RESUMO

Triatomine assassin bugs comprise hematophagous insect vectors of Trypanosoma cruzi, the causative agent of Chagas disease. Although the microbiome of these species has been investigated to some extent, only one virus infecting Triatoma infestans has been identified to date. Here, we describe for the first time seven (+) single-strand RNA viruses (RpV1-7) infecting Rhodnius prolixus, a primary vector of Chagas disease in Central and South America. We show that the RpVs belong to the Iflaviridae, Permutotetraviridae and Solemoviridae and are vertically transmitted from the mothers to the progeny via transovarial transmission. Consistent with this, all the RpVs, except RpV2 that is related to the entomopathogenic Slow bee paralysis virus, established persistent infections in our R. prolixus colony. Furthermore, we show that R. prolixus ovaries express 22-nucleotide viral siRNAs (vsiRNAs), but not viral piRNAs, that originate from the processing of dsRNA intermediates during viral replication of the RpVs. Interestingly, the permutotetraviruses and sobemoviruses display shared pools of vsiRNAs that might provide the basis for a cross-immunity system. The vsiRNAs are maternally deposited in the eggs, where they likely contribute to reduce the viral load and protect the developing embryos. Our results unveil for the first time a complex core virome in R. prolixus and begin to shed light on the RNAi-based antiviral defenses in triatomines.

BACKGROUND: SARS-CoV-2 emerged in China and spread throughout the world due to its rapid transmission. The exposure rate in the healthy population is unknown, mainly in resource-limited countries. Herein, we estimated the seroprevalence of anti-SARS-CoV-2 antibodies and risk factors among blood donors in Luanda, the capital city of Angola. METHODS: This was a retrospective study conducted with 343 blood donors. Chi-square and logistic regression were calculated to predict the independent variable for SARS-CoV-2 infection and deemed significant when p < 0.05. RESULTS: Seroprevalence of anti-SARS-CoV-2 was 4.7%. Positivity rates varied to age groups (3.5-14.3%), gender (0-5%), area of residence (3.1-.6%), educational level (5.1-10.2%), occupation (4.4-7.7%), and the blood donor category (2.0-5.1%). Past and recent infections were detected in 3.2% and 1.5%, respectively. Blood donors under the age of 20 years (OR: 4.58, p = 0.241) and from non-urbanized areas (OR: 1.86, p = 0.293) presented a high risk related to infection. The infection was higher in blood group A and lower in blood group O. The risk of SARS-CoV-2 infection has increased from January 2020 (OR: 0.03, p = 0.001) to August 2020 (OR: 0.57, p = 0.426). CONCLUSIONS: We provide an estimate of the exposure of healthy blood donors in Luanda. Also, we detected anti-SARS-CoV-2 in January 2020, indicating that the SARS-CoV-2 could have been imported during the first month of 2020. Further studies should be performed to assess the exposure rate in different groups from Angola.

Jambolan is an unexplored fruit rich in bioactive compounds like anthocyanins, catechin, and gallic acid. Thus, the extraction of bioactive compounds allows adding value to the fruit. In this context, the present study reports the recovery and concentration of jambolan fruit extract by ultra and nanofiltration for the first time. Acidified water was used to extract polyphenols from the pulp and peel of jambolan. The extracts were concentrated using ultrafiltration and nanofiltration membranes with nominal molecular weight cut-off ranging from 180 to 4000 g mol-1. Total monomeric anthocyanin, total phenolic compounds, and antioxidant capacity were analyzed. Phenolic compounds were quantified, and anthocyanins were identified by high-performance liquid chromatography coupled to diode-array detection and mass spectrometry (HPLC-DAD-MS). Concentration factors higher than 4.0 were obtained for anthocyanins, gallic acid, and catechin after nanofiltration of the extracts. Other compounds such as epicatechin, p-Coumaric acid, and ferulic acid were quantified in the concentrated extract, and the main anthocyanins identified were 3,5-diglucoside: petunidin, malvidin, and delphinidin. Therefore, jambolan extract showed a high potential to be used as a natural dye and antioxidant in food products.

PURPOSE: Some studies have explored the relationship between music and cortical activities; however, there are just few studies investigating guitar performance associated with different sensory stimuli. Our aim was to evaluate alpha and beta activity during guitar playing. MATERIALS AND METHOD: Twenty healthy right-handed people participated in this study. Cortical activity was measured by electroencephalogram (EEG) during rest and 4 tasks (1: easy music with an auditory stimulus; 2: easy music with an audiovisual stimulus; 3: complex music with an auditory stimulus; 4: complex music with an audiovisual stimulus). The peak frequency (PF), median frequency (MF) and root mean square (RMS) of alpha and beta EEG signals were assessed. RESULTS: A higher alpha PF at the T3-P3 was observed, and this difference was higher between rest and task 3, rest and task 4, tasks 1 and 3, and tasks 1 and 4. For beta waves, a higher PF was observed at C4-P4 and a higher RMS at C3-C4 and O1-O2. At C4-P4, differences between rest and tasks 2 and 4 were observed. The RMS of beta waves at C3-C4 presented differences between rest and task 3 and at O1-O2 between rest and task 2 and 4. CONCLUSION: The action observation of audiovisual stimuli while playing guitar can increase beta wave activity in the somatosensory and motor cortexes; and increase in the alpha activity in the somatosensory and auditory cortexes and increase in the beta activity in the bilateral visual cortexes during complex music execution, regardless of the stimulus type received. BACKGROUND: Aseptic loosening (AL) of hip prosthesis presents inflammation and pain as sign and symptom similarly to arthritis pathologies. Still, the immune and innervation profiles in hip AL remain unclear and their interplay is poorly explored. Herein, local tissue inflammatory response, sensory and sympathetic innervation as well as associated local mediators were assessed in hip joint microenvironment underlying AL and compared to osteoarthritis (OA). METHODS: Histopathological analysis, immune cells (macrophages, T, B cells and PMNs) as well as sensory and sympathetic nerve fibers (SP(+), CGRP(+), TH(+)) distribution and profiles were analyzed on tissues retrieved from patients with failed hip prostheses due to AL (n = 20) and hip OA (n = 15) by immunohistochemistry. Additionally, transcriptional levels of pro-inflammatory cytokines (TNF-α, IL-1ß, IL-6, IL-12a, iNOS), anti-inflammatory cytokine (IL-10), osteoclastic factor (RANKL) and bone remodeling factor (TGF-ß1) were locally evaluated by qRT-PCR. Serum TGF-ß1 levels were assessed preoperatively by ELISA. RESULTS: Histopathological analysis revealed that tissues, aseptic interface membranes of AL patients had distinct tissue architecture and immune cells profile when compared to OA synovial tissues. Macrophages, T cells and B cells showed significant differences in tissue distribution. In OA, inflammation is mostly confined to the vicinity of synovial membrane while in AL macrophages infiltrated throughout the tissue. This differential immune profile is also accompanied with a distinct pattern of sensory and sympathetic innervation. Importantly, in AL patients, a lack of sympathetic innervation aseptic interface membranes without compensation mechanisms at cellular levels was observed with simultaneous reorganization of sensorial innervation. Despite the different histopathological portrait, AL and OA patients exhibited similar transcriptional levels of genes encoding key proteins in local immune response. Nevertheless, in both pathologies, TGF-ß1 expression was prominent in sites where the inflammation is occurring. However, at systemic level no differences were found. CONCLUSION: These findings indicate that AL patients exhibit different local inflammatory response and innervation signatures from OA patients in hip joint. These insights shed the light on neuro-immune interplay in AL and highlight the need to better understand this crosstalk to unravel potential mechanisms for targeted-therapies to improve hip joint lifetime and treatment.

Background and Aims: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection is a public health concern. Until 2021, more than 2 million cumulative deaths were reported worldwide. Herein, we investigated the immune profile of healthcare professionals 6 months after vaccination or exposure to SARS-CoV-2 in Angola. Methods: This was a prospective study conducted with 1068 Angolan healthcare professionals between August and December 2021. Participants were screened for the presence of IgG and IgM against SARS-CoV-2. Results: About 9.6% and 98.2% of the participants had prior exposure to SARS-CoV-2 or vaccination against it, respectively. Participants aged between 20 and 40 years (11.2%), female (12.4%), with higher educational level (12.8%), from Luanda (60.3%), and nonhealthcare professionals (8.1%) were the most affected by the SARS-CoV-2. Gender, education, and local residence were related to SARS-CoV-2 exposure (p < 0.05). About 7.3% and 98% of the exposed population developed IgM and IgG after 3 months of exposure, respectively. The AstraZeneca vaccine was the most used, followed by the Jonhson & Johnson and Sputinik. Almost all (98%) participants vaccinated with AstraZeneca had immunity >3 months. Individuals who received only the first dose regardless of the type of vaccine had a higher immunity duration (>3 months) than those who received two doses. For individuals who received the Sputnik and Johnson, the average immunity was lower (<3 months), especially among those who were older (over 40 years old) and exposed to SARS-CoV-2. Conclusion: We observed a high adherence rate to vaccination and a long immunity duration. The immunity duration depended on the type of vaccine. Further studies on the immunity profile in the population exposed to SARS-CoV-2 must be carried out in the general population from Angola to assess antibody-waning periods.

Background and Aims: SARS-CoV-2 infection is a public health concern. Several aspects related to the pattern of infection remain unclear. This study aimed to investigate the blood pressure pattern among blood donors exposed to SARS-CoV-2 in Luanda, Angola, a sub-Saharan African country. Methods: We performed a retrospective analysis containing 343 blood donors from December 2019 to September 2020. Parametric tests compared means while χ 2 and logistic regression checked features associated with high blood pressure and were considered significant when p < 0.05. Results: The mean age of blood donors was 32.2 ± 8.81 years (ranging from 18 to 61 years) and 93% of the men's gender. Overall, 4.7% of the studied population had been exposed to SARS-CoV-2. High blood pressure prevalence increased from unexposed to exposed SARS-CoV-2 (6.7%-18.8%, p = 0.071). SARS-CoV-2 exposure increase systole (131 ± 12.2 mmHg to 136 ± 14.2 mmHg, p = 0.098), diastole (79.9 ± 9.53 mmHg to 84.2 ± 12.7 mmHg, p = 0.086), pulse in beats per minute (72.0 ± 11.1 to 73.7 ± 8.50, p = 0.553), and decrease donating time (6.31 ± 3.72 min to 5.48 ± 1.61 min, p = 0.371). Chances of having high blood pressure were high [OR: 3.20 (95% confidence interval [CI]: 0.85-12.1), p = 0.086] in exposed SARS-CoV-2. Donors exposed to SARS-CoV-2 with abnormal donation time increased from the donor up to 40 years to over 40 years (from 35.7% to 50%, p = 0.696). The mean systolic, diastolic, and pulse pressure were higher for non-O donors (p > 0.05). A significant link was observed, between the Rhesus factor and blood pressure status (p = 0.032). Conclusion: We showed important variations in blood pressure indices of the Angolan population exposed to SARS-CoV-2. Older age and non-O blood groups appear to be important biological factors for SARS-CoV-2 infection, as well as the risk of developing cardiovascular disease after or during SARS-CoV-2 exposure. Further studies assessing the impact on cardiovascular functions with ongoing or long-term SARS-CoV-2 exposure in individuals from resource-limited countries should be considered.

Tuberculosis (TB) is a major cause of illness and public health concern, especially in resource-limited countries. This study analyzed the characteristics related to anti-TB drug resistance. Moreover, we examined the evidence-based indications for the treatment of active TB in Angola. This study evaluated the medical records of 176 patients screened for TB from January to September 2016 in Luanda, the capital city of Angola. Approximately 66.5% of the patients were newly diagnosed with active TB. The residence area showed a significant relationship with TB (P = 0.025), whereas age group (P = 0.272), gender (P = 0.853), and HIV status (P = 0.284) did not showed any relationship with TB. Overall, 72.4% of TB patients had resistance to at least one of the anti-TB drugs. The risk of anti-TB drug resistance was higher in males (odds ratio [OR]: 1.22; 95% confidence interval [CI]: 0.42-3.58, P = 0.685] and in TB-HIV coinfected patients [OR: 1.39; (95% CI: 0.26-7.28), P = 0.700], whereas it was lower in patients aged 30 years or older (OR: 0.56; 95% CI: 0.18-1.69) P = 0.303) and in patients living in urbanized areas (OR: 0.74; 95% CI: 0.17-3.25; P = 0.685). Our findings showed that drug-resistant TB is emerging in Angola. Further studies on factors related to anti-TB drug resistance are urgently needed to ascertain the magnitude of the problem and to proffer strategies toward TB control in Angola.

RESUMO

The Siegert relation relates field and intensity temporal correlations. After a historical review of the Siegert relation and the Hanbury Brown and Twiss effect, we discuss the validity of this relation in two different domains. We first show that this relation can be used in astrophysics to determine the fundamental parameters of stars, and that it is especially important for the observation with stellar emission lines. Second, we check the validity of this relation for moving quantum scatterers illuminated by a strong driving field.

BACKGROUND AND PURPOSE: Pain is a common condition among people with hemophilia (PWH), negatively impacting quality of life. However, effective treatment remains a challenge. This two-arm, parallel randomized controlled pilot trial aimed to examine the three-month effects of hypnosis intervention on clinical and psychosocial variables, and on the inflammatory profile of PWH. MATERIALS AND METHODS: The study was conducted between January and October 2018, in a Reference Center for Congenital Coagulopathies. Adult (age ≥18) patients were randomized to experimental group (EG) or control group (CG). The EG received four weekly hypnosis sessions plus treatment-as-usual, and the CG maintained treatment-as-usual only. Outcomes were evaluated at one week and three months post-intervention and included pain, joint status, health-related quality of life (HRQoL), emotional state and inflammatory profile (leukocytes, C-reactive protein, cytokines). The randomization sequence was computer-generated, and allocation was concealed until enrolment. The outcome assessor was blind to allocation, but blinding of the participants was not possible due to the differences in procedure. RESULTS: Twenty patients were randomized to EG (n = 10; 8 analyzed) or CG (n = 10; 10 analyzed). Two-way mixed ANOVA showed significant time × group interactions on pain interference with normal work and with relations with other people, and on perception of health status. The EG significantly improved in pain interference with normal work and perception of health status. There was no report of harm. CONCLUSION: Hypnosis may be a promising intervention to manage hemophilia-related pain and promote HRQoL, with benefits lasting up to three months.

Rhodnius prolixus is a Triatominae insect species and a primary vector of Chagas disease. The genome of R. prolixus has been recently sequenced and partially assembled, but few transcriptome analyses have been performed to date. In this study, we describe the stage-specific transcriptomes obtained from previtellogenic stages of oogenesis and from mature eggs. By analyzing ~ 228 million paired-end RNA-Seq reads, we significantly improved the current genome annotations for 9206 genes. We provide extended 5' and 3' UTRs, complete Open Reading Frames, and alternative transcript variants. Strikingly, using a combination of genome-guided and de novo transcriptome assembly we found more than two thousand novel genes, thus increasing the number of genes in R. prolixus from 15,738 to 17,864. We used the improved transcriptome to investigate stage-specific gene expression profiles during R. prolixus oogenesis. Our data reveal that 11,127 genes are expressed in the early previtellogenic stage of oogenesis and their transcripts are deposited in the developing egg including key factors regulating germline development, genome integrity, and the maternal-zygotic transition. In addition, GO term analyses show that transcripts encoding components of the steroid hormone receptor pathway, cytoskeleton, and intracellular signaling are abundant in the mature eggs, where they likely control early embryonic development upon fertilization. Our results significantly improve the R. prolixus genome and transcriptome and provide novel insight into oogenesis and early embryogenesis in this medically relevant insect.

The yerba mate (Ilex paraguariensis A.St.-Hil.) is abundant on phenolic compounds and their preservation by encapsulation can promote increased stability. The objective was to produce mate extract with a high concentration of bioactive compounds, microencapsulate and characterize them. A commercial yerba mate was used to produce the extracts and select the best extraction solvent (ethanol, water, and ethanol:water (1:1). The ethanol:water extract was encapsulated at 1:1 (w/w) by fructooligosaccharide (FOS) and gum arabic (GA), in different combinations: FOS, GA:FOS (1:3), GA:FOS (1:1), GA:FOS (3:1) and GA. The encapsulation of chlorogenic acids was around 80%, considering the major phenolic compounds by HPLC. Thermal stability, by DSC, increased at treatments FOS, GA:FOS (1:3), and GA:FOS (1:1) compare to the unencapsulated extract, and changes were noticed in ATR-FTIR bands and antioxidant activity. The encapsulated phenolic compounds from I. paraguariensis can be stable in food under heat processing.

ABSTRACT: The objective of this study was to evaluate the effects of garlic (Allium sativum) essential oil on the rates of eggs fertilization and hatching and on the normal development of larvae of artificially incubated silver catfish (Rhamdia quelen). The experiment was carried out using a completely randomized experimental design with four treatments and five replicates. The treatments consisted of introducing garlic essential oil into the incubators, which used a closed system of water recirculation, at concentrations of 0, 1, 3, and 5mg L-1. Rates of eggs fertilization and larval hatching were determined at 10 and 24h after the beginning of eggs hydration. Morphology of larvae was assessed with the aid of a stereo microscope to determine the rate of normal development. The concentration of 5mg L-1 promoted better rates of eggs fertilization and larval hatching. Concentrations of garlic essential oil had a linear effect on eggs fertilization rates and larval hatching. Garlic oil did not influence the morphology of the larvae (P>0.05). It is recommended to use 5mg L-1 of garlic essential oil to promote better rates of eggs fertilization and larval hatching of artificially incubated silver catfish.


RESUMO: O objetivo deste trabalho foi avaliar o efeito da utilização do óleo essencial de alho (Allium sativum) sobre as taxas de fertilização dos ovos e eclosão e normalidade de larvas de jundiá cinza, Rhamdia quelen, incubados artificialmente. O experimento foi realizado utilizando delineamento experimental inteiramente casualizado com quatro tratamentos e cinco repetições. Os tratamentos consistiram na introdução de óleo essencial de alho nas incubadoras, utilizando um sistema fechado de recirculação de água, com concentrações de 0; 1; 3 e 5mg L-1 de óleo essencial de alho. As taxas de fertilização dos ovos e eclosão das larvas foram determinadas, respectivamente as 10 e 24h após o início da hidratação dos ovos. Utilizou-se do padrão morfológico de larvas para determinar a taxa de normalidade, com o auxílio de um microscópio estereoscópio. A concentração de 5mg L-1 promoveu melhores taxas de fertilização de ovos e eclosão de larvas. As concentrações de óleo essencial de alho apresentaram efeito linear sobre as taxas de fertilização dos ovos e eclosão das larvas. O óleo de alho não influenciou na normalidade das larvas (P>0,05). RESUMO

This paper focuses on the starting point of various studies that are being carried out in two possible locations being considered to host a hypothetical site for a repository for low and intermediate level radioactive waste (LILW) produced in Portugal in compliance with international requirements on the long-term safety of this kind of repository. Previous studies concerning the geology of the much larger geographical areas where these locations are included were fundamental in the choice of these locations and for the design of the survey strategy. One of the fundamental assessment studies during the site-selection is the overall radiological characterization of the locations and its relation to the geology. This paper pretends to show the adequability of using a fast and reasonably inexpensive survey technique such as in situ gamma-ray portable detectors, to access the radiometric response of the systems in study by providing the radiometric mapping of the areas. The existence of adequate radiometric maps represents a critical pre-requisite to constrain both the number and spatial distribution of samples to be collected for further analysis, sustaining as well the subsequent extrapolation of results needed to fully characterise the surveyed system. Both areas were surveyed using portable gamma-ray spectrometers with NaI(Tl) detectors. In situ gamma-ray measurements have clearly shown not only the poorly contrasting geological features, but also their differences representing: (i) a deformed/metamorphosed ophiolite complex and (ii) a monotonous meta-sedimentary sequence. The radiometric maps obtained have show heterogeneities that reflect mostly changes in rock-forming mineral assemblages, even in the presence of small variations of gamma radiation. These maps support objective criteria about the number/distribution of samples to be collected for subsequent comprehensive studies and reinforce the valuable contribution of in situ gamma spectrometry to assess, in radiological terms, the prevalent geological features.
