RESUMO

Ratoon stunt (RS) caused by bacterium Leifsonia xyli subsp. xyli (Lxx) results in substantial yield losses in sugarcane (Saccharum sp. L. hybrid). Since RS does not produce reliable symptoms in the field, laboratory-based techniques are necessary for detection. Loop-mediated isothermal amplification (LAMP) assay overcomes the limitations of laboratory-based techniques which are costly, time consuming and cannot be used for near-field detection. A sensitive LAMP assay was developed to detect Lxx at 65°C in 30 min. However, carry-over contamination affected the reliability of the assay. In the present study, contaminants were successfully eliminated by incorporation of uracil nucleoside glycosylase (1 U µl-1 ) into the LAMP assay and incubation for 10 min at 37°C. To avoid the use of colorimetric reagents, lateral flow devices were successfully used for the detection of LAMP products and were equally sensitive to detection by agarose gel electrophoresis. The use of exudate from leaf sheath discs as an alternate template for the LAMP assay was found to be less sensitive than xylem sap. The preprepared master mix could be stored for up to 4 months at -20°C without any reduction in performance. These changes make the assay suitable for near-field detection in laboratories with basic facilities. SIGNIFICANCE AND IMPACT OF THE STUDY: This study presents a modified loop-mediated isothermal amplification (LAMP) assay for the detection of Leifsonia xyli subsp. xyli. Modifications include incorporation of uracil nucleoside glycosylase to eliminate carry-over contamination and substitution of colorimetric detection for the use of lateral flow devices. LAMP master mix was preprepared and was stably stored up to 4 months at -20°C. Sugarcane leaf sheaths worked well as a substitute to xylem sap as template, although the sensitivity was lower. These modifications allow the assay to be conducted without contamination concerns and reduction in set up time, making it ideal for near-field diagnosis.

The understanding of popular beliefs about mental health care and the pathways clients take prior to admission to a mental health institution is vital in planning to reduce delays in seeking treatment. The objectives of this exploratory survey were to determine pathways of care the clients with mental illness take, which ultimately lead to the mental health institution, the effects of socio-cultural and economic factor on the pathways to mental health care and the satisfaction with different service providers consulted. Data was gathered through semi-structured interviews. The results indicate that African clients interpret mental illness as bewitchment. Delays in seeking appropriate mental health care are experienced because traditional and faith healers are the first port of call. The short pathways are used when the first signs of psychotic features are severe, including like aggressive or violent behaviour. Financial constraints seem to be the problem for most of the clients in accessing mental health care. Furthermore, defaulting treatment was also observed due to the fact that mental illnesses are stigmatised in African communities.

OBJECTIVES: To determine the prevalence of depression in a university student population attending the Unitra Health Service and further compare the prevalence rate according to gender, age group and presenting complaint. METHOD: The sampling was carried out at the University of Transkei Health Service (UHS), Umtata. Two hundred and fifty students were randomly selected over a two month period during their first visit to the health centre for various ailments including family planning. Each student was given the 21-item Beck's Depression Inventory (BDI) questionnaire to fill at their own leisure time. RESULTS: The results of the study demonstrated a high prevalence of depression among the selected student population. BDI scores showed a total percentage of mild to severe depression to be 53%. BDI scores also showed that depression affects all age groups in this population with females being more affected than males at the ratio 3:1. All moderately and severely depressed students (14%) presented with somatic symptoms of headache and stomachache. None of these depressed students were diagnosed as being depressed by the UHS staff. CONCLUSION: Depression occurs more in females than males; it affects all age groups; is difficult to diagnose, even when it is severe, because of its somatic presentation. RECOMMENDATION: UHS nursing staff as well as students should be educated about depression to improve recognition and diagnosis. This recommendation applies to all nurses working at primary health care centres.
