Polar bears (Ursus maritimus) in the wild are under threat due to climate change, primarily loss of sea ice, and experience poor reproductive success in zoos. The polar bear is a seasonally polyestrous species that exhibits embryonic diapause and pseudopregnancy, complicating characterization of reproductive function. Fecal excretion of testosterone and progesterone have been studied in polar bears, but accurately predicting reproductive success remains difficult. Dehydroepiandrosterone (DHEA) is a steroid hormone precursor correlated with reproductive success in other species, but has not been well studied in the polar bear. The purpose of the present study was to characterize the longitudinal excretion of DHEAS, the sulfated form of DHEA, from zoo-housed polar bears using a validated enzyme immunoassay. Lyophilized fecal samples from parturient females (n = 10), breeding non-parturient females (n = 11), a non-breeding adult female, a juvenile female, and a breeding adult male were investigated. Five of the breeding non-parturient females had been previously contracepted, while six were never contracepted. DHEAS concentrations were closely associated with testosterone concentrations (p < 0.05, rho > 0.57) for all reproductive statuses. Breeding females exhibited statistically significant (p < 0.05) increases in DHEAS concentration on or near breeding dates, which were not observed outside of the breeding season, or in the non-breeding or juvenile animals. Breeding non-parturient females exhibited higher median and baseline DHEAS concentrations than parturient females over the course of the breeding season. Previously contracepted (PC) breeding non-parturient females also exhibited higher season-long median and baseline DHEAS concentrations than non-previously (NPC) contracepted breeding non-parturient females. These findings suggest that DHEA is related to estrus or ovulation in the polar bear, that there is an optimal DHEA concentration window, and concentrations exceeding that threshold may be associated with reproductive dysfunction.

The goal of the present study was to carry out a thorough methodological validation and describe baseline profiles for glucocorticoid hormones (cortisol and corticosterone) in blubber from blue (n = 77) and gray (n = 103) whales from the eastern North Pacific Ocean. For each species, we modelled cortisol and corticosterone concentrations in response to life history parameters (age, sex, reproductive status) and season or geographic location. In blue whales, cortisol concentrations did not vary significantly by age class, sex, or reproductive status, whereas corticosterone was significantly lower in immature than in adult females (p < .001). In gray whales, cortisol concentrations were significantly higher in lactating whales (p < .05), while corticosterone was significantly different between females and males (p = .001) and elevated in calves (p = .003). In gray whales, corticosterone concentrations were significantly lower in males sampled later in the year (August to November) compared to both sexes sampled between March and August (p = .05), but no seasonal trend occurred in blue whales. Our results indicate that glucocorticoid actions vary between species and sex in large whales. Analysis of multiple hormones improves our understanding of the physiology of maintaining metabolic homeostasis or coping with chronic stressors.

The goal of the present study was to complement existing data of testosterone and progesterone in blue whale (Balaenoptera musculus) blubber from the eastern North Pacific Ocean to evaluate effects of seasonality and location on these hormones and to better assess reproductive status of individuals. Physiological parameters regarding reproduction are fundamental for describing population dynamics, and hormones can be a valid tool to estimate those for wildlife populations. In this study, blubber tissue was validated for testosterone and progesterone assays. Hormone concentrations were measured in 69 (35 males and 34 females) blubber samples from live (n = 66) and stranded (n = 3) animals collected between 2002 and 2016 from a known winter reproductive ground in the Gulf of California (GoC) and summer feeding areas along the United States West Coast (USWC), specifically off the states of California and Oregon. Results were combined with sighting histories as a tool to determine reproductive status of individual whales. Testosterone concentrations in adult male blue whales were significantly higher (p < 0.05) in blubber biopsies sampled off the USWC between the months of June and November compared to those sampled in the GoC between February and April. Elevated testosterone concentrations likely indicate physiological preparation for reproductive activity while the animals were present off the USWC. Progesterone concentrations were significantly elevated in pregnant females, confirming progesterone as an indicator of pregnancy in blue whales. Probabilities of being pregnant were estimated for adult females with unknown sighting histories based on progesterone concentrations. Testosterone in females was detected and measured only in pregnant whales suggesting its biosynthesis or metabolism is altered during gestation. These results provide updated and new information on the reproductive cycle of blue whales in the eastern North Pacific, posing new milestones to better estimate the timing of the mating season for this endangered population.

Developing a better understanding of the stress response is critical to ensuring the health and sustainability of marine mammal populations. However, accurately measuring and interpreting a stress response in free-ranging, large cetaceans is a nascent field. Here, an enzyme immunoassay for corticosterone was validated for use in biopsy samples from male humpback whales (Megaptera novaeangliae). Analyses were conducted on 247 male North Pacific humpback whale blubber samples, including 238 non-calves and 9 calves that were collected on the Hawaiian breeding and Southeast Alaskan feeding grounds from 2004 to 2006. Significant relationships were found when corticosterone concentrations were examined by year, age class and distribution between locations. When examined by year, corticosterone concentrations for male humpback whales were higher in Hawaii in 2004 than in 2005 and 2006 (p < 0.05). Corticosterone concentration also varied by age class with initially high concentrations at birth which subsequently tapered off and remained relatively low until sexual maturity was reached around age 8-10 years. Corticosterone concentrations appeared to peak in male humpback whales around 15-25 years of age. Blubber biopsies from Alaska and Hawaii had similar mean corticosterone concentrations, yet the variability in these samples was much greater for whales located in Hawaii. It is clear that much work remains to be done in order to accurately define or monitor a stress response in male humpback whales and that specific attention is required when looking at age, sex, and yearly trends. Our results suggest that a stress response may be most impacted by age and yearly oceanographic conditions and needs to be initially examined at the individual level.

Understanding reproductive profiles and timing of reproductive events is essential in the management and conservation of humpback whales (Megaptera novaeangliae). Yet compared to other parameters and life history traits, such as abundance, migratory trends, reproductive rates, behavior and communication, relatively little is known about variations in reproductive physiology, especially in males. Here, an enzyme immunoassay (EIA) for testosterone was validated for use in biopsy samples from male humpback whales. Analyses were conducted on 277 North Pacific male humpback whale blubber samples, including 268 non-calves and 9 calves that were collected in the Hawaiian breeding grounds and the Southeast Alaskan feeding grounds from 2004 to 2006. Testosterone concentrations (ng/g) were significantly different between non-calves sampled in Hawaii (n = 182) and Alaska (n = 86, p < 0.05) with peak testosterone concentrations occurring in the winter (January-March) and the lowest concentrations occurring in the summer (June-September). Fall and spring showed increasing and decreasing trends in testosterone concentrations, respectively. Blubber testosterone concentrations in non-calves and calves sampled in Alaska were not significantly different. Blubber and skin from the same individual biopsies (n = 37) were also compared, with blubber having significantly higher testosterone concentrations (p < 0.05) than skin samples. We found variability in testosterone concentration with age, suggesting that male humpbacks reach peak lifetime testosterone concentrations in the breeding grounds around age 8-25 years. The testosterone profile of male humpback whales follows a predictable pattern for capital breeders, where testosterone begins to increase prior to the breeding season, stimulating the onset of spermatogenesis. Incorporation of reproductive hormonal profiles into our overall understanding of humpback whale physiology will shed additional light on the timing of reproduction and overall health of the recently delisted Hawaii distinct population segment (DPS).

Steroid hormone assessment using non-invasive sample collection techniques can reveal the reproductive status of aquatic mammals and the physiological mechanisms by which they respond to changes in their environment. A portion of the eastern North Pacific blue whale (Balaenoptera musculus) population that seasonally visits the Gulf of California, Mexico has been monitored using photo-identified individuals for over 30 years. The whales use the area in winter-early spring for nursing their calves and feeding and it therefore is well suited for fecal sample collection. Using radioimmunoassays in 25 fecal samples collected between 2009 and 2012 to determine reproductive state and stress, we validated three steroid hormones (progesterone, corticosterone and cortisol) in adult female blue whales. Females that were categorized as pregnant had higher mean fecal progesterone metabolite concentrations (1292.6 ±â€¯415.6 ng·g-1) than resting and lactating females (14.0 ±â€¯3.7 ng·g-1; 23.0 ±â€¯5.4 ng·g-1, respectively). Females classified as pregnant also had higher concentrations of corticosterone metabolites (37.5 ±â€¯9.9 ng·g-1) than resting and lactating females (17.4 ±â€¯2.0 ng·g-1; 16.8 ±â€¯2.8 ng·g-1, respectively). In contrast, cortisol metabolite concentrations showed high variability between groups and no significant relationship to reproductive state. We successfully determined preliminary baseline parameters of key steroid hormones by reproductive state in adult female blue whales. The presence of pregnant or with luteal activity and known lactating females confirms that the Gulf of California is an important winter-spring area for the reproductive phase of these blue whales. The baseline corticosterone levels we are developing will be useful for assessing the impact of the increasing coastal development and whale-watching activities on the whales in the Gulf of California.

As part of a long-term research program, Cook Inlet beluga (Delphinapterus leucas) presence was acoustically monitored with two types of acoustic sensors utilized in tandem in moorings deployed year-round: an ecological acoustic recorder (EAR) and a cetacean and porpoise detector (C-POD). The EAR was used primarily to record the calls, whistles, and buzzes produced by belugas and killer whales (Orcinus orca). The C-POD was used to log and classify echolocation clicks from belugas, killer whales, and porpoises. This paper describes mooring packages that maximized the chances of successful long-term data collection in the particularly challenging Cook Inlet environment, and presents an analytical comparison of odontocete detections obtained by the collocated EAR and C-POD instruments from two mooring locations in the upper inlet. Results from this study illustrate a significant improvement in detecting beluga and killer whale presence when the different acoustic signals detected by EARs and C-PODs are considered together. Further, results from concurrent porpoise detections indicating prey competition and feeding interference with beluga, and porpoise displacement due to ice formation are described.

Monthly blood samples, daily mating observations from Galapagos tortoises (Chelonoidis nigra), and local rainfall and temperature were collected at the Honolulu Zoo as part of a fertility evaluation. Testosterone concentrations were measured for males (n = 6), two of which were seen copulating and were considered sexually active. Estrone sulfate and progesterone concentrations were measured for female tortoises (n = 9), two of which nested and only one had laid eggs. Testosterone profiles were similar for both sexually active and sexually inactive males, both of which were positively correlated with temperature but not rainfall. Peak testosterone concentrations (12.0 ± 1.4 ng/ml sexually active animals vs. 14.4 ± 2.4 ng/ml sexually inactive animals) occurred at the end of the nesting season, from April to July. Estrone sulfate concentrations were similar for nesting (n = 2) and non-nesting (n = 7) female tortoises, rising from non-detectable concentrations (September), and increasing to peak concentrations during the nesting season. Progesterone concentrations remained low and spiked (9.44 ng/ml) only for the female that nested and laid eggs. Testosterone was negatively correlated with mating behavior, and the male tortoises were likely capable of spermatogenesis even though only two of them engaged in mating behavior. The female tortoises were not senescent, as the estrone sulfate concentrations likely reflected waves of ovarian follicular activity. Endocrine parameters were not in synchrony with rainfall, and a disconnect between the timing of reproductive events and the environmental milieu may help to explain the poor fertility of these tortoises. There is increasing interest in measuring endocrine and immune parameters in free-ranging seals and sea lions, but there is a lack of understanding in how an acute stress response, often associated with capture and handling, influences these parameters of interest. The main objective of this study was to assess the impact of a simulated stressor on both endocrine and immune parameters. During two seasons, exogenous adrenocorticotrophic hormone (ACTH) was administered to seven female juvenile harbor seals and the response of several hormones (cortisol, aldosterone, total and free thyroxine and total triiodothyronine) and immunological parameters (total and differential leukocyte counts and peripheral blood mononuclear cells (PBMC) proliferation) were assessed. Cortisol peaked at 165 min (winter 203.1±84.7 ng/ml; summer 205.3±65.7 ng/ml) and remained significantly elevated 240 min after ACTH infusion in both seasons. Aldosterone peaked at 90 min (winter 359.3±249.3 pg/ml; summer 294.1±83.7 pg/ml) and remained elevated 240 min after administration of ACTH in both seasons. An increase in circulating total white blood cells was driven primarily by the increase in neutrophils which occurred simultaneously with a decrease in lymphocytes leading to an overall increase in neutrophil to lymphocyte ratio. These findings demonstrate that a simulated stress response in juvenile harbor seals results in a predictable increase in both cortisol and aldosterone concentrations, and were associated with altered immunological parameters.

From the 1970s to the 1990s, the breeding population of spectacled eiders (Somateria fischeri) in western Alaska declined by 96%, which led to the listing of this species as threatened under the Endangered Species Act in 1993. Since then, the population has stabilized, but has not recovered to pre-decline numbers. While little is known about reproductive endocrinology in spectacled eiders, in other avian species, estrogen and testosterone are known to initiate and modulate various reproductive processes including yolk protein synthesis, reproductive behaviors and secondary sex characteristics. Measurement of the metabolites of estrogen and testosterone (EM and TM, respectively) in excrement reflect circulating hormone concentrations and provide a non-invasive method to monitor reproductive physiology. We measured concentrations of excreted EM in captive females and TM in males to (1) determine the efficacy of commercially available radioimmunoassay kits to detect EM and TM, (2) describe annual profiles of EM and TM concentrations, and (3) define the reproductive season of captive spectacled eiders using endocrine status. Excrement samples were collected from captive female and male spectacled eiders three times per week throughout 1 year. Female EM and male TM levels were quantified using radioimmunoassay. Mean female EM profile exhibited values exceeding the threshold for "peak" values (EM>193.3 ng/g) from mid-February to early July, and again in September. Additionally, the highest average concentrations of EM were seen in March, May and September. Elevated TM concentrations occurred in mid March, mid May and late June. These data suggest that levels of excreted sex steroids reflect patterns predicted by breeding landmarks in the annual cycle and will assist in field monitoring and captive breeding programs for spectacled eiders.

Body condition indices have been useful in assessing the health of domestic and free ranging populations of terrestrial mammals. Given the high energy demand and rapid growth during the postnatal period of Steller sea lion (Eumetopias jubatus) (SSL) pups, body condition was expected to be related to concentrations of a suite of hormones (cortisol, aldosterone, thyroxine, triiodothyronine, leptin) previously associated with changes in body mass and composition in developing pinnipeds. Blood samples were collected from free ranging SSL pups of known ages and sex. A body condition index (BCI) previously developed for SSL pups based on a mass and length relationship was applied to 61 SSL pups ranging in age from 5 to 38days old. BCI was not related to pup age. Overall, male pups were larger than females and older pups were larger than younger pups. Aldosterone was negatively correlated with BCI only in female pups, whereas no relationship was observed between aldosterone and BCI in males. Further, male pups had higher aldosterone concentrations than females. Concentrations of cortisol, total and free thyroxine (T4), and total triiodothyronine (T3) decreased when regressed against the elapsed time between researchers' arrival on the rookery and removal of pup from the holding corral for blood collection. While the overall variation attributed to the rookery disturbance was low (r(2)<0.293), it may be of significance for future studies on free ranging pinnipeds. This study adds to the current knowledge of the postnatal changes in free ranging SSL pups by providing ranges of the BCI and several hormone concentrations from an apparently stable population.

Identifying markers of individual quality is a central goal of life-history theory and conservation biology. The 'corticosterone (CORT)-fitness hypothesis' postulates that low fitness signals impaired ability to cope with the environment, resulting in elevated baseline CORT levels. CORT can, however, be negatively, positively or neutrally related to fitness, depending on the context. In order to clarify this controversial issue, we elucidate the utility of using baseline CORT as a correlate of individual fitness in incubating female eiders across variable environments. An increase in serum CORT with decreasing body condition was evident in older, more experienced breeders, while increased clutch mass was associated with elevated serum CORT in females breeding late in the season. For faecal CORT, the expected negative association with body condition was observed only in early breeders. We found a strong increase in faecal CORT with increasing baseline body temperature, indicating the utility of body temperature as a complementary stress indicator. Females in good body condition had a lower baseline body temperature, but this effect was only observed on open islands, a harsher breeding habitat less buffered against weather variability. Females with higher reproductive investment also maintained a lower baseline body temperature. Nest success strongly decreased with increasing serum and faecal CORT concentrations, and individual stress hormone and body temperature profiles were repeatable over years. Although our data support the tenet that baseline CORT is negatively related to fitness, the complex context-dependent effects call for cautious interpretation of relationships between stress physiology and phenotypic quality.

The endangered beluga whale (Delphinapterus leucas) population in Cook Inlet, AK faces threats from a variety of anthropogenic factors, including coastal development, oil and gas exploration, vessel traffic, and military activities. To address existing gaps in understanding about the occurrence of belugas in Cook Inlet, a project was developed to use passive acoustic monitoring to document the year-round distribution of belugas, as well as killer whales (Orcinus orca), which prey on belugas. Beginning in June 2009, ten moorings were deployed throughout the Inlet and refurbished every two to eight months. Despite challenging conditions consisting of strong tidal currents carrying debris and seasonal ice cover, 83% of mooring deployments were successfully recovered. Noise from water flow, vessel traffic, and/or industrial activities was present at several sites, potentially masking some signals. However, belugas were successfully detected at multiple locations. Detections were relatively common in the upper inlet and less common or absent at middle and lower inlet locations. Killer whale signals were also recorded. Some seasonal variability in the occurrence of both belugas and killer whales was evident.

Rehabilitating pinniped pups are often admitted to care centers as neonates and generally lack maternal investment and are in poor body condition. Upon admittance to a rehabilitation facility, pups are typically fed a milk replacement formula via gavage, which is switched to frozen fish upon weaning. While rehabilitation has been successful in terms of recovery and release, preweaning growth rates in captivity are consistently lower than in the wild. Indicators of stress (cortisol and total thyroxine; TT4), and standard morphometrics, of harbor seal pups in rehabilitation (n = 20) were determined for both preweaned and weaned pups. Hormone concentrations and standard morphometrics from pups in care were compared with free-ranging harbor seal pups (n = 59). Pups in rehabilitation gained mass on both milk and fish diets. Preweaned pups had greater mean serum cortisol and similar TT4 concentrations than weaned pups. Free-ranging harbor seal pups were heavier and longer than preweaned and weaned pups in rehabilitation. The free-ranging pups had the lowest cortisol and highest TT4 concentrations of any of the pups. These results suggest that weaned pups that have undergone rehabilitation are not physiologically equivalent to free-ranging weaned pups. Additional research is needed regarding physiological changes in endocrine values during early development under captive care conditions. This information should be useful to marine mammal rehabilitation centers in their development of care protocols and release criteria for rehabilitating harbor seal pups.

Surviving prolonged fasting implies closely regulated alterations in fuel provisioning to meet metabolic requirements, while preserving homeostasis. Little is known, however, of the endocrine regulations governing such metabolic adaptations in naturally fasting free-ranging animals. The hormonal responses to natural prolonged fasting and how they correlate to the metabolic adaptations observed, were investigated in subantarctic fur seal (Arctocephalus tropicalis) pups, which, because of the intermittent pattern of maternal attendance, repeatedly endure exceptionally long fasting episodes throughout their development (1-3 mo). Phase I fasting was characterized by a dramatic decrease in plasma insulin, glucagon, leptin, and total l-thyroxine (T(4)) associated with reductions in mass-specific resting metabolic rate (RMR), plasma triglycerides, glycerol, and urea-to-creatine ratio, while nonesterified fatty acids (NEFA) and ß-OHB increased. In contrast, the metabolic steady-state of phase II fasting reached within 6 days was associated with minimal concentrations of insulin, glucagon, and leptin; unchanged cortisol and triiodothyronine (T(3)); and moderately increased T(4). The early fall in insulin and leptin may mediate the shift to the strategy of energy conservation, protein sparing, and primary reliance on body lipids observed in response to the cessation of feeding. In contrast to the typical mammalian starvation response, nonelevated cortisol and minimal glucagon levels may contribute to body protein preservation and downregulation of catabolic pathways, in general. Furthermore, thyroid hormones may be involved in a process of energy conservation, independent of pups' nutritional state. These original hormonal settings might reflect an adaptation to the otariid repeated fasting pattern and emphasize the crucial importance of a tight physiological control over metabolism to survive extreme energetic constraints.

Survival depends on an animal's ability to find and acquire prey. In diving vertebrates, this ability is directly related to their physiological capability (e.g. oxygen stores). We studied the seasonal variation in oxygen stores, body temperature and body condition in California sea lions (Zalophus californianus) (CSL) as a function of seasonal variation in temperature, primary productivity, diving behavior and reproductive stage. During summer, blood oxygen stores were significantly greater and muscle oxygen stores were significantly lower than in winter. Total oxygen stores, body condition and body temperature did not change between seasons but variations in body temperature were greater during summer. Changes in oxygen stores are partly attributed to diving behavior, temperature and pregnancy that could increase oxygen consumption. Blood and muscle oxygen stores appear to be influenced by reproductive state. Blood oxygen stores are more likely influenced by diving behavior and temperature than muscle oxygen stores.

Long-term monitoring programs to evaluate climate-driven changes to tidewater glaciers, an important habitat for harbor seals (Phoca vitulina) in Alaska, are primarily carried out by costly, weather-dependent aerial surveys from fixed-winged aircraft. Unmanned aerial systems (UAS) can be an alternative cost-effective solution for gathering image data to quantify, monitor, and manage these habitats. However, there is a paucity of information related to the accuracy of using imagery collected by UAS for purposes of measuring floating icebergs. We evaluated the accuracy of using a UAS with a built-in 20 megapixel (MP) camera as well as a consumer-grade digital 16 MP camera to capture images of floating and stationary icebergs for the purpose of collecting vertical height measurements. Images (n=869) were captured of simulated icebergs (Cuboidal foam boxes) "Cb" (n=5) and real icebergs (n=5) that were either grounded or floating. The mean error ratios obtained were less than 10% and derived by comparing the mean calculated measurements of heights of Cb obtained from images captured by UAS with the physically measured heights of these Cb. The mean error ratio for height measurements of grounded icebergs (n=4) and one floating iceberg was also less than 10%. Within an object-image distance range of 6-25 m, the cameras captured images that were suitable to accurately calculate the heights of floating and grounded objects, and drift or uncontrolled movement of the UAS caused by wind or temporary loss of GPS did not appear to have any significant effects on measurement error. Our study provides substantial evidence of the high accuracy associated with using images captured by UAS for measuring dimensions of structures positioned on water and land surfaces. Ultimately, accurate surveys of glacial ice used by harbor seals will improve our understanding of the role of decreasing habitat in explaining population variability between different tidewater glaciers.

Most of our knowledge on reproductive biology of gray whales dates back to scientific research conducted during commercial whaling in the late 1950s and 1960s. The goal of the present study was to provide updated insights on reproductive physiology of gray whales, using progesterone and testosterone as biomarkers. We measured hormone concentrations using enzyme immunoassay (EIA) techniques in blubber biopsies collected from 106 individual whales from March to November over a span of 12 years (2004-2016) between California and Alaska. We found testosterone concentrations in males to increase significantly with age (P = 0.03). Adult males showed significantly elevated testosterone concentrations when sampled in the fall compared to the summer (P = 0.01), likely indicating physiological preparation for mating. We measured testosterone concentrations in females of different age classes, but no statistical differences were found. We found significantly higher progesterone concentrations in pregnant females compared to non-pregnant females and adult males (P< 0.001), indicating progesterone is a valid biomarker for pregnancy in gray whales. Both female and male calves had elevated progesterone concentrations, suggesting maternal transfer via lactation. We fit a mixture of two normal distributions to progesterone data from all non-calf females to identify clusters of high and low progesterone and estimated the probability of being pregnant for whales of unknown reproductive status. With this approach we identified likely pregnant and non-pregnant animals. This study represents an important milestone on reproductive profiles in this population, that can be used to estimate more accurate and precise reproductive parameters to be used for better understanding population dynamics of gray whales.

Southeast Alaska sea otters (Enhydra lutris) have had a rapid rise in their population. As they feed primarily on sessile prey, they are excellent sentinels for examining metals contamination. Objectives of this study on sea otters were to determine: (1) concentrations of metals in different tissues; (2) whether metals biomagnify from stomach contents (i.e., the prey) to other tissues; (3) whether selenium and mercury concentrations indicate an overall health benefit or risk; and (4) if metals concentrations in tissues vary with body size. Brain, kidney, gonad, liver, and stomach contents were collected from freshly harvested sea otters in Icy Strait, Alaska, and analyzed for arsenic (As), cadmium (Cd), copper (Cu), lead (Pb), total mercury (THg), and selenium (Se). Metals concentrations varied significantly, and some were biomagnified, with livers and kidneys harboring the highest concentrations. Lead and arsenic appeared to be readily excreted. This study represents baseline metals concentrations to assist in monitoring the health of sea otters.

The role of stress in the decline of Steller sea lions has become of interest in the identification of factors leading to their drastic decline and subsequent failure to recover. Serum cortisol was validated as a potential indicator of stress in three sub-populations inhabiting the majority of the Steller sea lions range (n=941). Additionally, seasonal variability in serum cortisol measured in captive animals (n=9) of different age classes and sex was investigated. Significant interactions were found between region and year, as well as year and sex (p=0.001 and p=0.005, respectively). Samples from pups in Russia (151.5+/-3.2 ng ml(-1)) and southeast Alaska (152.7+/-2.7 ng ml(-1)) were consistently higher than concentrations measured in pups from southwest Alaska (133.0+/-3.2 ng ml(-1)). Serum cortisol concentrations measured in 2001 (134.9+/-2.0 ng ml(-1)) were significantly lower than 2002 (156.5+/-2.4 ng ml(-1), p<0.001) for all regions. Seasonal effects indicated significant interactions between age, year and sex (p=0.011). Serum cortisol was elevated in months associated with the breeding season and annual molt (86.5+/-5.0 and 92.4+/-5.1 ng ml(-1), respectively) compared to the other seasons. Our results suggest that cortisol concentrations in Steller sea lions can be a useful diagnostic tool to compare the physiology between groups of sea lions, but factors such as animal sex, age, season and year must be considered.
