BACKGROUND: Renal damage is one of the typical clinical manifestations of systemic lupus erythematosus (SLE) and few effective markers can be used to predict SLE with renal damage. Additionally, the relationship among AhR, B cells and SLE with renal damage is poorly understood. METHOD: A case-control study was performed, and the clinical and laboratory data were acquired from medical records. Flow cytometry was used to analyze the expression of AhR in B cells. RESULTS: Both the proportion of B cells in peripheral blood lymphocytes and AhR in B cells were significantly higher in SLE patients than in healthy controls. AhR in B cells was negatively correlated with complement 3 and 4 levels but was positively correlated with SLEDAI-2 K, erythrocyte sedimentation rate, C-reactive protein and absolute lymphocyte count. Furthermore, more SLE patients suffered from renal damage and arthritis in the high-AhR B-cell group than in the low-AhR B-cell group. Interestingly, AhR in B cells was significantly increased in SLE with renal damage compared with SLE without renal damage, but no significant difference was observed between SLE with arthritis and SLE without arthritis. In addition, the area under the curve for AhR in B cells was 0.815, and its optimal cut-off level was 3.05 %, which provided a 83.3 % sensitivity and a 70.0 % specificity in predicting SLE with renal damage. CONCLUSION: AhR in B cells was correlated with SLE disease activity, and it may be a potential marker for predicting SLE with renal damage.

The conventional chemical methods of nanoparticles synthesis have been effectively replaced by nanoparticle synthesis mediated by plants. The current study describes the environmental friendly synthesis of chromium oxide nanoparticles (Cr2O3 NPs) using Erythrophleum guineense plant extract. The synthesis of Cr2O3 NPs was validated by UV/VIS spectroscopy, Energy Dispersive X-Ray (EDX), Scanning Electron Microscopy (SEM), and X-ray diffraction (XRD) studies. The appearance of the Sharpe peak at 460 nm in the UV/Vis spectrum and the colour change caused by surface plasma resonance confirmed the formation of Cr2O3 NPs. The EDX spectrum of Cr2O3 nanoparticles revealed the presence of carbon, oxygen, and chromium, while SEM analysis revealed an irregular round morphology (with a size below 400 nm). In addition, XRD studies suggested their crystalline nature by the characteristic peaks at 34° and 36° and 42° (2Ɵ), respectively. The green synthesized Cr2O3 NPs showed promise as in-vitro cholinesterase inhibitor at tested concentrations (62.5-1,000 µg/ml), with IC50 values of 120 and 100 µg/ml against Acetylcholinesterase (AChE) and Butyrylcholinesterase (BChE), respectively. The results suggested that the green synthesized Cr2O3 NPs could be used in the future to stop enzyme from working and for other biological activities.

Infection caused by antibiotic-resistant microorganisms (ARMs) has been declared a global threat to public health. Polymeric nanoparticles (PNPs) formed by antimicrobial peptides (AMPs) and synthetic PNPs against ARM infections are emerging. PNPs are also considered to be a promising natural biological preservative that prevents microbial spoilage through food processing and preservation. We engineered CNMs, a novel nanocomposite antibacterial agent based on chitosan nanoparticles and AMP microcin J25. In this study, we aimed to evaluate the comprehensive antimicrobial activity, potential antimicrobial mechanism, and anti-inflammatory activity of CNMs. We demonstrated that CNMs harbor excellent bactericidal activity against clinical foodborne pathogens and ARMs. CNMs caused fast mortality against different growth phases of tetracycline (Tet)-resistant enterotoxigenic E. coli (ETEC) and significantly killed Tet-resistant ETEC in food biological environments. Mechanistically, CNMs have the ability to bind lipopolysaccharides (LPS), neutralize endotoxin, and promote diaphragm permeability by damaging the cell membrane. CNMs did not cause mouse RAW264.7 cell cytotoxicity. Notably, CNMs significantly reduced the cytotoxicity of RAW264.7 macrophages induced by LPS. The LPS-induced inflammatory response was significantly ameliorated by CNMs by reducing the levels of nitric oxide and proinflammatory cytokines, including tumor necrosis factor α, interleukin (IL)-6, IL-8, IL-1ß, Toll-like receptor 4, and nuclear factor κB (NF-κB), in LPS-challenged RAW264.7 macrophages. CNMs downregulated the NF-κB and mitogen-activated protein kinase signaling pathways, thereby inhibiting inflammatory responses upon LPS stimulation. Taken together, CNMs could be applied as effective antimicrobial/anti-inflammatory agents with lower cytotoxicity in food, medicine, and agriculture to prevent bacterial contamination and infection, respectively.

BACKGROUND: Surgical resection is the only widely accepted curative method for intrahepatic cholangiocarcinoma (ICC). However, little is known about the efficacy of laparoscopic liver resection for ICC, especially in patients with early-stage disease. The aim of this study was to compare the short-term and long-term effects of laparoscopy and open surgery for the treatment of ICC. METHODS: Data from 1,084 patients treated at three hospitals from January 2011 to December 2018 were selected and analyzed. Propensity score matching was performed to compare the long-term outcomes (overall survival and recurrence-free survival) and short-term outcomes (perioperative outcomes) of all-stage and early-stage patients. RESULTS: After matching, 244 patients (122 vs. 122) in the all-stage group and 65 patients (27 vs. 38) in the early-stage group were included. The baseline of the two groups was balanced, and no significant differences were found in sex or age. The short-term results of the laparoscopic group were better than those of the open group, including less blood loss [blood loss ≥400 ml 27 (22.1%) vs. 6 (4.92%), p<0.001 for all-stage, 12 (31.6%) vs. 2 (7.41%), p=0.042 for early stage), shorter surgery [200 (141; 249) min vs. 125 (115; 222) min, p=0.025 for early stage] and shorter hospital stay [11.0 (9.00; 16.0) days vs. 9.00 (7.00; 12.0) days, p=0.001 for all stage, 11.0 (8.50; 17.8) days vs. 9.00 (6.50; 11.0) days, p=0.011 for early stage]. Regarding long-term outcomes, no significant differences were found for all-stage patients, while there were significant differences observed for the early-stage group (p=0.013 for OS, p=0.014 for RFS). For the early-stage patients, the 1-, 3-, and 5-year OS rates of the OLR group were 84.2, 65.8, and 41.1%, respectively, and those of the LLR group were 100, 90.9, and 90.9%, respectively. The RFS rates of the OLR group were 84.2, 66.7, and 41.7%, respectively, and those of the LLR group were and 92.3, 92.3, and 92.3%, respectively. CONCLUSION: Patients treated with laparoscopy seemed to have better short-term outcomes, such as less blood loss, shorter operation duration, and shorter hospital stay, than patients undergoing open surgery. Based on the long-term results, laparoscopic treatment for early ICC may have certain advantages.

Neuronal communication between different brain areas is achieved in terms of spikes. Consequently, spike-time regularity is closely related to many cognitive tasks and timing precision of neural information processing. A recent experiment on primate parietal cortex reports that spike-time regularity increases consistently from primary sensory to higher cortical regions. This observation conflicts with the influential view that spikes in the neocortex are fundamentally irregular. To uncover the underlying network mechanism, we construct a multilayered feedforward neural information transmission pathway and investigate how spike-time regularity evolves across subsequent layers. Numerical results reveal that despite the obviously irregular spiking patterns in previous several layers, neurons in downstream layers can generate rather regular spikes, which depends on the network topology. In particular, we find that collective temporal regularity in deeper layers exhibits resonance-like behavior with respect to both synaptic connection probability and synaptic weight, i.e., the optimal topology parameter maximizes the spike-timing regularity. Furthermore, it is demonstrated that synaptic properties, including inhibition, synaptic transient dynamics, and plasticity, have significant impacts on spike-timing regularity propagation. The emergence of the increasingly regular spiking (RS) patterns in higher parietal regions can, thus, be viewed as a natural consequence of spiking activity propagation between different brain areas. Finally, we validate an important function served by increased RS: promoting reliable propagation of spike-rate signals across downstream layers.
