Blooms of Aureococcus anophagefferens, referred to as brown tides are responsible for massive mortalities and recruitment failure of some bivalves. However, the molecular mechanisms underlying the toxicity remain elusive despite its biological significance, and the information currently available on the molecular effects is still insufficient. In this study, to evaluate the toxicity and associated mechanism of A. anophagefferens on bivalves, we analyzed the protein expression profiles in digestive glands of the A. anophagefferens-exposed Perna viridis by using iTRAQ. A total of 3138 proteins were identified in the digestive glands of A. anophagefferens-exposed P. viridis based on iTRAQ. Amongst, a repertoire of 236 proteins involved in cell, cell part, catalytic activity, metabolic process, biological regulation, immune system process, and response to stimulus were found to be differentially expressed. Functional analysis of the differentially expressed proteins demonstrated that innate immune system of P. viridis was activated, and some proteins associated with stress response and lipid metabolism were induced after exposure to A. anophagefferens. Additionally, MDA content, SOD activity and GSH-Px activity was increased significantly in the digestive gland of A. anophagefferens-exposed P. viridis. Taken together, our results indicated that the A. anophagefferens could induce oxidative stress, activate complement system and alter fat acid metabolism of P. viridis.

A series of gramine modified montmorillonites (gramine-MMTs) were prepared and characterized by thermogravimetric analysis, X-ray diffraction and zeta potential measurements. These modified clays with various amounts of gramine all exhibited higher removal efficiencies of both Chattonella marina and Alexandrium tamarense than did natural clay. With the augmentation of gramine content in the modified clays, the removal efficiency increased. The 24 h LC50 values of gramine-MMT with 12.68 % of gramine on C. marina and A. tamarense were 9.16 and 10.21 mg L(-1), respectively. These data suggest gramine-MMT might be a promising material to remediate harmful algal blooms.

Given that harvesting time has a great impact on the quality of herbal medicine, knowing the ontogenesis in the chemical profile aspect is essential to determine the optimal harvesting season. A high-throughput and versatile approach (herbal infrared macro-fingerprinting) harmonizing with the character of herbal medicine and providing the whole chemical profile (entirety), group analogues (part), and single compounds (major components) is developed to rapidly disclose the variation rule of the full chemical profile of herbal medicine over a growing season without extraction pretreatments, and thus to determine the optimal harvesting period in respect to groups of chemical compounds using Scutellaria baicalensis as a demonstration. IR macro-fingerprints of Scutellaria baicalensis harvested in the same period have a high similarity (> 0.91) despite small variations, suggesting that IR macro-fingerprinting can faithfully reflect the spectacle of "disordered order" in nature. From Year-1 spring to Year-3 autumn, general contents (%, w/w) of total flavonoids fluctuate up and down with a maximum value in Year-2 spring, and that of saccharides is relatively stable except for the attenuation from Year-2 autumn to Year-3 spring. From Year-1 autumn to Year-2 spring, flavonoid aglycones initially produced in Scutellaria baicalensis are extensively transformed to responding flavonoid glycosides. From Year-2 spring to Year-3 autumn, flavonoid glycosides are converted back to their corresponding aglycones. The best seasons for collecting Scutellaria baicalensis with a high content of flavonoid glycosides and aglycones would be Year-2 spring and Year-3 spring, respectively.

In the present paper, a tri-step infrared (IR) spectroscopy was used to study Scutellaria baicalensis Georgi (SBG) harvested in spring and autumn. The positions of peaks in the IR spectra of SBG harvested in spring and autumn were rather similar. However, according to the differences in the relative intensities of those characteristic peaks which include the ester carbonyl C=O absorption peak at 1 740 cm(-1), the peak near 1 614 cm(-1) assigned to the flavonoids, and the peak near 1 071 cm(-1) assigned to the carbohydrates, the amount of flavonoids and esters of spring SBGs was higher than that of autumn SBGs. Their carbohydrates were different by comparing the second derivative infrared spectra in the range of 1 300-400 cm(-1). Furthermore, the above differences were visually validated by two-dimensional correlation spectroscopy (2D-COS). Moreover, the FTIR spectra of 16 batches of SBG harvested in spring and autumn were analyzed with principal component analysis, and subsequently they were exactly identified and classified. Therefore, tri-step identification of IR spectroscopy combined with principal component analysis can be employed to fast and accurately identify the SBG harvested in spring and autumn and differentiate the differences of their chemical constituents.

OBJECTIVE: This study aimed to determine the expression of lactate dehydrogenase (LDH)-A and LDH-D in patients with uterine myoma, cellular leiomyoma (CLM), and uterine sarcoma and to evaluate their prognostic significance. METHODS: Protein expression levels of LDH-A and LDH-D were determined in tissue samples from 86 patients (26 uterine myoma, 10 CLM, 50 uterine sarcoma) by immunohistochemistry and their associations with clinicopathologic parameters and outcomes were analyzed in patients with uterine sarcoma. RESULTS: The positivity rates for LDH-A and LDH-D were significantly higher in patients with uterine sarcoma compared with those with uterine myoma or CLM (P < .05). Patients with uterine sarcoma were classified as having uterine leiomyosarcoma (LMS), malignant endometrial stromal sarcoma, and malignant mixed Mullerian tumor, with 5-year overall survival rates of 59%, 71%, and 29%, respectively (P < .05). Univariate analysis showed that patients younger than 50 years and with stage I-II had better clinical prognoses. LDH-A-positive LMS patients had a poorer prognosis than LDH-A-negative patients (P = .03). The median survival time of LDH-A-positive patients was 35 months. CONCLUSIONS: We demonstrated that LDH-D was expressed in patients with uterine sarcoma. Furthermore, the overexpressions of LDH-A and LDH-D in uterine sarcoma patients may contribute to further understanding of the mechanism of LDH in tumor metabolism in uterine sarcoma. Positive expression of LDH-A in patients with LMS may act as a potential prognostic biomarker in these patients.

In this paper, a new PVC-based liquid-membrane anion-selective electrode based on a copper(II) of N,N'-bis(salicylidene)-1,2-bis(p-aminophenoxy)ethane tetradentate complex (Cu(II)BBAP) is described, which displays a preferential potentiometric response to iodide ion at pH 2.0 and an anti-Hofmeister selectivity sequence: I->SCN->ClO4->NO2->H2PO4->NO3->SO4(2-)>Br->Cl-. The electrode exhibits a near-Nernstian potential linear range of 8.2x10(-7)-1.0x10(-1) M with a detection limit of 5.3x10(-7) M and a slope of -58.8 mV per decade. The A.C. impedance technique and the UV/Vis spectroscopy technique were used to analyze the response mechanism. The electrode could be applied to determine iodide in medicine analysis, and the obtained results were fairly satisfactory.

P-glycoprotein (P-gp or ABCB1) belongs to the family of ATP-binding cassette (ABC) transporters responsible for multixenobiotic resistance (MXR) in aquatic organisms. To provide more information of P-gp in shellfish, in this study, complete cDNA of P-gp in three bivalve species including Ruditapes philippinarum, Scapharca subcrenata and Tegillarca granosa were cloned and its expressions in gill, digestive gland, adductor muscle and mantle of the three bivalves were detected after exposure to Prorocentrum lima, a toxogenic dinoflagellate. The complete sequences of R. philippinarum, S. subcrenata and T. granosa P-gp showed high homology with MDR/P-gp/ABCB proteins from other species, having a typical sequence organization as full transporters from the ABCB family. Phylogenetic analyses revealed that the amino acid sequences of P-gp from S. subcrenata and T. granosa had a closest relationship, forming an independent branch, then grouping into the other branch with Mytilus californianus, Mytilus galloprovincialis and Crassostrea gigas. However, P-gp sequences from R. philippinarum were more similar to the homologs from the more distantly related Aplysia californica than to homologs from S. subcrenata and T. granosa, suggesting that bivalves P-gp might have different paralogs. P-glycoprotein expressed in all detected tissues but there were large differences between them. After exposure to P. lima, the expression of P-gp changed in the four tissues in varying degrees within the same species and between different species, but the changes in mRNA and protein level were not always synchronous.

A novel thiocyanate (SCN-)-selective PVC membrane electrode based on a zinc-phthalocyanine (ZnPc) complex as neutral carrier is described. The membrane electrode containing ZnPc with 5.1% (w/w) ionophore, 29.2% (w/w) PVC, and 65.7% (w/w) 2-nitrophenyl octyl ether (o-NPOE) as plasticizer displayed an anti-Hofmeister selectivity sequence, SCN- > Sal- > I- > ClO4- > Br- > Cl- > NO3- > NO2- > H2PO4- > SO4(2-)), and exhibited near-Nernstian potential response to thiocyanate ranging from about 1.0 x 10(-1) to 1.0 x 10(-6) mol L(-1) with a detection limit of 7.5 x 10(-7) mol L(-1) and a slope of 58.1+/-0.5 mV per decade in pH 3.0 phosphate buffer solution at 25 degrees C. This preferential response is believed to be associated with the unique coordination between the central metal of the carrier and thiocyanate.
