RESUMO

The surface of aboveground plant parts, known as the phyllosphere, is a habitat for various microorganisms called epiphytes establishing biotrophic interactions with their hosts. However, these communities can be affected by environmental and anthropogenic variations such as the application of agrochemicals. Thus, epiphytes have the capacity to survive in such environments. In this study, we obtained the genome of Pseudomonas sp. 14A, an epiphyte isolated from the pepper phyllosphere. The phylogenomic analyses suggested that Pseudomonas sp. 14A may be novel species closely related to P. moraviensis R28-S. Notably, the metabolic pathways proposed consistent with epiphytic lifestyle in Pseudomonas sp. 14A, were shared with other species displaying a different degree of phylogenetic relatedness. Furthermore, variations in configuration of metabolic gene clusters were observed, that could expand microbial metabolic diversity in close relatedness species, highlighting the relevance of microbial diversity associated with plants.

Among Boletales, the family Boletaceae has the highest diversity worldwide. Additionally, this fungal group has great ecological relevance because it not only includes mainly ectomycorrhizal but also saprotrophic species. Furthermore, some species are used as food and have sociocultural and economic importance worldwide. In Mexico, the Boletaceae family boasts a substantial number of species, yet our understanding of these species remains far from comprehensive. In this work, by using macro- and micromorphological and phylogenetic analyses of DNA sequences from multi-gene analyses based on ITS, nrLSU, rpb1, rpb2, and tef1, we report five new species belonging to the genera Aureoboletus and Chalciporus: A. ayuukii and A. elvirae from a Quercus scytophylla forest, A. readii from a mixed forest, C. perezsilvae from cloud forest, and C. piedracanteadensis from both a mixed coniferous forest and a Quercus-Pinus forest. In Mexico, four species of Aureoboletus are used as a food source, and in this work, we add another one, A. readii, which is traditionally consumed by members of the Tlahuica-Pjiekakjoo culture, who are located in the central part of the country. This work contributes to our knowledge of two genera of Boletaceae in a geographical area that is scarcely studied, and thus, our understanding of its biocultural relevance is enriched.

The present study describes Hemiaustroboletus gen. nov. in the subfamily Austroboletoideae (Boletaceae). Hemiaustroboletus is supported by morphological and molecular data using LSU and RPB2 regions. Additionally, its geographic distribution and intraspecific variation were inferred using ITS sequences. The genus is characterised by pileate-stipitate basidiomata; purple, brown, reddish-brown, orange-brown to dark brown vinaceous pileus; whitish or lilac to vinaceous context and a subclavate stipe. Microscopically, it is characterised by ornamented, slightly verrucose, cracked to perforated brown basidiospores. Two species are described within the genus, Hemiaustroboletusvinaceobrunneus sp. nov. and H.vinaceus sp. nov. Hemiaustroboletusvinaceus sp. nov. is morphologically similar to Austroboletusgracilis, which suggests they may have been confused in the past. This study presents the phylogenetic placement, microscopic structures, detailed morphological descriptions and illustrations of both new species.

Anaerobic digestion (AD) has recently been studied to obtain products of greater interest than biogas, such as volatile fatty acids (VFAs) and phytoregulators. The effect of the initial pH of cow manure and the fermentation time of the AD on the microbial composition, VFAs, indole-3-acetic acid (IAA) and gibberellic acid (GA3) production was evaluated. The cow manure (7% solids) was adjusted to initial pH values of 5.5, 6.5, 7.5, and 8.5, and the AD products were analyzed every four days until day 20. The initial pH and the fermentation time had an important effect on the production of metabolites. During AD, only the hydrolytic and acidogenic stages were identified, and the bacteria found were from the phyla Firmicutes, Bacteroidetes, Actinobacteria, and Spirochaetes. The most abundant genera produced in the four AD were Caproiciproducens, Clostridium sensu stricto 1, Romboutsia, Paeniclostridium, Turicibacter, Peptostreptococcaceae, Ruminococcaceae and Fonticella. The highest amount of VFAs was obtained at pH 8.5, and the production of the acids was butyric > acetic > propionic. The maximum production of GA3 and IAA was at an initial pH of 6.5 on day 20 and a pH of 5.5 on day 4, respectively. There was a strong correlation (> 0.8) between the most abundant microorganisms and the production of VFAs and GA3. The anaerobic digestion of cow manure is a good alternative for the production of VFAs, GA3 and IAA.
