RESUMO

Fasciolosis caused by Fasciola hepatica is a common parasitic disease of livestock especially sheep and cattle. In this study molecular characterization of ß-tubulin isotype 3 gene in Fasciola hepatica isolates from cattle and sheep in Turkey was carried out. For this purpose a total of 80 adult Fasciola hepatica isolates were collected from 20 sheep and 20 cattle in Kayseri and Erzurum provinces. PCR-RFLP was performed on ß-tubulin isotype 3 gene and MboII revealed two fragments of approximately 350 bp and 390 bp, whereas HphI enzyme yielded 210, 340 and 540 bp bands, HindII yielded 380 and 450 bp bands in all samples. A total of 80 isolates were tested by SSCP and all of them presented the same band profiles. Six samples (4 sheep and 2 cattle) were randomly selected and DNA sequence of a 935 bp coding fragment of ß-tubulin isotype 3 was performed. Sheep samples were more polymorphic than the cattle. This ß-tubulin isotype 3 gene polymorphism of F.hepatica isolates from sheep and cattle of two distinct geographical areas of Turkey have been investigated for the first time.

BACKGROUND: Cystic echinococcosis (CE), caused by hydatid cysts, is a widespread and hazardous disease in humans and animals worldwide. The disease is very common in Turkey, causing serious economic losses and public health problem. In this study, the seroprevalence of equine CE was determined by enzyme-linked immunosorbent assay (ELISA). METHODS: Partially purified cyst fluid antigen from sheep hydatid cyst fluid was used as antigen in ELISA. A total of 250 equids consisting of 206 donkeys and 44 horses from various regions of Erzurum province of Eastern Turkey. RESULTS: Anti- Echinococcus granulosus antibodies were detected in 78 (31.2%) of 250 equids. The prevalence rate was 20.4% for horses and 33.5% for donkeys. There was no statistically difference between sex and ages groups for both horses and donkeys (P>0.05). CONCLUSION: Equine CE is quite endemic in Eastern Turkey. The high prevalence of CE suggests that equids in the transmission cycles is possible as a source of infection for definitive hosts.

This study was conducted to determine the prevalence and molecular characteristics of Coenurus cerebralis, the metacestode of Taenia multiceps in cattle from Erzurum province located in eastern region of Turkey between November 2009 and April 2010. Five of 1045 brains of cattle (0.47%) were found to be infected with Coenurus cysts. The characteristics and morphology of C. cerebralis were seen in all the cysts. The cyts from three infected cattle were genetically analyzed and confirmed to be T. multiceps metacestodes by NAD1 and COX1 mitochondrial gene sequence analysis. Pairwise comparison between the NAD1 sequences of the T. multiceps isolates from Erzurum and other T. multiceps isolates available in GenBank showed differences ranging from 0.6 to 2.9%, while COX1 sequences showed differences ranging from 0.2 to 2.6%. Considering the two genes, it was seen that all of the three isolates from Erzurum province were in the same group according to phylogenetic analyses. The present findings could provide a stimulus for future studies on the systematic relationships and epidemiology of lesser-known taeniid cestodes in the region, employing mitochondrial sequence data sets.
