basin during the Triassic, allowing tentative correlations to its central part and to neighbouring basins. Moreover, we reviewed the fossil material previously collected by Gordon M. Stockley, confirming that the remains correspond to a valid species, Supradapedon stockleyi, which was incorporated into a comprehensive phylogeny of rhynchosaurs and found to represent an Hyperodapedontinae with a set of mostly plesiomorphic traits for the group. Data gathered form the revision and phylogenetic placement of Su. stockleyi helps understanding the acquisition of the typical dental traits of Late Triassic rhynchosaurs, corroborating the potential of hyperodapedontines as index fossils of the Carnian-earliest Norian.

In this contribution we report the first occurrence of the enigmatic African probainognathian genus Aleodon in the Middle-early Late Triassic of several localities from the state of Rio Grande do Sul in southern Brazil. Aleodon is unusual among early probainognathians in having transversely-expanded postcanine teeth, similar to those of gomphodont cynognathians. This genus was previously known from the Manda Beds of Tanzania and the upper Omingonde Formation of Namibia. The Brazilian record of this genus is based upon multiple specimens representing different ontogenetic stages, including three that were previously referred to the sectorial-toothed probainognathian Chiniquodon theotonicus. We propose a new species of Aleodon (A. cromptoni sp. nov.) based on the specimens from Brazil. Additionally, we tentatively refer one specimen from the upper Omingonde Formation of Namibia to this new taxon, strengthening biostratigraphic correlations between these strata. Inclusion of A. cromptoni in a phylogenetic analysis of eucynodonts recovers it as the sister-taxon of A. brachyrhamphus within the family Chiniquodontidae. The discovery of numerous specimens of Aleodon among the supposedly monospecific Chiniquodon samples of Brazil raises concerns about chiniquodontid alpha taxonomy, particularly given the extremely broad geographic distribution of Chiniquodon. The discovery of Brazilian Aleodon and new records of the traversodontid Luangwa supports the hypothesis that at least two subzones can be recognized in the Dinodontosaurus Assemblage Zone.

Liposome gels bearing an antineoplastic agent, 5-fluorouracil, intended for topical application have been prepared and drug release properties in vitro have been evaluated. Different formulations of liposomes were prepared by the film hydration method by varying the lipid phase composition (PL 90H/cholesterol mass ratio) and hydration conditions of dry lipid film (drug/aqueous phase mass ratio). Topical liposome gels were prepared by incorporation of lyophilized liposomes into a structured vehicle (1%, m/m, chitosan gel base). Also, hydrogels containing different concentrations of 5-fluorouracil were prepared and drug release properties were investigated. The rate of drug release from liposome gels was found to be dependent on the bilayer composition and the dry lipid film hydration conditions. Also, liposomes embedded into a structured vehicle of chitosan showed significantly slower release than hydrogels. The drug release obeyed the Higuchi diffusion model, while liposomes acted as reservoir systems for continuous delivery of the encapsulated drug.

O nível habitual de atividade física (NHAF) vem sendo utilizado para estabelecer uma relação entre estilo de vida ativo e saúde cardiovascular. No entanto, a avaliação da aptidão física permitiria a avaliação real das condições físicas do indivíduo, podendo assim apresentar melhores relações com a saúde cardiovascular. Objetivos: Relacionar o NHAF e aptidão física com a saúde cardiovascular de adultos e idosos e verificar a associação da prática regular de exercícios físicos supervisionada com os diferentes níveis de aptidão física. Método: Foram avaliados 213 adultos e idosos (> 50 anos) participantes de projetos comunitários. Os participantes realizaram as seguintes avaliações: NHAF por meio do questionário IPAQ, aptidão física por meio de uma bateria de testes motores que permitiu o cálculo do Índice de Aptidão Funcional Geral (IAFG), pressão arterial, perfil lipídico e índice de massa corporal. Resultados: O modelo linear generalizado evidenciou um maior número de diferenças com relação às variáveis relacionadas à saúde cardiovascular quando os participantes foram subdivididos em grupos tendo o IAFG como variável independente, comparado ao NHAF. O teste exato de Fisher evidenciou que os grupos de IAFG classificados como “bom” e “muito bom” apresentaram maior proporção de indivíduos com prática regular de exercícios físicos superior a 6 meses, com destaque para um maior número realizando a prática com supervisão (p<0,0001). Conclusão: O IAFG apresenta melhor associação com a saúde cardiovascular do que o NHAF e o tempo de prática e a supervisão estão associados ao nível de aptidão física...


The habitual level of physical activity (HLPA) has been used to establish the relation between active lifestyle and cardiovascular health. However, the assessment of physical fitness would review the actual physical condition of the individual, and thus can have better relations with cardiovascular health. Objectives: Relate HLPA and physical fitness with cardiovascular health of adults and elderly and to verify the association of supervised regular physical exercise with different levels of fitness. Method: 213 adults and elderly (> 50 years) participating in community projects were evaluated. The HLPA were assessed by the IPAQ and physical fitness through a battery of motor tests that allowed the calculation of the General Functional Fitness Index (GFFI). Blood pressure, lipid profile and body mass index were considered as indicators of cardiovascular health. Results: The generalized linear model showed a greater number of differences with respect to variables related to cardiovascular health when participants were subdivided having GFFI as an independent variable, compared to HLPA. The Fisher exact test showed that the groups GFFI classified as “good” and “very good” showed a higher proportion of individuals with regular practice for more than 6 months exercise, especially for a larger number performing the practice under supervision (p <0.0001). Conclusion: The GFFI has better association with cardiovascular health than HLPA and time of practice and supervision are associated with the level of physical fitness...

AIM: To determine the pharmacokinetics and bioavailability of florfenicol in the plasma of healthy Japanese quail (Coturnix japonica). METHODS: Sixty-five quail were given an I/V and I/M dose of florfenicol at 30 mg/kg bodyweight (BW). A two-period sequential design was used, with a wash-out period of 2 weeks between the different routes of administration. Concentrations of florfenicol in plasma were determined using high-performance liquid chromatography (HPLC). RESULTS: A naïve pooled data analysis approach for the plasma concentration-time profile of florfenicol was found to fit a non-compartmental open model. After I/V administration, the mean residence time (MRT), mean volume of distribution at steady state (Vss), and total body clearance of florfenicol were 12.0 (SD 0.37) h, 8.7 (SD 0.22) L/kg, and 1.3 (SD 0.08) L/h/ kg, respectively. After I/M injection, the MRT, mean absorption time (MAT), and bioavailability were 12.3 (SD 0.37) h, 0.2 (SD 0.02) h, and 79.1 (SD 1.79)%, respectively. CONCLUSIONS: The time for the concentration of florfenicol to fall below the probable effective concentration of 1 microg/ ml of approximately 10 h is sufficient for the minimum inhibitory concentration needed for many bacterial isolates. Further pharmacodynamic studies in quail are needed to evaluate a suitable dosage regimen.

A adoção de um estilo de vida ativo através da prática da caminhada é altamente recomendada pelos profissionais de saúde com o intuito de evitar, minimizar ou reverter diversos agravos de saúde que podem comprometer a qualidade de vida da população. Objetivo: Verificar se a prática da caminhada não supervisionada possui estímulo suficiente para a redução de fatores de risco de doenças cardiovasculares em adultos e idosos. Métodos: Participaram deste estudo 225 homens e mulheres saudáveis que foram divididos em 4 grupos: sedentário (G0), caminhada (G1), caminhada e uma atividade extra (G2) e, caminhada e duas atividades extras (G3). Todos os participantes realizaram as seguintes avaliações: International Physical Activity Questionnaire (IPAQ) - versão curta, bateria de testes de aptidão física (AAHPERD), pressão arterial, perfil lipídico e composição corporal. Resultados: Os resultados apontaram que a prática da caminhada não supervisionada (G1) promoveu benefícios limitados à saúde quando comparado ao grupo G3 (índice de aptidão funcional geral = 273,4±111 vs340,6±92; pressão arterial diastólica = 80,0±8 vs 75,4±7 mmHg; HDL colesterol = 44,3±10 vs 50,1±10mg/dL respectivamente). Remain physically active through the practice of walking is highly recommended by health professionals in order to avoid, minimize or reverse various health disorders that can compromise a good quality of life for the population. Objective: To verify if the non supervised walk has enough stimulation to reduce risk factors of cardiovascular disease in adults and the elderly. Methods: This study included 225 healthy men and women who were divided in 4 groups: sedentary (G0), walk (G1), walk an extra activity (G2), and walk and 2 extra activities (G3). All participants performed the following assessments: questionnaire IPAQ, AAHPERD battery tests, blood pressure, lipid profile and body composition. Results: The results suggest that practice of non supervised walk (G1) promotes limited benefits to the health when compared to G3 (general functional fitness index = 273.4±111 vs 340.6±92; diastolic blood pressure = 80.0±8vs 75.4±7 mmHg; HDL cholesterol = 44.3±10 vs 50.1±10 mg/dL respectively).
