RESUMO

ACTIVITY is a database on DNA/RNA site sequences with known activity magnitudes, measurement systems, sequence-activity relationships under fixed experimental conditions and procedures to adapt these relationships from one measurement system to another. This database deposits information on DNA/RNA affinities to proteins and cell nuclear extracts, cutting efficiencies, gene transcription activity, mRNA translation efficiencies, mutability and other biological activities of natural sites occurring within promoters, mRNA leaders, and other regulatory regions in pro- and eukaryotic genomes, their mutant forms and synthetic analogues. Since activity magnitudes are heavily system-dependent, the current version of ACTIVITY is supplemented by three novel sub-databases: (i) SYSTEM, measurement systems; (ii) KNOWLEDGE, sequence-activity relationships under fixed experimental conditions; and (iii) CROSS_TEST, procedures adapting a relationship from one measurement system to another. These databases are useful in molecular biology, pharmacogenetics, metabolic engineering, drug design and biotechnology. The databases can be queried using SRS and are available through the Web, http://wwwmgs. bionet.nsc.ru/systems/Activity/.

RESUMO

rSNP_Guide is a novel curated database system for analysis of transcription factor (TF) binding to target sequences in regulatory gene regions altered by mutations. It accumulates experimental data on naturally occurring site variants in regulatory gene regions and site-directed mutations. This database system also contains the web tools for SNP analysis, i.e., active applet applying weight matrices to predict the regulatory site candidates altered by a mutation. The current version of the rSNP_Guide is supplemented by six sub-databases: (i) rSNP_DB, on DNA-protein interaction caused by mutation; (ii) SYSTEM, on experimental systems; (iii) rSNP_BIB, on citations to original publications; (iv) SAMPLES, on experimentally identified sequences of known regulatory sites; (v) MATRIX, on weight matrices of known TF sites; (vi) rSNP_Report, on characteristic examples of successful rSNP_Tools implementation. These databases are useful for the analysis of natural SNPs and site-directed mutations. The databases are available through the Web, http://wwwmgs.bionet.nsc.ru/mgs/systems/rsnp/.

RESUMO

A model has been developed that describes the dependence of a quantitative selective trait on the animal homo- and heterozygosity for the genes that control the biochemical reaction rate (isoenzyme systems). The model includes any cases of gene control of a quantitative trait and is applicable to real, genetically heterogeneous populations. The results of testing the model show that it can be used for identifying the genes involved in additive polygenic determination of quantitative commercially valuable traits.

RESUMO

An aberrant chromosome 1 carrying an inverted fragment with two amplified DNA regions was isolated from natural populations of Mus musculus. A meiotic drive favouring the aberrant chromosome was previously demonstrated for heterozygous females. The cause for this was the preferential passage of the chromosome 1 to the oocyte. Genetic analysis made it possible to identify a two-component system conditioning the deviation from equal segregation of the homologues. The system consists of the postulated distorter and a responder. The distorter is located on the chromosome 1 distally to the responder, between the 1n and Pep 3 genes, the former acting on the responder when in the trans position. Polymorphism of the distorters was manifested as variation in their effect on the meiotic drive level in the laboratory strain and mice from natural populations.

This work is devoted to a linkage study of the gene controlling esterase D and a lethal factor in domestic pig. Segregation for two alleles EsDB, which originated from populations of miniature swine and pigs of a large white breed, and the effect of these variants on the number of offspring in a litter were compared. The results showed a linkage of locus EsD with a lethal factor. This lethal factor causes the mortality of homozygotes and half of the heterozygotes in a litter, which depends on the litter size. The lethal factor was found to be absent in miniature pigs, and in the large white breed it is predominantly linked with allele EsDB. The effect of the lethal factor on population structure and fecundity is discussed.

RESUMO

Associations between the alleles of nonlinked polymorphic genetic systems were studied in the population of the Semirechenskaya breed of pigs. The study of marker pairs of 21 genetic systems revealed that 7% of associations are statistically significant. Most of the associations detected are assumed to arise from breeding by commercial productivity traits and from the formation of an integral genotype adapted to conditions of artificial selection in the population.

The combinations of genetic markers (allotypes, blood-group systems, and polymorphic proteins) were analyzed in pigs of two phylogenetically related Siberian populations: the newly selected Siberian specialized meat breed SM-1 (daughter breed) and the reproductive core of the Kemerovo breed (maternal breed). Statistically significant interlocus associations were found between the genetic markers that are known not to be linked, being assigned to different linkage groups, and the main selected meat-productivity traits. Some of these associations were shared by both breeds; others were found to be specific to either maternal or daughter populations. Possible genetic mechanisms underlying these associations and their significance are discussed.

RESUMO

The study was designed to analyze vanadium content in urine samples of 87 workers engaged in the production of high-vanadium oil and 50 persons of the control group. Statistical data verified an elevated vanadium content in the urine of oil miners and especially of operators of the shop of oil and gas production. Urine concentration of vanadium grew along with the increase in the length of service.

RESUMO

SELEX_DB is a novel curated database on selected randomized DNA/RNA sequences designed for accumulation of experimental data on functional site sequences obtained by using SELEX and SELEX-like technologies from the pools of random sequences. This database also contains the programs for DNA/RNA functional site recognition within arbitrary nucleotide sequences. The first release of SELEX_DB has been installed under SRS and is available through the WWW at http://wwwmgs.bionet.nsc.ru/mgs/systems/selex/
